1Transcriptional landscape of rice roots at the single-cell resolution显示文摘There are two main types of root systems in flowering plants,namely taproot systems of dicots and fibrous root systems found in monocots.Despite this fundamental split,our current knowledge of cellular and molecular mechanism driving root development is mainly based on studies of the dicot model Arabidopsis.However,the world major crops are monocots and little is known about the transcriptional programs underlying cell-type specification in this clade.Here,we report the transcriptomes of more than 20000 single cells derived from root tips of two agronomically important rice cultivars.Using combined computational and experimental analyses we were able to robustly identify most of the major cell types and define novel cell-type-specific marker genes for both cultivars.Importantly,we found divergent cell types associated with specific regulatory programs,including phytohormone biosynthesis,signaling,and response,which were well conserved between the two rice cultivars.In addition,we detected substantial differences between the cell-type transcript profiles of Arabidopsis and rice.These species-specific features emphasize the importance of analyzing tissues across diverse model species,including rice.Taken together,our study provides insight into the transcriptomic landscape of major cell types of rice root tip at single-cell resolution and opens new avenues to study cell-type specification,function,and evolution in plants.Qing Liu Zhe Liang Dan Feng Sanjie Jiang Yifan Wang Zhuoying Du Ruoxi Li Guihua Hu Pingxian Zhang Yanfei Ma Jan ULohmann Xiaofeng Gu 2021Molecular Plant2021,14,3:9

3Simulation of EOM-based frequency-chirped laser slowing of MgF radicals显示文摘Here we propose a scheme to slow MgF molecules by using EOM-based frequency-chirped radiation pressure slowing.The scheme well addresses the need for a rapid chirp rate while light molecules are being laser slowed,whose scattering rate and recoil velocity are large.Two EOMs are used to compensate the rapidly changing Doppler shifts arised from the movement of molecules,and to cover the hyperfine energy structure of MgF,respectively.Based the scattering rate maps calculated from an optical Bloch equation model,individual molecule trajectories are simulated by using a semi-classical three-dimensional Monte Carlo approach.We show how the modulation configuration of EOM and the magnetic field influence the slowing results.The study shows that a cryogenic buffer gas-cooled MgF beam source is possible to be slowed down with a number of∼1.4×10^(6)–10^(7),and the final forward speed peaks at∼10 m/s near the capture velocity of a molecular MOT.Kang Yan RuoXi Gu Di Wu Jin Wei Yong Xia Jianping Yin 2022Frontiers of physics2022,17,4:1
4Discovery of small-molecule activators of nicotinamide phosphoribosyltransferase(NAMPT)and their preclinical neuroprotective activity显示文摘The decline of nicotinamide adenine dinucleotide(NAD)occurs in a variety of human pathologies including neurodegeneration.NAD-boosting agents can provide neuroprotective benefits.Here,we report the discovery and development of a class of potent activators(NATs)of nicotinamide phosphoribosyltransferase(NAMPT),the rate-limiting enzyme in the NAD salvage pathway.We obtained the crystal structure of NAMPT in complex with the NAT,which defined the allosteric action of NAT near the enzyme active site.The optimization of NAT further revealed the critical role of K189 residue in boosting NAMPT activity.NATs effectively increased intracellular levels of NAD and induced subsequent metabolic and transcriptional reprogramming.Importantly,NATs exhibited strong neuroprotective efficacy in a mouse model of chemotherapy-induced peripheral neuropathy(CIPN)without any overt toxicity.These findings demonstrate the potential of NATs in the treatment of neurodegenerative diseases or conditions associated with NAD level decline.Hong Yao Minghui Liu Leibo Wang Yumeng Zu Chou Wu Chenyu Li Ruoxi Zhang Haigen Lu Feifei Li Shuang Xi Shuangquan Chen Xuanyu Gu Tianya Liu Jie Cai Shirong Wang Maojun Yang Guo-Gang Xing Wei Xiong Lan Hua Yefeng Tang Gelin Wang 2022Cell Research2022,32,6:0
