1Characterization and Evaluation of OsLCT1 and OsN ramp5 Mutants Generated Through CRISPR/Cas9-Mediated Mutagenesis for Breeding Low Cd Rice显示文摘To explore how rice(Oryza sativa L.) can be safely produced in Cd-polluted soil, OsLCT1 and OsNramp5 mutant lines were generated by CRISPR/Cas9-mediated mutagenesis. One of OsLCT1 mutant(lct1×1) and two of OsNramp5 mutants(nramp5×7 and nramp5×9) were evaluated for grain Cd accumulation and agronomic performances. In paddy field soil containing approximately 0.9 mg/kg Cd, lct1×1 grains contained approximately 40%(0.17 mg/kg) of the Cd concentration of the wild type parental line, less than the China National Food Safety Standard(0.20 mg/kg). Both OsNramp5 mutants showed low grain Cd accumulation(< 0.06 mg/kg) in the paddy(approximately 0.9 mg/kg Cd) or in pots in soil spiked with 2 mg/kg Cd. However, only nramp5×7 showed normal growth and yield, whereas the growth of nramp5×9 was severely impaired. 2Fine mapping and candidate gene analysis of purple pericarp gene Pb in rice (Oryza sativa L.)显示文摘Purple rice is a type of rice with anthocyanins deposited in its grain pericarp. The rice Pb gene control-ling purple pericarp character is known to be on chromosome 4,and the purple color is dominant over white color. In this study,we fine mapped the Pb gene using two F2 segregating populations,i.e. Pei'ai 64S(white) × Yunanheixiannuo(purple) and Pei'ai 64S × Chuanheinuo(purple) . In the first-pass map-ping,the Pb gene was located in the region downstream the SSR marker RM3820. In the fine mapping,the candidate region was saturated with InDel and CAPS markers developed specifically for this study. Eventually,the Pb gene was mapped within the 25-kb region delimited by the upstream marker RID3 and the downstream marker RID4. The delimited region contained two annotated genes,Ra and bhlh16(TIGR Rice Genome,R.5) . The former is a homologue of the Myc transcription factor Lc controlling anthocyanin biosynthesis in maize,and the latter is a homologue of the TT8 gene,which is also an Myc transcription factor gene controlling the pericarp pigmentation in Arabidopsis thaliana. Sequence analysis showed that the exon 7 of the Ra gene of Yunanheixiannuo and Chuanheinuo had a 2-bp(GT) deletion compared with those of the white rice varieties Pei'ai 64S,9311 and Nipponbare. A CAPS marker,CAPSRa,was developed according to the GT deletion for analysis of the two F2 segregating populations and 106 rice lines. The results showed that all F2 plants with white pericarp,and all non-purple rice lines(63 white and 22 red) contained no GT deletion,but all 20 purple rice lines con-tained the GT deletion. 4Rice Aroma: A Natural Gift Comes with Price and the Way Forward显示文摘Aromatic rice belongs to a small but important sub-group of rice,which is highly regarded for its excellent aroma and superior grain quality.Aromatic rice,especially Basmati-and Jasmine-type rice,is being traded at a high price in the local and global markets.Genetically,rice aroma is a phenotypical expression of spontaneous recessive mutations of the OsBadh2 gene(also known as fgr/badh2/osbadh2/os2AP gene).These mutations inhibit the flow ofγ-aminobutyraldehyde(GAB-ald)toγ-aminobutyric acid(GABA),and consequently,the accumulated GAB-ald is diverted to a potent flavour component 2-acetyl-1-pyrroline(2AP)by a non-enzymatic reaction with methylglyoxal.The natural incidence of non-functional osbadh2 mutation along with selection and nursing by the farmer from the ancient time makes rice aroma as a prominent natural gift.As GABA and methylglyoxal play significant roles in stress tolerance,and their biosynthesis is strictly regulated in rice plants,the accumulation of 2AP in aromatic rice depends on the interaction of various genetic and environmental factors,and its production may come at some costs of sacrificing tolerance.This review focused on some potential underlying genes in the 2AP and GABA biosynthesis pathways,and analyzed most aspects of aroma formation in rice,and summarized the molecular mechanism of aroma production together with its genetic and non-genetic influencing factors.The present review also stated approaches to produce high-quality aromatic rice via developing novel cultivars and with good agronomic knowledge-based practice.Zakaria Hossain PRODHAN SHU Qingyao 2020Rice science2020,27,2:8
5Enhanced antitumor efficacy on hepatoma-bearing rats with adriamycin-loaded nanoparticles administered into hepatic artery显示文摘AIM: To investigate the antitumor activity of adriamycin (ADR) encapsulated in nanoparticles (NADR) and injected into the hepatic artery of hepatoma-bearing rats.METHODS: NADR was prepared by the interfacial polymerization method. Walker-256 carcinosarcomas were surgically implanted into the left liver lobes of 60 male Wistar rats, which were divided into 4 groups at random (15 rats per group). On the 7th day after implantation, normal saline (NS), free ADR (FADR), NADR, or ADR mixed with unloaded nanoparticles (ADR+NP) was respectively injected via the hepatic artery (i.a.) of rats in different groups. The dose of ADR in each formulation was 2.0 mg/kg body weight and the concentration was 1.0 mg/mL. Survival time, tumor enlargement ratio, and tumor necrosis degree were compared between each group.RESULTS: Compared with the rats that received NS i.a.,the rats that received FADR or ADR+NP acquired apparent inhibition on tumor growth, as well as prolonged their life span. Further significant anticancer efficacy was observed in rats that received i.a. administration of NADR. Statistics indicated that NADR brought on a more significant tumor inhibition and more extensive tumor necrosis, as compared to FADR or ADR+NP. The mean tumor enlargement ratio on the 7th day after NADR i.a. was 1.106. The mean tumorbearing survival time was 39.50 days. 6Ultra-sensitive Nanoprobe Modified with Tumor Cell Membrane for UCL/MRI/PET Multimodality Precise Imaging of Triple-Negative Breast Cancer显示文摘Triple-negative breast cancer(TNBC)is a subtype of breast cancer in which the estrogen receptor and progesterone receptor are not expressed,and human epidermal growth factor receptor 2 is not amplified or overexpressed either,which make the clinical diagnosis and treatment very challenging.Molecular imaging can provide an effective way to diagnose TNBC.Upconversion nanoparticles(UCNPs),are a promising new generation of molecular imaging probes.However,UCNPs still need to be improved for tumor-targeting ability and biocompatibility.This study describes a novel probe based on cancer cell membrane-coated upconversion nanoparticles(CCm-UCNPs),owing to the low immunogenicity and homologous-targeting ability of cancer cell membranes,and modified multifunctional UCNPs.This probe exhibits excellent performance in breast cancer molecular classification and TNBC diagnosis through UCL/MRI/PET tri-modality imaging in vivo.By using this probe,MDA-MB-231 was successfully differentiated between MCF-7 tumor models in vivo.Based on the tumor imaging and molecular classification results,the probe is also expected to be modified for drug delivery in the future,contributing to the treatment of TNBC.The combination of nanoparticles with biomimetic cell membranes has the potential for multiple clinical applications.Hanyi Fang Mengting Li Qingyao Liu Yongkang Gai Lujie Yuan Sheng Wang Xiao Zhang Min Ye Yongxue Zhang Mingyuan Gao Yi Hou Xiaoli Lan 2020Nano-Micro Letters2020,12,5:5

8Distribution Features of the Nanhua-Sinian Rifts and their Significance to Hydrocarbon Accumulation in the Tarim Basin显示文摘On the basis of reprocessing 34 new two-dimensional spliced long sections(20,191 km) in the Tarim Basin, the deep structure features of the Tarim Basin were analyzed through interpreting 30,451 km of two-dimensional seismic data and compiling basic maps. Seismic interpretation and geological analysis conclude that the Nanhua-Sinian strata are a set of rift-depression depositional systems according to their tectonic and depositional features. The rift valley formed in the Nanhua Period, and the transformation became weaker during the late Sinian Period, which eventually turned into depression. From bottom to top, the deposited strata include mafic igneous, tillite, mudstone, and dolomite. Three major depocenters developed inside this basin during the rift stage and are distributed in the eastern Tarim Basin, the Awati area, and the southwestern Tarim Basin. Among them, the rift in the eastern Tarim Basin strikes in the near east-west direction on the plane and coincides with the aeromagnetic anomaly belt. This represents a strong magnetic zone formed by upwelling basic volcanic rock along high, steep normal faults of the Nanhua Period. Controlled by the tectonic background, two types of sedimentary systems were developed in the rift stage and depression stage, showing two types of sequence features in the Sinian depositional stage. The Nanhua System appears as a wedge-shaped formation, with its bottom in unconformable contact with the base. The rifting event has a strong influence on the current tectonic units in the Tarim Basin, and affects the distribution of source rock in the Yuertus Formation and reservoir beds in the Xiaoerbulake Formation in Lower Cambrian, as well as the gypseous cap rock in Middle Cambrian. The distribution features of the rifts have important and realistic significance for determining the direction of oil and gas exploration in the deep strata of the Tarim Basin. 10Identification of a major quantitative trait locus and its candidate underlying genetic variation for rice stigma exsertion rate显示文摘Stigma exsertion in male sterile lines of hybrid rice is important for seed yield.In the present study, ZS616 [Oryza sativa subsp.Xian(indica)], a male sterile line with a stigma exsertion rate(SER) as high as 94.5%, was crossed to DS552, a japonica line with almost no exserted stigmas.F3 plants with extremely low and high SER were sequenced to identify SER-associated quantitative trait loci(QTL).A major QTL for SER, qSER-3.1, was identified along with other QTL on chromosome 3 in a 3.9 Mb region.A total of 307 nonsynonymous single-nucleotide polymorphisms(SNPs) and 27 frame-shift insertion/deletions(InDels)differentiating ZS616 and DS552 were identified in the region containing qSER-3.1.Most SNPs(294) and InDels(25) were excluded after further analysis because they were shared by ZS616 and low(<2.0%) SER accessions in the Huazhong Agricultural University(HAU) core rice collection.Association analysis using the full HAU collection identified a 17-bp InDel in OS03 G0689400 as the most likely causal genetic variant underlying qSER-3.1.ZS616-type accessions(n = 54, with the 17-bp insertion) in the HAU collection had minimum(16.5%)and mean(39.6%) SERs significantly greater than those(n = 424) without the insertion(with minimum and mean SERs of 0.2% and 20.6%, respectively).Thus, this study identified a major QTL for stigma exsertion and revealed the mutation in a candidate gene for the QTL.Shouling Xu Yunchao Zheng Yang Liu Xiaohao Guo Yuanyuan Tan Qiuping Qian Qingyao Shu Jianzhong Huang 2019The Crop Journal2019,7,3:2
11Nuclear translocation of OsMFT1 that is impeded by OsFTIP1 promotes drought toleranee in rice显示文摘Drought is the leading environmental threat affecting crop productivity,and plants have evolved a series of mechanisms to adapt to drought stress.The FT-interacting proteins(FTIPs)and phosphatidylethanola mine.binding proteins(PEBPs)play key roles in developmental processes,whereas their roles in the regulation of stress response are still largely unknown.Here,we report that OsFTIP1 negatively regulates drought response in rice.We showed that OsFTIP1 interacts with rice MOTHER OF FT AND TFL1(OsMFT1),a PEBP that promotes rice tolerance to drought treatment.Further studies discovered that OsMFT1 interacts with two key drought-related transcription factors,OsbZIP66 and OsMYB26,regulating their binding capacity on drought-related genes and thereby enhancing drought toleranee in rice.Interestingly,we found that OsFTIP1 impedes the nucleocytoplasmic translocation of OsMFT1,implying that dynamic modulation of drought-responsive genes by the OsMFT1-OsMYB26 and OsMFT1-OsbZIP66 complexes is integral to OsFTIP1-modulated nuclear accumulation of OsMFT1.Our findings also suggest that OsMFT1 might act as a hitherto unknown nucleocytoplasmic trafficking signal that regulates drought tolerance in rice in response to environmental signals.Ying Chen Jun Shen Liang Zhangi Haoyue Qi Lijia Yang Huanyu Wang Jiaxuan Wang Yuexing Wang Hao Du Zeng Tao Ting Zhao Pingchuan Deng Qingyao Shu Qian Qian Hao Yu Shiyong Song 2021Molecular Plant2021,14,8:2

18Surveillance of enteric pathogens in imported seafood and environmental surfaces in five seafood markets before the outbreak of COVID-19显示文摘To monitor the presence of enteric pathogens in imported seafood,a total of 140 seafood samples imported from eight overseas countries were collected from Beijing,Dalian,Shanghai,Guangzhou,and Wuhan seafood markets from June to November 2019.Additionally,116 viral,environmental swab samples were also collected from the Wuhan and Guangzhou seafood markets.Five typical enteric bacterial pathogens(Aeromonas spp.,Shigella spp.,Salmonella spp.,Vibrio spp.,and Listeria monocytogenes)and four viruses(Rotavirus,Norovirus,Astrovirus,and Sapovirus)were detected positive.Results showed that eight Vibrio parahaemolyticus isolates appeared in seafood imported to Dalian,Wuhan,Shanghai,Guangzhou,and Beijing.In contrast,Vibrio fluvialis and Aeromonas were isolated in another two samples.Norovirus was detected in one oyster sample imported from France and environmental surface in Guangzhou.The remaining pathogens were negative in all the samples being tested.With 120 V.parahaemolyticus isolates from the above countries,the genomic analysis revealed that sequence type ST1152 isolates imported from Canada were clustered with two V.parahaemolyticus isolates from Canada.This study presented the first microbiological analysis of the Wuhan seafood market before the outbreak of COVID-19,which demonstrated that supervision should be strengthened to prevent enteric pathogens via imported seafood.Songzhe Fu Wei Wang Qingyao Wang Fenglan He Jingwei Hao Bo Pang 2021Biosafety and Health2021,3,4:1
