| 1 | Reduced protein diet with near ideal amino acid profile improves energy efficiency and mitigate heat production associated with lactation in sows显示文摘Background:The study objective was to test the hypothesis that 1)lowering dietary crude protein(CP)increases dietary energetic efficiency and reduces metabolic heat associated with lactation,and 2)excessive dietary leucine(Leu)supplementation in a low CP diet decreases dietary energetic efficiency and increases metabolic heat associated with lactation.Methods:Fifty-four lactating multiparous Yorkshire sows were allotted to 1 of 3 isocaloric diets(10.80 MJ/kg net energy):1)control(CON;18.75%CP),2)reduced CP with a near ideal or optimal AA profile(OPT;13.75%CP)and 3)diet OPT with excessive Leu(OPTLEU;14.25%CP).Sow body weight and backfat were recorded on day 1 and 21 of lactation and piglets were weighed on day 1,4,8,14,18,and 21 of lactation.Energy balance was measured on sows during early(day 4 to 8)and peak(day 14 to18)lactation,and milk was sampled on day 8 and 18.Results:Over 21-day lactation,sows fed OPT lost body weight and body lipid(P<0.05).In peak lactation,sows fed OPT had higher milk energy output(P<0.05)than CON.Sows fed OPTLEU tended(P=0.07)to have less milk energy output than OPT and did not differ from CON.Maternal energy retention was lower(P<0.05)in OPT and OPTLEU compared to CON sows,and did not differ between OPTLEU and OPT sows.Sows fed OPT had higher(P<0.05)apparent energy efficiency for milk production compared to CON.Heat production associated with lactation was lower(P<0.05)or tended to be lower(P=0.082),respectively,in OPT and OPTLEU compared to CON sows.Conclusion:The OPT diet,in peak lactation,improved dietary energy utilization for lactation due to less urinary energy and metabolic heat loss,and triggered dietary energy deposition into milk at the expense of maternal lipid mobilization.Leucine supplementation above requirement may reduce dietary energy utilization for lactation by decreasing the energy partitioning towards milk,partially explaining the effectiveness of OPT diet over CON diets. | Sai Zhang Jay S.Johnson Mu Qiao Nathalie L.Trottier | 2020 | Journal of Animal Science and Biotechnology2020,11,2: | 3 |
| 2 | Strawberry sweetness and consumer preference are enhanced by specific volatile compounds显示文摘Breeding crops for improved flavor is challenging due to the high cost of sensory evaluation and the difficulty of connecting sensory experience to chemical composition.The main goal of this study was to identify the chemical drivers of sweetness and consumer liking for fresh strawberries(Fragaria×ananassa).Fruit of 148 strawberry samples from cultivars and breeding selections were grown and harvested over seven years and were subjected to both sensory and chemical analyses.Each panel consisted of at least 100 consumers,resulting in more than 15,000 sensory data points per descriptor.Three sugars,two acids and 113 volatile compounds were quantified.Consumer liking was highly associated with sweetness intensity,texture liking,and flavor intensity,but not sourness intensity.Partial least square analyses revealed 20 volatile compounds that increased sweetness perception independently of sugars;18 volatiles that increased liking independently of sugars;and 15 volatile compounds that had positive effects on both.Machine learning-based predictive models including sugars,acids,and volatiles explained at least 25%more variation in sweetness and liking than models accounting for sugars and acids only.Volatile compounds such asγ-dodecalactone;5-hepten-2-one,6-methyl;and multiple medium-chain fatty acid esters may serve as targets for breeding or quality control attributes for strawberry products.A genetic association study identified two loci controlling ester production,both on linkage group 6 A.Co-segregating makers in these regions can be used for increasing multiple esters simultaneously.This study demonstrates a paradigm for improvement of fruit sweetness and flavor in which consumers drive the identification of the most important chemical targets,which in turn drives the discovery of genetic targets for marker-assisted breeding. | Zhen Fan Tomas Hasing Timothy S.Johnson Drake M.Garner Michael L.Schwieterman Christopher R.Barbey Thomas A.Colquhoun Charles A.Sims Marcio F.R.Resende Vance M.Whitaker | 2021 | Horticulture Research2021,8,1: | 2 |

| 13 | A behavior and physiology-based decision support tool to predict thermal comfort and stress in non-pregnant,mid-gestation,and late-gestation sows显示文摘Background:Although thermal indices have been proposed for swine,none to our knowledge differentiate by reproductive stage or predict thermal comfort using behavioral and physiological data.The study objective was to develop a behavior and physiology-based decision support tool to predict thermal comfort and stress in multiparous(3.28±0.81)non-pregnant(n=11),mid-gestation(n=13),and late-gestation(n=12)sows.Results:Regression analyses were performed using PROC MIXED in SAS 9.4 to determine the optimal environmental indicator[dry bulb temperature(TDB)and dew point]of heat stress(HS)in non-pregnant,mid-gestation,and lategestation sows with respiration rate(RR)and body temperature(TB)successively used as the dependent variable in a cubic function.A linear relationship was observed for skin temperature(T_(S))indicating that TDB rather than the sow HS response impacted T_(S)and so T_(S)was excluded from further analyses.Reproductive stage was significant for all analyses(P<0.05).Heat stress thresholds for each reproductive stage were calculated using the inflections points of RR for mild HS and TB for moderate and severe HS.Mild HS inflection points differed for non-pregnant,mid-gestation,and late gestation sows and occurred at 25.5,25.1,and 24.0℃,respectively.Moderate HS inflection points differed for non-pregnant,mid-gestation,and late gestation sows and occurred at 28.1,27.8,and 25.5℃,respectively.Severe HS inflection points were similar for non-pregnant and mid-gestation sows(32.9℃)but differed for late-gestation sows(30.8℃).These data were integrated with previously collected behavioral thermal preference data to estimate the TDB that non-pregnant,mid-gestation,and late-gestation sows found to be cool(TDB | Betty R.McConn Allan P.Schinckel Lindsey Robbins Brianna N.Gaskill Angela R.Green‑Miller Donald C.Lay Jr Jay S.Johnson | 2023 | Journal of Animal Science and Biotechnology2023,14,2: | 0 |

| 15 | Development of a unilateral ureteral obstruction model in cynomolgus monkeys显示文摘Background:Chronic kidney disease(CKD)has a high global prevalence and large unmet need.Central to developing new CKD therapies are in vivo models in CKD.However,next-generation antibody,protein,and gene therapies are highly specific,meaning some do not cross-react with rodent targets.This complicates preclinical development,as established in vivo rodent models cannot be utilized unless tool therapeutics are also developed.Tool compounds can be difficult to develop and,if available,typically have different epitopes,sequences,and/or altered affinity,making it unclear how efficacious the lead therapeutic may be,or what dosing regimen to investigate.To address this,we aimed to develop a nonhuman primate model of CKD.Methods:In vivo rodent unilateral ureteral obstruction(UUO)models kidney fibrosis and is commonly used due to its rapidity,consistency,and ease.We describe translation of this model to the cynomolgus monkey,specifically optimizing the model duration to allow adequate time for assessment of novel therapeutics prior to the fibrotic plateau.Results:We demonstrated that disease developed more slowly in cynomolgus monkeys than in rodents post-UUO,with advanced fibrosis developing by 6 weeks.The tubulointerstitial fibrosis in cynomolgus monkeys was more consistent with human obstructive disease than in rodents,having a more aggressive tubular basement expansion and a higher fibroblast infiltration.The fibrosis was also associated with increased transglutaminase activity,consistent with that seen in patients with CKD.Conclusion:This cynomolgus monkey UUO model can be used to test potential human-specific therapeutics in kidney fibrosis. | Linghong Huang Jia Ni Tanika Duncan Zhizhan Song Timothy S.Johnson | 2021 | Animal Models and Experimental Medicine2021,4,4: | 0 |
