1From knowledge based software engineering to knowware based software engineering显示文摘The first part of this paper reviews our efforts on knowledge-based software engi- neering, namely PROMIS, started from 1990s. The key point of PROMIS is to gen- erate applications automatically based on domain knowledge as well as software knowledge. That is featured by separating the development of domain knowledge from the development of software. But in PROMIS, we did not find an appropriate representation for the domain knowledge. Fortunately, in our recent work, we found such a carrier for knowledge modules, i.e. knowware. Knowware is a commercial- ized form of domain knowledge. This paper briefly introduces the basic definitions of knowware, knowledge middleware and knowware engineering. Three life circle models of knowware engineering and the design of corresponding knowware im- plementations are given. 2In ovo leptin administration affects hepatic lipid metabolism and microRNA expression in newly hatched broiler chickens显示文摘Background: A leptin-like immunoreactive substance has been found in chicken eggs and has been implicated in serving as a maternal signal to program offspring growth and metabolism. In the present study, we investigated the effects of in ovo leptin administration on hatch weight, serum and hepatic concentrations of metabolites and hormones, as well as on the expression of genes involved in hepatic lipid metabolism and the predicted microRNAs (miRNAs) targeting the affected genes. To this end we injected fertile eggs with either 0.5 μg of recombinant murine leptin or vehicle (PBS) before incubation. Results: Prenatally leptin-exposed chicks showed lower hatch weight, but higher liver weight relative to the body weight, compared to the control group. In ovo leptin treatment increased the hepatic content and serum concentration of leptin in newly hatched chickens. The hepatic contents of triglycerides (TG) and total cholesterol (Tch) were decreased, whereas the serum levels of TG, Tch and apolipoprotein B (ApoB) were increased. The hepatic mRNA expression of sterol regulator element binding protein 1 (SREBP-1c), SREBP-2, hydroxy-3-methylglutaryl coenzyme A reductase (HMGCR) and cholesterol 7α-hydroxylase 1 (CYP7A1) was significantly up-regulated, as was the protein content of both SREBP-1c and SREBP-2 in hepatic nuclear extracts of leptin-treated chickens. Moreover, out of 12 miRNAs targeting SREBP-1c and/or HMGCR, five were significantly up-regulated in liver of leptin-treated chicks, including gga-miR-200b and gga-miR-429, which target both SREBP-1c and HMGCR. 3Locality preserving projection on SPD matrix Lie group: algorithm and analysis显示文摘Symmetric positive definite(SPD) matrices used as feature descriptors in image recognition are usually high dimensional. Traditional manifold learning is only applicable for reducing the dimension of high-dimensional vector-form data. For high-dimensional SPD matrices, directly using manifold learning algorithms to reduce the dimension of matrix-form data is impossible. The SPD matrix must first be transformed into a long vector, and then the dimension of this vector must be reduced. However, this approach breaks the spatial structure of the SPD matrix space. To overcome this limitation, we propose a new dimension reduction algorithm on SPD matrix space to transform high-dimensional SPD matrices into low-dimensional SPD matrices. Our work is based on the fact that the set of all SPD matrices with the same size has a Lie group structure, and we aim to transform the manifold learning to the SPD matrix Lie group. We use the basic idea of the manifold learning algorithm called locality preserving projection(LPP)to construct the corresponding Laplacian matrix on the SPD matrix Lie group. Thus, we call our approach Lie-LPP to emphasize its Lie group character. 4Sexual maturation in hens is not associated with increases in serum leptin and the expression of leptin receptor mRNA in hypothalamus显示文摘Background: In mammals, leptin is an attractive candidate for mediating the metabolic signal and the reproductive function via the specific receptor in hypothalamus. However, till now, the role of leptin on reproduction in birds is less well established. This experiment was conducted to elucidate the role of leptin on the onset of reproduction in bird, as a first step, to detect the changes of peripheral leptin and leptin receptor mRNA expression in hypothalamus between mature and immature hens at the same age. 120 ISA brown pullets at D60 were allocated randomly into two groups, long light(LL) group being raised under artificial light regimes with incrementally increased light phase(from 8 L:16D to 14 L:12D) and short light(SL) group raised on consistent light(8 L:16D) for 12 wk. Results: The results showed that pullets in LL group reached sexual maturation 15 d earlier than those in SL group. Serum E2 showed a significant increase with age, but no difference was observed between two groups. Serum leptin concentration decreased significantly from D112 to D136 in LL, and was markedly higher in LL group than that in SL at D112, while there was no significant difference between two groups at D136. Leptin receptor and GnRH-I mRNA expression in hypothalamus were significantly increased with age, yet there was no significant difference between SL and LL chickens at the same age. The expression of FSH-β and LH-β mRNA in pituitary was increased with age but did not show significant difference between LL and SL group. GnRH-I mRNA expression was very rich in pineal gland, and decreased from D112 to D136 in LL but not in SL group, and there was no difference between two groups at the same age. 5Heat exposure impairs porcine oocyte quality with suppressed actin expression in cumulus cells and disrupted F-actin formation in transzonal projections显示文摘Background:Transzonal projections(TZPs)constitute a structural basis for the communication between the oocyte and its surrounding cumulus cells(CCs),which play critical roles in promoting the oocyte maturation.Previously we found that heat stress(HS)causes loss of TZPs in porcine cumulus-oocyte complexes(COCs)with decreased density of filamentous actin(F-actin).However,the time-course responses of F-actin and its monomeric actins(β-actin andγ-actin)during the in vitro maturation of oocytes remain unclear.Results:In this study,excised porcine ovaries were exposed to HS at 41.5°C for 1 h before COCs were isolated and matured in vitro for 44 h.HS significantly reduced oocyte quality,characterized by impaired cumulus expansion,delayed meiotic resumption and lower survival rate and polar body extrusion rate,as well as decreased expression of mitochondrial DNA-encoded genes and elevated mitochondrial reactive oxygen species concentration.Expression ofβ-actin andγ-actin in CCs increased gradually with oocytes maturation,which was significantly reduced in HS group,especially at 24 h and/or 44 h of in vitro maturation.By contrast,the number of TZPs and the fluorescence intensity of F-actin in zona pellucida decreased gradually during oocytes maturation,which were significantly reduced by HS at 24 h of in vitro maturation.Moreover,colocalization analyses revealed bothβ-actin andγ-actin contribute to the F-actin formation in porcine TZPs,and the colocalization of F-actin with GJ protein connexin 45 was significantly reduced in heat-exposed COCs.Conclusions:The results indicate that the suppression of actin expressions in CCs,which may lead to the F-actin unstabilization in TZPs,will subsequently contribute to the compromised quality of oocytes under HS.Chao Yin Jie Liu Zhanglin Chang Bin He Yang Yang Ruqian Zhao 2020Journal of Animal Science and Biotechnology2020,11,3:2
6Dietary betaine activates hepatic VTGⅡexpression in laying hens associated with hypomethylation of GR gene promoter and enhanced GR expression显示文摘Background: Vitellogenin(VTG) is a precursor of egg yolk proteins synthesized within the liver of oviparous vertebrates. Betaine is an important methyl donor that is reported to improve egg production of laying hens with an unclear mechanism. In the present study, we fed betaine-supplemented diet(0.5%) to laying hens for 4 wk and investigated its effect on VTGII expression in the liver.Results: Betaine did not affect chicken weight, but significantly(P < 0.05) increased egg laying rate accompanied with a significant(P < 0.05) increase in hepatic concentration and plasma level of VTGI. Plasma estrogen level did not change,but the hepatic expression of estrogen receptor α(ERα) mR NA was significantly(P < 0.05) up-regulated. Betaine did not affect the protein content of ERα, but significantly(P < 0.05) increased hepatic expression of glucocorticoid receptor(GR)at both mR NA and protein levels. Also, ERα/GR interaction tended to be enhanced in the liver nuclear lysates of betainesupplemented hens as determined by co-immunoprecipitation. Furthermore, dietary betaine supplementation significantly increased(P < 0.05) the hepatic expression of methyl-transfer enzymes, such as BHMT, GNMT, and DNMT1,which was associated with higher SAM/SAH ratio and hypomethylation of GR promoter regions.Conclusions: Betaine activates hepatic VTGII expression in association with modified DNA methylation of GR gene promoter, GR expression and ERα/GR interaction. 7Comparative proteomic analysis of the effects of high-concentrate diet on the hepatic metabolism and inflammatory response in lactating dairy goats显示文摘Background: To understand the impact of feeding a high-concentrate diet to mid-lactating goats for a long time on liver metabolism and inflammatory response, two dimensional polyacrylamide gel electrophoresis(2-DE) and real-time PCR method were employed to detect proteins differentially expressed in liver and their m RNAs expression in goats fed high concentrate diet(HC) or low concentrate diet(LC). Twelve lactating dairy goats were randomly assigned to either a HC diet group(65 % concentrate of dry matter; n = 6) or a LC diet group(35 % concentrate of dry matter; n = 6) for 10 wk.Results: Twenty differentially expressed spots(≥2.0-fold changes) in the hepatic tissues were excised and successfully identified using MALDI TOF/TOF. Of these, 8 proteins were up-regulated, while the rest 12 proteins were down-regulated in HC goats compared to LC. Differential expressed proteins including alpha enolase 1(ENO1), glutamate dehydrogenase 1(GLUD1), glutathione S-transferase A1(GSTA1), ATP synthase subunit 5β(ATP5β), superoxide dismutase [Cu-Zn](SOD1), cytochrom c oxidase subunit Via(COX6A1) and heat shock protein 60(HSP60) were further verified by real-time PCR and/or western blot at m RNA or protein expression level. Consistent with the 2-DE results, a significant decrease of β-actin protein expression and SOD enzyme activity was observed in liver of HC goats(P < 0.05), while ENO1 protein expression was significantly up-regulated in HC compared to LC goats(P < 0.05). However, western blot analysis did not show a significant difference of hepatic HSP60 protein between HC and LC group, which did not match the decrease of HSP60 content detected by 2-DE analysis. Real-time PCR showed that glutathione S-transferase P1(GSTP1) and SOD1 m RNA expression was significantly decreased in liver of HC goats, while cytochrom c oxidase(COX3) and ATPase 8(ATP8) m RNAs expression were markedly increased compared to LC(P < 0.05). Gene Ontology(GO) analysis revealed that HC diet resulted in altered expression of proteins related to catalytic and mitochondrial metabolism in the liver, and may increase the stress response with up-regulating the expression of differentiation 14(CD14) cluster and serum amyloid A(SAA) as well as C-reactive protein(CRP) in the liver.Conclusions: These results suggest that feeding high concentrate diet to lactating goats for 10 wk leads to the activation of the inflammatory response, and decreases the anti-oxidant capacity, and subsequently impairs the mitochondrial function in the liver.Yongqian Duanmu Rihua Cong Shiyu Tao Jing Tian Haibo Dong Yuanshu Zhang Yingdong Ni Ruqian Zhao 2016Journal of Animal Science and Biotechnology2016,7,2:2
8Sodiophilic skeleton based on the packing of hard carbon microspheres for stable sodium metal anode without dead sodium显示文摘The propensity of metallic Na dendrites from uneven electrodeposits and the low Coulombic efficiency due to the inevitable existence of 'dead sodium' are crucial barriers to realizing the Na metal anode.Herein,we report a multifunctional sodiophilic skeleton based on the packing of hard carbon(HC)microspheres for stable sodium metal electrodeposition without 'dead sodium'.Firstly,HC is sodiophilic substrate due to the intrinsic heteroatoms or defects which is a favor for the nucleation of Na.Secondly,silver nanoparticles electroplating on HC(Ag-HC)was adopted to boost the Na diffusion and further regulate the uniform Na metal epitaxial deposition due to well compatibility with AIMD simulation.Finally,the packing of HC microspheres provides the inner space for Na plating.Importantly,it was first found by Cryo-TEM that Na metal deposition in nanoscale is achieved by oriented attachment along[110]direction,leading to the formation of polycrystalline Na metal film on Ag-HC.Such epitaxial deposition can efficiently reduce the formation of 'dead sodium' as revealed by chromatography tests,allowing the high Coulombic efficiency and good cycling stability robust kinetics.Finally,HC-Ag||Na_(3)V_(2)(PO_(4))_(3)full cell with a low negative/positive ratio of 0.6 is firstly achieved and displays good cycling stability.This finding provides a new practical strategy without pre-plating of Na metals and demonstrates a highly reversible polycrystalline Na metal anode toward a high-energy Na-based battery.Ruoxue Qiu Si Zhao Zhijin Ju Yiyin Huang Lituo Zheng Ruqian Lian Xinyong Tao Zhensheng Hong 2022Journal of Energy Chemistry2022,31,10:2

13Multifunctional two-dimensional van der Waals Janus magnet Cr-based dichalcogenide halides显示文摘Two-dimensional van der Waals Janus materials and their heterostructures offer fertile platforms for designing fascinating functionalities.Here,by means of systematic first-principles studies on van der Waals Janus monolayer Cr-based dichalcogenide halides CrYX (Y = S,Se,Te;X = Cl,Br,I),we find that CrSX (X = Cl,Br,I) are the very desirable high T_(C) ferromagnetic semiconductors with an out-of-plane magnetization.Excitingly,by the benefit of the large magnetic moments on ligand S^(2−) anions,the sought-after large-gap quantum anomalous Hall effect and sizable valley splitting can be achieved through the magnetic proximity effect in van der Waals heterostructures CrSBr/Bi_(2)Se_(3)/CrSBr and MoTe_(2)/CrSBr,respectively.Additionally,we show that large Dzyaloshinskii–Moriya interactions give rise to skyrmion states in CrTeX (X = Cl,Br,I) under external magnetic fields.Our work reveals that two-dimensional Janus magnet Cr-based dichalcogenide halides have appealing multifunctionalities in the applications of topological electronic and valleytronic devices.Yusheng Hou Feng Xue Liang Qiu Zhe Wang Ruqian Wu 2022npj Computational Materials2022,,1:1
14GR-mediated transcriptional regulation of m^(6)A metabolic genes contributes to dietinduced fatty liver in hens显示文摘Background:Glucocorticoid receptor(GR)mediated corticosterone-induced fatty liver syndrome(FLS)in the chicken by transactivation of Fat mass and obesity associated gene(FTO),leading to demethylation of N6-methyladenosine(m^(6)A)and post-transcriptional activation of lipogenic genes.Nutrition is considered the main cause of FLS in the modern poultry industry.Therefore,this study was aimed to investigate whether GR and m^(6)A modification are involved in high-energy and low protein(HELP)diet-induced FLS in laying hens,and if true,what specific m^(6)A sites of lipogenic genes are modified and how GR mediates m^(6)A-dependent lipogenic gene activation in HELP diet-induced FLS in the chicken.Results:Laying hens fed HELP diet exhibit excess(P<0.05)lipid accumulation and lipogenic genes activation in the liver,which is associated with significantly increased(P<0.05)GR expression that coincided with global m^(6)A demethylation.Concurrently,the m^(6)A demethylase FTO is upregulated(P<0.05),whereas the m^(6)A reader YTHDF2 is downregulated(P<0.05)in the liver of FLS chickens.Further analysis identifies site-specific demethylation(P<0.05)of m^(6)A in the mRNA of lipogenic genes,including FASN,SREBP1 and SCD.Moreover,GR binding to the promoter of FTO gene is highly enriched(P<0.05),while GR binding to the promoter of YTHDF2 gene is diminished(P<0.05).Conclusions:These results implicate a possible role of GR-mediated transcriptional regulation of m^(6)A metabolic genes on m^(6)A-depenent post-transcriptional activation of lipogenic genes and shed new light in the molecular mechanism of FLS etiology in the chicken.Yue Feng Yanlin Li Wenduo Jiang Yun Hu Yimin Jia Ruqian Zhao 2022Journal of Animal Science and Biotechnology2022,13,2:1

17Effects of chronic dexamethasone administration on hyperglycemia and insulin release in goats显示文摘Background: Dexamethasone(Dex), a synthetic glucocorticoid, is among the most commonly used drugs worldwide in animals and humans as an anti-inflammatory and immunosuppressive agent. GC has profound effects on plasma glucose level and other metabolic conditions. However, the effect of prolonged use of Dex on glucose metabolism in ruminants is still unclear.Results: Ten goats were randomly assigned to two groups: the control goats were injected with saline, and the Dex-treated goats were intramuscularly injected daily for 21 d with 0.2 mg/kg Dex. The results showed that plasma glucose and insulin concentrations were significantly increased after Dex administration(P < 0.05). Additionally, the content of hepatic glycogen was also markedly increased in Dex-treated goats(P < 0.01), while the content of glycogen in dorsal longissimus was unchanged by Dex(P > 0.05). The expression of several key genes, involved in blood glucose regulation, was detected by real-time PCR in the small intestine, skeletal muscle and liver. The expression of glucose transporter type 2(GLUT2), sodium-glucose transporter 1(SGLT1) and sodium-potassium ATPase(Na-K/ATPase) in the small intestine were generally increased by Dex, and GLUT2 m RNA expression was significantly up-regulated(P < 0.05). In liver, the expression of genes involved in gluconeogenesis including glucose-6-phosphatase catalytic subunit(G6 PC), cytosolic form of phosphoenolpyruvate carboxykinase(PCK1) and pyruvate carboxylase(PC), were significantly down-regulated by Dex. However, the protein expression levels of PCK1 & PCK2 were significantly increased by Dex, suggesting a post-transcriptional regulation. In dorsal longissimus, the m RNA expression of genes associated with gluconeogenesis and the insulin signaling pathway were generally upregulated by Dex, but the m RNA expression of two markers of muscle atrophy, namely F-box protein 32(FBXO32/Atrogin1) and muscle RING-finger protein 1(Mu RF1), was not altered by Dex.Conclusions: Taken together, these results indicate that chronic administration of a low dosage of Dex induces hyperglycemia mainly through gluconeogenesis activation in the goat liver.Liqiong Niu Qu Chen Canfeng Hua Yali Geng Liuping Cai Shiyu Tao Yingdong Ni Ruqian Zhao 2018Journal of Animal Science and Biotechnology2018,9,3:1

20Negative effects of long-term feeding of high-grain diets to lactating goats on milk fat production and composition by regulating gene expression and DNA methylation in the mammary gland显示文摘Background: It is well known that feeding a high concentrate(HC) diet to lactating ruminants likely induces subacute ruminal acidosis(SARA) and leads to a decrease in milk fat production. However, the effects of feeding a HC diet for long periods on milk fatty acids composition and the mechanism behind the decline of milk fat still remains poorly understood. The aim of this study was to investigate the impact of feeding a HC diet to lactating dairy goats on milk fat yield and fatty acids composition with an emphasis on the mechanisms underlying the milk fat depression. Seventeen mid-lactating dairy goats were randomly allocated to three groups. The control treatment was fed a low-concentrate diet(35% concentrate, n = 5, LC) and there were two high-concentrate treatments(65% concentrate, HC), one fed a high concentrate diet for a long period(19 wks, n = 7, HL); one fed a high concentrate diet for a short period of time(4 wk, n = 5, HS). Milk fat production and fatty acids profiles were measured. In order to investigate the mechanisms underlying the changes in milk fat production and composition,the gene expression involved in lipid metabolism and DNA methylation in the mammary gland were also analyzed.Results: Milk production was increased by feeding the HC diet in the HS and HL groups compared with the LC diet(P < 0.01), while the percentage of milk fat was lower in the HL(P < 0.05) but not in the HS group. The total amount of saturated fatty acids(SFA) in the milk was not changed by feeding the HC diet, whereas the levels of unsaturated fatty acids(UFA) and monounsaturated fatty acids(MUFA) were markedly decreased in the HL group compared with the LC group(P < 0.05). Among these fatty acids, the concentrations of C15:0(P < 0.01), C17:0(P < 0.01), C17:1(P < 0.01), C18:1 n-9 c(P < 0.05), C18:3 n-3 r(P < 0.01) and C20:0(P < 0.01) were markedly lower in the HL group, and the concentrations of C20:0(P < 0.05) and C18:3 n-3 r(P < 0.01) were lower in the HS group compared with the LC group. However, the concentrations of C18:2 n-6 c(P < 0.05) and C20:4 n-6(P < 0.05) in the milk fat were higher in the HS group. Real-time PCR results showed that the m RNA expression of the genes involved in milk fat production in the mammary gland was generally decreased in the HL and HS groups compared with the LC group. Among these genes, ACSL1, ACSS1 & 2, ACACA, FAS, SCD, FADS2, and SREBP1 were downregulated in the mammary gland of the HL group(P < 0.05), and the expressions of ACSS2, ACACA, and FADS2 m RNA were markedly decreased in the HS goats compared with the LC group(P < 0.05). In contrast to the gene expression, the level of DNA methylation in the promoter regions of the ACACA and SCD genes was increased in the HL group compared with the LC group(P < 0.05). The levels of ACSL1 protein expression and FAS enzyme activity were also decreased in the mammary gland of the HL compared with the LC group(P < 0.05).Conclusions: Long-term feeding of a HC diet to lactating goats induced milk fat depression and FAs profile shift with lower MUFAs but higher SFAs.
