4Laboratory-based flight performance of the fall armyworm,Spodoptera frugiperda显示文摘In late 2018,the fall armyworm(FAW)Spodoptera frugiperda Smith(Lepidoptera:Noctuidae)made its arrival in China and its populations have since proliferated across most of eastern Asia.While S.frugiperda exhibits a considerable dispersal capacity and engages in long-distance migration,there’s only scant information on the species’flight capability.Here,we empirically assessed S.frugiperda flight activity under varying climatic conditions using a flight mill.More specifically,under laboratory conditions,FAW exhibited superior flight performance at 20-25℃ and 60-90%relative humidity(RH).When quantifying flight performance over five consecutive nights(i.e.,10 h/night),all flight parameters initially increased and then gradually dropped and FAW adults attained a total flight distance,duration and velocity equal to 63.73 km(48.42-94.12 km)(median,quartile range),24.12 h(20.87-27.73 h)and 2.73 km h^(-1)(2.13-3.33 km h^(-1)),respectively.Our work constitutes a first comprehensive assessment of S.frugiperda flight performance and provides baseline information for future efforts to forecast spatiotemporal changes in its geographical distribution,movement patterns and invasion trajectories.Such can ultimately permit a timely and targeted deployment of area-wide pest management measures against this newly-invasive pest in China and across eastern Asia.GE Shi-shuai HE Li-mei HE Wei YAN Ran Kris AGWYCKHUYS WU Kong-ming 2021Journal of Integrative Agriculture2021,20,3:10

6Mycoplasma pneumoniae Macrolide Resistance and MLVA Typing in Children in Beijing,China,in 2016:Is It Relevant?显示文摘Objective The aim of this study is to investigate the macrolide resistance rate and molecular type with multiple-locus variable-number tandem-repeat analysis(MLVA)of Mycoplasma pneumoniae of Beijing in 2016 in pediatric patients.Methods Real-time quantitative polymerase chain reaction(PCR)was used to identify M.pneumoniae,and MLVA was performed.The domain V of the 23 S rRNA was sequenced to detect macrolide-resistant point mutations.We also investigated the activities of antibiotics against M.pneumoniae isolates in vitro.Results The PCR detection rate of M.pneumoniae in children in Beijing was 40%,and the macrolide resistance rate was 66%.The A2063 G mutation in the 23 S rRNA V region is the dominant mutation(137/146,93.84%),whereas the A2064 G mutation is rare(9/146,6.16%).Seventy-three samples were typed successfully by MLVA typing,including 86.3%(63/73)were MLVA type 4-5-7-2,and 13.7%(10/73)were MLVA type 3-5-6-2.No other types were found.No strains were resistant to levofloxacin or tetracycline.Conclusion In 2016,a specific decrease in the macrolide resistance rate occurred in Beijing.The detection rate and macrolide resistance rate of outpatients are lower than those of inpatients.The A2063 G mutants M.pneumoniae have high levels of resistance to erythromycin and azithromycin.The primary MLVA type is 4-5-7-2,followed by 3-5-6-2.No other MLVA types were detected.No strains resistant to tetracycline or levofloxacin were found in vitro.DOU Hai Wei TIAN Xiu Jun XIN De Li WEI Ran ZHOU Wei WANG Hong QIN Xuan Guang SHAO Jun Yan XU Bao Ping GE Li Xia SHI Da Wei 2020Biomedical and Environmental Sciences2020,33,12:9

8Flight activity promotes reproductive processes in the fall armyworm,Spodoptera frugiperda显示文摘The fall armyworm(FAW),Spodoptera frugiperda(Lepidoptera:Noctuidae),has invaded many countries in Africa and Asia since 2016,posing a major threat to world food security.Long-distance migration and strong reproductive ability form the biological basis of its rapid population expansion,but the relationship between the flight and reproduction of FAW has not been studied in depth.Here,an empirical assessment of this relationship in an invasive FAW population in China found that 1-3-day-old adults which had undergone 10-h tethered flights had a significantly shorter pre-oviposition period and greater oviposition synchronization,but did not show any differences in fecundity,oviposition period,mating percentage or other reproductive variables.Further studies on moths after 1.25-15-h tethered flights indicated that the reproductive process of adults could be fully triggered by flight activity longer than 2.5 h.Dissection of the reproductive organs also showed that tethered flight promoted ovarian and testicular development of FAW.These results show that appropriate moth flight activity significantly speeds up the reproductive process of FAW,which increases our knowledge on its migratory biology in relation to regional outbreaks.GE Shi-shuai HE Wei HE Li-mei YAN Ran ZHANG Hao-wen WU Kong-ming 2021Journal of Integrative Agriculture2021,20,3:4
9SciFinder-guided rational design of fluorescent carbon dots for ratiometric monitoring intracellular pH fluctuations under heat shock显示文摘Intracellular pH plays a significant role in various biological processes, including cell proliferation,apoptosis, metabolism, enzyme activity and homeostasis. In this work, a novel design strategy for the preparation of pH responsive carbon dots(CDs-pH) for ratiometric intracellular imaging was reported. By using SciFinder database, fluorescent CDs-pH with the required p Kavalue of 6.84 were rationally designed, which is vital important for precise sensing of intracellular pH. As a result, the synthesized CDspH demonstrated robust ability to test pH fluctuations within the physiological range of 5.4-7.4. The CDspH was further utilized for fluorescent ratiometric imaging of pH in living HeLa cells, effectively avoided the influence of autofluorescence from native cellular species. Moreover, real-time monitoring of intracellular pH fluctuation under heat shock was successfully realized. 13Identifying critically ill patients at risk of death from coronavirus disease显示文摘BACKGROUND:A pandemic of coronavirus disease(COVID-19)has been declared by the World Health Organization(WHO)and caring for critically ill patients is expected to be at the core of battling this disease.However,little is known regarding an early detection of patients at high risk of fatality.METHODS:This retrospective cohort study recruited consecutive adult patients admitted between February 8 and February 29,2020,to the three intensive care units(ICUs)in a designated hospital for treating COVID-19 in Wuhan.The detailed clinical information and laboratory results for each patient were obtained.The primary outcome was in-hospital mortality.Potential predictors were analyzed for possible association with outcomes,and the predictive performance of indicators was assessed from the receiver operating characteristic(ROC)curve.RESULTS:A total of 121 critically ill patients were included in the study,and 28.9%(35/121)of them died in the hospital.The non-survivors were older and more likely to develop acute organ dysfunction,and had higher Sequential Organ Failure Assessment(SOFA)and quick SOFA(qSOFA)scores.Among the laboratory variables on admission,we identifi ed 12 useful biomarkers for the prediction of in-hospital mortality,as suggested by area under the curve(AUC)above 0.80.The AUCs for three markers neutrophilto-lymphocyte ratio(NLR),thyroid hormones free triiodothyronine(FT3),and ferritin were 0.857,0.863,and 0.827,respectively.The combination of two easily accessed variables NLR and ferritin had comparable AUC with SOFA score for the prediction of in-hospital mortality(0.901 vs.0.955,P=0.085).CONCLUSIONS:Acute organ dysfunction combined with older age is associated with fatal outcomes in COVID-19 patients.Circulating biomarkers could be used as powerful predictors for the in-hospital mortality.Wei Guo Lin-yu Ran Ji-hong Zhu Qing-gang Ge Zhe Du Fei-long Wang Wei-bo Gao Tian-bing Wang 2021World Journal of Emergency Medicine2021,12,1:2
14Risk factors of recurrent acute pancreatitis after conservative treatment in pediatric population: a single-center study显示文摘Background Although complete resolution and recovery occurs in most children with an initial attack of acute pancreatitis(AP),a subset of children may progress to recurrent AP(RAP).RAP has serious effects to the individual and the socioeconomic burden.The aim of this project was to identify the independent risk factors for pediatric RAP so as to provide evidence for its prevention,early diagnosis and treatment.Methods A retrospective cohort study of children discharged from Tianjin Children’s Hospital from June 2017 to January 2020 was performed.Demographic and clinical variables,treatment strategies,clinical course and outcomes were collected.Independent risk factors of RAP were identified using the logistic regression model.Results Of the total 96 enrolled children,30(31.3%)developed RAP during the follow-up period.The majority(27/30,90%)of the children with AP developed RAP within 6 months of their first AP attack.The presence of systemic inflammatory response syndrome(SIRS)[odds ratio(OR)=6.652,95%confidence interval(CI)1.989 to 22.247],fasting time(OR=1.267,95%CI 1.104 to 1.583),whether meet all three AP diagnostic criteria(OR=7.438,95%CI 1.346 to 41.103)and abnormal amylase/lipase value on the seventh day of hospitalization(OR=3.601,95%CI 0.972 to 13.342)were independent risk factors of RAP in children.Conclusions Most children who developed RAP had progressed within 6 months after their first episode of AP.RAP was more common in children who met all three AP diagnostic criteria at initial attack and in children with SIRS,long fasting time and abnormal amylase/lipase value on the seventh day of hospitalization.Qipeng Zheng Xueqiang Yan Liang Ge Shujian Zhang Yan Sun Jinfu Jia Shengqiao Zhao Xiaodan Xu Ran Dou Qingyun Gou Jinfeng Zhao Jianghua Zhan 2021World Journal of Pediatric Surgery2021,4,1:1
15KRT17 Promotes the Activation of HSCs via EMT in Liver Fibrosis显示文摘Background and Aims:Although activation of hepatic stellate cells(HSCs)plays a central role in the development of liver fibrosis,the mechanism underlying the activation of HSCs remains unclear.Keratin 17(KRT17),a member of the intermediate filament family,can regulate tumor cell proliferation and migration.The current study aimed to elucidate the role of KRT17 in the activation of HSCs and the mechanisms underlying liver fibrosis.Methods:The expression of KRT17 was determined using immunohistochemistry in tissue microarray.Western blotting and qRT-PCR assays were used to determine the KRT17 expression in fibrotic liver tissues obtained from human subjects and mice.LX-2 cells were treated with TGF-β1 recombinant protein and adipocyte differentiation mixture(MDI)mix to induce and reverse LX-2 cell activation,respectively,in order to explore the correlation between KRT17 and HSC activation.Additionally,cell proliferation and migration abilities of LX-2 cells transfected with KRT17-overexpressing plasmid or small interfering RNA were determined using CCK-8,flow cytometry,Transwell,and wound healing assays.Finally,rescue assay was used to explore the role of KRT17 in HSC activation and epithelial-mesenchymal transition(EMT).Results:The expression of KRT17 was higher in the hu-man and mouse fibrotic liver tissues than in healthy liver tissues,and it was positively correlated with HSC activa-tion.Upregulated KRT17 enhanced proliferation,migration,HSC activation and EMT in LX-2 cells,while knockdown of KRT17 reversed these effects.TGF-β1 recombinant protein accelerated KRT17-mediated EMT,HSC activation and proliferation,while TGF-β1 inhibitor counteracted the effect of KRT17 in vitro.Conclusions:KRT17 promoted HSC activation,proliferation and EMT in hepatic fibrosis probably via TGF-β1 signaling,and KRT17 might serve as a therapeutic target for the treatment of liver fibrosis.Jing Chen Si-Jia Ge Hai-Juan Feng Shu-Zhen Wu Ran Ji Wei-Rong Huang Wei Huang Cui-Hua Lu 2022Journal of Clinical and Translational Hepatology2022,10,2:1
