An improved Tet-on system in micro RNA overexpression and CRISPR/Cas9-mediated gene editing显示文摘Background: Tetracycline(Tet)-regulated expression system has become a widely applied tool to control gene activity. This study aimed to improve the Tet-on system with superior regulatory characteristics. Results: By comprehensively comparing factors of transactivators, Tet-responsive elements(TREs), orientations of induced expression cassette, and promoters controlling the transactivator, we developed an optimal Tet-on system with enhanced inducible efficiency and lower leakiness. With the system, we successfully performed effective inducible and reversible expression of micro RNA, and presented a more precise and easily reproducible fine-tuning for confirming the target of a mi RNA. Finally, the system was applied in CRISPR/Cas9-mediated knockout of nuclear factor of activated T cells-5(NFAT5), a protective transcription factor in cellular osmoregulation. Conclusions: This study established an improved Tet-on system for powerful and stringent gene regulation in functional genetic studies. | Kang Kang Lian Huang Qing Li Xiaoyun Liao Quanjin Dang Yi Yang Jun Luo Yan Zeng Li Li Deming Gou | 2019 | Journal of Animal Science and Biotechnology2019,10,4: | 2
Phase separation in cGAS-STING signaling显示文摘Biomolecular condensates formed by phase separation are widespread and play critical roles in many physiological and pathological processes.cGAS-STING signaling functions to detect aberrant DNA signals to initiate anti-infection defense and antitumor immunity. At the same time,cGAS-STING signaling must be carefully regulated to maintain immune homeostasis.Interestingly, exciting recent studies have reported that biomolecular phase separation exists and plays important roles in different steps of cGAS-STING signaling, including cGAS condensates,STING condensates, and IRF3 condensates. In addition, several intracellular and extracellular factors have been proposed to modulate the condensates in cGAS-STING signaling. These studies reveal novel activation and regulation mechanisms of cGAS-STING signaling and provide new opportunities for drug discovery. Here,we summarize recent advances in the phase separation of cGAS-STING signaling and the development of potential drugs targeting these innate immune condensates. | Quanjin Li Pu Gao | 2023 | Frontiers of Medicine2023,17,5: | 0
First extraction of the proton mass radius and scattering length |αρ0p|from ρ^(0) photoproduction显示文摘As it involves the lightest physical states excited from the vacuum by the vector quark current,near-threshold ρ^(0)photoproduction is considered a possible way to research the proton radius and the absolute value of the scattering lengths of the ρ^(0)-proton interaction. In this study, under the assumption of a scalar form factor of dipole form, the value of the proton mass radius is calculated as(0.85±0.06) fm by fitting the differential cross section of the γp→ρ^(0)p reaction at near-threshold energy. For light vector meson photoproduction, because the exchange of a scalar quark-antiquark pair is not suppressed and should dominate the scalar gluon exchange, the radius we extract from ρ^(0)photoproduction is likely to represent the quark radius of the proton. This fact may explain why the value obtained in this study is very near the proton charge radius. Moreover, the absolute value of the ρ^(0)-proton scattering length |αρ^(0)p|=(0.31±0.06) fm is obtained for the first time within the vector meson dominance model. This result disobeys the rule that the absolute value of the vector meson and proton scattering length |αVp| increases with the meson’s mass, which can be attributed to treating the ρ^(0)meson as a point in the analysis. These results provide useful theoretical information for an in-depth understanding of proton structure and the proton-vector meson interaction. | Xiao-Yun Wang Fancong Zeng Quanjin Wang Li Zhang | 2023 | Science China(Physics,Mechanics & Astronomy)2023,66,3: | 0
Theoretical Study of the Energies of the Oscillating System with a Well-Distributed Mass of the Spring显示文摘The energy of a spring with a well-distributed mass ms is theoretically studied in this paper. The solution of the wave equation is derived in detail, and then the kinetic energy and potential energy of the spring are studied with the wave equation,as well as the kinetic energy of the oscillating mass M.The kinetic energy and potential energy of the spring, and total energy are numerically simulated for different ratios ms/M with considering the spring’s mass, which makes the property of energy of the oscillating system understood easily. | Yewan Ma Hansen Chang Zhaowang Wu Yanyan Jiang Juan Li Xunchang Yin Quanjin Liu Lihua Zhang | 2019 | Sound & Vibration2019,53,4: | 0
Research on PM_(2.5) Concentration Model Based on MODIS Remote Sensing Retrieval显示文摘Aiming at the complex variation of haze and the influence of various factors,Xi'an is taken as the research area to study the qualitative and quantitative issues between aerosol optical depth(AOD)and haze before and after correction.Combining atmospheric water vapor content(PWV)and meteorological factor data,it is proposed to use'backward screening'method to carry out regression modeling and verification of the revised PM_(2.5) mass concentration,AOD,PWV and meteorological factors. The results show that the correlation between AOD and PM_(2.5) is significantly improved after vertical correction and humidity correction. From the model's decision coefficient R^(2) and the relative error of the estimated PM_(2.5) mass concentration,it can be seen that the estimation model of PM_(2.5) mass concentration based on multiple impact factors is better than the estimation model solely based on AOD. | Quanjin LI Min MAO Guangting LIU Yawen SHAO | 2023 | Meteorological and Environmental Research2023,14,5: | 0
