| 1 | Activation of mTORC1 in subchondral bone preosteoblasts promotes osteoarthritis by stimulating bone sclerosis and secretion of CXCL12显示文摘Increasing evidences show that aberrant subchondral bone remodeling plays an important role in the development of osteoarthritis(OA).However,how subchondral bone formation is activated and the mechanism by which increased subchondral bone turnover promotes cartilage degeneration during OA remains unclear.Here,we show that the mechanistic target of rapamycin complex 1(mTORC1)pathway is activated in subchondral bone preosteoblasts(Osterix+)from OA patients and mice.Constitutive activation of mTORC1 in preosteoblasts by deletion of the mTORC1 upstream inhibitor,tuberous sclerosis 1,induced aberrant subchondral bone formation,and sclerosis with little-to-no effects on articular cartilage integrity,but accelerated posttraumatic OA development in mice.In contrast,inhibition of mTORC1 in preosteoblasts by disruption of Raptor(mTORC1-specific component)reduced subchondral bone formation and cartilage degeneration,and attenuated post-traumatic OA in mice.Mechanistically,mTORC1 activation promoted preosteoblast expansion and Cxcl12 secretion,which induced subchondral bone remodeling and cartilage degeneration during OA.A Cxcl12-neutralizing antibody reduced cartilage degeneration and alleviated OA in mice.Altogether,these findings demonstrate that mTORC1 activation in subchondral preosteoblasts is not sufficient to induce OA,but can induce aberrant subchondral bone formation and secrete of Cxcl12 to accelerate disease progression following surgical destabilization of the joint.Pharmaceutical inhibition of the pathway presents a promising therapeutic approach for OA treatment. | Chuangxin Lin Liangliang Liu Chun Zeng Zhong-Kai Cui Yuhui Chen Pinling Lai Hong Wang Yan Shao Haiyan Zhang Rongkai Zhang Chang Zhao Hang Fang Daozhang Cai Xiaochun Bai | 2019 | Bone Research2019,7,1: | 12 |
| 2 | FABP4 secreted by M1-polarized macrophages promotes synovitis and angiogenesis to exacerbate rheumatoid arthritis显示文摘Increasing evidence shows that adipokines play a vital role in the development of rheumatoid arthritis(RA).Fatty acid-binding protein 4(FABP4),a novel adipokine that regulates inflammation and angiogenesis,has been extensively studied in a variety of organs and diseases.However,the effect of FABP4 on RA remains unclear.Here,we found that FABP4 expression was upregulated in synovial M1-polarized macrophages in RA.The increase in FABP4 promoted synovitis,angiogenesis,and cartilage degradation to exacerbate RA progression in vivo and in vitro,whereas BMS309403(a FABP4 inhibitor)and anagliptin(dipeptidyl peptidase 4 inhibitor)inhibited FABP4 expression in serum and synovial M1-polarized macrophages in mice to alleviate RA progression.Further studies showed that constitutive activation of mammalian target of rapamycin complex 1(mTORC1)by TSC1 deletion specifically in the myeloid lineage regulated FABP4 expression in macrophages to exacerbate RA progression in mice.In contrast,inhibition of mTORC1 by ras homolog enriched in brain(Rheb1)disruption specifically in the myeloid lineage reduced FABP4 expression in macrophages to attenuate RA development in mice.Our findings established an essential role of FABP4 that is secreted by M1-polarized macrophages in synovitis,angiogenesis,and cartilage degradation in RA.BMS309403 and anagliptin inhibited FABP4 expression in synovial M1-polarized macrophages to alleviate RA development.Hence,FABP4 may represent a potential target for RA therapy. | Dong Guo Chuangxin Lin Yuheng Lu Hong Guan Weizhong Qi Hongbo Zhang Yan Shao Chun Zeng Rongkai Zhang Haiyan Zhang Xiaochun Bai Daozhang Cai | 2022 | Bone Research2022,10,4: | 5 |

| 5 | Brain organoid-on-chip system to study the effects of breast cancer derived exosomes on the neurodevelopment of brain显示文摘Early human brain development can be affected by multiple prenatal factors that involve chemical exposures in utero,maternal health characteristics such as psychiatric disorders,and cancer.Breast cancer is one of the most common cancers worldwide arising pregnancy.However,it is not clear whether the breast cancer might influence the brain development of fetus.Exosomes secreted by breast cancer cells play a critical role in mediating intercellular communication and interplay between different organs.In this work,we engineered human induced pluripotent stem cells(hiPSCs)-derived brain organoids in an array of micropillar chip and probed the influences of breast cancer cell(MCF-7)derived-exosomes on the early neurodevelopment of brain.The formed brain organoids can recapitulate essential features of embryonic human brain at early stages,in terms of neurogenesis,forebrain regionalization,and cortical organization.Treatment with breast cancer cell derived-exosomes,brain organoids exhibited enhanced expression of stemness-related marker OCT4 and forebrain marker PAX6.RNA-seq analysis reflected several activated signaling pathways associated with breast cancer,medulloblastoma and neurogenesis in brain organoids induced by tumor-derived exosomes.These results suggested that breast cancer cell-derived exosomes might lead to the impaired neurodevelopment in the brain organoids and the carcinogenesis of brain organoids.It potentially implies the fetus of pregnant women with breast cancer has the risk of impaired neurodevelopmental disorder after birth. | Kangli Cui Wenwen Chen Rongkai Cao Yingying Xie Peng Wang Yunsong Wu Yaqing Wang Jianhua Qin | 2022 | Cell Regeneration2022,11,1: | 1 |

| 7 | Stattic sensitizes osteosarcoma cells to epidermal growth factor receptor inhibitors via blocking the interleukin 6-induced STAT3 pathway显示文摘Osteosarcoma(OS),the most common malignant bone tumor with high metastatic potential,frequently affects children and adolescents.Epidermal growth factor receptor(EGFR)-targeted tyrosine kinase inhibitors exhibit encouraging anti-tumor activity for patients with solid tumors,whereas their effects on OS remain controversial.In the present study,we aimed to elucidate the anti-tumor activity of gefitinib for OS,as well as to explore the underlying mechanisms.Gefitinib inhibits cell viability,tumor growth,cell migration,and invasion and promotes cell apoptosis and G1 cycle arrest in OS at a relatively high concentration via suppressing the PI3K/Akt and ERK pathways.However,gefitinib treatment results in the feedback activation of signal transducer and activator of transcription 3(STAT3)induced by interleukin 6(IL-6)secretion.Combined treatment with gefitinib and stattic,an inhibitor for STAT3 phosphorylation,engenders more evident inhibitory effects on cell proliferation,migration,and invasion and promotive effects on cell apoptosis and G1 phase arrest in OS,compared with the single exposure to gefitinib or stattic.Western blot analysis demonstrates that stattic treatment in gefitinib-treated OS abrogates the IL-6-induced STAT3 activation and subsequently further restrains the activities of EGFR,Akt,and ERK pathways in tumor cells.This study confirms that the EGFR inhibitor of gefitinib has moderate anti-tumor effects on OS through IL-6 secretion-mediated STAT3 activation.Additional administration of stattic in EGFR-targeted therapies may contribute to improve the efficacy for OS. | Shenglin Wang Yunqing Wang Zhen Huang Hongxiang Wei Xinwen Wang Rongkai Shen Wenbin Lan Guangxian Zhong Jianhua Lin | 2021 | Acta Biochimica et Biophysica Sinica2021,53,12: | 1 |

| 10 | Shifting the Paradigm:A Fresh Look at Physical Activity Frequency and Its Impact on Mental Health,Life Satisfaction,and Self-Rated Health in Adolescents显示文摘As adolescent mental health problems are becoming a more serious issue globally,this paper explores the relationship of physical activity in adolescents and its frequency on mental health as well as examines the mediating effects of life satisfaction and self-rated health in order to provide a reference for the promotion of mental health in adolescents.A sample of 3578 Chinese high school students completed questionnaires assessing their mental health,physical activity frequency,life satisfaction,and self-rated health.The mean SCL-90 value for adolescents was found to be 1.629%,and 24.73%of adolescents had varying degrees of mental health issue.Increased physical activity frequency is positively associated with improved mental health(p<0.001).Additionally,life satisfaction and self-rated health were found to play significant mediating roles in the relationship between physical activity frequency and mental health.Specifically,low-frequency physical activity had the most pronounced mediating effect on mental health through life satisfaction,while high-frequency physical activity exerted the most significant mediating effect on mental health through self-rated health.These findings underscore the importance of promoting physical activity among adolescents and highlight the distinct pathways through which physical activity frequency can influence mental health outcomes.Further research is needed to explore these relationships in diverse populations and settings,as well as to develop targeted intervention. | Wenjie Li Yucheng Gao Guoqing Liu Rongkai Hao Meijie Zhang Xiaotian Li | 2024 | International Journal of Mental Health Promotion2024,26,2: | 0 |

| 16 | Epidermal growth factor receptor promotes tumor progression and contributes to gemcitabine resistance in osteosarcoma显示文摘Osteosarcoma(OS)is the most common type of primary malignant tumors that originate in the bone.Resistance to chemotherapy confers a poor prognosis on OS patients.Dysregulation of the epidermal growth factor receptor(EGFR)signaling has been reported in sarcomas.However,the functional contribution of EGFR hyperactivation to the tumor biology and chemoresistance remains largely unexplored in OS.In this study,we aimed to investigate the role of EGFR in OS progression and in the response of OS to gemcitabine treatment.The EGFR expression was found to be upregulated in fibroblastic OS cell lines.EGFR knockdown suppressed OS cell proliferation,migration,and invasion in vitro and tumor formation in vivo.Conversely,EGFR overexpression promoted the growth and motility of OS cells.In terms of mechanism,the levels of phospho-Akt and phospho-ERK were decreased upon EGFR knockdown but increased as a result of EGFR overexpression,implying a possible involvement of PI3K/Akt and ERK pathways in mediating the effects of EGFR on OS cells.Moreover,the level of phospho-EGFR was increased in OS cells when exposed to gemcitabine treatment.A more profound proliferative inhibition and a higher rate of apoptosis were obtained in OS cells via inducing cell cycle arrest at G1 phase upon gemcitabine treatment combined with EGFR knockdown,as compared to gemcitabine alone.On the contrary,EGFR overexpression counteracted the growth-inhibiting and pro-apoptotic effects of gemcitabine in OS cells.The present study suggests that EGFR promotes tumor progression and contributes to gemcitabine resistance in OS. | Shenglin Wang Hongxiang Wei Zhen Huang Xinwen Wang Rongkai Shen Zhaoyang Wu Jianhua Lin | 2021 | Acta Biochimica et Biophysica Sinica2021,53,3: | 0 |
