1Chimeric antigen receptor T cell targeting EGFRvIII for metastatic lung cancer therapy显示文摘Lung cancer is the most common incident cancer and the leading cause of cancer death. In recent years, the development of tumor immunotherapy especially chimeric antigen receptor T (CAR-T) cell has shown a promising future. Epidermal growth factor receptor variant III (EGFRvlll) is a tumor-specific mutation expressed in various types of tumors and has been detected in non-small cell lung cancer with a mutation rate of 10%. Thus, EGFRvIII is a potential antigen for targeted lung cancer therapy. In this study, CAR vectors were constructed and transfected into virus-packaging cells. Then, activated T cells were infected with retrovirus harvested from stable virus-producing single clone cell lines. CAR expression on the surfaces of the T cells was detected by flow cytometry and Western blot. The function of CAR-T targeting EGFRvIII was then evaluated. The EGFRvIII-CAR vector was successfully constructed and confirmed by DNA sequencing. A stable virus-producing cell line was produced from a single clone by limited dilution. The culture conditions for the cell line, including cell density, temperature, and culture medium were optimized. After infection with retrovirus, CAR was expressed on more than 90% of the T cells. The proliferation of CAR-T cells were induced by cytokine and specific antigen in vitro. More importantly, EGFRvIII-CART specifically and efficiently recognized and killed A549-EGFRvIII cells with an effector/target ratio of 10:1 by expressing and releasing cytokines, including perforin, granzyme B, IFN-γ, and TNF-α. The in vivo study indicated that the metastasis of A549-EGFRvIII cells in mice were inhibited by EGFRvIII-CART cells, and the survival of the mice was significantly prolonged with no serious side effects. EGFRvIII-CART showed significantly efficient antitumor activity against lung cancer cells expressing EGFRvlll in vivo and in vitro. 2Expression of hIGF-I in the silk glands of transgenic silkworms and in transformed silkworm cells显示文摘To express human insulin-like growth factor-I (hIGF-I) in transformed Bombyx mori cultured cells and silk glands, the transgenic vector pigA3GFP-hIGF-ie-neo was constructed with a neomycin resistance gene driven by the baculovirus ie-1 promoter, and with the hIGF-I gene under the control of the silkworm sericin promoter Ser-1. The stably transformed BmN cells expressing hIGF-I were selected by using the antibiotic G418 at a final concentration of 700-800 μg/mL after the BmN cells were transfected with the piggyBac vector and the helper plasmid. The specific band of hIGF-I was detected in the transformed cells by Western blot. The expression level of hIGF-I, determined by ELISA, was about 7800 pg in 5×105 cells. Analysis of the chromosomal insertion sites by inverse PCR showed that exogenous DNA could be inserted into the cell genome randomly or at TTAA target sequence specifically for piggyBac element transposition. The transgenic vector pigA3GFP-hIGF-ie-neo was transferred into the eggs using sperm-mediated gene transfer. Finally, two transgenic silkworms were obtained after screening for the neo and gfp genes and verified by PCR and dot hybridization. 3Changing the strategy and culture of stroke awareness education in China: implementing Stroke 1-2-0显示文摘This project implemented the Stroke1-2-0 stroke awareness programme across China and investigated its impact over a 2-year period.We initiated the Stroke1-2-0 educational campaign and Stroke1-2-0 special task forces(STF)across the nation.Massive media coverage,community-based educational sessions with videos and other related materials and induction of Stroke1-2-0 STF were the major means of promotion.We delivered a survey at the end of 2016 and 2018 to evaluate the impact of our effort.A total of 3066 participants responded to the first survey in 2016,and 15207 participants responded in 2018 across China.The acceptance rate for Stroke1-2-0 versus FAST(an English-language stroke awareness tool)was 50.2%versus 19.1%in 2016,and changed significantly to 82.2%versus 8.0%in 2018(p<0.001).Stroke1-2-0 was well accepted by all ages and by people with different academic qualifications.Only 6.5%of survey respondents were aware that there was a therapeutic window for thrombolytic therapy in 2016,but this awareness increased significantly to 32.8%in 2018.Only 12.6%of people in 2016 indicated that they would send patients with stroke to the nearest hospital capable of performing thrombolytic therapy,but there was a nearly threefold increase(52.5%)in this number by 2018.More than 1000 major hospitals joined the Stroke1-2-0 STF,and more than 20000‘stroke warriors’have joined our stroke awareness improvement effort so far.Stroke1-2-0 stroke awareness programme is well-implemented and accepted,and is generating profound improvement in stroke awareness in China.Jing Zhao Xutong Li Xiaochuan Liu Yuming Xu Jihong Xu Anding Xu Yongjun Wang Renyu Liu 2020Stroke & Vascular Neurology2020,5,4:6
4Bake hardening behavior of TRIP and DP steels显示文摘The bake hardening (BH) behavior of transformation-induced plasticity (TRIP) and dual-phase (DP) steels after different prestrains was studied. The experimental results indicate that TRIP steel exhibits good BH ability while DP steel does not, and pre-strain displays a strong effect on the BH values of both steels. The comparison of microstructures of the two steels showed that the hard second phase in the matrix might be harmful to the BH ability. 5How Does Trade Openness Affect Regional Demographic Transitions? Evidence from China's Provincial Panel Data显示文摘This paper investigates the effect of trade openness and other variables on thedemographic transition in China using the instrumental variables regression methodbased on provincial panel data for the period between 1981 and 2013. The resultsindicate that trade openness is one of the determinants of China's demographictransition and has two distinct effects: (i) an income effect that accelerated thedemographic transition by increasing per capita income; and (ii) a human capital effectthat suppressed the demographic transition by reducing human capital accumulation.The effects of trade on demographic transition vary across different regions. 6Femtosecond infrared optical vortex lasers based on optical parametric amplification显示文摘Infrared femtosecond optical vortices open up many new research fields,such as optical micro–nano manipulation,time-resolved nonlocal spectroscopy in solids,vortex secondary radiation and particle generations.In this article,we demonstrate a femtosecond optical vortex laser system based on a two-stage optical parametric amplifier.In our experiment,1.45µm vortex signal pulses with energy of 190µJ and 1.8µm vortex idler pulses with energy of 158µJ have been obtained,and the pulse durations are 51 and 48 fs,respectively.Both the energy fluctuations of the signal and idler pulses are less than 0.5%(root mean square),and the spectral fluctuations are less than 1.5%within 1 hour.This type of highly stable femtosecond optical vortex laser has a wide range of applications for vortex strong-field physics.Renyu Feng Junyu Qian Yujie Peng Yanyan Li Wenkai Li Yuxin Leng Ruxin Li 2022High Power Laser Science and Engineering2022,10,5:1

10Scientific objectives of the Hot Universe Baryon Surveyor(HUBS) mission显示文摘The Hot Universe Baryon Surveyor(HUBS) is a proposed space-based X-ray telescope for detecting X-ray emissions from the hot gas content in our universe. With its unprecedented spatially-resolved high-resolution spectroscopy and large field of view,the HUBS mission will be uniquely qualified to measure the physical and chemical properties of the hot gas in the interstellar medium, the circumgalactic medium, the intergalactic medium, and the intracluster medium. These measurements will be valuable for two key scientific goals of HUBS, namely to unravel the AGN and stellar feedback physics that governs the formation and evolution of galaxies, and to probe the baryon budget and multi-phase states from galactic to cosmological scales. In addition to these two goals, the HUBS mission will also help us solve some problems in the fields of galaxy clusters, AGNs, difuse X-ray backgrounds, supernova remnants, and compact objects. 12Genetic architecture of embryo size and related traits in maize显示文摘The embryo in maize has a critical role in controlling kernel nutrition components and grain yield.We measured five embryo weight and size traits,six kernel weight and size traits,and five embryo-tokernel ratio traits in a nested association mapping(NAM)population of 611 recombinant inbred lines(RILs)derived from four inbred lines including the high-oil,giant-embryo line BY815 as the common parent.Using three statistical methods,we identified 5–22 quantitative trait loci(QTL)for each trait,explaining 4.7%–46.7%of the phenotypic variation.The genetic architecture of maize embryo size and its related traits appeared to be dominated by multiple small-effect loci with little epistasis,and the genetic control underlying embryo size appeared to be distinct from that underlying kernel size.A trait–QTL association network included 205 nodes and 439 edges and revealed 28 key loci associated with at least three traits.Cloned maize genes including Zm Urb2,Emp12 and Zm BAM1 d,maize orthologs of known rice genes that control seed size including BG1,XIAO and GS9,and 11 maize orthologs of Arabidopsis EMBRYO-DEFECTIVE(EMB)genes were identified as underlying these key loci.Further,the phenotypic and genetic relationships between embryo size and kernel size were evaluated,and genetic patterns for identified loci that control embryo size and its related traits were proposed.Our findings reveal distinct genetic architectures for embryo size,kernel size,and embryo-to-kernel ratio traits and establish a foundation for the improvement of embryo-size-mediated kernel nutrition and grain yield.Xiaowei Li Min Wang Renyu Zhang Hui Fang Xiuyi Fu Xiaohong Yang Jiansheng Li 2022The Crop Journal2022,10,1:1
13Identifying QTL and candidate genes for prolificacy in maize显示文摘In maize,prolificacy,the number of ears per plant,is a trait of interest to maize breeders for breeding high grain-yielding cultivars or specialty corn,as well as being a model trait for decoding the molecular mechanism of maize evolution.Its genetic basis remains largely unknown.We identified a stable quantitative trait locus,qEN7,for ear number on chromosome 7 in both F2 and F2:3 populations derived from a single cross between the nonprolific inbred line Mo17 and the prolific inbred line LAN404 derived from the landrace PI217404.qEN7 explained 10.7%-11.9%of phenotypic variation,and the LAN404 allele at this locus was associated with an increase of around one ear per plant.qEN7 was confined by fine-mapping to a 0.56-Mb region containing eight annotated genes.Analysis of selection,gene expression patterns in various maize tissues,and sequence polymorphisms between the two parental lines suggested that Zm00001d020683,which encodes a putative INDETERMINATE DOMAIN(IDD)transcription factor,is the most likely candidate gene underlying qEN7.Zm00001d020683 is expressed mainly in the vegetative meristem,immature ears,and internodes and has undergone selection during maize improvement.The identification of qEN7 and the prediction of its candidate gene sheds some light on the evolution of maize ear number and provides a novel resource for breeding of multi-ear maize cultivars.Min Wang Renyu Zhang Yangzi Zhao Jieyuan Yao Weiya Li Zhixing Yang Fei Sun Xiaohong Yang 2023The Crop Journal2023,11,2:0

16Internal Friction on the Bake-Hardening Behavior of 0.11C-1.67Mn-1.19Si TRIP Steel显示文摘The bake-hardening (BH) values and the 'internal friction-temperature' spectrums were studied for the baked 0.11C-1.67Mn-1.19Si TRIP (transformation induced plasticity) specimens with 0%, 2% and 6% prestrain. Results show that the experimental TRIP steel deserves good bake-hardening ability and Cottrell atmosphere is the reason for its bake hardening characteristic.
