1Flood-illuminated adaptive optics ophthalmoscope with a single curved relay mirror显示文摘For decreasing light loss and diminishing the aberrations of the optical system,an open-loop adaptive optics(AO)system for retinal imaging in vivo is introduced.Taking advantage of the ability of young human eyes to accommodate,there was only one single curved mirror to make the pupil of the eye conjugate with the wavefront corrector and the wavefront sensor.A liquid crystal spatial light modulator(LC-SLM)was adopted as the wavefront corrector because the LC-SLM can be made in a small size to match the sensor.To reduce a pair of lenses or focusing mirrors,the wavefront corrector and sensor are positioned in the noncommon path.The system adopts open-loop control and the high-precision LC-SLM guarantees the effectiveness of the AO system.The designed field of view is 1°on the retina(about 300μm).The image quality was simulated with different mirror surface types,including circular,parabolic,and hyperbolic.A hyperbolic mirror with conic constant−1.07,which is close to−1,could best eliminate the aberrations.Theoretical analysis showed that the optical throughput of this system was at least 22.4%higher than that of a standard transmission AO system.In a practical experiment,a parabolic mirror was positioned in the optical path.Images of the cone photoreceptors and the capillary vessels were obtained successfully.This system simplifies the optical setup in comparison to the commonly used 4F systems while still guaranteeing the effectiveness of AO to correct the ocular aberrations.Ruixue Liu Yue Qi Xianliang Zheng Mingliang Xia Li Xuan 2013Photonics Research2013,1,3:3
2Role of Eclipta prostrata extract in improving spatial learning and memory deficits in D-galactose-induced aging in rats显示文摘OBJECTIVE: To investigate the role of Eclipta prostrata (E. prostrata) extract in improving spatial learning and memory deficits in D-galactose-induced aging in rats. METHODS: Rats were divided into five groups, with 10 animals in each group. Aging rats were produced by treatment with 100 mg·kg-1·d-1 of D-galactose for 6 weeks. Rats in the E. prostrata treatment groups received an aqueous extract of E. prostrata orally at a concentration of 50, 100, or 200 mg·kg-1· d-1 for 3 weeks. Animals in both the normal and model groups were treated with similar volumes of saline. Spatial memory performance was measured using the Morris water maze. The mRNA levels and enzyme activities of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx) and glutathione reductase (GR) were analyzed using real- time quantitative PCR and spectrophotometry,respectively. The levels of induced nitric oxide synthase (iNOS), nitric oxide (NO), dopamine (DA), norepinephrine (NE), and serotonin (5-HT) were determined using enzyme-linked immunosorbent assay and spectrophotometry. RESULTS: Compared with the normal group, rats in the D-galactose-treated model group exhibited significant memory loss. There was severe damage to the hippocampal CA1 area, and expression levels of SOD, CAT, GPx, and GR were significantly decreased in the model group compared with the normal group. In the model group, levels of iNOS and NO were significantly increased compared with the normal group. However, treatment with E. prostrata extract reversed the conditions caused by D-galactose- induced aging, especially in the groups with higher treatment concentrations. Compared with the normal group, the levels of DA, NE, and 5-HT were significantly lower in the D-galactose-treated model group. In the E. prostrata extract-treated groups, however, there was a dose-dependent upregulation of DA, NE, and 5-HT expression. CONCLUSION: Our results suggest that administration of E. prostrata extract can result in an improvement in the learning and memory impairments that are induced by D-galactose treatment in rats. 3Evidence base of randomized controlled trials and guideline recommendations of patent Traditional Chinese Medicines for uncomplicated acute lower respiratory tract infections in adults显示文摘OBJECTIVE:To compare clinical practice guideline recommendations on the use of oral patent Traditional Chinese Medicines(PTCMs) for uncomplicated acute lower respiratory tract infections(ALRTIs)in adults with the existing evidence using results of a systematic review of randomized controlled trials(RCTs).METHODS:A systematic review on RCTs and a systematic review of current guidelines on orally taken PTCMs for uncomplicated ALRTIs were performed.Pub Med,Cochrane Library,EMBASE and four Chinese databases were searched from inception to September 2016 for RCTs testing orally taken PTCMs for uncomplicated ALRTIs(excluding pneumonia).Two reviewers independently screened each study,extracted study data,and assessed risk of bias.Disagreements were resolved through discussion or by consultation with a third reviewer.Clinical practice guidelines for uncomplicated ALRTIs containing PTCM recommendations were identified and quality appraised.The quality of pooled evidence of the RCTs and the guidelines was assessed with GRADE and AGREE Ⅱ respectively.The consistency of the evidence base in RCTs and the guideline recommendations were then compared.RESULTS:For the systematic review of RCTs,4810 papers were identified,among which 29 RCTs(5093 patients) were included in the review.PTCMs compared to placebo increased the effective treatment rate of cough(3 trials,949 patients,risk ratio(RR) 2.50,1.16 to 5.43;low certainty);improved assessment of global health(3 trials,948 patients,RR1.70,1.44 to 2.01;low certainty);and increased the effective rate of specific symptom relief(1 trial,478 patients,RR 4.01,2.76 to 5.81;moderate certainty).21 trials(3432 patients) compared effects of different PTCMs.For the guideline evaluation,29 PTCMs were recommended for the use of uncomplicated ALRTIs,of which27 had no supportive evidence from RCTs.CONCLUSION:The evidence base of PTCMs for uncomplicated ALRTIs is weak and the guideline recommendations were based on almost no clinical trial evidence.Rigorous clinical research is urgently needed to inform the clinical use of these herbal medicines.Further training in evidence-based medicine methods for Traditional Chinese Medicine guideline developers is essential.Xia Ruyu Hu Xiaoyang Wang Di Wang Ying Merlin Willcox Liu Jianping Michael Moore Andrew Flower Li Xun Lily Lai Hu Ruixue Wen Lingzi Zhang Lishan Wang Qi Fei Yutong 2018Journal of Traditional Chinese Medicine2018,38,4:1

5Variation in concentration of dissolved silicate in the Eastern Philippine deep sea显示文摘Upper Circumpolar Deep Water(UCDW)and North Pacifi c Deep Water(NPDW)coexist in the upper deep layer(i.e.,with a 1.2-2.0-℃potential temperature range and a 2000-4100-dbar pressure range)of the Eastern Philippine Sea.They have similar properties in potential temperature and salinity,while have a signifi cant diff erence in dissolved silicate.Based on the repeated observations along a 137°E transect from the World Ocean Database(WOD18),this study revealed the interannual variability of dissolved silicate in the upper deep layer of the Eastern Philippine Sea.Dissolved silicate increased in 1995,1996,2005,2006,and 2007,and decreased in 1997,2000,2001,2002,and 2004.Composition analysis showed that the large diff erence between positive and negative dissolved silicate anomalies occurred mainly at~15°N and north of 25°N,with the concentration reaching 4.25μmol/g.Further analysis indicated that the interannual dissolved silicate variability was related to the zonal current variation in the upper deep layer.The relatively strong(weak)westward current transport increased(decreased)NPDW to the Eastern Philippine Sea,thereby resulting in increased(decreased)dissolved silicate.Ruixue XIA Qiang XIE Weiqiang WANG Hongzhou XU Xuekun SHANG Yeqiang SHU 2023Journal of Oceanology and Limnology2023,41,4:0
6Identification of oleic acid as an endogenous ligand of GPR3显示文摘Although GPR3 plays pivotal roles in both the nervous system and metabolic processes,such as cold-induced thermogenesis,its endogenous ligand remains elusive.Here,by combining structural approach(including cryo-electron microscopy),mass spectrometry analysis,and functional studies,we identify oleic acid(OA)as an endogenous ligand of GPR3.Our study reveals a hydrophobic tunnel within GPR3 that connects the extracellular side of the receptor to the middle of plasma membrane,enabling fatty acids to readily engage the receptor.Functional studies demonstrate that OA triggers downstream Gs signaling,whereas lysophospholipids fail to activate the receptor.Moreover,our research reveals that cold stimulation induces the secretion of OA in mice,subsequently activating Gs/cAMP/PKA signaling in brown adipose tissue.Notably,brown adipose tissues from Gpr3 knockout mice do not respond to OA during cold stimulation,reinforcing the significance of GPR3 in this process.Finally,we propose a“born to be activated and cold to enhance”model for GPR3 activation.Our study provides a starting framework for the understanding of GPR3 signaling in cold-stimulated thermogenesis.Yangjie Xiong Zhenmei Xu Xinzhi Li Yuqin Wang Jing Zhao Na Wang Yaning Duan Ruixue Xia Zhengbin Han Yu Qian Jiale Liang Anqi Zhang Changyou Guo Asuka Inoue Yu Xia Zheng Chen Yuanzheng He 2024Cell Research2024,34,3:0
7Sex-biased single-cell genetic landscape in mice with autism spectrum disorder显示文摘Autistic spectrum disorder(ASD)is a male-biased,heterogeneous neurodevelopmental disorder that affects approximately 1%e2%of the population.Prenatal exposure to valproic acid(VPA)is a recognized risk factor for ASD,but the cellular and molecular basis of VPA-induced ASD at the single-cell resolution is unclear.Here,we aim to compare the cellular and molecular differences in the hippocampus between male and female prenatal mice with ASD at the single-cell transcriptomic level.The transcriptomes of more than 45,000 cells are assigned to 12 major cell types,including neurons,glial cells,vascular cells,and immune cells.Cell type-specific genes with altered expression after prenatal VPA exposure are analyzed,and the largest number of differentially expressed genes(DEGs)are found in neurons,choroid plexus epithelial cells,and microglia.In microglia,several pathways related to inflammation are found in both males and females,including the tumor necrosis factor(TNF),nuclear factor kappa B(NF-kB),toll-like receptor(TLR),and mitogen-activated protein kinase(MAPK)signaling pathways,which are important for the induction of autistic-like behavior.Additionally,we note that several X-linked genes,including Bex1,Bex3,and Gria3,were among the male-specific DEGs of neurons.This pioneering study describes the landscape of the transcriptome in the hippocampus of autistic mice.The elucidation of sexual differences could provide innovative strategies for the prevention and treatment of ASD.Qian Zhang Yongjie Wang Jie Tao Ruixue Xia Yijie Zhang Zhirui Liu Jiwei Cheng 2024Journal of Genetics and Genomics2024,51,3:0

9The synergistic effects of PRDX5 and Nrf2 on lung cancer progression and drug resistance under oxidative stress in the zebrafish models显示文摘Previous studies have shown that PRDX5 and Nrf2 are antioxidant proteins related to abnormal reactive oxidative species(ROS).PRDX5 and Nrf2 play a critical role in the progression of inflammations and tumors.The combination of PRDX5 and Nrf2 was examined by Co-immunoprecipitation,western blotting and Immunohistochemistry.H2O2 was applied to affect the production of ROS and induced multi-resistant protein 1(MRP1)expression in NSCLC cells.The zebrafish models mainly investigated the synergistic effects of PRDX5 and Nrf2 on lung cancer drug resistance under oxidative stress.We showed that PRDX5 and Nrf2 form a complex and significantly increase the NSCLC tissues compared to adjacent tissues.The oxidative stress improved the combination of PRDX5 and Nrf2.We demonstrated that the synergy between PRDX5 and Nrf2 is positively related to the proliferation and drug resistance of NSCLC cells in the zebrafish models.In conclusion,our data indicated that PRDX5 could bind to Nrf2 and has a synergistic effect with Nrf2.Meanwhile,in the zebrafish models,PRDX5 and Nrf2 have significant regulatory impacts on lung cancer progression and drug resistance activities under oxidative stress.SITONG QIAN YING FANG CHENGYUN YAO YONGSHENG WANG ZHI ZHANG XIAOHUA WANG JIN GAO YONG FENG LEI SUN RUNYUE ZOU GUOREN ZHOU JINJUN YE RUIXUE XIA HONGPING XIA 2022Oncology Research2022,30,2:0
