| 1 | p53 upregulated by HIF-la promotes hypoxiainduced G2/M arrest and renal fibrosis in vitro and in vivo显示文摘Hypoxia plays an important role in the genesis and progression of renal fibrosis.The underlying mechanisms, however, have not been sufficiently elucidated. We examined the role of p53 in hypoxia-induced renal fibrosis in cell culture (human and rat renal tubular epithelial cells) and a mouse unilateral ureteral obstruction (UUO) model. Cell cycle of tubular cells was determined by flow cytometry, and the expression of profibrogenic factors was determined by RT-PCR, immunohistochemistry, and western blotting. Chromatin immunoprecipitation and luciferase reporter experiments were performed to explore the effect of HIF-lα on p53 expression. We showed that, in hypoxic tubular cells, p53 upregulation suppressed the expression of CDK1 and cyclins Bl and DI, leading to cell cycle (G2/M) arrest (or delay) and higher expression of TGF-β, CTGF, collagens, and fibronectin. p53 suppression by siRNA or by a specific p53 inhibitor (PIF-α) triggered opposite effects preventing the G2/M arrest and profibrotic changes. In vivo experiments in the UUO model revealed similar antifibrotic results following intraperitoneal administration of PIF-α(2.2 mg/kg). Using gain-of-function, loss-of-function, and luciferase assays, we further identified an HRE3 region on the p53 promoter as the HIF-lα-binding site. The HIF-la-HRE3 binding resulted in a sharp transcriptional activation of p53. Collectively, we show the presence of a hypoxia-activated, p53-responsive profibrogenic pathway in the kidney. | 2 | Effect of Yang-activating and stasis-eliminating decoction from Traditional Chinese Medicine on intestinal mucosal permeability in rats with ulcerative colitis induced by dextran sulfate sodium显示文摘OBJECTIVE: To evaluate the effect of decoction prepared with Yang-activating and stasis-eliminating medicinals from Traditional Chinese Medicine(TCM) on the intestinal mucosal permeability in rats with ulcerative colitis induced by dextran sulfate sodium(DSS).METHODS: Totally 55 male Wistar rats(body weight of 170-190 g) were randomly divided into the blank group(n = 10) and the model duplication group(n = 45). The blank group was not intervened, while the other was modeled with 5% dextran sulfate sodium by gavaging in a dosage of4 m L per day to induce ulcerative colitis, a total of7 days. Then, the model rats were divided into model blank group, mesalazine group and TCM group,and each group was consisted of 15 rats. They were given retention enema 10 min with normal saline,mesalazine enema(0.036 g/m L), and Yang-activating and stasis-eliminating decoction [0.54 g/m L of a decoction boiled by Puhuang(Pollen Typhae), Xiebai(Bulbus Allii Macrostemonis) and Wulingzhi(Faeces Trogopteri)] for 10 days respectively. Afterwards,all of the rats were evaluated by disease activity index(DAI), histological changes of distal colon, expression of occludin protein and ultrastructure of intestinal epithelial cells. Furthermore, ratio of lactulose to mannitol(L/M) discharged in urine was evaluated.RESULTS: Comparing the results between TCM and model control groups, scores of DAI and histological lesions decreased significantly(P = 0.000 <0.01), ultrastructures of intestinal epithelial cells and tight junctions were more complete. The expression of occludin protein(P = 0.001 < 0.01) increased while the L/M value decreased significantly(P = 0.000 < 0.01) in TCM group. | 3 | Stochastic modelling of soil moisture dynamics in a grassland of Qilian Mountain at point scale显示文摘Stochastic modeling of soil moisture dynamics is crucial to the quantitative understanding of plant responses to water stresses,hydrological control of nutrient cycling processes,water competition among plants,and some other ecological dynamics,and thus has become a hotspot in ecohydrology at present.In this paper,we based on the continuously monitored data of soil moisture during 2002―2005 and daily precipitation date of 1992―2006,and tried to make a probabilistic analysis of soil moisture dynamics at point scale in a grassland of Qilian Mountain by integrating the stochastic model improved by Laio and the Monte Carlo method.The results show that the inter-annual variations for the soil moisture patterns at different depths are very significant,and that the coefficient of variance(CV) of surface soil moisture(20 cm) is almost continually kept at about 0.23 whether in the rich or poor rainy years.Interestingly,it has been found that the maximal CV of soil moisture has not always appeared at the surface layer.Comparison of the analytically derived soil moisture probability density function(PDF) with the statistical distribution of the observed soil moisture data suggests that the stochastic model can reasonably describe and predict the soil moisture dynamics of the grassland in Qilian Mountain at point scale.By extracting the statistical information of the historical precipitation data in 1994―2006,and inputting them into the stochastic model,we analytically derived the long-term soil moisture PDF without considering the inter-annual climate fluctuations,and then numerically derived the one when considering the inter-annual fluctuation effects in combination with a Monte-Carlo procedure.It was found that,though the peak position of the probability density distribution significantly moved towards drought when considering the disturbance forces,and its width was narrowed,accordingly its peak value was increased,no significant bimodality was observed in the soil moisture dynamics under the given intensity of random fluctuation disturbance. | LIU Hu,ZHAO WenZhi HE ZhiBin & ZHANG LiJie Linze Inland River Basin Research Station,Chinese Ecosystem Research Network Laboratory of Watershed Hydrology and Ecol-ogy,Cold and Arid Regions Environmental and Engineering Research Institute,Chinese Academy of Sciences,Lanzhou 730000,China | 2007 | Science China Earth Sciences2007,50,12: | 7 |

| 5 | The oxygen cycle and a habitable Earth显示文摘As an important contributor to the habitability of our planet, the oxygen cycle is interconnected with the emergence and evolution of complex life and is also the basis to establish Earth system science. Investigating the global oxygen cycle provides valuable information on the evolution of the Earth system, the habitability of our planet in the geologic past, and the future of human life. Numerous investigations have expanded our knowledge of the oxygen cycle in the fields of geology,geochemistry, geobiology, and atmospheric science. However, these studies were conducted separately, which has led to onesided understandings of this critical scientific issue and an incomplete synthesis of the interactions between the different spheres of the Earth system. This review presents a five-sphere coupled model of the Earth system and clarifies the core position of the oxygen cycle in Earth system science. Based on previous research, this review comprehensively summarizes the evolution of the oxygen cycle in geological time, with a special focus on the Great Oxidation Event(GOE) and the mass extinctions, as well as the possible connections between the oxygen content and biological evolution. The possible links between the oxygen cycle and biodiversity in geologic history have profound implications for exploring the habitability of Earth in history and guiding the future of humanity. Since the Anthropocene, anthropogenic activities have gradually steered the Earth system away from its established trajectory and had a powerful impact on the oxygen cycle. | 11 | A robust polyacrylic acid/chitosan cryogel for rapid hemostasis显示文摘Currently,it is a challenge to develop hemostatic materials with high water absorption capacity and anti-fatigue properties for quickly preventing massive hemorrhage from arteries and visceral organs.A series of polyacrylic acid/chitosan(PAA/CS)cryogels were prepared by a cryostructurization technique to improve mechanical performance and hemostatic efficiency of chitosan(CS).In this system,the chemically cross-linked PAA network was used as a framework to improve water absorption behaviors and mechanical strength.The CS network was co-blended by hydrogen bonding and electrostatic interactions,both of which synergistically promoted hemostasis.These cryogels had high porosity(>94%),rapid water absorption rate(<3 s),high blood absorption capacity(>2000%),outstanding mechanical strength,and fatigue resistance.Moreover,the results of cytotoxicity and hemolysis demonstrated that the cryogels had good biocompatibility.Notably,the PAA/CS cryogels exhibited superior whole blood coagulation ability and red blood cell and platelet adhesion ability compared to those of commercial hemostatic dressing(gauze,gelatin sponges,and CS sponges).Based on these results,mouse femoral artery hemorrhage models and liver hemorrhage models were prepared to investigate the hemostatic ability of the prepared PAA/CS cryogels.Results suggested that the hemostatic ability of PAA5/CS cryogels was superior to that of commercial hemostatic materials.Therefore,the PAA/CS cryogels showed potential application in preventing massive hemorrhage from arteries and visceral organs. | SHI MingYue JIANG LiJie YU ChaoJie DONG XiaoRu YU QingYu YAO MengMeng HE ShaoShuai YUE ZhiWei YAO FangLian ZHANG Hong SUN Hong LI JunJie | 2022 | Science China(Technological Sciences)2022,65,5: | 1 |

| 13 | Integration of coal pyrolysis process with iron ore reduction: Reduction behaviors of iron ore with benzene-containing coal pyrolysis gas as a reducing agent显示文摘An integrated coal pyrolysis process with iron ore reduction is proposed in this article. As the first step, iron oxide reduction is studied in a fixed bed reactor using simulated coal pyrolysis gas with benzene as a model tar compound. Variables such as reduction temperature, reduction time and benzene concentration are studied. The carbon deposition of benzene results in the retarded iron reduction at low temperatures. At high temperatures over800 °C, the presence of benzene in the gas can promote iron reduction. The metallization can reach up to 99% in20 min at 900 °C in the presence of benzene. Significant increases of hydrogen and CO/CO2 ratio are observed in the gas. It is indicated that iron reduction is accompanied by the reforming and decomposition of benzene. The degree of metallization and reduction increases with the increasing benzene concentration. Iron oxide can nearly completely be converted into cementite with benzene present in the gas under the experimental conditions.
