| 2 | Global gene expression profiles for the growth phases of Trichophyton rubrum显示文摘Trichophyton rubrum (T.rubrum) is a common superficial fungus.Molecular and genetic studies of T.rubrum are still limited.In this paper,we report the global analysis of gene expression profiles at different growth phases using cDNA microarray technology.A total of 2044 differentially expressed genes were obtained and clustered into three expression patterns.Our data confirmed previous results that many mRNAs were pre-stored in the conidia of T.rubrum.Transcriptional profiling and function analysis showed that some glycolytic enzymes share similar expression patterns and may be coregulated during the transition of growth phases.Some genes involved in small GTPase signaling pathways,and in cAMP-dependent and MAPK regulation pathways were induced in response to the growth dynamics of T.rubrum.Although the detailed biological roles of these T.rubrum genes are still unknown,our results suggest that these genes may be involved in regulation mechanisms in the life cycle of the fungus. | XU XingYe LIU Tao LENG WenChuan DONG Jie XUE Ying YANG HanChuna JIN Qi | 2011 | Science China(Life Sciences)2011,54,7: | 2 |

| 4 | Effect of Tuina along“bladder meridian”alleviating intervertebral disc degeneration by regulating the transforming growth factor-β1/Smad signaling pathway in a rabbit model显示文摘OBJECTIVE:The aim of this study was to investigate the protective effects of Tuina(a traditional Chinese massage therapy)on intervertebral disc(IVD)degeneration and the regulatory mechanisms of the transforming growth factor-β1(TGF-β1)/small mothers against decapentaplegic(Smad)signaling pathway.METHODS:Thirty New Zealand white rabbits were randomized into five groups:the control group,model group,model+Tuina group(Tuina group),model+TGF-β1 group(TGF-β1 group),and model+TGF-β1 inhibitor SB431542 group(SB431542 group).The model was established by posterolateral annulus fibrosus puncturing(AFP).Recombinant TGF-β1 and inhibitor SB431542 was injected into the TGF-β1 group and SB431542 group with a microsyringe,respectively.The rabbits in the Tuina group received Tuina treatment along the bladder meridian for 4 weeks.Magnetic resonance imaging(MRI)was performed on rabbits before AFP and after 4 weeks of intervention.Lumbar IVDs(L2-L3 to L4-L5)were harvested after intervention.Histopathological changes in the IVDs were measured by hematoxylin and eosin(HE)staining.Type I collagen was analyzed by immunohistochemistry detection.The expression level of matrix metalloproteinase-3(MMP3)was determined by enzyme-linked immunosorbent assay.Cell apoptosis was evaluated by terminal deoxynucleotidyl transferasemediated nick end labeling and Western blotting.Realtime polymerase chain reaction and Western blotting were used to analyze the expression of TGF-β1 and Smad2/3/4 and a disintegrin and metalloproteinase with thrombospondin motifs 5.RESULTS:Posterolateral AFP induced IVD degeneration in rabbits with histopathological damage and noticeable changes in MRI images.Tuina alleviated histopathological changes and reversed the expression of extracellular matrix degeneration-related molecules and apoptosis-related proteins.Furthermore,AFP induced the activation of TGF-β1 and Smad2/3/4,whereas Tuina therapy markedly reduced the protein expression of Smad2/3 and the gene expression of TGF-β1 and Smad2/3/4.Additionally,the TGF-β1/Smad signaling pathway was activated in the TGF-β1 group,while the TGF-β1/Smad signaling pathway was inhibited in the SB431542 group.CONCLUSION:Posterolateral AFP induced disc degeneration as determined by MRI assessment and histological analysis.Tuina alleviated disc degeneration,possibly by inhibiting the fibrotic response mediated by the TGF-β1/Smad pathway,thus alleviating extracellular matrix degeneration and reducing cell apoptosis. | SU Chengguo ZHAO Xiaoyan YE Jiangnan ZHANG Xin JIANG Yuqing GUO Junjie ZHANG Xiyuan QI Wenchuan ZHU Jun | 2023 | Journal of Traditional Chinese Medicine2023,43,5: | 0 |
| 5 | Immunogenicity and safety of a booster COVID-19 vaccination in patients with chronic liver disease:A multicenter study显示文摘Aim:Patients with chronic liver disease(CLD),especially cirrhosis,are at a high risk of severe illness or death from coronavirus disease-2019(COVID-19)and may have a suboptimal immune response to the severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)vaccine.This study aimed to evaluate the safety and immunogenicity of the COVID-19 booster vaccination in patients with CLD.Methods:The study protocol was prospectively registered at ClinicalTrials.gov(No.NCT05204602)after approval by the Ethics Committee.Adult participants with CLD were enrolled in this multicenter prospective study.They completed two doses of the inactivated COVID-19 vaccine and received booster doses at least 6 months later.Adverse reactions were recorded within 14 days after the booster dose.Serum samples of the enrolled patients were collected before and after booster vaccination and tested for SARS-CoV-2 receptor-binding domain(RBD)immunoglobulin G and neutralizing anti-bodies.The chi-squared or Fisher's exact test was used to compare categorical data,and the Mann-Whitney U test was used to compare continuous variables.Two-sided p<0.05 were considered statistically significant.Results:In total,63 patients were enrolled from four hospitals in China,including 29 patients with cirrhosis.The median age of all patients was 55 years,and 61.9%(39/63)were male.The vaccines were well tolerated;most adverse reactions were mild and transient,and injection site pain(6.4%;4/63)and fatigue(3.2%,2/63)were the most frequent local and systemic adverse events.Following the booster vaccination,our results showed that in the whole cohort,the levels and positive rates of anti-RBD IgG and neutralizing antibodies were significantly higher than baseline levels(all p<0.05).Conclusions:The inactivated COVID-19 booster vaccine was safe and significantly increased antibody levels and positivity rates following standard vaccination regimens in patients with CLD,especially those with cirrhosis. | Jitao Wang Jingwen Ai Huiling Xiang Yanliang Zhang Zhiyun Hou Qiran Zhang Jiaojian Lv Shubo Chen Chuan Liu Qianqian Li Jing Liang Faren Xie Shujun Jiang Nina Zhang Aiguo Zhang Xiaolin Lan Xuying Zhang Jinlong Li Dengxiang Liu Wenchuan Wang Wei Rao Zhang Qun Qiuju Tian Xiaolong Qi Wenhong Zhang | 2023 | Portal Hypertension & Cirrhosis2023,2,3: | 0 |
