Clinically relevant factors associated with quantitative optical coherence tomography angiography metrics in deep capillary plexus in patients with diabetes显示文摘Background:To test clinically relevant factors associated with quantitative artifact-free deep capillary plexus(DCP)metrics in patients with diabetes mellitus(DM).Methods:563 eligible eyes(221 with no diabetic retinopathy[DR],135 with mild DR,130 with moderate DR,and 77 with severe DR)from 334 subjects underwent optical coherence tomography-angiography(OCT-A)with a swept-source OCT(Triton DRI-OCT,Topcon,Inc.,Tokyo,Japan).Strict criteria were applied to exclude from analysis those DCP images with artifacts and of poor quality, including projection artifacts, motion artifacts, blurriness, signal loss,B-scan segmentation error,or low-quality score.A customized MATLAB program was then used to quantify DCP morphology from the artifact-free DCP images by calculating three metrics:foveal avascular zone(FAZ),vessel density(VD),and fractal dimension(FD).Results:166(29.5%)eyes were excluded after quality control, leaving in the analysis 397 eyes(170 with no DR,101 with mild DR,90 with moderate DR,36 with severe DR)from 250 subjects. In the multiple regression models, larger FAZ area was associated with more severe DR(β=0.687;p=0.037),shorter axial length(AL)(β=−0.171;p=0.003),thinner subfoveal choroid thickness(β=−0.122;p=0.031),and lower body mass index(BMI)(β=−0.090;p=0.047).Lower VD was associated with more severe DR(β=−0.842;p=0.001),shorter AL(β=0.107;p=0.039),and poorer visual acuity(VA)(β=−0.133;p=0.021).Lower FD was associated with more severe DR(β=−0.891;p<0.001)and with older age(β=−0.142;p=0.004).Conclusions:Quantitative artifact-free DCP metrics are associated with VA,DR severity,AL,subfoveal choroidal thickness, age, and BMI in diabetic patients. The effects of ocular and systemic factors should be considered for meaningful interpretations of DCP changes in DM patients. | Fang Yao Tang Erica O.Chan Zihan Sun Raymond Wong Jerry Lok Simon Szeto Jason C.Chan Alexander Lam Clement C.Tham Danny S.Ng Carol Y.Cheung | 2020 | Eye and Vision2020,7,1: | 7
Incorporation of a nanotechnology-based additive in cementitious products for clay stabilisation显示文摘The mechanical performances and water retention characteristics of clays, stabilised by partial substitution of cement with by-products and inclusion of a nanotechnology-based additive called RoadCem(RC),are studied in this research. The unconfined compression tests and one-dimensional oedometer swelling were performed after 7 d of curing to understand the influence of addition of 1%of RC material in the stabilised soils with the cement partially replaced by 49%,59%and 69%of ground granulated blast furnace slag(GBBS)or pulverised fuel ash(PFA).The moisture retention capacity of the stabilised clays was also explored using the soil-water retention curve(SWRC)from the measured suctions. Results confirmed an obvious effect of the use of RC with the obtained strength and swell properties of the stabilised clays suitable for road application at 50%replacement of cement. This outcome is associated with the in-depth and penetrating hydration of the cementitious materials by the RC and water which results in the production of needle-like matrix with interlocking filaments e a phenomenon referred to as the‘wrapping’effect. On the other hand, the SWRC used to describe the water holding capacity and corresponding swell mechanism of clays stabilised by a proportion of RC showed a satisfactory response. The moisture retention of the RC-modified clays was initially higher but reduced subsequently as the saturation level increased with decreasing suction. This phenomenon confirmed that clays stabilised by including the RC are water-proof in nature, thus ensuring reduced porosity and suction even at reduced water content.Overall, the stabilised clays with the combination of cement,GGBS and RC showed a better performance compared to those with the PFA included. | E.U.Eyo S.Ng’ambi S.J.Abbey | 2020 | Journal of Rock Mechanics and Geotechnical Engineering2020,12,5: | 2

Suppressing tumor growth of nasopharyngeal carcinoma by hTERTC27 polypeptide delivered through adeno-associated virus plus adenovirus vector cocktail显示文摘Nasopharyngeal carcinoma(NPC) is a metastatic carcinoma that is highly prevalent in Southeast Asia. Our laboratory has previously demonstrated that the C-terminal 27-kDa polypeptide of human telomerase reverse transcriptase (hTERTC27) inhibits the growth and tumorigenicity of human glioblastoma and melanoma cells. In this study, we investigated the antitumor effect of hTERTC27 in human C666-1 NPC cells xenografted in a nude mouse model. A cocktail of vectors comprising recombinant adeno-associated virus (rAAV) and recombinant adenovirus (rAdv) that each carry hTERTC27 (rAAV-hTERTC27 and rAdv- hTERTC27; the cocktail was abbreviated to rAAV/rAdv-hTERTC27) was more effective than either rAAV- hTERTC27 or rAdv-hTERTC27 alone in inhibiting the growth of C666-1 NPC xenografts. Furthermore, we established three tumors on each mouse and injected rAAV/rAdv-hTERTC27 into one tumor per mouse. Although hTERTC27 expression could only be detected in the injected tumors, reduced tumor growth was observed in the injected tumor as well as the uninjected tumors, demonstrating that the vector cocktail could provoke an antitumor effect on distant, metastasized tumors. Further studies showed the observed antitumor effects included inducing necrosis and apoptosis and reducing microvessel density. Together, our data suggest that the rAAV/rAdv-hTERTC27 cocktail can potently inhibit NPC tumor growth in both local and metastasized tumors and should be further developed as a novel gene therapy strategy for NPC. | Xiong Liu Xiang-Ping Li Ying Peng Samuel S.Ng Hong Yao Zi-Feng Wang Xiao-Mei Wang Hsiang-Fu Kung Marie C.M. | 6 | A pan-cancer fingerprint:common molecular denominators of the human tumor microenvironment显示文摘In a recent study published in Cancer Cell,Bagaev et al.developed a pan-cancer tumor microenvironment(TME)classification that can be predictive of response to immune checkpoint inhibitor(ICI)therapy.1 The authors presented a detailed visualization that can be generated for each tumor, which combines the 29 functional gene expression signatures(Fges)used to subtype the TME with targetable genomic alterations.Understanding the compartments and pathways within the TME could aid decision-making in patient-specific treatment of cancer, especially in the context of immunotherapies.2 The increasing use of ICIs have shown promising clinical outcomes in several cancer types. However, response to treatment is heterogenous and often results in immune-related adverse events.3 Therefore, efforts are focused on the development of prognostic tools to predict response to ICI.Unsupervised analysis of 468 melanoma patients resulted in grouping of the TME into four major subsets based on manually curated Fges. | Susanna S.Ng Sonia Leonardelli Michael Hölzel | 2021 | Signal Transduction and Targeted Therapy2021,6,12: | 0
Origin of Unliberated Bitumen in Athabasca Oil Sands显示文摘Oil sands contain a so-called organic rich solids component (ORS), i.e., solids whose surfaces are strongly associated with toluene insoluble organic matter (TIOM). Typically, humic material is the major component of TIOM. It provides sites for adsorption and chemical fixation of bitumen. This bound bitumen is “unliberated”, and considerable mechanical or chemical energy may be required to release it. In order to establish a correlation between bitumen recovery and ORS content, a few selected oil sands were processed in a Batch Extraction Unit (BEU). Analysis of the middlings and coarse tailings streams from these tests indicated a relatively constant bitumen to ORS ratio of 2.8±0.7. This value allows the liberated-unliberated bitumen balance (LUBB) to be calculated for any given oil sands. The amounts of bitumen recovered as primary froth during the BEU experiments are close to the estimated liberated bitumen contents in each case tested. This observation indicates that the liberated-unliberated bitumen calculation is an important quantitative parameter for prediction of bitumen recovery under specific recovery conditions. Preliminary results indicate that the ORS content of an oil sands may be estimated from the carbon content of bitumen free oil sands solids. | TuYun J.B.O'Carroll B.D.Sparks L.S.Kotlyar S.Ng K.H.Chung G.Cuddy | 2005 | Petroleum Science2005,2,1: | 0
A Causality-guided Statistical Approach for Modeling Extreme Mei-yu Rainfall Based on Known Large-scale Modes——A Pilot Study显示文摘Extreme Mei-yu rainfall(MYR)can cause catastrophic impacts to the economic development and societal welfare in China. While significant improvements have been made in climate models, they often struggle to simulate local-to-regional extreme rainfall(e.g.,MYR).Yet,large-scale climate modes(LSCMs)are relatively well represented in climate models.Since there exists a close relationship between MYR and various LSCMs,it might be possible to develop causality-guided statistical models for MYR prediction based on LSCMs.These statistical models could then be applied to climate model simulations to improve the representation of MYR in climate models. In this pilot study,it is demonstrated that skillful causality-guided statistical models for MYR can be constructed based on known LSCMs.The relevancy of the selected predictors for statistical models are found to be consistent with the literature. The importance of temporal resolution in constructing statistical models for MYR is also shown and is in good agreement with the literature. The results demonstrate the reliability of the causality-guided approach in studying complex circulation systems such as the East Asian summer monsoon(EASM).Some limitations and possible improvements of the current approach are discussed. The application of the causality-guided approach opens up a new possibility to uncover the complex interactions in the EASM in future studies. | Kelvin S.NG Gregor C.LECKEBUSCH Kevin I.HODGES | 2022 | Advances in Atmospheric Sciences2022,39,11: | 0
