| 1 | The CAMS Climate System Model and a Basic Evaluation of Its Climatology and Climate Variability Simulation显示文摘A new coupled climate system model(CSM) has been developed at the Chinese Academy of Meteorological Sciences(CAMS) by employing several state-of-the-art component models. The coupled CAMS-CSM consists of the modified atmospheric model [ECmwf-HAMburg(ECHAM5)], ocean model [Modular Ocean Model(MOM4)], sea ice model [Sea Ice Simulator(SIS)], and land surface model [Common Land Model(CoLM)]. A detailed model description is presented and both the pre-industrial and 'historical' simulations are preliminarily evaluated in this study.The model can reproduce the climatological mean states and seasonal cycles of the major climate system quantities,including the sea surface temperature, precipitation, sea ice extent, and the equatorial thermocline. The major climate variability modes are also reasonably captured by the CAMS-CSM, such as the Madden–Julian Oscillation(MJO), El Ni?o–Southern Oscillation(ENSO), East Asian Summer Monsoon(EASM), and Pacific Decadal Oscillation(PDO).The model shows a promising ability to simulate the EASM variability and the ENSO–EASM relationship. | 2 | Treatment of coke plant wastewater by A/O fixed biofilm system显示文摘Coke plant wastewater was treated by an anaerobic–aerobic (A/O) biofilm system. A lab-scale experiment, during which semi-soft media were packed in both the anaerobic and the aerobic reactors, was carried out. The influence parameters, such as HRT, C/N ratio, OLR, and the recycling flow rate on the performance of the system, were studied. The results showed that a sufficient carbon source was important to the performance of bio-degradation system. The COD removal rate increased from 64.15% to 83.28% by raising C/N in wastewater from 2.5 (no ex-ternal carbon source) to 5. But the effluent COD concentration was still a little higher than the discharge standard. In order to make it meet the discharge standard, a deep treatment, coagula-tion, was applied, which was proved as an effective method. Then the effluent COD concentra-tion can be brought to lower than 200 mg·L?1. The NH4-N removal rate in this system was sat-isfactory, being higher than 99%. In addition, it was almost not affected by both C/N ratio and coagulation treatment. | 3 | Characterization of mechanical properties of aluminum cast alloy at elevated temperature显示文摘The tensile response, the low cycle fatigue(LCF) resistance, and the creep behavior of an aluminum(Al) cast alloy are studied at ambient and elevated temperatures.A non-contact real-time optical extensometer based on the digital image correlation(DIC)is developed to achieve strain measurements without damage to the specimen. The optical extensometer is validated and used to monitor dynamic strains during the mechanical experiments. Results show that Young's modulus of the cast alloy decreases with the increasing temperature, and the percentage elongation to fracture at 100℃ is the lowest over the temperature range evaluated from 25℃ to 300℃. In the LCF test, the fatigue strength coefficient decreases, whereas the fatigue strength exponent increases with the rising temperature. The fatigue ductility coefficient and exponent reach maximum values at 100℃. | 4 | Boreal Summer Intraseasonal Oscillation in the Asian–Pacific Monsoon Region Simulated in CAMS-CSM显示文摘The boreal summer intraseasonal oscillation(BSISO) is simulated by the Climate System Model(CSM) developed at the Chinese Academy of Meteorological Sciences(CAMS), China Meteorological Administration. Firstly, the results indicate that this new model is able to reasonably simulate the annual cycle and seasonal mean of the precipitation, as well as the vertical shear of large-scale zonal wind in the tropics. The model also reproduces the eastward and northward propagating oscillation signals similar to those found in observations. The simulation of BSISO is generally in agreement with the observations in terms of variance center, periodicity, and propagation, with the exception that the magnitude of BSISO anomalous convections are underestimated during both its eastward propagation along the equator and its northward propagation over the Asian–Pacific summer monsoon region. | 5 | A Carboxymethyl Cellulase from a Marine Yeast(Aureobasidium pullulans 98): Its Purification, Characterization, Gene Cloning and Carboxymethyl Cellulose Digestion显示文摘We have reported that A. pullulans 98 produces a high yield of cellulase. In this study, a carboxymethyl cellulase(CMCase) in the supernatant of the culture of A. pullulans 98 was purified to homogeneity, and the maximum production of CMCase was 4.51 U(mg protein)-1. The SDS-PAGE analysis showed that the molecular mass of the purified CMCase was 67.0 k Da. The optimal temperature of the purified enzyme with considerable thermosensitivity was 40℃, much lower than that of the CMCases from other fungi. The optimal p H of the enzyme was 5.6, and the activity profile was stable in a range of acidity(p H 5.0–6.0). The enzyme was activated by Na+, Mg2+, Ca2+, K+, Fe2+ and Cu2+, however, it was inhibited by Fe3+, Ba2+, Zn2+, Mn2+ and Ag+. Km and Vmax values of the purified enzyme were 4.7 mg m L-1 and 0.57 μmol L-1 min-1(mg protein)-1, respectively. Only oligosaccharides with different sizes were released from carboxymethylcellulose(CMC) after hydrolysis with the purified CMCase. The putative gene encoding CMCase was cloned from A. pullulans 98, which contained an open reading frame of 954 bp(EU978473). The protein deduced contained the conserved domain of cellulase superfamily(glucosyl hydrolase family 5). The N-terminal amino acid sequence of the purified CMCase was M-A-P-H-A-E-P-Q-S-Q-T-T-E-Q-T-S-S-G-Q-F, which was consistent with that deduced from the cloned gene. | 15 | Programmed nanoparticle-loaded microparticles for effective antigen/adjuvant delivery显示文摘A microscale vaccine containing SiO_(2)nanoparticles loaded in CaC〇3 microparticles was constructed using the co-precipitation method.The antigen ovalbumin(OVA)was covalently conjugated with SiO_(2)nanoparticles,and these nanoparticles and CpG were co-encapsulated into CaCO_(3)microparticles,generating a vaccine with a size of approximately 5.2μm.Scanning electron microscopy(SEM),energy-dispersive X-ray(EDX),elemental mapping,and Fourier transform infrared(FTIR)analyses confirmed the successful preparation of the microscale vaccine;the vaccine had good storage stability without sustained antigen release,and negligible cytotoxicity to dendritic cells(DCs)and macrophages.Compared to SiO_(2)nanoparticles,the microscale vaccine can significantly improve antigen/adjuvant uptake.DCs internalized the entire microscale vaccine into lysosomes via macropinocytosis,and an increase in antigen endo/lysosomal escape was observed by confocal User scanning microscopy(CLSM).Specifically,DCs pulsed with the vaccine were fully mature,expressing high levels of costimulatory molecules(CD40,CD80,and CD86),MHCⅡ,and MHCⅠand secreting high levels of proinflammatory cytokines(IL-12,TNF-α,IL-1β,and IL-6).In addition,the vaccine had good in vivo biocompatibility,could protect the antigen from rapid degradation,and increased the retention time in lymph nodes.SiO_(2)nanoparticles-in-CaCO_(3)microparticles were an excellent carrier for antigen and adjuvant delivery.Hopefully,this study can provide some information on the design of microscale carriers for vaccine delivery systems. | Rong Xu Ying Dong Yajing Zhang Xiaoli Wang Chuangnian Zhang Yanjun Jiang | 2022 | Particuology2022,20,1: | 0 |
| 16 | 凋亡抑制蛋白Survivin和血管内皮生长因子高表达是喉鳞状细胞癌生存预后的独立预测因子(英文)显示文摘Objective: To study the relationship between Survivin and VEGF proteins in a subgroup of patients with squa-mous carcinoma of larynx. Methods: 108 cases of squamous carcinoma of larynx with clinical data were collected and expressions of Survivin and VEGF in peripheral blood were investigated by enzyme-linked immunosorbent assay (ELISA). Results: Expressions of Survivin and VEGF were significantly associated with T stage, N stage and metastasis of squamous carcinoma of larynx. The patients with Survivin or VEGF over-expressions presented lower survival rate, respectively, as compared to those of low-expression (P < 0.05). The survival rate in squamous carcinoma of larynx patients with Survivin and VEGF dual over-expressions was significantly lower than that of patients with dual low-expression (P < 0.05). Multivariate analysis indicated that both Survivin and VEGF over-expressions in squamous carcinoma of larynx peripheral blood samples were strong independent factors of poor prognosis in squamous carcinoma of larynx patients. Conclusion: Survivin and VEGF over-expressions are independent prognostic factors for the patients with squamous carcinoma of larynx.
