1Electroacupuncture alters pain-related behaviors and expression of spinal prostaglandin E_2 in a rat model of neuropathic pain显示文摘OBJECTIVE: To investigate the role of spinal prostaglandin E_2(PGE_2) in electroacupuncture(EA) analgesia and assess the theoretical basis for selection of acupoints in the treatment of neuropathic pain.METHODS: A rat model of neuropathic pain was established. Rats were randomly divided into normal,model, sham, EA 1, EA 2, and EA 3 groups. In EA 1group, the rats were needled at bilateral L5 Jiaji(EX-B2), Dachangshu(BL 25), Weizhong(BL 40) and Kunlun(BL 60). In EA 2 group, the rats were needled at bilateral Weizhong(BL 40) and Kunlun(BL60). In EA 3 group, the rats were needled at bilateral L5 Jiaji(EX-B2) and Dachangshu(BL 25). EA stimulation was administered once daily over 7 days. Motor function and thermal withdrawal latencies were evaluated at 1 day preoperatively and at 3, 5, and 7 days postoperatively. After 7 days of intervention,enzyme-linked immunosorbnent assay(ELISA) was used to quantify the expression of the spinal PGE_2.RESULTS: Rats in the model group exhibited evident hyperalgesia in responses to thermal withdrawal latencies compared with those in the control group(P < 0.01), and EA reversed thermal withdrawal latencies(P < 0.01). The expression level of the spinal PGE_2 was significantly higher in the model group than that in the control group and was reversed by EA(P < 0.01; P < 0.05).CONCLUSION: The effect of EA on neuropathic pain might alleviate the hyperalgesia state by an inhibition of local prostaglandin E_2 secretion.Jiang Huili Yu Xue Ren Xiujun Tu Ya 2016Journal of Traditional Chinese Medicine2016,36,1:19
2Genetically modified CD7-targeting allogeneic CAR-T cell therapy with enhanced efficacy for relapsed/refractory CD7-positive hematological malignancies:a phase I clinical study显示文摘Chimeric antigen receptor(CAR)-T cell therapy against T cell malignancies faces major challenges including fratricide between CAR-T cells and product contamination from the blasts.Allogeneic CAR-T cells,generated from healthy donor T cells,can provide ready-to-use,blast-free therapeutic products,but their application could be complicated by graft-versus-host disease(GvHD)and host rejection.Here we developed healthy donor-derived,CD7-targeting CAR-T cells(RD13-01)with genetic modifications to resist fratricide,GvHD and allogeneic rejection,as well as to potentiate antitumor function.A phase I clinical trial(NCT04538599)was conducted with twelve patients recruited(eleven with T cell leukemia/lymphoma,and one with CD7-expressing acute myeloid leukemia).All patients achieved pre-set end points and eleven proceeded to efficacy evaluation.No dose-limiting toxicity,GvHD,immune effector cell-associated neurotoxicity or severe cytokine release syndrome(grade≥3)were observed.28 days post infusion,81.8%of patients(9/11)showed objective responses and the complete response rate was 63.6%(7/11,including the patient with AML).3 of the responding patients were bridged to allogeneic hematopoietic stem cell transplantation.With a median follow-up of 10.5 months,4 patients remained in complete remission.Cytomegalovirus(CMV)and/or Epstein-Barr virus(EBV)reactivation was observed in several patients,and one died from EBV-associated diffuse large B-cell lymphoma(DLBCL).Expansion of CD7-negative normal T cells was detected post infusion.In summary,we present the first report of a Phase I clinical trial using healthy donor-derived CD7-targeting allogeneic CAR-T cells to treat CD7^(+)hematological malignancies.Our results demonstrated the encouraging safety and efficacy profiles of the RD13-01 allogeneic CAR-T cells for CD7^(+)tumors.Yongxian Hu Yali Zhou Mingming Zhang Houli Zhao Guoqing Wei Wengang Ge Qu Cui Qitian Mu Gong Chen Lu Han Tingting Guo Jiazhen Cui Xiaoyan Jiang Xiujun Zheng Shuhui Yu Xiaolong Li Xingwang Zhang Mingxi Chen Xiuju Li Ming Gao Kang Wang Cheng Zu Hao Zhang Xiaohong He Yanbin Wang Dongrui Wang Jiangtao Ren He Huang 2022Cell Research2022,32,11:2
3Effect of electroacupuncture on E-prostanoid 2 expression in the spinal cord in rats with neuropathic pain显示文摘OBJECTIVE: To observe the effect of electroacupuncture(EA) treatment on the expression of E-prostanoid 2(EP2) in the spinal cord using a rat model of neuropathic pain, and to examine the effect of different acupuncture prescriptions.METHODS: Rats were randomly divided into six groups: a normal group, model group, sham group,EA 1 group(distal-proximal acupoints), EA 2 group(proximal acupoints), and EA 3 group(distal acupoints). Rats in the EA groups were treated for20 min, once daily for 7 days. The mechanical withdrawal threshold was evaluated at 1 day preoperatively and at 3, 5, and 7 days postoperatively. At 7days postoperatively, hematoxylin-eosin(HE) staining was used to investigate spinal morphological changes. Spinal EP2 messenger ribonucleic acid(m RNA) was detected using reverse-transcription polymerase chain reaction.RESULTS: After the EA treatment, the mechanical withdrawal threshold was up-regulated compared with the model group(P < 0.05). The expression of EP2 m RNA was up-regulated in the spinal cord on day 7 after surgery(P < 0.05). EA stimulation tended to down-regulate EP2 m RNA expression compared with the model group(P > 0.05). The EA 1and EA 2 groups showed a larger treatment effect than the EA 3 group. The HE results showed that EA can improve spinal histomorphology induced by neuropathic pain.CONCLUSION: EA had a strong analgesic and anti-inflammatory effect. 4Effect of electroacupuncture at distal-proximal acupoint combinations on spinal interleukin-1 beta in a rat model of neuropathic pain显示文摘Objective:Pain from herniated disc is a common type of neuropathic pain.This study investigated whether electroacupuncture (EA) stimulation at distal-proximal combinations of acupoints in the rat model of neuropathic pain modulates spinal interleukin-1 beta (IL-1β) to induce acupuncture analgesia and possibly serve as a pain-relief modality for herniated disc.Methods:A rat model of neuropathic pain was established.Rats were randomly divided into normal,model,sham,EA 1,EA 2,and EA 3 groups.EA 1 rats were needled at bilateral ExB2,BL25,BL40,and BL60 acupoints.EA 2 rats Were needled at bilateral BL40 and BL60.EA 3 rats were needled at bilateral L5 Ex-B2 and BL25.EA stimulation was administered once daily over 7 days.Mechanical withdrawal threshold from noxious mechanical stimulation was measured 1 day preoperatively and at 3,5,and7 days postoperatively.After 7 days of intervention,enzyme-linked immunosorbent assay (ELISA) was used to quantify IL-1β in the spinal cord.Results:Mechanical withdrawal threshold of rats in the model group decreased at 3 days postoperatively when compared with the normal group (P < 0.01),lasting 7 days postoperatively.Mechanical withdrawal thresholds in the EA 1,EA 2,and EA 3 groups were elevated over the model group (P < 0.05;P < 0.01).No obvious differences were found between EA 1,EA 2,and EA 3 groups.ELISA demonstrated an increase in IL-1β in the spinal cord of rats in the model group compared with the normal group (P < 0.01).EA treatment attenuated the increase in spinal IL-1β in the model group.Expression of spinal IL-1β was significantly lower in EA 1,EA 2,and EA 3 groups.Conclusion:EA at distal + proximal acupoints,distal points,as well as proximal points attenuated upregulation of spinal IL-1β,alleviated the extent of neuropathic pain hypersensitivity,and promoted mechanical withdrawal threshold,resulting in EA analgesia.Huili Jiang Xue Yu Xiujun Ren Tingyu Fang Ya Tu 2015Journal of Traditional Chinese Medical Sciences2015,2,1:1
5Impact of seepage flow on sediment resuspension by internal solitary waves:parameterization and mechanism显示文摘Sediment incipient motion is the first step in sediment resuspension.Previous studies ignored the effect of seepage flow on the mobility of sediment particles and simplified the seabed surface as a rigid boundary.A flume experiment was designed to innovatively divide the seabed into two parts to control the dynamic response of the seabed and control the seepage conditions.In the experiment,the seabed sediments and the amplitude of internal solitary waves(ISWs)were changed to compare and analyze the impact of seepage flow on the sediment resuspension by shoaling ISWs.Moreover,parametric research and verification were carried out.Results indicate that seepage flow can greatly influence fine sand,promote sediment resuspension,and increase the amount of suspension by two times on average.However,seepage flow had a little effect on the suspension of clayey silt and sandy silt.Besides,seepage force was added to the traditional gravity,drag force,and uplift force,and the parameterization of threshold starting shear stress of coarse-grained sediments was developed.The results of this parameterization were verified,and seepage force was critical to parameterization.The threshold starting shear stress was reduced by 54.6%after increasing the seepage force.The physical mechanism of this process corresponded to the vertical reciprocating transient seepage in and out the seabed interface caused by the wave-induced transient excess pore water pressure.This quantitative study on seepage flow for shear stress of coarse-grained sediments induced by ISWs is critical to geohazard assessment.Zhuangcai TIAN Chao LIU Ziyin REN Xiujun GUO Mingwei ZHANG Xiuhai WANG Lei SONG Yonggang JIA 2023Journal of Oceanology and Limnology2023,41,2:0
