1Nanoparticle-immersed paper imprinting mass spectrometry imaging reveals uptake and translocation mechanism of pesticides in plants显示文摘Design and discovery of carrier-mediated modified pesticides are vital for reducing pesticide dosage and increasing utilization,yet it remains a great challenge due to limited insights into plant translocation mechanisms.Nanostructure/nanoparticle assisted laser desorption/ionization strategy has established itself as a preferential analytical tool for biological tissue analysis,whereas potential applications in plant sciences are hindered with regard to the inability to slice plant leaves and petals.Herein,we report gold nanoparticle(AuNP)-immersed paper imprinting mass spectrometry imaging(MSI)for the spatiotemporal visualization of pesticide translocation in plant leaves.This approach plays a dual role in preserving spatial information and improving ionization efficiency for pesticides regardless of imaging artifacts due to homogenous AuNP deposition.Using this MSI platform,we proposed the elaborate plant translocation mechanism of agrochemicals for the first time,which is currently poorly understood.The dynamic processes of carrier-mediated pesticides can be clearly visualized,including crossing of plasma membranes by transporters,translocation downward in stems through the phloem,diffusion to the xylem and,conversely,accumulation at margins of the treated leaves.Moreover,this AuNP-assisted paper imprinting method could be highly compatible with laser-based MSI instruments,expediting researches across a broad range of fields,especially in nanomaterial development and life sciences.Xinzhou Wu Run Qin Hanxiang Wu Guangkai Yao Yue Zhang Ping Li Yizhu Xu Zhixiang Zhang Zhibin Yin Hanhong Xu 2020Nano Research2020,13,3:8
2Biochemical sensing in graphene-enhanced microfiber resonators with individual molecule sensitivity and selectivity显示文摘Photonic sensors that are able to detect and track biochemical molecules offer powerful tools for information acquisition in applications ranging from environmental analysis to medical diagnosis.The ultimate aim of biochemical sensing is to achieve both quantitative sensitivity and selectivity.As atomically thick films with remarkable optoelectronic tunability,graphene and its derived materials have shown unique potential as a chemically tunable platform for sensing,thus enabling significant performance enhancement,versatile functionalization and flexible device integration.Here,we demonstrate a partially reduced graphene oxide(prGO)inner-coated and fiber-calibrated Fabry-Perot dye resonator for biochemical detection.Versatile functionalization in the prGO film enables the intracavity fluorescent resonance energy transfer(FRET)to be chemically selective in the visible band.Moreover,by measuring the intermode interference via noise canceled beat notes and locked-in heterodyne detection with Hz-level precision,we achieved individual molecule sensitivity for dopamine,nicotine and single-strand DNA detection.This work combines atomic-layer nanoscience and high-resolution optoelectronics,providing a way toward high-performance biochemical sensors and systems.Zhongxu Cao Baicheng Yao Chenye Qin Run Yang Yanhong Guo Yufeng Zhang Yu Wu Lei Bi Yuanfu Chen Zhenda Xie Gangding Peng Shu-Wei Huang Chee Wei Wong Yunjiang Rao 2019Light(Science & Applications)2019,8,1:8
3Vacuum therapy prevents corporeal veno-occlusive dysfunction and penile shrinkage in a cavernosal nerve injured rat model显示文摘Erectile dysf un ction and penile shri nkage are the comm on complications after radical prostatectomy.Penile rehabilitati on is widely applied after the surgery.Vacuum therapy is one of the three penile rehabilitation methods used in the clinical setting,but its mechanism is not well known.This study was designed to investigate whether vacuum erectile device(VED)can prevent corporeal veno-occlusive dysfunction and penile shrinkage in the bilateral cavernous nerve crush(BCNC)rat model.Adult male Sprague-Dawley rats were randomly assigned into three groups:sham group,BCNC group,and BCNC+VED group.After 4 weeks,penile length and intracavernosal pressure(ICP)were measured,and then the middle part of the penis was harvested after dynamic inf usion caver no sometry to complete the followi ng items:smooth muscle/collage n ratios and collagenⅠ/Ⅲratios;ultramicrostructure of the tunica albuginea,endothelial cell,and smooth muscle cell;and the expression of calponin・l and osteopontin.The penile shortening,peak ICP and ICP drop rate after alprostadil injection were significantly improved with vacuum therapy after 4-week treatme nt.Compared with BCNC group,VED sign ifica ntly in creased smooth muscle/collagen ratios,decreased collagenⅠ/Ⅲratios,and preserved the ultramicrostructure of the tunica albuginea,endothelial cell,and smooth muscle cell.The data also showed that an imals exposed to VED could partially reverse the expression of calp onin・l and osteop ontin induced by BCNC.In conclusion,vacuum therapy is effective to prevent penile shrinkage and veno-occlusive dysfunction in penile rehabilitation,which may be associated with well-preserved structure and function of the tunica albuginea,endothelial cell,and smooth muscle cell.Sheng-Qiang Qian Feng Qin Shuang Zhang Yang Yang Qiang Wei Run Wang Jiu-Hong Yuan 2020Asian Journal of Andrology2020,22,3:3
4A LexA-based yeast two-hybrid system for studying light- switchable interactions of phytochromes with their interacting partners显示文摘Phytochromes are a family of photoreceptors in plants that perceive the red(R)and far-red(FR)components of their light environment.Phytochromes exist in vivo in two forms,the inactive Pr form and the active Pfr form,that are interconvertible by treatments with R or FR light.It is believed that phytochromes transduce light signals by interacting with their signaling partners.A GAL4-based lightswitchable yeast two-hybrid(Y2H)system was developed two decades ago and has been successfully employed in many studies to determine phytochrome interactions with their signaling components.However,several pairs of interactions between phytochromes and their interactors,such as the phyACOP1 and phyA-TZP interactions,were demonstrated by other assay systems but were not detected by this GAL4 Y2H system.Here,we report a modified LexA Y2H system,in which the LexA DNA-binding domain is fused to the C-terminus of a phytochrome protein.The conformational changes of phytochromes in response to R and FR light are achieved in yeast cells by exogenously supplying phycocyanobilin(PCB)extracted from Spirulina.The well-defined interaction pairs,including phyA-FHY1 and phyB-PIFs,are well reproducible in this system.Moreover,we show that our system is successful in detecting the phyA-COP1 and phyA-TZP interactions.Together,our study provides an alternative Y2H system that is highly sensitive and reproducible for detecting light-switchable interactions of phytochromes with their interacting partners.Hong Li Xinyan Qin Pengyu Song Run Han Jigang Li 2021aBIOTECH2021,2,2:2

6Optimization of Soundscape in Theme Park Based on Soundscape Comfortable Degree——A Case Study of China Dinosaur Land in Changzhou显示文摘Tourism soundscape is a new field of tourism landscape research, soundscape designs offer new concepts for the landscape research and development. Taking China Dinosaur Land in Changzhou City for example, visitors' perception about soundscape comfort in theme parks was studied. The researches found that acoustic comfort was infl uenced by objective parameters of sound and subjective perception of visitors, and when the soundscape volume stayed at a certain threshold value, visitors would gain the most satisfi ed acoustic comfort experience, and they were most sensitive to sound elements that able to convey cultural connotations of the theme park. 7Nanograins on Ti-25Nb-3Mo-2Sn-3Zr alloy facilitate fabricating biological surface through dual-ion implantation to concurrently modulate the osteogenic functions of mesenchymal stem cells and kill bacteria显示文摘Surface mechanical attrition treatment(SMAT)method is an effective way to generate nanograined(NG)surface on Ti-25 Nb-3 Mo-2 Sn-3 Zr(wt.%)(named as TLM),a kind ofβ-type titanium alloy,and the achieved nanocrystalline surface was proved to promote positive functions of osteoblastic cells.In this work,to further endow the NG TLM alloy with both good osteogenic and antibacterial properties,magnesium(Mg),silver(Ag)ion or both were introduced onto the NG TLM surface by ion implantation process,as a comparison,the Mg and Ag ions were also co-implanted onto coarsegrained(CG)TLM surface.The obtained results show that subsequent ion implantation does not remarkably induce the surface roughness and topography alteration of the SMAT-treated layers,and it also has little impact on the microstructure of the SMAT-derivedβ-Ti nanograins.In addition,the implanted Mg and Ag ions are observed to exist as MgO and metallic Ag na noparticles(NPs)embedding tightly in theβ-Ti matrix with grain size of about 15 and 7 nm,respectively.Initial cell adhesion and functions(including proliferation,osteo-differentiation and extracellular matrix mineralization)of rabbit bone marrow mesenchymal stem cells(rBMMSCs)and the bacterial colonization of Staphylococcus aureus(S.aureus)on the different surfaces were investigated.The in-vitro experimental results reveal that the Mg and Ag single-ion implanted NG surface either significantly promotes the rBMMSCs response or inhibits the growth ofS.aureus,whereas the Mg/Ag coimplanted NG surface could concurrently enhance the rBMMSCs functions as well as inhibit the bacterial growth compared to the NG surface,and this efficacy is more pronounced as compared to the Mg/Ag co-implantation in the CG surface.The SMAT-achieved nanograins in the TLM surface layer are identified to not only play a leading role in determining the fate of rBMMSCs but also facilitate fabricating dualfunctio nal surface with both good osteogenic and antibacterial activities through co-implantation of Mg and Ag ions.Our investigation provides a new strategy to develop high-performance Ti-based implants for clinical application.Run Huang Lei Liu Bo Li Liang Qin Lei Huang Kelvin W.K.Yeung Yong Han 2021Journal of Materials Science & Technology2021,,14:1

13Proteomics Study on the Differentially Expressed Proteins in c-fos-silenced Cells Exposed to PM2.5显示文摘Objective To investigate the effect of c-fos gene silencing on differentially expressed proteins(DEPs) in human bronchial epithelial(HBE) cells after exposure to fine particulate matter(PM2.5).Methods HBE cells and c-fos-silenced HBE cells were exposed to 50 μg/mL PM2.5, LC-MS/MS and tandem mass tag(TMT) labeling methods were combined with bioinformatics methods, and DEPs and interaction networks were identified.Results In the HBE group, 414 DEPs were screened, of which 227 were up-regulated and 187 downregulated. In the c-fos silenced HBE group, 480 DEPs were screened, including 240 up-regulated proteins and 240 down-regulated proteins. KEGG annotations showed that DEPs in the HBE group are mainly concentrated in the glycolysis/gluconeogenesis pathway and those in the c-fos silenced group are concentrated mainly in endoplasmic reticulum and the processing of proteins. Additionally, the abnormal expression of GPRC5 C, DKK4, and UBE2 C was identified in top 15 DEPs. After constructing the protein interaction network, 20 Hub proteins including HNRNPA2 B1, HNRNPL, RPS15 A, and RPS25 were screened from the HBE group and the c-fos silenced HBE group.Conclusion c-fos gene affected the expression of cancer-related proteins. 14Photo-responsive nanoparticles for β-lapachone delivery in vitro显示文摘Drug efficiency delivery and release to target site play an important role in the treatment of cancer. To achieve these goals, developing drug delivery systems is the key step. The common materials used for drug delivery like liposomes, micelles, dendrimers cannot control the delivery and releasing by spatial and temporal. In this paper, we design a photoresponsive drug delivery system to reduce the side effects of traditional chemotherapy on normal cells and improve therapeutic efficacy in treating cancers. We synthesized two kinds of photo-responsive polymers using two different monomers containing two photoresponsive groups. The two monomers were crosslinked by poly(ethylene glycol) dimethacrylate to form polymers and then encapsulated b-lapachone into the nanoparticles, after UV irradiation, the particle will degrade and release the drug. 16Expression of Bcl-2 and Survivin in uterine cervical carcinogenesis and correlation between the expression and infection of HPV_(16/18)显示文摘To investigate the expression of Bcl-2 and Survivin and HPV_ 16/18 infection in uterine cervical carcinogenesis, formalin-fixed, paraffin-embedded specimens from cervical carcinomas, cervical intraepithelial neoplasia (CIN) and normal cervical tissues were studied. Using streptavidin-biotin peroxidase (S-P) immunohistochemical technique, the authors examined the expression of Bcl-2 and Survivin in these specimens. The number of apoptosis cells was assessed in situ by TdT-mediated dUTP-biotin end labeling (TUNEL) method. The infection of HPV type 16, 18 DNA were determined by PCR. It was found that there were significant differences in Bcl-2, Survivin and apoptotic index (AI) between cervical carcinomas, CIN and normal cervical tissues, respectively. Expression of Bcl-2 and AI were correlated with tumor grades, clinical stages and lymph node metastasis and expression of Survivin was associated with tumor grades and lymph node metastasis. There were different positive rate of HPV_ 16/18 between cervical carcinomas, CIN and normal tissues and were not associated with tumor grades, clinical stages and lymph node metastasis. The infection of HPV_ 16/18 was associated with the expression of Bcl-2, Survivin and AI, respectively. It was concluded that the abnormal expression of Bcl-2, Survivin and infection of HPV_ 16/18 were associated with cervical carcinomas. 17Construction of hierarchical In_(2)O_(3)/In_(2)S_(3) microsphere heterostructures for TEA detection显示文摘Construction of heterojunction has been considered as an efficient strategy to enhance the gas-sensing performances of metal oxide semiconductors.On this basis,hierarchical In_(2)O_(3)/In_(2)S_(3) microsphere heterostructures were synthesized by partial oxidation of In_(2)S_(3) precursors which were obtained via a facile hydrothermal method.Besides,gas sensors based on the acquired materials were fabricated to investigate their sensing performances toward triethylamine(TEA).The results reveal that the gas sensor based on In_(2)O_(3)/In_(2)S_(3) exhibits a high response of 37 at 300℃ toward 0.45 mg/L TEA,which is 3.7 times higher than that of bare In_(2)O_(3).Meanwhile,it also possesses fast response/recovery time(19 s/154 s),good repeatability,selectivity and long-term stability.The excellent sensing performances toward TEA are mainly attributed to the massive oxygen vacancy defects and heterojunction formed between In_(2)O_(3) and In_(2)S_(3).This work provides a facile temperature-dependent route to controllably synthesize hierarchical In_(2)O_(3)/In_(2)S_(3) microsphere heterostructures,and the In_(2)O_(3)/In_(2)S_(3) sensor shows great application prospects in TEA detection.Yongjie Zhang Cong Qin Run Zhang Yan Zhang Yan Wang Zhanying Zhang Jianliang Cao 2022Journal of Materiomics2022,8,6:0

19Anti-lysyl oxidase combined with a vacuum device induces penile lengthening by remodeling the tunica albuginea显示文摘This study aimed to explore whether and how anti-lysyl oxidase(anti-LOX)combined with a vacuum device(VD)could promote penile lengthening and to evaluate the effect on erectile function.This study was performed on four groups of adult rats:control,anti-LOX,VD(negative pressure value of-300 mmHg),and anti-LOX+VD.Penile length was measured by a modified VD method and verified on exposed length data.Intracavernous pressure(ICP)and maximum ICP/mean arterial pressure(MAP)ratio were recorded to assess erectile function.For corpus cavernosum,LOX activity and concentrations of pyridinoline,desmosine,hydroxyproline,and elastin were analyzed;transmission electron microscope and Hart’s elastin staining were performed to monitor microstructural changes.Anti-LOX and VD significantly lengthened the penis by 10.8%(3.75 mm)and 8.2%(2.48 mm)compared with the control group,respectively,while anti-LOX+VD achieved the longest penile size(40.58±0.40 mm)which was 17.4%longer than the control group(34.58±0.54 mm).After 1-week washout,no penile retraction was observed.Meanwhile,exposed penile length data confirmed that the penis in the anti-LOX+VD group was also significantly longer.Anti-LOX inhibited LOX activity to reduce pyridinoline level,which led the penile tunica albuginea remodeling.However,it had no effect on hydroxyproline,desmosine,and elastin levels.Moreover,anti-LOX had no impact on erectile function,which was determined by ICP and ICP/MAP ratio.These results suggest that anti-LOX elongates the penis by reducing pyridinoline,which induces tunica albuginea remodeling.This lengthening effect was more obvious when combined with a VD.All procedures had no impact on erectile function.Tao Li Fu-Dong Fu Chang-Jing Wu Feng Qin Run Wang Jiu-Hong Yuan 2020Asian Journal of Andrology2020,22,5:0
20Oxidation of diclofenac by permanganate: Kinetics, products and effect of inorganic reductants显示文摘The large consumption and discharge of diclofenac(DCF) lead to its frequent detection in surface water and groundwater, posing great threats to humans and ecosystems. This study explored the oxidation kinetics of DCF by permanganate(Mn(Ⅶ)), and expounded the underlying reason for the unusual p Hdependency that was unclear in previous studies. The kinetics of DCF analogues(i.e., aromatic secondary amines) by Mn(Ⅶ) oxidation were comparatively investigated. Then, a tentative kinetic model involving the formation of an intermediate between Mn(Ⅶ) and DCF or its analogues was proposed to fit the p H-rate profile. Since DCF contained two chloro groups, and a carboxyl group which could be ionized by negative electrospray ionization, a precursor ionization scanning approach was used for the first time for detection of N-containing chlorinated oxidation products. New degradation pathways of DCF containing ring opening, carboxylation, carbonylation, electrophilic addition, hydroxylation and dehydrogenation were proposed based on the identified oxidation products. Moreover, it was demonstrated that the introduction of various reducing agents such as Mn(Ⅱ), Fe(Ⅱ) and bisulfite significantly improved the oxidation kinetics of DCF by Mn(Ⅶ). The positive effects of Mn(Ⅱ) and Fe(Ⅱ) were mainly attributed to the accelerated formation of MnO_(2)that acted as a catalyst or co-oxidizer contributing to DCF degradation. The presence of bisulfite caused two-stage kinetics, where a sharp drop of DCF concentration followed by a slowdown of DCF removal. In the first stage, potent reactive manganese species(e.g., Mn(Ⅲ), Mn(V), and Mn(VI)) and sulfate radical were generated during reaction of bisulfite with Mn(Ⅶ), whereas bisulfite was depleted fast due to excess Mn(Ⅶ) concentrations and the system became the Mn(Ⅶ)/MnO_(2)system in the second stage.
