3MDR1/P-Glycoprotein Overexpression in Bladder Transitional Cell Carcinoma and its Correlation with Expression of Survivin and Fas显示文摘OBJECTIVE To explore the expression of the MDR1/P-glycoprotein, Fas and survivin and to examine their correlation with the biologic behavior of bladder transitional cell carcinoma (BTCC). METHODS Immunohistochemistry was used to examine the expression of P-gp, survivin and Fas in BTCC (n=64) and normal bladder mucosa (n=12). RESULTS The expression level of P-gp and survivin in BTCC was higher compared to normal bladder mucosa (P<0.01) and their expression was strongly correlated with clinical grading (P<0.01). In BTCC and normal bladder mucosa Fas expression was 50% and 100%, respectively (P< 0.01). Recurrent BTCC showed higher expression than primary BTCC (P< 0.01) and the expression of P-gp in BTCC had a reverse correlation with Fas expression but no correlation with survivin expression. CONCLUSUON The MDR of BTCC was strongly correlated with the ex- pression of P-gp and Fas, but was not correlated with survivin expres- sion. 4Synergistic Catalysis of Enzymes and Biomimetic MOFs: Immobilizing Cyt c on Two-dimensional MOFs to Enhance the Performance of Peroxidase显示文摘Metal-organic frameworks(MOFs) have been widely regarded as promising carriers for enzyme immobilization owing to their advantages in improving loading and regulating interaction with enzymes. However, they are still suffering from the problems of slow mass transfer and compromising activity. In this paper, the active two-dimensional(2D) MOF of Cu-TCPP(Fe)[TCPP=tetrakis(4-carboxy-phenyl)porphyrin], which possesses the biomimetic architecture of peroxidase, was adopted to anchor cytochrome(Cyt c) for the enhancement of catalytic activity. The atomic/nanometer thickness and micrometer lateral dimension of 2D MOFs can ensure the full exposure of immobilized enzymes and a shorter diffusion distance for the reactant molecules. Besides, the active carrier can provide synergistic catalysis and activity compensation during the reaction. 5Study of recombinant human IFN-α-2b bacilli Calmette–Guerin activated killer cells and against bladder cancer cell in vitro显示文摘Presently,one of the most potent immunothera-pies is the application of bacillus Calmette Guerin(BCG)to prevent recurrences of the superficial bladder cancer.Despite its successful use,nonresponders and certain side effects remain a major obstacle.Therefore,current studies aim at developing recombinant BCG(rBCG)strains secreting Th1-like cytokines to improve the effectiveness of the therapy.In this study,a new type of rBCG strain constructed by Tianjin institute of Urology was tested for its immunostimulatory capacity in vitro.Peripheral blood monocytes(PBMC)were stimulated by recombinant BCG and transformed into bacilli Calmette–Guerin activated killer(BAK)cells,and the effect of anticancer BAK cells was studied.Recombinant IFN-a-2b-BCG,wild-type BCG(wBCG),wild-type BCG and IFN-a-2b were coincubated with PBMCs respectively in vitro,and the proliferation of PBMC was detected with MTT in different time.BAK cells have the ability to kill bladder tumor cells,and the antitumor activity of effecter cells was determined by LDH release assay.The result of MTT showed that the proli-feration of PBMC in the recombinant BCG group was more powerful than in the other two groups(P<0.05).The result of LDH release assay showed that the antitumor activity of BAK cells stimulated by Recombinant BCG was the highest in all groups.We conclude that the recombinant BCG can activate more PBMCs to anti-bladder cancer in vitro than wild-type BCG does.FAN Xiaodong HAN Ruifa 2007Frontiers of Medicine2007,1,4:0
6A meta-analysis of randomized trials of maintenance bacillus Calmette-Guerin instillation efficacy against recurrence of T1G3 bladder tumor显示文摘Meta-analysis was used to determine whether maintenance intravesical bacillus Calmette-Guerin(BCG)could reduce recurrence after transurethral resection of tumor 1 grade 3(T1G3)superficial bladder cancer.All available published data of randomized clinical trials comparing transurethral resection plus intravesical BCG to either resection alone or resection plus another treat-ment on the treatment results in patients with superficial bladder cancer of T1G3 were selected for analysis.Both the fixed effects model and random effects model were applied,and the odds ratio(OR)with its 95%confidence interval(CI)was used as the effect size estimate.Sensitivity analysis and publication bias determination were performed by funnel plots and comparing ORs of different models.Within the follow-up period,375 of 915(41.0%)BCG-treated patients and 332 of 733(45.3%)non-BCG-treated patients developed tumor recurrence.In the combined results,a statistically signifi-cant difference in the ORs for tumor recurrence between the two treatment groups was found(randomized model combined effect OR 0.58,95%CI 0.41 to 0.83,P 50.003).The stratified meta-analysis did not show any statistically significant confounding effects on the results when strati-fied by BCG strains.The randomized model combined effect OR of Pasteur F and other strains were 0.50(95%CI 0.26 to 0.95,P 50.04)and 0.63(95%CI 0.40 to 0.99,P 50.04),respectively.Therefore,we came to the conclu-sion that adjuvant maintenance instillation BCG com-bined with transurethral resection of bladder tumor(TURBT)is an effective conservative treatment for pre-venting recurrence of T1G3 bladder cancer.Jiangang PAN Xing ZHOU Ruifa HAN Zhiguang CHEN 2008Frontiers of Medicine2008,2,3:0
