| 2 | Therapeutic efficacy and safety of various botulinum toxin A doses and concentrations in spastic foot after stroke: a randomized controlled trial显示文摘No recommended guidelines currently exist for the therapeutic concentration or dose of botulinum toxin type A(BTXA) injected into the muscle to treat limb spasticity. Therefore, in this randomized controlled trial, we explored the safety and efficacy of two concentrations and two doses of BTXA in the treatment of spastic foot after stroke to optimize this treatment in these patients. Eligible patients(n = 104) were randomized into four groups. The triceps surae and tibialis posterior on the affected side were injected with BTXA at one of two doses(200 U or 400 U) and two concentrations(50 U/m L or 100 U/m L). The following assessments were conducted before as well as 4 days and 1, 2, 4, and 12 weeks after treatment: spasticity, assessed using the modified Ashworth scale; basic functional mobility, assessed using a timed up and go test; pace, assessed using a 10-meter timed walking test; and the ability to walk, assessed using Holden's graded scale and a visual analog scale. The reported results are based on the 89 patients that completed the study. We found significant differences for the two doses and concentrations of BTXA to improve the ability of patients to walk independently, with the high-dose/low-concentration combination providing the best effect. Onset and duration of the ameliorating effects of BTXA were 4–7 days and 12 weeks, respectively. | 4 | A new tool for comprehensive geriatric assessment in elderly patients with acute myeloid leukemia:a pilot study from China显示文摘Background:Acute myeloid leukemia(AML)is a common type of hematological malignancy in elderly people.Geriatricians developed comprehensive geriatric assessment(CGA)methods for elderly patients;however,the tools used for CGA in AML are not uniform.Thus,we aimed to validate the instrumental activities of daily living(IADL)scales,age,comorbidities(Charlson Comorbidity Index),and albumin(IACA)index,which is a new tool for CGA,in elderly patients with AML.Methods:Patients aged>60 years who had been diagnosed with AML were screened for eligibility.Among the IACA low-,intermediate-,and high-risk groups,continuous variables were compared using the Mann-Whitney U test,and categorical variables were compared using/2 and Fisher exact tests.In addition,probabilities of overall survival(OS)were estimated using the Kaplan-Meier method.Results:A total of 21,34,and 6 patients were categorized into IACA low-risk(0 point),intermediate-risk(1-2 points),and high-risk(>3 points)groups,respectively.The rates of relapse/progression-related mortality were 23.8%,58.8%,and 100.0%in the IACA low-,intermediate-,and high-risk groups,respectively(χ^2=12.81,P<0.001).The 2-year probabilities of OS were 47.7%(95%confidence interval[CI]22.8%-72.6%)and 20.2%(95%CI 5.9%-34.5%)in the IACA low-and intermediate-risk groups,respectively(χ^2=5.99,P=0.014),which were significantly higher than those in the high-risk group(low-risk[47.7%(95%CI 22.8%-72.6%)]vs.high-risk[0],/=20.80,P<0.001;intermediate-risk[20.2%(95%CI 5.9%-34.5%)]vs.high-risk[0],χ^2=7.56,P=0.006;respectively).In the IACA low-risk group,the 2-year probability of OS in patients receiving induction chemotherapy(50.8%[95%CI 24.1%-77.5%])was significantly higher than that in those receiving best supportive care(0,χ^2=25.74,P<0.001).Conclusion:We suggest that the IACA index might be a simple and effective tool for comprehensive geriatric assessment in elderly AML patients. | Chun-Li Zhang Ru Feng Jiang-Tao Li Ting Wang Jie-Fei Bai Hui Liu | 2020 | Chinese Medical Journal2020,,4: | 5 |
| 5 | Inhibitory effects of lapachol on rat C6 glioma in vitro and in vivo by targeting DNA topoisomerase Ⅰ and topoisomerase Ⅱ显示文摘OBJECTIVE The aim of this study is to investigate the inhibitory effects of lapachol on rat C6 glioma both in vitro and in vivo,as well as the potential mechanisms.METHODS First,the model of C6 glioma in Wistar rats was established and verified by hemotoxylin and eosin staining,immunohistochemical staining and magnetic resonance imaging(MRI).Then different doses of lapachol were gavaged and tumor volumes of the C6 glioma were detected by MRI.The effects of lapachol on C6 cell proliferation,apoptosis and DNA damage were detected by 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium(MTS)/phen-azinemethosulfate(PMS)assay,Hoechst33358 staining,AnnexinⅤ-FITC/PI staining,and comet assay.Effects of lapachol on topoisomeraseⅠ(TOPⅠ)and topoisomeraseⅡ(TOPⅡ)activities were detected by TOPⅠand TOPⅡmediated supercoiled p BR322 DNA relaxation assay.Molecular docking was used to predict the interaction of lapachol-TOPⅠand lapachol-TOPⅡ.TOP I and TOPⅡexpression levels in C6 cells were determined by Enzymelinked immunosorbent assay kits and real-time polymerase chain reaction(RT-PCR).RESULTS The rat C6 glioma model was successfully established.High dose lapachol showed significant inhibitory effect on the C6 glioma in Wistar rats(P<0.05).MTS/PMS assay,Hoechst 33258 staining,AnnexinⅤ-FITC/PI staining,and comet assay showed that lapachol could inhibit proliferation,induce apoptosis and DNA damage of C6 cells in dose dependent manners.Lapachol could inhibit the activities of both TOPⅠandⅡ.Molecular docking showed that lapachol-TOPⅠshowed relatively stronger interaction than that of lapachol-TOPⅡ.Enzyme-linked immunosorbent assay and RT-PCR showed that lapachol could inhibit TOPⅡexpression levels,but not TOPⅠexpression levels.CONCLUSION These results showed that lapachol could significantly inhibit C6 glioma both in vivo and in vitro,which might be related with inhibiting TOPⅠand TOPⅡactivities,as wel as TOPⅡexpression. | XU Huan-li CHEN Qun-ying WANG Hong XU Ping-xiang YUAN Ru LI Xiao-rong BAI Lu XUE Ming | 2016 | 中国药理学与毒理学杂志2016,30,10: | 3 |

| 7 | Effect of sintering temperature on the electrolysis of TiO_2显示文摘The effects of sintering temperature on the microstructure and the conductivity of TiO 2 cathodes were studied by examining the phase composition, microstructure, and element contents of the sintered cathodes and the cathodic products using X-ray diffraction and scan-ning electronic microscopy-energy dispersive spectrometry. The oxygen vacancy, conductivity, average pore diameter, and specific surface area of the sintered cathodes were detected by X-ray photoelectron spectroscopy, four-point probe, and ASPA 2010. The results showed that TiO 2 phase transformations occurred, and oxygen vacancies formed with the increase of sintering temperature. The cathodic conductivity improved, but the average pore diameter and the effective response area of the TiO 2 cathode were reduced when the sintering temperature increased. These phenomena could weaken the contact between reaction ions and electrons and also had the same effect on the cathodes and the molten salt. Moreover, they were disadvantageous to ion migration, so a lower sintering temperature was favorable for the microstructure of electrolysis. Consequently, the cathodic conductivity may be improved, but the microstructure became compact with the increase of sintering temperature.
