1Dynamic property tests of frozen red sandstone using a split hopkinson pressure bar显示文摘In this study, frozen red sandstone specimens were impacted by a Split Hopkinson bar (SHPB), with a velocity of 4.558 ~ 6.823 ms-1. The temperature of the specimens was maintained at -15℃ during the experiment. For comparison purposes, static uniaxial compression tests were conducted in advance using a freezing triaxial test machine. Four stress-strain curves were obtained in diff erent average strain rates. The test results suggested that when the average strain rate is low, the specimen strength changes gradually;but when it is high, its strength changes rapidly. When the average strain rate is 120.73 s-1, the peak value of stress is as high as 82.96 MPa, which is about two times that of the static compressive strength of 44.1 MPa. A constitutive model was established that was composed of the damaged, viscoelastic and spring bodies, and revealed the variations of compressive strength and strain for the frozen red sandstone under diff erent high strain rates. The test results also showed that the failure form was correlated to the average strain rate of the frozen red sandstone. When the average strain rate is low, the damage was only distributed on the specimen’s edges. However, as the average strain rate increases, the damage range extended to the central parts of the specimen. 2Impact of dissolved humic acid on the bioavailability of acenaphthene and chrysene assessed by membrane-based passive samplers显示文摘Dissolved organic carbon (DOC) is known to reduce the bioavailability of hydrophobic organic compounds (HOCs) in aqueous environments. This reduction occurs as a result of adsorption to DOC, apparently reducing the freely dissolved concentration of HOCs. In the present study, triolein-embedded cellulose acetate membrane (TECAM) and Japanese medaka (Oryzias latipes) were used to measure the uptake of acenaphthene and chrysene in the presence of commercial humic acid (HA) at different concentrations (0―15 mg C·L-1) under controlled laboratory conditions. Apparent uptake rate constants for PAHs in TECAM and medaka were compared and DOC-water partition coefficients (KDOCs) of two PAHs were calculated with different sampling methods by model fit. Results showed that HA present in water significantly reduced the uptake of PAHs in TECAM and medaka. The obtained values of log KDOC of acenaphthene and chrysene measured by TECAM were 4.63 and 5.83, respectively, whereas biologically determined values were 4.52 and 5.76, respectively. These log KDOC values were comparable to earlier published KDOCs toward commercial HA, thereby indicating that TECAM accumulated only the freely dissolved fraction of chemicals and uptake PAHs in a manner similar to that of fish. 4Application of in situ cultivation in marine microbial resource mining显示文摘Microbial communities in marine habitats are regarded as underexplored reservoirs for discovering new natural products with potential application.However,only a few microbes in nature can be cultivated in the laboratory.This has led to the development of a variety of isolation and cultivation methods,and in situ cultivation is one of the most popular.Diverse in situ cultivation methods,with the same basic principle,have been applied to a variety of environmental samples.Compared with conventional approaches,these new methods are able to cultivate previously uncultured and phylogenetically novel microbes,many with biotechnological potential.This review introduces the various in situ cultivation methods for the isolation of previously uncultured microbial species and their potential for marine microbial resource mining.Furthermore,studies that investigated the key and previously unidentified mechanisms of growing uncultivated microorganisms by in situ cultivation,which will shed light on the understanding of microbial uncultivability,were also reviewed.Dawoon Jung Liwei Liu Shan He 2021Marine Life Science & Technology2021,3,2:1

8Screening of Host Proteins Interacting with PorcineEpidemic Diarrhea Virus (PEDV) N Protein by YeastTwo-hybrid System显示文摘[Objective] The paper was to obtain host proteins interacting with porcine epidemic diarrhea virus(PEDV) N protein. [Method] The recombinant vector pGBKT7-N of PEDV N gene was constructed and used as the bait plasmid to screen the proteins interacting with N protein of PEDV from the cDNA library of porcine alveolar macrophage(PAM) by yeast two-hybrid method. [Result] There was no toxicity and self activation of bait protein in yeast hybridization system, and six proteins(FTH1, LGALS3, CORO1C, SNRPG, KRTAP5-3, ZNF598) interacting with N protein were indentified. It was confirmed that LGALS3 and SNRPG had specific interaction with N protein by return experiment and co-immunoprecipitation(CoIP) test.
