| 4 | Virtual screening of flavonoids from Jatropha gossypiifolia L.as potential drugs for diabetic complications显示文摘Background:Diabetes mellitus is a chronic metabolic disease that is a risk factor for epidemic pathologies.Under hyperglycemic conditions,the enzyme aldose reductase catalyzes the formation of sorbitol in the metabolism of glucose via polyols,leading to the development of diabetic complications.Therefore,inhibitors of this enzyme are therapeutic targets for the prophylaxis and treatment of these conditions.Methods:In this study,a generalized linear regression model was developed to analyze flavonoids-obtained from a database-that have been tested as inhibitors of aldose reductase.In this sense,the molecular descriptors implemented in DRAGON and MATLAB software were used to determine the correlation between the chemical structure of the inhibitors and their pharmacological activity.The model was validated according to the Organisation for Economic Co-operation and Development Standards and subsequently used for the virtual screening of the flavonoids identified in Jatropha gossypiifolia L.Results:The proposed model showed a good fit for its statistical parameters(R2=0.95).In addition,it showed good predictive power(R2 ext=0.94)and robustness(Q2 LOO=0.92).The experimental chemical space wherein the predictions were reliable(domain of application)was also defined.Finally,the model was used to identify 10 flavonoids from Jatropha gossypiifolia L.as candidates for natural drugs.Compounds with a low probability of oral absorption were identified,among which the elagic acid biflavonoid showed the greatest promise(pIC50 predicted=9.75).Conclusion:The Jatropha gossypiifolia L.species harbors flavonoids with high potential as inhibitors of the aldose reductase enzyme,in which the biflavonoid ellagic acid was shown to be the most promising inhibitor of the aldose reductase enzyme,suggesting its possible use in the treatment of the late complications of diabetes mellitus. | Yudith Cañizares-Carmenate Roberto Díaz-Amador Mirtha Mayra Gonzalez-Bedia Tan Tran Quang Nhat Francisco Torrens Juan Alberto Castillo-Garit | 2022 | Traditional Medicine Research2022,7,2: | 0 |

| 6 | Granulocyte colony-stimulating factor reduces biliary fibrosis and ductular reaction in a mouse model of chronic cholestasis显示文摘Background:Biliary atresia is a rare congenital bile duct disease that is the leading cause of liver fibrosis in neonates.Granulocyte colony-stimulating factor(GCSF)is a potential therapy for hepatocellular diseases,but data on GCSF for cholestatic conditions remain limited.Materials and methods:The current study examines the role of GCSF in improving bile duct obstruction in mice.Two doses were administered:10.0 mg/kg/day and 61.5 mg/kg/day,which is the animal equivalent dose of 5.0 mg/kg in humans.Seven days(D7)after bile duct ligation(BDL),Swiss mice were treated with phosphate buffered saline or GCSF for 5 days.The intrahepatic adaptive response of BDL mice was evaluated on postsurgical days D12,D19,and D26.Results:Treatment with 61.5 mg/kg of GCSF resulted in a significant increase in circulating leukocytes and neutrophils on D12.Amelioration of liver injury,as shown by reduced aspartate aminotransferase levels,increased albumin levels and survival rate,as well as reduced intrahepatic inflammation and hepatic myeloperoxidase expression,downregulated ductular proliferation,periportal fibroblast activation,and fibrosis,enhanced expressions of hepatocyte growth factor,peroxisome proliferator-activated receptoralpha,and ki67,and suppressed expression of cleaved caspase-3 protein,was noted after treatment with 61.5 mg/kg of GCSF.Additionally,GCSF treatment was associated with an increased number of intrahepatic cd3-Sca1tc-Kitt bone marrow cells. | Trinh Van Le Thanh Minh Dang Huy Quang Do Ai-Xuan Le Holterman Hong-Thuy Phan-Thi Thong Tan Tran Nhung Hai Truong | 2023 | Liver Research2023,7,1: | 0 |
| 7 | Marker-Assisted Selection of Xa21 Conferring Resistance to Bacterial Leaf Blight in indica Rice Cultivar LT2显示文摘Bacterial leaf blight of rice (BLB), caused by Xanthomonas oryzae pv. oryzae, is one of the most destructive diseases in Asian rice fields. A high-quality rice variety, LT2, was used as the recipient parent.IRBB21, which carries the Xa21 gene, was used as the donor parent. The resistance gene Xa21 was introduced into LT2 by marker-assisted backcrossing. Three Xoo races were used to inoculate the improved lines following the clipping method. Eleven BC_3F_3 lines carrying Xa21 were obtained based on molecular markers and agronomic performance. The 11 lines were then inoculated with the three Xoo races. All the 11 improved lines showed better resistance to BLB than the recipient parent LT2. Based on the level of resistance to BLB and their agronomic performance, five lines (BC_3F_3 5.1.5.1, BC_3F_3 5.1.5.12, BC_3F_3 8.5.6.44, BC_3F_3 9.5.4.1 and BC_3F_3 9.5.4.23) were selected as the most promising for commercial release. | 8 | Long-Term Improvement in Precautions for Flood Risk Mitigation:A Case Study in the Low-Lying Area of Central Vietnam显示文摘Local actors appear as inseparable components of the integrated flood risk mitigation strategy in Vietnam.Recognizing this fact,this study examined the long-term improvement in precautions taken by commune authorities and households between two major floods in 1999 and 2017 by applying both quantitative and qualitative methods.Two flood-prone villages were selected for a survey;one in a rural area and the other in a suburban area of Thua Thien Hue Province,central Vietnam.The findings indicate that most villagers doubted the structural works’efficacy and were dissatisfied with the current efforts of local authorities.Households’self-preparation thus became the decisive factor in mitigating risk.While most households have paid greater attention to flood precautions in 2017,others seem to be lagging.Poverty-related barriers were the root causes restraining households in both rural and suburban villages.The suburban riverine residents were further identified as vulnerable by their limitations in upgrading structural measures,which was ascribed to the inconsistency in the ancient town’s preservation policy.This multidimensional comparison,in terms of vulnerability,emphasized the importance of space-function links in the suburb and the contradictions of different policy initiatives,such as landscape rehabilitation,disaster prevention,and livelihood maintenance. | Cong Dinh Nguyen Funiikazu Ubukata Quang Tan Nguyen Hoang Ha Vo | 2021 | International Journal of Disaster Risk Science2021,12,2: | 0 |
