| 2 | A rare Waxy allele coordinately improves rice eating and cooking quality and grain transparency显示文摘In rice(Oryza sativa), amylose content(AC) is the major factor that determines eating and cooking quality(ECQ). The diversity in AC is largely attributed to natural allelic variation at the Waxy(Wx)locus. Here we identified a rare Wx allele, Wx^(mw) ,which combines a favorable AC, improved ECQ and grain transparency. Based on a phylogenetic analysis of Wx genomic sequences from 370 rice accessions, we speculated that Wx^(mw) may have derived from recombination between two important natural Wx alleles, Wx^(in) and Wx^(b). We validated the effects of Wx^(mw) on rice grain quality using both transgenic lines and near-isogenic lines(NILs). When introgressed into the japonica Nipponbare(NIP) background, Wx^(mw) resulted in a moderate AC that was intermediate between that of NILs carrying the Wx^(b)allele and NILs with the Wx^(mp) allele. Notably, mature grains of NILs fixed for Wx^(mw) had an improved transparent endosperm relative to soft rice. Further, we introduced Wx^(mw) into a high-yielding japonica cultivar via molecular marker-assisted selection: the introgressed lines exhibited clear improvements in ECQ and endosperm transparency. | 3 | Entropy Generation Analysis in a Mixed-Flow Compressor with Casing Treatment显示文摘Casing treatments(CT) can effectively extend compressors flow ranges with the expense of efficiency penalty. Compressor efficiency is closely linked to loss. Only revealing the mechanisms of loss generation can design a CT with high aerodynamic performance. In the paper, a highly-loaded mixed-flow compressor with tip clearance of 0.4 mm was numerically studied at a rotational speed of 30,000 r/min to reveal the effects of axial slot casing treatment(ASCT) on the loss mechanisms in the compressor. The results showed that both isentropic efficiency and stall margin were improved significantly by the ASCT. The local entropy generation method was used to analyze the loss mechanisms and to quantify the loss distributions in the blade passage. Based on the axial distributions of entropy generation rate, for both the cases with and without ASCT, the peak entropy generation rate increased in the rotor domain and decreased in the stator domain during throttling the compressor. The peak entropy generation in rotor was mainly caused by the tip leakage flow and flow separations near the rotor leading edge for the mixed-flow compressor no matter which casing was applied. The radial distributions of entropy generation rate showed that the reduction of loss in the rotor domain from 0.4 span to the rotor casing was the major reason for the efficiency improved by ASCT. The addition of ASCT exerted two opposite effects on the losses generated in the compressor. On the one hand, the intensity of tip leakage flow was weakened by the suction effect of slots, which alleviated the mixing effect between the tip leakage flow and main flow, and thus reduced the flow losses;On the other hand, the extra losses upstream the rotor leading edge were produced due to the shear effect and to the heat transfer. The aforementioned shear effect was caused by the different velocity magnitudes and directions, and the heat transfer was caused by temperature gradient between the injected flow and the incoming flow. For case with smooth casing(SC), 61.61% of the overall loss arose from tip leakage flow and casing boundary layer. When the ASCT was applied, that decreased to 55.34%. | 5 | Composition design of superhigh strength maraging stainless steels using a cluster model显示文摘The composition characteristics of maraging stainless steels were studied in the present work investigation using a cluster-plus-glue-atom model. The least solubility limit of high-temperature austenite to form martensite in basic Fe–Ni–Cr corresponds to the cluster formula [NiFe12]Cr3,where NiFe12is a cuboctahedron centered by Ni and surrounded by 12 Fe atoms in FCC structure and Cr serves as glue atoms. A cluster formula [NiFe12](Cr2Ni) with surplus Ni was then determined to ensure the second phase(Ni3M) precipitation,based on which new multicomponent alloys [(Ni,Cu)16Fe192](Cr32(Ni,Mo,Ti,Nb,Al,V)16) were designed. These alloys were prepared by copper mould suction casting method,then solid-solution treated at 1273 K for 1 h followed by water-quenching,and finally aged at 783 K for 3 h. The experimental results showed that the multi-element alloying results in Ni3M precipitation on the martensite,which enhances the strengths of alloys sharply after ageing treatment. Among them,the aged [(Cu4Ni12)Fe192](Cr32(Ni8.5Mo2Ti2Nb0.5Al1V1)) alloy(Fe74.91Ni8.82Cr11.62Mo1.34Ti0.67Nb0.32Al0.19V0.36Cu1.78wt%) has higher tensile strengths with YS?1456 MPa and UTS?1494 MPa. | 14 | Brassinosteroid and gibberellin coordinate rice seed germination and embryo growth by regulating glutelin mobilization显示文摘Seed germination is the beginning of a new lifecycle,and involves many complex physiological and biochemical reactions including seed reserve mobilization in the endosperm and nutrient transport and reuse in the embryo.Although glutelin is a dominant storage protein in rice,its contribution to seed germination and its regulatory mechanisms are mostly unknown.Gibberellin (GA) and brassinosteroid (BR),two major growth-promoting phytohormones,also play positive roles in controlling seed germination.However,how GA and BR interact and coordinate seed germination and facilitate glutelin mobilization remains unclear.In the present study,biochemical and physiological analyses of seed germination indicated that both GA and BR promote seed germination and post-germination growth.Exogenous application of GA restored germination defects caused by BR deficiency or insensitivity.Proteomic and q RT-PCR results showed that the expression of several glutelin proteins and their encoding genes was induced by BR and GA in the embryo.Expression assays suggested that the increased accumulation of glutelin protein in the embryo was due to the accelerated degradation of glutelin by a cysteine proteinase (REP-1) in the endosperm.The breakdown of glutelin in the endosperm showed a strict positive correspondence with the length of the shoot.The GluA2 mutation led to reduced degradation rate of glutelin and defects in seed germination,and the promotion effect of GA on seed germination was weakened in the glua2mutant.In vitro culture assay of rice embryos showed that glutelin mobilization functioned downstream of the GA and BR pathways to promote shoot elongation.These findings suggest a mechanism that mediates crosstalk between BR and GA in co-regulating rice seed germination and embryo growth. | Min Xiong Lingyi Chu Qianfeng Li Jiawen Yu Yihao Yang Peng Zhou Yong Zhou Changquan Zhang Xiaolei Fan Dongsheng Zhao Changjie Yan Qiaoquan Liu | 2021 | The Crop Journal2021,9,5: | 1 |
