| 1 | p53 upregulated by HIF-la promotes hypoxiainduced G2/M arrest and renal fibrosis in vitro and in vivo显示文摘Hypoxia plays an important role in the genesis and progression of renal fibrosis.The underlying mechanisms, however, have not been sufficiently elucidated. We examined the role of p53 in hypoxia-induced renal fibrosis in cell culture (human and rat renal tubular epithelial cells) and a mouse unilateral ureteral obstruction (UUO) model. Cell cycle of tubular cells was determined by flow cytometry, and the expression of profibrogenic factors was determined by RT-PCR, immunohistochemistry, and western blotting. Chromatin immunoprecipitation and luciferase reporter experiments were performed to explore the effect of HIF-lα on p53 expression. We showed that, in hypoxic tubular cells, p53 upregulation suppressed the expression of CDK1 and cyclins Bl and DI, leading to cell cycle (G2/M) arrest (or delay) and higher expression of TGF-β, CTGF, collagens, and fibronectin. p53 suppression by siRNA or by a specific p53 inhibitor (PIF-α) triggered opposite effects preventing the G2/M arrest and profibrotic changes. In vivo experiments in the UUO model revealed similar antifibrotic results following intraperitoneal administration of PIF-α(2.2 mg/kg). Using gain-of-function, loss-of-function, and luciferase assays, we further identified an HRE3 region on the p53 promoter as the HIF-lα-binding site. The HIF-la-HRE3 binding resulted in a sharp transcriptional activation of p53. Collectively, we show the presence of a hypoxia-activated, p53-responsive profibrogenic pathway in the kidney. | 2 | Dynamic behavior of gas bubble in single bubble sonoluminescence—vibrator model显示文摘Single bubble sonoluminescence is a process of energy transformation from sound to light. Therefore the motion equations of near spherical vibration of a gas bubble in an incompressible and viscous liquid can be deduced by Lagrangian Equation with dissipation function when the bubble is considered as a vibrator surrounded by liquid. The analytical solutions in the bubble expanding, collapsing and rebounding stages can be obtained by solving these motion equations when some approximations are adopted. | 3 | The thermoelastic excitation of air-solid interface waves using the pulsed laser显示文摘Applying the integral transform to the coupled problem of thermoelastic and heat conduction equations, the integral representation of the normal displacement field for the air-solid interface waves excited by a pulsed laser line source is obtained. The pole residues of the integrand are performed analytically, and the transient displacement field is calculated by using FFT technique. The thermoelastic excitation and detection of the air-solid interface waves is carried out by a laser ultrasonic system, on which the pulsed laser is focused into a line source onto the interface to excite the air-solid interface waves: leaky Rayleigh wave and Scholte wave, and the interface waves displacement signal is detected successfully by a SH130 laser heterodyne interferometer. | 4 | Ultrasonic displacement field generated by laser pulse line source显示文摘Applying the Fourier transform to the wave equations of elastic medium at its surface a laser pulse line source is acted, the integral representations of solutions are obtained. Displacement waveforms are calculated numerically by using double FFT. The calculated results include the displacements of the elastic half space of Aluminum medium, and epicenter and off-epicenter of an Aluminum plate. The two exciting sources of thermoelastic and ablating generation are considered respectively. The experiment was made on the Aluminum medium with a Nd:YAG laser and the normal displacement signals are detected by a laser interferometer. | 6 | Measurement of acoustic field radiated by low frequency power ultrasonic transducer with laser-interferometer显示文摘Based on the piezo-optic effect of medium, the refractive index of medium is the function of its density, and so it’s also the function of acoustic pressure. Therefore, acoustic pressure in the optical path everywhere can be determined absolutely by laser-interferometric technique and relative distribution of pressure in the middle and far acoustic field, which can be obtained from theory or experiment respectively. Theory and experiment of measurement of pressure in acoustic field with laser-interferometer are introduced. Distribution of pressure radiated by a power ultrasonic transducer is determined by laser interferometric technique. The theoretical and experimental results are in good agreement. The effects of rice husk ash(RHA) on cement hydration product phases and interfacial transition zone(ITZ) in mortar were investigated from the nano-scale structure perspective. The experimental results indicate that, with the increase of RHA dosages of samples, the volume fraction of high-density calcium-silicate-hydrate(HD C-S-H) in porosity and hydration product phases increases. The volume fractions of HD C-S-H in C-S-H of samples show an increasing trend with the increase of RHA dosages.
