| 1 | PARylation regulates stress granule dynamics, phase separation, and neurotoxicity of disease-related RNA-binding proteins显示文摘Mutations in RNA-binding proteins (RBPs) localized in ribonucleoprotein (RNP) granules, such as hnRNP A1 and TDP-43, promote aberrant protein aggregation, which is a pathological hallmark of various neurodegenerative diseases, such as amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD). Protein posttranslational modifications (PTMs) are known to 佗gulate RNP granules. In this study, we investigate the function of poly(ADP-ribosyl)ation (PARylation), an important PTM involved in DNA damage repair and cell death, in RNP granule-related neurodegeneration. We reveal that PARylation levels are a major regulator of the assembly-disassembly dynamics of RNP granules containing disease-related RBPs, hnRNP A1 and TDP-43. We find that hnRNP A1 can both be PARylated and bind to PARylated proteins or poly(ADP-ribose)(PAR). We further uncover that PARylation of hnRNP A1 at K298 controls its nucleocytoplasmic transport, whereas PAR-binding via the PAR-binding motif (PBM) of hnRNP Al regulates its association with stress granules. Moreover, we reveal that PAR not only dramatically enhances the liquid-liquid phase separation of hnRNP A1, but also promotes the co-phase separation of hnRNP A1 and TDP-43 in vitro and their interaction in vivo. Finally, both genetic and pharmacological inhibition of PARP mitigates hnRNP Al- and TDP-43-mediated neurotoxicity in cell and Drosophila models of ALS. | 2 | Microbial Characterization of Denitrifying Sulfide Removal Sludge Using High-Throughput Amplicon Sequencing Method显示文摘The denitrifying sulfide removal(DSR) process has recently been studied extensively from an engineering perspective. However, the importance of microbial communities of this process was generally underestimated. In this study, the microbial community structure of a lab-scale DSR reactor was characterized in order to provide a comprehensive insight into the key microbial groups in DSR system. Results from high-throughput sequencing analysis revealed that the fraction of autotrophic denitrifiers increased from 2.34 % to 10.93% and 44.51% in the DSR system when the influent Na Cl increased from 0 g/L, to 4 g/L and 30 g/L, respectively. On the contrary, the fraction of heterotrophic denitrifiers decreased from 61.74% to 39.57%, and 24.12%, respectively. Azoarcus and Thiobacillus were the main autotrophic denitrifiers, and Thauera was the main hetetrophic denitrifier during the whole process. | 3 | A numerical modelling approach to assess the behaviour of underground cavern subjected to blast loads显示文摘The paper gives an insight into the behaviour of large underground caverns which are subjected to blast loads. Caverns are generally constructed in hard rock formation which compels us to use blasting methods for the excavation works. Comparative study was done between models with intact rock mass and discontinuities to assess the stability of cavern as a result of blast loads. Numerical modelling was performed with 3 dimensional distinct element code(3 DEC) to analyse the performance of cavern walls in terms of displacement and to compute peak particle velocities(PPV) both around the cavern periphery and at surface of models. Results showed that the velocity wave with higher frequency exhibited large displacements around the periphery of cavern. Computation of PPV showed that model with horizontal joint sets showed lower PPV in comparison to model with intact rock mass. PPV values were also analysed on the surface for model consisting vertical joints spaced at 4 m intervals. Comparative study of PPV on surface vertically above the blast location between models with horizontal joints spaced at 4 m and vertical joints at 4 m intervals were conducted. | 8 | Effects of Fe^(2+), Co^(2+) and Ni^(2+) Ions on Biological Methane Production from Residual Heavy Oil显示文摘On the basis of single factor tests, the effect of trace elements—Fe2+, Co2+ and Ni2+ ions—on biological methane production from heavy oil was investigated by the response surface method. A three-level Box-Behnken design was employed to study the relationship between the independent variables and the dependent variable by applying initial Fe2+, Co2+ and Ni2+ concentration as the independent variables(factors) and using the methane production after 270 days of cultivation as the dependent variable(response). A prediction model of quadramatic polynomial regression equation was obtained. The results showed that the methane production could be as high as 240.69 μmol after optimization compared with 235.74 μmol obtained under un-optimized condition. Furthermore, the microbial communities before and after biodegradation were analyzed by PCR-DGGE method. The dominant bands were recovered and sequenced. | 9 | Contrasting on Hydrocarbon Generation Modeland Characteristics of Pyrolysis of Modern Aquatic Plant(Gloeocapsa) vs MarineAnimal(Mantis Shrimp)显示文摘A hydrocarbon model of the modern aquatic plant Gloeocapsa and the aquatic animal mantis shrimp was found in thermal simulating experiment. The results show that the modern aquatic plant Gloeocapsa is characterized by late generation, late termination and a long duration of oil generation, while the aquatic animal mantis shrimp is characterized by early generation, early termination and a short duration of oil generation. The nalkanes from Gloeocapsa and mantis shrimp are characterized by peak carbon C 15 -C 17 . With increasing thermal simulating temperature, the peak carbon changes from C 17 to C 15 , and the odd-even predominance of nalkanes becomes less clear. The products from Gloeocapsa contain abundant phenanthrene and naphthalene compounds, and even a little retene, while those from mantis shrimp are dominated by naphthalene compounds, and are poor in phenanthrene compounds. Gloeocapsa and mantis shrimp are rich in C 27 sterane, relatively rich in tricyclic terpanes (C 19 -C 29 ) and hopanes (C 27 -C 35 ), poor in 5α,14β,17β sterane, and coprostane does not disappear until 450 ℃. The tricyclic terpanes in Gloeocapsa are characterized by peak carbon C 23 and C 23 >C 21 , and in mantis shrimp by C 21 and C 21 >C 23 .
