| 3 | Expression of voltage.gated sodium channel Nav1.5 in non.metastatic colon cancer and its associations with estrogen receptor(ER)-βexpression and clinical outcomes显示文摘Background: Voltage-gated sodium channel 1.5(Nav1.5) potentially promotes the migratory and invasive behaviors of colon cancer cells. Hitherto, the prognostic significance of Nav1.5 expression remains undetermined. The present study aimed to explore the associations of Nav1.5 expression with clinical outcomes and estrogen receptor-β(ER-β)expression in non-metastatic colon cancer patients receiving radical resection.Methods: A total of 269 consecutive patients with pathologically confirmed stages Ⅰ-Ⅲ colon cancer who underwent radical resection were selected. Nav1.5 and ER-β expression was detected by using immunohistochemistry(IHC)on tissue microarray constructed from paraffin-embedded specimens. IHC score was determined according to the percentage and intensity of positively stained cells. Statistical analysis was performed with the X-tile method, k coefficient, Chi square test or Fisher's exact test, logistic regression, log-rank test, and Cox proportional hazards models.Results: We found that Nav1.5 was commonly expressed in tumor tissues with higher mean IHC score as compared with matched tumor-adjacent normal tissues(5.1 ± 3.5 vs. 3.5 ± 2.7, P < 0.001).The high expression of Nav1.5 in colon cancer tissues was associated with high preoperative carcinoembryonic antigen level [odds ratio(OR) = 2.980;95% confidential interval(CI)1.163-7.632; P = 0.023] and high ER-β expression(OR = 2.808; 95% CI 1.243-6.343;p = 0.00 3). Log-rank test results showed that high Nav1.5 expression contributed to a low 5-year disease-free survival(DFS) rate in colon cancer patients(77.2% vs. 92.1%, P = 0.048), especially in patients with high ER-β expression tumor(76.2% vs. 91.3%, P = 0.032). | 4 | Preparation of polymer-supported hydrated ferric oxide based on Donnan membrane effect and its application for arsenic removal显示文摘In the present study a novel technique was proposed to prepare a polymer-supported hydrated ferric oxide (D201-HFO) based on Donnan membrane effect by using a strongly basic anion exchanger D201 as the host material and FeCl3-HCl-NaCl solution as the reaction environment. D201-HFO was found to exhibit higher capacity for arsenic removal than a commercial sorbent Purolite ArsenX. Furthermore, it presents favorable adsorption selectivity for arsenic removal from aqueous solution, as well as satis- factory kinetics. Fixed-bed column experiments showed that arsenic sorption on D201-HFO could re- sult in concentration of this toxic metalloid element below 10 μg/L, which was the new maximum con- centration limit set recently by the European Commission and imposed by the US EPA and China. | 5 | Effect of a combined inversion and plantarflexion surface on ankle kinematics and EMG activities in landing显示文摘目的:探寻下落在内翻结合跖屈表面时踝关节运动学参数和腓肠肌内侧、腓骨长肌、胫骨前肌的肌电活动情况。方法:12名运动员从30 cm高度下落时3种不同条件的表面:平面、25°内翻表面、25°内翻结合25°跖屈表面,每种条件分别采集5次落地。用单因素方差分析运动学参数,同时采用3×3双因素方差分析3种肌肉的积分肌电值(p<0.05)。结果:积分肌电值结果显示不同肌肉和表面之间存在交互作用。下落于平面时胫骨前肌的肌电活动高于另两种表面。相比较平面,下落于内翻表面的最大内翻角度和角速度明显提高,但腓骨长肌的肌电活动却相似。此外,下落于内翻结合跖屈的表面,其腓肠肌的积分肌电、踝关节的跖屈角度和内翻的关节活动度明显高于25°的内翻表面。结论:与单纯的内翻表面相比,内翻结合跖屈的表面在下落时会产生更不稳定的落地状况,从而更易造成踝关节扭伤。 | Divya Bhaskaran Michael Wortley Qingjian Chen Clare E. | 6 | Differential Inhibition of Nav1.7 and Neuropathic Pain by Hybridoma-Produced and Recombinant Monoclonal Antibodies that Target Nav1.7显示文摘The voltage-gated Na^+ channel subtype Nav1.7 is important for pain and itch in rodents and humans. We previously showed that a Nav1.7-targeting monoclonal antibody(SVmab) reduces Na+ currents and pain and itch responses in mice. Here, we investigated whether recombinant SVmab(rSVmab) binds to and blocks Nav1.7 similar to SVmab. ELISA tests revealed that SVmab was capable of binding to Nav1.7-expressing HEK293 cells,mouse DRG neurons, human nerve tissue, and the voltagesensor domain Ⅱ of Nav1.7. In contrast, rSVmab showed no or weak binding to Nav1.7 in these tests. Patch-clamp recordings showed that SVmab, but not rSVmab, markedly inhibited Na+ currents in Nav1.7-expressing HEK293 cells. Notably, electrical field stimulation increased the blocking activity of SVmab and rSVmab in Nav1.7-expressing HEK293 cells. SVmab was more effective than rSVmab in inhibiting paclitaxel-induced mechanical allodynia. SVmab also bound to human DRG neurons and inhibited their Na^+ currents. | 13 | Does scatter-hoarding of seeds benefit cache owners or pilferers?显示文摘The scatter-hoarding behavior of granivorous rodents plays an important role in seed dispersal and seedling regeneration of trees,as well as the evolution of several well-known mutualisms between trees and rodents in forest ecosystems.Because it is difficult to identify seed hoarders and pilferers under field conditions by traditional methods,the full costs incurred and benefits accrued by scatter-hoarding have not been fully evaluated in most systems.By using infrared radiation camera tracking and seed tagging,we investigated the benefits and losses of scatter-hoarded seeds(Camellia oleifera)for 3 sympatric rodent species(Apodemus draco,Niviventer confucianus and Leopoldamys edwardsi)in a subtropical forest of Southwest China during 2013 to 2015.We established the relationships between the rodents and the seeds at the individual level.For each rodent species,we calculated the cache recovery rate of cache owners,as well as conspecific and interspecific pilferage rates.We found that all 3 sympatric rodent species had a cache recovery advantage with rates that far exceeded average pilferage rates over a 30-day tracking period.The smallest species(A.draco)showed the highest rate of scatter-hoarding and the highest recovery advantage compared with the other 2 larger species(N.confucianus and L.edwardsi).Our results suggest that scatter-hoarding benefits cache owners in food competition,supporting the pilferage avoidance hypothesis.Therefore,scatter-hoarding behavior should be favored by natural selection,and plays a significant role in species coexistence of rodent community and in the formation of mutualism between seeds and rodents in forest ecosystems. | Haifeng GU Qingjian ZHAO Zhibin ZHANG | 2017 | Integrative Zoology2017,12,6: | 1 |

| 19 | Mechanosensitive Ion Channel TMEM63A Gangs Up with Local Macrophages to Modulate Chronic Post-amputation Pain显示文摘Post-amputation pain causes great sufering to amputees,but still no efective drugs are available due to its elusive mechanisms.Our previous clinical studies found that surgical removal or radiofrequency treatment of the neuroma at the axotomized nerve stump efectively relieves the phantom pain aficting patients after amputation.This indicated an essential role of the residual nerve stump in the formation of chronic post-amputation pain(CPAP).However,the molecular mechanism by which the residual nerve stump or neuroma is involved and regulates CPAP is still a mystery.In this study,we found that nociceptors expressed the mechanosensitive ion channel TMEM63A and macrophages infltrated into the dorsal root ganglion(DRG)neurons worked synergistically to promote CPAP.Histology and qRT-PCR showed that TMEM63A was mainly expressed in mechanical pain-producing non-peptidergic nociceptors in the DRG,and the expression of TMEM63A increased signifcantly both in the neuroma from amputated patients and the DRG in a mouse model of tibial nerve transfer(TNT).Behavioral tests showed that the mechanical,heat,and cold sensitivity were not afected in the Tmem63a-/-mice in the naïve state,suggesting the basal pain was not afected.In the infammatory and post-amputation state,the mechanical allodynia but not the heat hyperalgesia or cold allodynia was signifcantly decreased in Tmem63a-/-mice.Further study showed that there was severe neuronal injury and macrophage infltration in the DRG,tibial nerve,residual stump,and the neuromalike structure of the TNT mouse model,Consistent with this,expression of the pro-infammatory cytokines TNFα,IL-6,and IL-1βall increased dramatically in the DRG.Interestingly,the deletion of Tmem63a signifcantly reduced the macrophage infltration in the DRG but not in the tibial nerve stump.Furthermore,the ablation of macrophages signifcantly reduced both the expression of Tmem63a and the mechanical allodynia in the TNT mouse model,indicating an interaction between nociceptors and macrophages,and that these two factors gang up together to regulate the formation of CPAP.This provides a new insight into the mechanisms underlying CPAP and potential drug targets its treatment. | Shaofeng Pu Yiyang Wu Fang Tong Wan-Jie Du Shuai Liu Huan Yang Chen Zhang Bin Zhou Ziyue Chen Xiaomeng Zhou Qingjian Han Dongping Du | 2023 | Neuroscience Bulletin2023,39,2: | 1 |
