1Isolation and Identification of Cancer Stem-Like Cells from Murine Melanoma Cell Lines显示文摘In current study,cancer stem-like cells in the murine melanoma B16F10 cells were investigated.CD phenotypes of the B16F10 cells were analyzed by flow cytometry,and the specific CD phenotype cells from the B16F10 cells were isolated by MACS.Then we used colony formation assay in soft agar media,the cell growth assay in serum-free culture media as well as the tumorigenicity investigation of the specific CD phenotype cells in C57BL/6 mice, respectively,to identify cancer stem-like cells in the B16F10 cells.The results showed that the B16F10 cells could form spherical clones in serum-free culture media,and the rate of clonegenesis of CD133 + ,CD44 + and CD44 + CD133 + cells was higher than that of CD133 - ,CD44 - and CD44 + CD133 - cells in soft agar media,respectively. The tumorigenic potential of CD133 + ,CD44 + ,CD44 + CD133 + cells and CD44 + CD133 + CD24 + cells was stronger than that of CD133 - ,CD44 - ,CD44 + CD133 - cells and CD44 + CD133 + CD24 - cells in mice,respectively.In conclusion,the CD44 + CD133 + CD24 + cells have some biological properties of cancer stem-like cells or are highly similar to the characteristics of cancer stem cells(CSC).These results provide an important method for identifying cancer stem-like cells in B16F10 cells and for further cancer target therapy.Jun Dou Meng Pan Ping Wen Yating Li Quan Tang Lili Chu Fengshu Zhao Chuilian Jiang Weihua Hu Kai Hu Ning Gu 2007Cellular & Molecular Immunology2007,4,6:25
2IntegrinαEβ7^(+)T cells direct intestinal stem cell fate decisions via adhesion signaling显示文摘Intestinal stem cell(ISC)differentiation is regulated precisely by a niche in the crypt,where lymphocytes may interact with stem and transient amplifying(TA)cells.However,whether and how lymphocyte–stem/TA cell contact affects ISC differentiation is largely unknown.Here,we uncover a novel role of T cell–stem/TA cell contact in ISC fate decisions.We show that intestinal lymphocyte depletion results in skewed ISC differentiation in mice,which can be rescued by T cell transfer.Mechanistically,integrinαEβ7 expressed on T cells binds to E-cadherin on ISCs and TA cells,triggering E-cadherin endocytosis and the consequent Wnt and Notch signaling alterations.BlockingαEβ7−E-cadherin adhesion suppresses Wnt signaling and promotes Notch signaling in ISCs and TA cells,leading to defective ISC differentiation.Thus,αEβ7^(+)T cells regulate ISC differentiation at single-cell level through cell–cell contact-mediatedαEβ7−E-cadherin adhesion signaling,highlighting a critical role of the T cell–stem/TA cell contact in maintaining intestinal homeostasis.Shiyang Chen Yajuan Zheng Xiaojuan Ran Hui Du Hua Feng Lei Yang Yating Wen Changdong Lin Shihui Wang Mengwen Huang Zhanjun Yan Dianqing Wu Hongyan Wang Gaoxiang Ge An Zeng Yi Arial Zeng Jianfeng Chen 2021Cell Research2021,31,12:3

5Effectiveness of a multifaceted intervention for the improvement of nutritional status and nutrition knowledge of children in poverty-stricken areas in Shaanxi Province, China显示文摘Objectives:Compare the differences between the intervention and control groups in changes of primary outcomes(body mass index[BMI]and waist-to-height ratio[WHtR])and secondary outcomes(nutrition knowledge of children and parents),from baseline to the 6-and 12-month follow-ups;examine the associations of changes in nutrition knowledge of children and their parents with children’s changes in BMI and WHtR.Data sources:Data collected from four schools in two poverty-stricken counties in northern Shaanxi Province in 2020‒2021.Methods:A multifaced intervention program targeted children(promoting healthy diet and nutrition education)and their parents(promoting nutrition knowledge)was conducted in the interventional group.Four schools,with two in each group,were randomly allocated to the intervention or control group,with 814 eligible children aged 7.1 to 12.8 years.The control group conducted myopia promotion.Differences in changes of BMI and WHtR between groups were compared with t-test.Mixed-effects model was used to examine the associations between changes in nutrition knowledge of children and parents with changes in children’s BMI and WHtR.Results:At the 6-month follow-up,the difference in changes in BMI between the intervention and control groups was 0.4 kg/m 2(P<0.001).At the 12-month follow-up,the difference in changes in BMI and WHtR between intervention and control groups was 0.1 kg/m^(2)(P<0.001)and 0.01 kg/m^(2)(P<0.001).In the intervention group,the nutrition knowledge awareness rate of children increased from 16.9%(69/409)at baseline to 21.3%(87/409)at 6-month,and 22.7%(93/409)at 12-month.The awareness rate of nutrition knowledge of parents also fluctuated,from 5.6%(23/409)to 6.6%(27/409)and 5.4%(22/409).However,the difference in changes in nutrition knowledge between intervention and control groups was non-significant.The 6-month follow-up changes in children’s nutrition knowledge scores were negatively associated with changes in BMI in girls(β=−0.26,95%confidence interval[CI]:−0.38 to−0.14,P<0.001),while positively associated with changes in WHtR in boys(β=0.003,95%CI:−0.0002 to 0.005,P=0.035).The 12-month follow-up changes in children’s nutrition knowledge scores were positively associated with changes in children’s WHtR(β=0.003,95%CI:0.0004 to 0.01,P=0.018).Conclusions:The intervention strategies did not reduce the BMI and WHtR of children,and the nutrition knowledge of children and parents increased after the intervention in intervention group.Changes in nutrition knowledge of children and their parents are associated with changes in children’s BMI or WHtR.Interventions with a new focus on obesity are needed to help improve children’s nutritional status in poverty-stricken areas in Shaanxi Province of China.Cai Zhao Lu Ma Liwang Gao Yan Wu Yating Yan Wen Peng Youfa Wang 2022Global Health Journal2022,6,3:1
6Engineering the thermostability of D-lyxose isomerase from Caldanaerobius polysaccharolyticus via multiple computer-aided rational design for efficient synthesis of D-mannose显示文摘D-Mannose is an attractive functional sugar that exhibits many physiological benefits on human health.The demand for low-calorie sugars and sweeteners in foods are increasingly available on the market.Some sugar isomerases,such as D-lyxose isomerase(D-LIase),can achieve an isomerization reaction between D-mannose and D-fructose.However,the weak thermostability of D-LIase limits its efficient conversion from D-fructose to D-mannose.Nonetheless,few studies are available that have investigated the molecular modification of D-LIase to improve its thermal stability.In this study,computer-aided tools including FireProt,PROSS,and Consensus Finder were employed to jointly design D-LIase mutants with improved thermostability for the first time.Finally,the obtained five-point mutant M5(N21G/E78P/V58Y/C119Y/K170P)showed high thermal stability and cat-alytic activity.The half-life of M5 at 65◦C was 10.22 fold,and the catalytic efficiency towards 600 g/L of D-fructose was 2.6 times to that of the wild type enzyme,respectively.Molecular dynamics simulation and intramolecular forces analysis revealed a thermostability mechanism of highly rigidity conformation,newly formed hydrogen bonds andπ-cation interaction between and within protein domains,and redistributed surface electrostatic charges for the mutant M5.This research provided a promising D-LIase mutant for the industrial production of D-mannose from D-fructose.Hao Wu Ming Yi Xiaoyi Wu Yating Ding Minghui Pu Li Wen Yunhui Cheng Wenli Zhang Wanmeng Mu 2023Synthetic and Systems Biotechnology2023,8,2:0
7Establishment of new convenient two-line system for hybrid production by targeting mutation of OPR3 in allopolyploid Brassica napus显示文摘The two-line pollination control system,which usually depends on the utilization of thermosensitive or photoperiod genic male-sterile lines,has been widely used in various crops.However,this system is susceptible to instability issues caused by uncontrollable weather fluctuations.A stable and handy two-line pollination control system is highly desirable in many crop species for heterosis exploitation.Oxophytodienoic acid reductase 3(OPR3)was proven to be involved in jasmonate biosynthesis.In the present study,CRISPR/Cas9(Clustered Regularly Interspaced Short Palindromic Repeat)was utilized to mutate two OPR3 homologs in Brassica napus.After two OPR3 homologs were simultaneously mutated,mutants exhibited complete male sterility,and fertility could be easily restored by exogenous MeJA treatment.Hybrids produced from crosses between the opr3 sterile lines and normal varieties exhibited heterosis.This new two-line system based on OPR3 mutation provides higher stability and convenience than traditional systems.By using exogenous MeJA treatment to restore fertility,the system enables more precise control of male fertility transition,which has great potential to significantly contribute to the maneuverable production of hybrid seeds in rapeseed as well as other Brassica species crops.Hongtao Cheng Mengyu Hao Shifei Sang Yunfei Wen Yating Cai Hui Wang Wenxiang Wang Desheng Mei Qiong Hu 2023Horticulture Research2023,10,12:0
8Thermally Activated Delayed Fluorescence of Aggregates Induced by Strongπ–πInteractions and Reversible Dual-Responsive Luminescence Switching显示文摘A reversible dual-responsive luminescent material was introduced by our group to show the simultaneous color and lifetime switching in response to external stimuli.Pristine crystalline powder of(E)-2-(benzo[d]thiazol-2-yl)-3-(pyren-1-yl)acrylonitrile(Py-BZTCN)shows the orderedπ–πstacking with only near-monomer-normal orange-yellow fluorescence,but it exhibits red emission with thermally activated delayed fluorescence(TADF)after grinding,which can be reversibly recovered by heating or fuming treatment.Grinding disturbs the orderedπ–πstacking of pristine powder,leading to the formation of small aggregates with compressed distance and increased overlap ofπ–πstacking between adjacent molecules.The cause of switching was verified by single-crystal X-ray diffraction experiments of two corresponding crystals.This strongπ–πinteraction effectively promotes the excited-state energy splitting and substantially decreases the singlet–triplet energy gap(ΔEST)of aggregates,resulting in the red TADF emission of aggregates through reverse intersystem crossing.This finding proposes a new route to realizing the TADF emission of aggregates through strong intermolecular interactions based on non-TADF monomer,thereby enabling a novel high-contrast dualresponsive luminescence switching.Xiangyu Zhang Tong Lu Changjiang Zhou Haichao Liu Yating Wen Yue Shen Bao Li Shi-Tong Zhang Bing Yang 2022CCS Chemistry2022,4,2:0

11Protein trap-engineered metal-organic frameworks for advanced enzyme encapsulation and mimicking显示文摘Immobilizing enzymes within metal-organic frameworks(MOFs)enables enzymes to against extreme environments.However,these MOF shells are just like armors,protective but heavy,which shield the enzymes from threats while locking them in the cage.The exploitation of immobilization strategy and intrinsic property of MOFs themselves is of great significance.Here,we proposed a functional protein trap strategy for efficient enzyme encapsulation.The ferrocenedicarboxylic acid(Fc)was used to induce the formation of defect-rich Co-based MOFs(CoBDC-Fc).As result,the engineered protein trap can not only improve the enzyme loading but also accelerate catalytic efficiency.Specifically,the atomically dispersed Fc sites serve as cocatalysts/cofactors and even change the conformation of enzymes in the construed microenvironment.Furthermore,the obtained CoBDC-Fc/enzyme exhibits excellent recyclability and tolerance to inhospitable conditions.Benefited by these,the CoBDC-Fc/enzyme/antigen composites were further prepared for cascade enzyme-linked immunosorbent assay of prostate-specific antigen with satisfactory sensitivity.Weiqing Xu Yu Wu Lei Jiao Meng Sha Xiaoli Cai Yating Wen Yifeng Chen Wenling Gu Chengzhou Zhu 2023Nano Research2023,16,2:0
