1Research on the Crack Detection of Conductive Components Using Pulsed Eddy Current Thermography显示文摘Crack of conductive component is one of the biggest threats to daily production. In order to detect the crack on conductive component,the pulsed eddy current thermography models were built according to different materials with the cracks based on finite element method(FEM) simulation. The influence of the induction heating temperature distribution with the different defect depths were simulated for the carbon fiber reinforced plastic(CFRP) materials and general metal materials. The grey value of image sequence was extracted to analyze its relationship with the depth of crack. Simulative and experimental results show that in the carbon fiber reinforced composite materials,the bigger depth of the crack is,the larger temperature rise of the crack during the heating phase is; and the bigger depth of the crack is,the faster the cooling rate of the crack during the cooling phase is. 2Near-Real-Time Carbon Emission Accounting Technology Toward Carbon Neutrality显示文摘Climate change is the greatest environmental threat to humans and the planet in the 21st century.Global anthropogenic greenhouse gas emissions are one of the main causes of the increasing number of extreme climate events.Cumulative carbon dioxide(CO_(2))emissions showed a linear relationship with cumulative temperature rise since the pre-industrial stage,and this accounts for approximately 80%of the total anthropogenic greenhouse gases.Therefore,accurate and reliable carbon emission data are the foundation and scientific basis for most emission reduction policymaking and target setting.Currently,China has made clear the ambitious goal of achieving the peak of carbon emissions by 2030 and achieving carbon neutrality by 2060.The development of a finer-grained spatiotemporal carbon emission database is urgently needed to achieve more accurate carbon emission monitoring for continuous implementation and the iterative improvement of emission reduction policies.Near-real-time carbon emission monitoring is not only a major national demand but also a scientific question at the frontier of this discipline.This article reviews existing annual-based carbon accounting methods,with a focus on the newly developed real-time carbon emission technology and its current application trends.We also present a framework for the latest near-real-time carbon emission accounting technology that can be widely used.The development of relevant data and methods will provide strong database support to the policymaking for China’s“carbon neutrality”strategy.Finally,this article provides an outlook on the future of real-time carbon emission monitoring technology.Zhu Liu Taochun Sun Ying Yu Piyu Ke Zhu Deng Chenxi Lu Da Huo Xiang Ding 2022Engineering2022,8,7:5
3Permeabilization of Escherichia coli with ampicillin for a whole cell biocatalyst with enhanced glutamate decarboxylase activity显示文摘The activity of whole-cell biocatalysts is strongly compromised by the cell envelope, which is a permeability barrier against the diffusion of substrates and products. Although common chemical or physical permeabilization methods used in cultured cells enhance cell permeability, these methods inevitably add several extra processing steps after cell cultivation, as well as impede large scale processing. To increase membrane permeability and cellbound glutamate decarboxylase(GAD) activity of recombinant Escherichia coli(BL21(DE3)-p ET28a-gad B) cells without the need for an additional permeabilization step, we investigated the permeabilizing effects of adding cell wall synthesis inhibitors or surfactants to the culture media. Ampicillin was the most effective at improving cell-bound GAD activity of the BL21(DE3)-p ET28a-gad B, although it decreased the cell biomass yield. The best permeabilization effect was observed using an ampicillin concentration of 5 μg·ml-1. Using this concentration,the cell biomass did decrease by 40.58%, but the cell-bound GAD activity of BL21(DE3)-p ET28a-gad B and total cell-bound GAD activity per milliliter of culture was enhanced by 6.24- and 3.64-fold, respectively. Treatment of BL21(DE3)-p ET28a-gad B cells with 5 μg·ml-1ampicillin resulted in structural changes to the cell envelope,but did not substantially affect GAD expression. By entrapping the ampicillin-treated cells in an open pore gelation matrix, which is a polymer derived from polyvinyl alcohol(PVA), alginate, and boric acid, the transformation rate of γ-aminobutyric acid(GABA) at the 10 th cycle produced by immobilized and permeabilized cells remained 46% of the first cycle.
