1Transcriptome characterization and SSR discovery in Squaliobarbus curriculus显示文摘Squaliobarbus curriculus is a eurythermic ?sh found mainly in East Asia. The natural germplasm resources of this species, an organism of potential economic aquacultural importance, have recently been threatened by over?shing and habitat degradation. The objective of this study was to obtain functional genomic resources and candidate microsatellite markers for future genetic observation and germplasm protection of S. curriculus. Illumina paired-end sequencing generated over 22.20 million clean reads, which were assembled into 204 027 transcripts with an average length of 1 457 bp. A total of 112 570 protein-coding sequences(55.17%) were predicted, 111 934 of which had signi?cant matches in the NCBI nonredundant protein database. Overall, 60 450, 34 830, and 77 755 transcripts were assigned to Gene Ontology, Clusters of Orthologous Groups, and Eukaryotic Orthologous Groups categories, respectively.In addition, 44 781 transcripts(21.95%) were mapped to 230 KEGG pathways. A total of 51 426 candidate simple sequence repeat(SSR) markers were identi?ed and used to design 36 149 PCR primer pairs. Fifty of these primer pairs were randomly selected for PCR validation in 32 S. curriculus specimens, which resulted in 25 pairs yielding the expected products. Further analysis revealed that 40%(10) of the 25 ampli?ed loci were polymorphic. 2Differential expression of miRNAs and mRNAs in the livers of largemouth bass Micropterus salmoides under heat stress显示文摘Global warming threatens freshwater ecosystems and compromises fish survival.To elucidate the role of miRNAs in the livers of heat stressed largemouth bass,juvenile fish was subject to heat stress under 37°C.Both mRNA-seq and miRNA-seq were conducted on the liver tissues under control and heat stress conditions.Differential gene expression analysis and enrichment analysis were performed on mRNA and miRNA expression profiles.A total of 406 differentially expressed genes(DEGs)were discovered,of which 212 were up-regulated and 194 were down-regulated.Most of the DEGs were significantly implicated in the regulation of“protein processing in endoplasmic reticulum”,“proteasome”,“steroid biosynthesis”,and“ornithine decarboxylase inhibitor activity”pathways.In addition,47 differentially expressed miRNAs(DEMs)were identified in largemouth bass livers under heat stress,including 21 up-regulated and 25 down-regulated miRNAs.A negatively regulated miRNA-mRNA network including 12 miRNAs and 19 mRNAs was constructed with DEMs involved in“protein degradation”,“calcium ion regulation”,“cell apoptosis”,and“lipid metabolism”.Moreover,this study indicated novel-miR-144 activated the IRE1 signaling pathway by targeting txndc5 to induce liver apoptosis in largemouth bass under heat stress.This study revealed the involvement of miRNA regulation in largemouth bass in response to heat stress.Xuqian ZHAO Zijie LIN Caijuan LI Hao ZHU Lingling LI Wenjia MAO Qufei LING 2024Journal of Oceanology and Limnology2024,42,2:0
