1Regulation of Pain and Itch by TRP Channels显示文摘Nociception is an important physiological process that detects harmful signals and results in pain perception. In this review, we discuss important experimental evidence involving some TRP ion channels as molecular sensors of chemical, thermal, and mechanical noxious stimuli to evoke the pain and itch sensations.Among them are the TRPA1 channel, members of the vanilloid subfamily(TRPV1, TRPV3, and TRPV4), and finally members of the melastatin group(TRPM2, TRPM3,and TRPM8). Given that pain and itch are pro-survival,evolutionarily-honed protective mechanisms, care has to be exercised when developing inhibitory/modulatory compounds targeting specific pain/itch-TRPs so that physiological protective mechanisms are not disabled to a degree that stimulus-mediated injury can occur. Such events have impeded the development of safe and effective TRPV1-modulating compounds and have diverted substantial resources. A beneficial outcome can be readily accomplished via simple dosing strategies, and also by incorporating medicinal chemistry design features during compound design and synthesis. 2Novel one-pot synthesis of 4H-chromene derivatives using amino functionalized silica gel catalyst显示文摘A simple, efficient, and environmentally benign method for the synthesis of 4H-chromene derivatives was developed using well ordered and recoverable amino functionalized silica gel as a base catalyst. 3Epalrestat improves motor symptoms by reducing oxidative stress and inflammation in the reserpine induced mouse model of Parkinson’s disease显示文摘Background:Parkinson's disease(PD)is a progressive neurodegenerative disorder affecting a large number of elderly people worldwide.The current therapies for PD are symptom-based;they do not provide a cure but improve the quality of life.Muscular dysfunction is the hallmark clinical feature of PD and oxidative stress and inflammation play a critical role in its pathogenesis.Epalrestat is used for the treatment of diabetic neuropathy and is known to improve antioxidative defense mechanisms in the CNS.Therefore,in this study,we investigated the role of Epalrestat in the reserpine induced mouse model of PD.Method:We used Swiss Albino mice for the PD model and tested for akinesia/bradykinesia,muscular rigidity,palpebral ptosis,and tremor,as well as conducting swim and open field tests.Brain samples were used to determine oxidative stress parameters and infiltration of immune cells.Results:Epalrestat treatment significantly improved akinesia and bradykinesia,muscular dysfunctions,tremor level,and gait functions compared to the reserpine group.It also improved the latency in the swim test.Eplarestat significantly reduced lipid peroxidation and NO concentration in different brain tissues and increased the activity of antioxidative enzymes,glutathione,catalase,and superoxide dismutase.Furthermore,Epalrestat reduced neuroinflammation by reducing the number of infiltrating immune cells.Conclusion:Eplarestat improves muscular dysfunction in PD by reducing oxidative stress and inflammation.Md.Mahbubur Rahman Rupali Rani Chakraborti Md.Abdullah Potol Ariful Haque Abir Ozayra Sharmin Mahabub Alam Md.Fazlur Rahman Khan Rownock Afrin Humayra Jannat Rasiqh Wadud Zaki Farhad Habib 2020Animal Models and Experimental Medicine2020,3,1:3
4Para-aortic lymph node involvement should not be a contraindication to resection of pancreatic ductal adenocarcinoma显示文摘BACKGROUND Para-aortic lymph nodes(PALN)are found in the aortocaval groove and they are staged as metastatic disease if involved by pancreatic ductal adenocarcinoma(PDAC).The data in the literature is conflicting with some studies having associated PALN involvement with poor prognosis,while others not sharing the same results.PALN resection is not included in the standard lymphadenectomy during pancreatic resections as per the International Study Group for Pancreatic Surgery and there is no consensus on the management of these cases.AIM To investigate the prognostic significance of PALN metastases on the oncological outcomes after resection for PDAC.METHODS This is a retrospective cohort study of data retrieved from a prospectively maintained database on consecutive patients undergoing pancreatectomies for PDAC where PALN was sampled between 2011 and 2020.Statistical comparison of the data between PALN+and PALN-subgroups,survival analysis with the Kaplan-Meier method and risk analysis with univariable and multivariable time to event Cox regression analysis were performed,specifically assessing oncological outcomes such as median overall survival(OS)and disease-free survival(DFS).RESULTS 81 cases had PALN sampling and 17(21%)were positive.Pathological N stage was significantly different between PALN+and PALN-patients(P=0.005),while no difference was observed in any of the other characteristics.Preoperative imaging diagnosed PALN positivity in one case.OS and DFS were comparable between PALN+and PALN-patients with lymph node positive disease(OS:13.2 mo vs 18.8 mo,P=0.161;DFS:13 mo vs 16.4 mo,P=0.179).No difference in OS or DFS was identified between PALN positive and negative patients when they received chemotherapy either in the neoadjuvant or in the adjuvant setting(OS:23.4 mo vs 20.6 mo,P=0.192;DFS:23.9 mo vs 20.5 mo,P=0.718).On the contrary,when patients did not receive chemotherapy,PALN disease had substantially shorter OS(5.5 mo vs 14.2 mo;P=0.015)and DFS(4.4 mo vs 9.8 mo;P<0.001).PALN involvement was not identified as an independent predictor for OS after multivariable analysis,while it was for DFS doubling the risk of recurrence.CONCLUSION PALN involvement does not affect OS when patients complete the indicated treatment pathway for PDAC,surgery and chemotherapy,and should not be considered as a contraindication to resection.Rupaly Pande Shafiq Chughtai Manish Ahuja Rachel Brown David C Bartlett Bobby V Dasari Ravi Marudanayagam Darius Mirza Keith Roberts John Isaac Robert P Sutcliffe Nikolaos A Chatzizacharias 2022World Journal of Gastrointestinal Surgery2022,14,5:2
5Bioengineering bacterial encapsulin nanocompartments as targeted drug delivery system显示文摘The development of Drug Delivery Systems(DDS)has led to increasingly efficient therapies for the treatment and detection of various diseases.DDS use a range of nanoscale delivery platforms produced from polymeric of inorganic materials,such as micelles,and metal and polymeric nanoparticles,but their variant chemical composition make alterations to their size,shape,or structures inherently complex.Genetically encoded protein nanocages are highly promising DDS candidates because of their modular composition,ease of recombinant production in a range of hosts,control over assembly and loading of cargo molecules and biodegradability.One example of naturally occurring nanocompartments are encapsulins,recently discovered bacterial organelles that have been shown to be reprogrammable as nanobioreactors and vaccine candidates.Here we report the design and application of a targeted DDS platform based on the Thermotoga maritima encapsulin reprogrammed to display an antibody mimic protein called Designed Ankyrin repeat protein(DARPin)on the outer surface and to encapsulate a cytotoxic payload.The DARPin9.29 chosen in this study specifically binds to human epidermal growth factor receptor 2(HER2)on breast cancer cells,as demonstrated in an in vitro cell culture model.The encapsulin-based DDS is assembled in one step in vivo by co-expressing the encapsulin-DARPin9.29 fusion protein with an engineered flavin-binding protein mini-singlet oxygen generator(MiniSOG),from a single plasmid in Escherichia coli.Purified encapsulin-DARPin_miniSOG nanocompartments bind specifically to HER2 positive breast cancer cells and trigger apoptosis,indicating that the system is functional and specific.The DDS is modular and has the potential to form the basis of a multi-receptor targeted system by utilising the DARPin screening libraries,allowing use of new DARPins of known specificities,and through the proven flexibility of the encapsulin cargo loading mechanism,allowing selection of cargo proteins of choice.Alexander Van de Steen Rana Khalife Noelle Colant Hasan Mustafa Khan Matas Deveikis Saverio Charalambous Clare M.Robinson Rupali Dabas Sofia Esteban Serna Diana A.Catana Konstantin Pildish Vladimir Kalinovskiy Kenth Gustafsson Stefanie Frank 2021Synthetic and Systems Biotechnology2021,6,3:2

14Enhanced Clustering Based OSN Privacy Preservation to Ensure k-Anonymity, t-Closeness, l-Diversity, and Balanced Privacy Utility显示文摘Online Social Networks (OSN) sites allow end-users to share agreat deal of information, which may also contain sensitive information,that may be subject to commercial or non-commercial privacy attacks. Asa result, guaranteeing various levels of privacy is critical while publishingdata by OSNs. The clustering-based solutions proved an effective mechanismto achieve the privacy notions in OSNs. But fixed clustering limits theperformance and scalability. Data utility degrades with increased privacy,so balancing the privacy utility trade-off is an open research issue. Theresearch has proposed a novel privacy preservation model using the enhancedclustering mechanism to overcome this issue. The proposed model includesphases like pre-processing, enhanced clustering, and ensuring privacy preservation.The enhanced clustering algorithm is the second phase where authorsmodified the existing fixed k-means clustering using the threshold approach.The threshold value is determined based on the supplied OSN data of edges,nodes, and user attributes. Clusters are k-anonymized with multiple graphproperties by a novel one-pass algorithm. After achieving the k-anonymityof clusters, optimization was performed to achieve all privacy models, suchas k-anonymity, t-closeness, and l-diversity. The proposed privacy frameworkachieves privacy of all three network components, i.e., link, node, and userattributes, with improved utility. The authors compare the proposed techniqueto underlying methods using OSN Yelp and Facebook datasets.
