1Particulate matter(PM(2.5)) exposure season-dependently induces neuronal apoptosis and synaptic injuries显示文摘Epidemiological studies have shown that particulate matter 2.5(PM_(2.5)) not only increases the incidence of cardiopulmonary illnesses but also relates to the development of neurodegenerative diseases. Considering that PM_(2.5)is highly heterogeneous with regional disparity and seasonal variation, we investigated whether PM_(2.5)exposure induced neuronal apoptosis and synaptic injuries in a season-dependent manner. The results indicated that PM_(2.5)altered the expression of apoptosis-related proteins(mainly bax and bcl-2), activated caspase-3 and caused neuronal apoptosis. Additionally, PM_(2.5)decreased the levels of synaptic structural protein postsynaptic density(PSD-95) and synaptic functional protein N-methyl-D-aspartate(NMDA) receptor subunit(NR2B) expression. These effects occurred in a season-dependent manner, and PM_(2.5)collected from the winter showed the strongest changes. Furthermore, the effect was coupled with the inhibition of phosphorylated extracellular signal-regulated kinase 1/2(p-ERK1/2) and phosphorylated c AMP-response element binding protein(p-CREB). 2Coking wastewater increases micronucleus frequency in mouse in vivo via oxidative stress显示文摘Coking wastewater has caused serious health risk in coal-producing areas of China, however its toxic efects have not been well understood. The genotoxicity induced by coking wastewater on mice in vivo and its possible oxidative mechanisms were investigated via observing the induction of micronuclei in polychromatic erythrocytes of mouse bone marrow, and subsequently determining the antioxidative enzyme activities(superoxide dismutase Cu, Zn-SOD, Se-dependent glutathione peroxidase, and catalase), thiobarbituric acid reactive substance contents and protein carbonyl levels in brains and livers of mice. Results showed that the tested coking wastewater caused a significant increase of micronucleus frequencies in a concentration-dependent manner. Also, the sample increased lipid peroxidation and protein oxidation levels, which was accompanied by changes in antioxidative status. Interestingly, pre-treatment with an antioxidant(vitamin C) led to a statistical reduction in the micronucleus frequency caused by coking wastewater. 3Effects of nitrogen dioxide and its acid mist on reactive oxygen species production and antioxidant enzyme activity in Arabidopsis plants显示文摘Nitrogen dioxide(NO2) is one of the most common and harmful air pollutants. To analyze the response of plants to NO2 stress, we investigated the morphological change, reactive oxygen species(ROS) production and antioxidant enzyme activity in Arabidopsis thaliana(Col-0) exposed to 1.7, 4, 8.5, and 18.8 mg/m3NO2. The results indicate that NO2 exposure affected plant growth and chlorophyll(Chl) content, and increased oxygen free radical(O2-)production rate in Arabidopsis shoots. Furthermore, NO2 elevated the levels of lipid peroxidation and protein oxidation, accompanied by the induction of antioxidant enzyme activities and change of ascorbate(As A) and glutathione(GSH) contents. Following this, we mimicked nitric acid mist under experimental conditions, and confirmed the antioxidant mechanism of the plant to the stress. Our results imply that NO2 and its acid mist caused pollution risk to plant systems. 4Abnormal energy metabolism and tau phosphorylation in the brains of middle-aged mice in response to atmospheric PM2.5 exposure显示文摘In light of the accelerated aging of the global population and the deterioration of the atmosphere pollution, we sought to clarify the potential mechanisms by which fine particulate matter(PM_(2.5)) can cause cognitive impairment and neurodegeneration through the alteration of mitochondrial structure and function. The results indicate that PM_(2.5) inhalation reduces ATP production by disrupting the aerobic tricarboxylic acid cycle and oxidative phosphorylation, thereby causing the hypophosphorylation of tau in the cortices of middle-aged mice. Furthermore, excessive reactive oxygen species generation was involved in the impairment. Interestingly, these alterations were partially reversed after exposure to PM_(2.5) ended. 5Identification of differential gene expressions in colorectal cancer and polyp by cDNA microarray显示文摘AIM:To screen the differential expressed genes in colorectal cancer and polyp tissue samples.METHODS:Tissue specimens containing 16 cases of colorectal adenocarcinoma and colorectal polyp vs normal mucosae were collected and subjected to cDNA microarray and bioinformatical analyses.Quantitative reverse transcription-polymerase chain reaction(qRTPCR) was used to confirm some of the cDNA microarray data.RESULTS:The experimental data showed that eight genes were differentially expressed,most of which were upregulated in adenomatous polyp lesions.Forty-six genes expressions were altered in colorectal cancers,of which 29 were upregulated and 17 downregulated,as compared to the normal mucosae.In addition,18 genes were similarly altered in both adenomatous polyps and colorectal cancer.qRT-PCR analyses confirmed the cDNA microarray data for four of those 18 genes:MTA1,PDCD4,TSC1 and PDGFRA.CONCLUSION:These differentially expressed genes likely represent biomarkers for early detection of colorectal cancer and may be potential therapeutic targets after confirmed by further studies.Yi-Chen Dai Xiao-San Zhu Qing-Zhen Nan Zhang-Xin Chen Jun-Pei Xie Yu-Ka Fu Yuan-Yuan Lin Qing-Na Lian Qiao-Fang Sang Xiao-Juan Zhan 2012World Journal of Gastroenterology2012,18,6:4
6Synergistic effects of particulate matter(PM_10) and SO_2 on human non-small cell lung cancer A549 via ROS-mediated NF-κB activation显示文摘Since a real atmospheric scenario usually represents a system involving multiple pollutants, air pollution studies typically focused on describing adverse effects associated with exposure to individual pollutants cannot reflect actual health risk. Particulate matter(PM10) and sulfur dioxide(SO2) are two major pollutants derived from coal combustion processes and co-existing in coal-smoke air pollution, but their potentially synergistic toxicity remains elusive thus far. In this study, we investigated the cytotoxic responses of PM10 and SO2, singly and in binary mixtures, using human non-small cell lung cancer A549 cells, followed by clarifying the possible mechanisms for their interaction. The results indicated that the concomitant treatment of PM10 and SO2 at low concentrations led to synergistic injury in terms of cell survival and apoptosis occurrence, while PM10 and SO2 alone at the same concentrations did not cause damage to the cells. Also, radical oxygen species(ROS) production followed by nuclear factor kappa B(NF-κB) activation was involved in the above synergistic cytotoxicity, which was confirmed by the repression of the actions by an ROS inhibitor(NAC). 7Morphological and physiological analysis of narrow and striped leaf 1(nsl1) mutant of rice(Oryza sativa L.) and the gene mapping显示文摘The shape and color of rice leaves are important agronomic traits that directly influence the proportion of sunlight energy utilization and ultimately affect the yield and quality.A new mutant exhibiting stable inheritance was identified as derived from ethyl methane sulfonate(EMS)-treated restorer Jinhui 10,tentatively named as narrow and striped leaf 1(nsl1).The nsl1 displayed pale white leaves at the seeding stage and then white striped leaves in parallel to the main vein at the jointing stage.Meanwhile,its leaf blades are significantly narrower than the control group of Jinhui 10.The chloroplast structures of cells in the white striped area of the nsl1 mutant break down,and the photosynthetic pigments are significantly lower than that of the wild type.Moreover,fluorescence parameters,such as F0,F v/F m,U psII,qp,and ETR,in the nsl1 mutant are significantly lower than those of the wild type,and the photosynthetic efficiency is also significantly decreased.These changes in leaf color and shape,together with physiological changes in the nsl1,result in smaller plant height and a decrease in the most important agronomic traits,such as the number of grains per panicle,grain weight,etc.Genetic analysis shows that the narrow and striped traits of the nsl1 mutant are controlled by a single recessive nuclear gene,which is located between InDel 16and InDel 12 in chromosome 3.The physical distance is204 kb.So far,no similar genes of such leaf color and shape in this area have been reported.This study has laid a solid foundation for the gene cloning and function analysis of NSL1.Fangming Zhao Yong Zhang Yanru Wu Tongming Wang Ling Ma Zhenglin Yang Xia Wei Xianchun Sang Yinghua Ling Nan Wang Changwei Zhang Guanghua He 2014Chinese Science Bulletin2014,59,9:3
8Genetic Analysis and Gene Mapping of a Rice Tiller Angle Mutant tac2显示文摘Tiller angle, a very essential agronomic trait, is significant in rice breeding, especially in plant type breeding. A tiller angle controlling 2 (tac2) mutant was obtained from a restorer line Jinhui 10 by ethyl methane sulphonate mutagenesis. The tac2 mutant displayed normal phenotype at the seedling stage and the tiller angle significantly increased at the tillering stage. A preliminary physiological research indicated that the mutant was sensitive to GA. Thus, it is speculated that TAC2 and TAC1 might control the tiller angle in the same way. Genetic analysis showed that the mutant trait was controlled by a major recessive gene and was located on chromosome 9 using SSR markers. 9Ultrasound-Guided Continuous Thoracic Paravertebral Block Improves Patientʼs Quality of Recovery After Open Hepatectomy: A Randomized, Double-Blind, Placebo-Controlled Trial显示文摘Background Ultrasound-guided continuous thoracic paravertebral block can provide pain-relieving and opioid-sparing effects in patients receiving open hepatectomy.We hypothesize that these effects may improve the quality of recovery(QoR)after open hepatectomy.Methods Seventy-six patients undergoing open hepatectomy were randomized to receive a continuous thoracic paravertebral block with ropivacaine(CTPVB group)or normal saline(control group).All patients received patient-controlled intravenous analgesia with morphine postoperatively for 48 hours.The primary outcome was the global Chinese 15-item Quality of Recovery score on postoperative day 7,which was statistically analyzed using Student’s t-test.Results Thirty-six patients in the CTPVB group and 37 in the control group completed the study.Compared to the control group,the CTPVB group had significantly increased global Chinese 15-item Quality of Recovery scores(133.14±12.97 vs.122.62±14.89,P=0.002)on postoperative day 7.Postoperative pain scores and cumulative morphine consumption were significantly lower for up to 8 and 48 hours(P<0.05;P=0.002),respectively,in the CTPVB group.Conclusion Perioperative CTPVB markably promotes patient’s QoR after open hepatectomy with a profound analgesic effect in the early postoperative period.Xulei Cui Nan Xu Zhiyong Zhang Bo Zhu Yuelun Zhang Yongchang Zheng Shunda Du Yilei Mao Xinting Sang Yuguang Huang 2022Chinese Medical Sciences Journal2022,37,1:3
10Association between cystatin C and diabetic retinopathy among type 2 diabetic patients in China: a Meta-analysis显示文摘AIM:To explore the correlation between cystatin C(Cys-C)and diabetic retinopathy(DR)in those patients with type 2 diabetes mellitus(DM)in China.METHODS:Articles were collected from China National Knowledge Infrastructure(CNKI),Wanfang,VIP,Pub Med,EMBASE,Cochrane Library,Clinical Trials.gov,and Google Scholar.Quality and risk of bias within included studies was assessed using the Newcastle-Ottawa scale(NOS).Heterogeneity was determined by using Cochran’s Q-test and Higgins I^(2) statistics.Mean differences(MDs)and 95%confidence intervals(CIs)of Cys-C within the diabetes without retinopathy(DWR)and DR,DWR and non-proliferative diabetic retinopathy(NPDR),NPDR and proliferative diabetic retinopathy(PDR)were collected by using random-effects model because of high heterogeneity.Meta-analysis was conducted based on 23 articles of 2331 DR including NPDR and PDR patients and 2023 DWR patients through Review Manager 5.3.Subgroup analyses were also performed according to DM duration,body mass index(BMI),total cholesterol(TC),total triglycerides(TG),low-density lipoprotein C(LDL-C),and high-density lipoprotein C(HDL-C),sample origins and methods.Publication bias was assessed by the funnel plot.RESULTS:Cys-C level in DR patients was increased compared with that of DWR(total MD:0.69,95%CI:0.41 to 0.97,Z=4.79,P<0.01).Besides,the synthesized results of the studies showed the similar findings in the DWR vs NPDR group(total MD:0.29,95%CI 0.20 to 0.39,Z=6.02,P<0.01)and the NPDR vs PDR group(total MD:0.63,95%CI 0.43 to 0.82,Z=6.33,P<0.01).Heterogeneity of most of the subgroup analyses was still obvious(I^(2)≥50%,P<0.1).Forest plots of different subgroups indicated that there was a slight increase of Cys-C during the period between DWR and DR,DWR and NPDR,NPDR and PDR.Funnel plot showed that there was no significant publication bias.CONCLUSION:The elevated Cys-C is closely related with DR and probably plays a critical role in its progression.Nan Yang Yun-Fei Lu Xiao Yang Kui Jiang Ai-Min Sang Hui-Qun Wu 2021International Journal of Ophthalmology(English edition)2021,14,9:2
11A comparison of the toxicity of landfill leachate exposure at the seed soaking and germination stages on Zea mays L.(maize)显示文摘To compare the toxicity of landfill leachate exposure at the early stages of seed soaking and germination on maize,a field experiment was conducted to evaluate the physiological aspects of growth,yield and potential clastogenicity of root-tip cells. The maizes were treated with leachate at levels of 2%,10%,20%,30% or 50%(V/V). First,the change of physiological indexes,including chlorophyll(Chl),Malondialdehyde(MDA)and Reactive oxygen species(ROS)levels,combined with yield all showed that soaking with leachate,but not germination,generated a greater ecological risk on maize. After a soaking treatment of maize with 50% leachate,the Chl,MDA and ROS levels during a vigorous growth period were 47.3%,149.8% and 309.7%,respectively,of the control,whereas the yield decreased to 68.6% of the control. In addition,our results demonstrated that the leachate at lower levels could promote growth. This is mainly embodied in that the yield of maize treated with 10%leachate at the soaking stage increased to 116.0% of the control. Moreover,the cytological analysis experiment also demonstrated that the ecological risk of leachate still exists in both cases. Furthermore,the gray relational analysis showed that the ear row number and tassel branch number were the major factors affecting the yield of maize treated with 50%leachate at the stages of soaking and germination,respectively. 16Immunotherapy prototype Mark 3.0 model in primary liver cancer:adding locoregional stereotactic therapy and prognostic factors classification management显示文摘Immune checkpoint inhibitors(ICIs)like programmed cell death-1(PD-1)/programmed death-ligand 1(PD-L1)inhibitor have shown considerable efficacy in several important cancers including primary liver cancer(PLC)like hepatocellular carcinoma and cholangiocarcinoma.However,only some patients with PLC will benefit,so combination therapy and biomarker classification detected by next-generation sequencing or immunohistochemistry are very important.Herein,we briefly summarize ICI-based therapies and stratify these evolving therapies for advanced PLC into three stages of immunotherapies Mark(Mk.)1.0,2.0,and 3.0.We illustrated the significance of ICI monotherapy(Mk.1.0),offering combinational approaches with traditional strategies(Mk.2.0)and additional locoregional therapy(Mk.3.0)to achieve longer survival and even meet the“No Evidence of Disease”status.We also highlight the importance of biomarkers and prognostic factors for patients with advanced PLC treated with ICI-based therapies.Multidisci-plinary team management should be investigated and collaborated closely to manage adverse events and sequential therapy suggestions for patients.Xu Yang Nan Zhang Yang Song Xiaobo Yang Xinting Sang Haitao Zhao 2022Medical Review2022,2,6:1
