2Effect of Different Vegetation Types on the Rhizosphere Soil Microbial Community Structure in the Loess Plateau of China显示文摘The Loess Plateau in China is one of the most eroded areas in the world.Accordingly,vegetation restoration has been implemented in this area over the past two decades to remedy the soil degradation problem.Understanding the microbial community structure is essential for the sustainability of ecosystems and for the reclamation of degraded arable land.This study aimed to determine the effect of different vegetation types on microbial processes and community structure in rhizosphere soils in the Loess Plateau.The six vegetation types were as follows:two natural grassland(Artemisia capillaries and Heteropappus altaicus),two artificial grassland(Astragalus adsurgens and Panicum virgatum),and two artificial shrubland(Caragana korshinskii and Hippophae rhamnoides)species.The microbial community structure and functional diversity were examined by analyzing the phospholipid fatty acids(PLFAs) and community-level physiological profiles.The results showed that rhizosphere soil sampled from the H.altaicus and A.capillaries plots had the highest values of microbial biomass C,average well color development of carbon resources,Gram-negative(G-)bacterial PLFA,bacterial PLFA,total PLFA,Shannon richness,and Shannon evenness,as well as the lowest metabolic quotient.Soil sampled from the H.rhamnoides plots had the highest metabolic quotient and Gram-positive(G+)bacterial PLFA,and soil sampled from the A.adsurgens and A.capillaries plots had the highest fungal PLFA and fungal:bacterial PLFA ratio.Correlation analysis indicated a significant positive relationship among the microbial biomass C,G-bacterial PLFA,bacterial PLFA,and total PLFA.In conclusion,plant species under arid climatic conditions significantly affected the microbial community structure in rhizosphere soil.Among the studied plants,natural grassland species generated the most favorable microbial conditions.ZHANG Chao LIU Guo-bin XUE Sha XIAO Lie 2013Journal of Integrative Agriculture2013,12,11:14
3Soil Microbial Community Composition During Natural Recovery in the Loess Plateau, China显示文摘This study aimed to determine the characteristics of soil microbial community composition and its relationship with soil chemical properties during natural recovery in the Loess Plateau.The soil microbial community composition was analyzed by comparing the soil microbial phospholipid fatty acids(PLFAs) of eight croplands abandoned for 1,3,5,10,13,15,20,and 30 yr in the Dunshan watershed,northern Loess Plateau,China.The results showed that soil organic carbon,total nitrogen,soil microbial biomass carbon,and soil microbial biomass nitrogen significantly increased with the abandonment duration,whereas the metabolic quotient significantly decreased.The Shannon richness and Shannon evenness of PLFAs significantly increased after 10 yr of abandonment.Gram-negative,Gram-positive,bacterial,fungal,and total PLFAs linearly increased with increased abandonment duration.Redundancy analysis showed that the abandonment duration was the most important environmental factor in determining the PLFA microbial community composition.The soil microbial PLFAs changed from anteiso-to iso-,unsaturated to saturated,and short-to long-chain during natural recovery.Therefore,in the Loess Plateau,cropland abandonment for natural recovery resulted in the increase of the soil microbial PLFA biomass and microbial PLFA species and changed the microbial from chemolithotrophic to a more heterotrophic community.XIAO Lie LIU Guo-bin XUE Sha ZHANG Chao 2013Journal of Integrative Agriculture2013,12,10:10
4Fractal features of soil profiles under different land use patterns on the Loess Plateau, China显示文摘Fractal theory is becoming an increasingly useful tool to describe soil structure dynamics for a better understanding of the performance of soil systems. Changes in land use patterns significantly affect soil physical, chemical and biological properties. However, limited information is available on the fractal characteristics of deep soil layers under different land use patterns. In this study, the fractal dimensions of particle size distribution(PSD) and micro-aggregates in the 0–500 cm soil profile and soil anti-erodibility in the 0–10 cm soil profile for 10 typical land use patterns were investigated in the Zhifanggou Watershed on the Loess Plateau, China. The 10 typical land use patterns were: slope cropland, two terraced croplands, check-dam cropland, woodland, two shrublands, orchard, artificial and natural grasslands. The results showed that the fractal dimensions of PSD and micro-aggregates were all significantly influenced by soil depths, land use patterns and their interaction. The plantations of shrubland, woodland and natural grassland increased the amount of larger micro-aggregates, and decreased the fractal dimensions of micro-aggregates in the 0–40 cm soil profile. And they also improved the aggregate state and aggregate degree and decreased dispersion rate in the 0–10 cm soil profile. The results indicated that fractal theory can be used to characterize soil structure under different land use patterns and fractal dimensions of micro-aggregates were more effective in this regard. 5Air-sea heat flux exchange over the South China Sea under different weather conditions before and after southwest monsoon onset in 2000显示文摘With the data observed from the Second SCS Air-Sea Flux Experiment on the Xisha air-sea flux research tower, the radiation budget, latent, sensible heat fluxes and net oceanic heat budgetswere caculated before and after summer monsoon onset. It is discovered that, after summer monsoononset, there are considerable changes in air-sea fluxes, especially in latent heat fluxes and net oceanicheat budget. Furthermre, the analyzed results of five synoptic stages are compared. And the character-istics of the flux transfer during different stages around onset of South China Sea monsoon are discussed.The flux change shows that there is an oceanic heat accumulating process during the pre-onset and thebreak period, as same as oceanic heat losing process during the onset period. Moreover, latent fluxes,the water vapor moving to the continent, even the rainfall appearance in Chinese Mainland also can beinfluenced by southwester. Comparing Xisha fluxes with those obtained from the Indian Ocean and thewestern Pacific Ocean, their differences may be obeerved. 8Purification and Characterization of a Low-temperature Hydroxylamine Oxidase from Heterotrophic Nitrifier Acinetobacter sp.Y16显示文摘Objective To purify a low-temperature hydroxylamine oxidase(HAO) from a heterotrophic nitrifying bacterium Acinetobacter sp.Y16 and investigate the enzyme property.Methods A HAO was purified by an anion-exchange and gel-filtration chromatography from strain Y16.The purity and molecular mass were determined by RP-HPLC and SDS-PAGE.The HAO activity was detected by monitoring the reduction of potassium ferricyanide using hydroxylamine as substrate and ferricyanide as electron acceptor.The partial amino acid sequence was determined by mass spectrometry.Results The low-temperature HAO with a molecular mass of 61 kDa was purified from strain Y16 by an anion-exchange and gel-filtration chromatography.The enzyme exhibited an ability to oxidize hydroxylamine in wide temperature range(4-40 ℃) in vitro using hydroxylamine as substrate and ferricyanide as electron acceptor.It was stable in the temperature range of 4 to 15 ℃ and pH range of6.0 to 8.5 with less than 30%change in its activity.The optimal temperature and pH were 15 ℃ and 7.5,respectively.Three peptides were determined by mass spectrometry which were shown to be not identical to other reported HAOs.Conclusion This is the first study to purify a low-temperature HAO from a heterotrophic nitrifier Acinetobacter sp.It differs from other reported HAOs in molecular mass and enzyme properties.The findings of the present study have suggested that the strain Y16 passes through a hydroxylamine-oxidizing process catalyzed by a low-temperature HAO for ammonium removal.ZHANG Shu Mei LI Wei Guang ZHANG Duo Ying HUANG Xiao Fei QIN Wen SHA Chang Qing 2014Biomedical and Environmental Sciences2014,27,7:6
9Comparative analysis of the clinical outcomes between wavefront-guided and conventional femtosecond LASIK in myopia and myopia astigmatism显示文摘AIM:To compare the clinical outcomes of wavefront guided femtosecond LASIK(WFG LASIK)and conventional femtosecond LASIK(NWFG LASIK)in eyes with myopia and myopia astigmatism.METHODS:This was a retrospective,nonrandomized,comparative investigation enrolling 236 eyes of 122 patients(18-50 y)with low&moderate and high myopia.The WFG group including 97 eyes(50 patients)undergone WFG LASIK and the NWFG group including 139 eyes(72 patients)undergone conventional LASIK.Mean efficacy index,high order aberrations(HOAs),pupil size and the quality of visual questionnaire were evaluated 6 mo postoperatively.RESULTS:There is no difference between WFG group(-0.054±0.049 in logMAR)and NWFG group(-0.040±0.056)in uncorrected distance visual acuity(UDVA)postoperatively.The myopia astigmatism is higher in WFG group than that in NWFG group(P<0.05).However,the mean efficacy index(MEI)in the WFG group(1.09±0.106)is better than that in the NWFG group(1.036±0.124;P<0.001).Increased HOAs were observed in NWFG group(0.30±0.196)than that in WFG group(0.146±0.188;P<0.001).The pupil size is larger in WFG group(5.15±0.76 mm)than that in NWFG group(4.32±0.52 mm).The patients are satisfied with the clinical surgery,yet WFG group showed better visual quality using the questionnaire survey.Meanwhile,high myopia would result in worse MEI,HOAs and visual quality than low&moderate myopia.CONCLUSION:WFG and NWFG FS-LASIK are both effective and safe procedures to correct low&moderate and high myopia,but WFG FS-LASIK gives a better postoperative MEI,aberrometric control and predictable outcome.Meanwhile,WFG FS-LASIK is better than NWFG FS-LASIK in correction of myopia astigmatism.Low&moderate myopia allow better clinical outcomes than high myopia using any surgical method.Xiao Gui Rui Zhang Sha Li Na Zhao Hao-Rui Zhang Yu-Kun Zhou Chen-Yang Huan Chun-Yan Zhao Hong-Ying Wang Hong-Yuan Song Wei Shen Jian-Hua Zhang 2021International Journal of Ophthalmology(English edition)2021,14,10:5
10LIN28A inhibits DUSP family phosphatases and activates MAPK signaling pathway to maintain pluripotency in porcine induced pluripotent stem cells显示文摘LIN28A,an RNA-binding protein,plays an important role in porcine induced pluripotent stem cells(piPSCs).However,the molecular mechanism underlying the function of LIN28A in the maintenance of pluripotency in piPSCs remains unclear.Here,we explored the function of LIN28A in piPSCs based on its overexpression and knockdown.We performed total RNA sequencing(RNA-seq)of piPSCs and detected the expression levels of relevant genes by quantitative real-time polymerase chain reaction(qRT-PCR),western blot analysis,and immunofluorescence staining.Results indicated that piPSC proliferation ability decreased following LIN28A knockdown.Furthermore,when LIN28A expression in the shLIN28A2 group was lower(by 20%)than that in the negative control knockdown group(shNC),the pluripotency of piPSCs disappeared and they differentiated into neuroectoderm cells.Results also showed that LIN28A overexpression inhibited the expression of DUSP(dual-specificity phosphatases)family phosphatases and activated the mitogen-activated protein kinase(MAPK)signaling pathway.Thus,LIN28A appears to activate the MAPK signaling pathway to maintain the pluripotency and proliferation ability of piPSCs.Our study provides a new resource for exploring the functions of LIN28A in piPSCs.Xiao-Long Wu Zhen-Shuo Zhu Xia Xiao Zhe Zhou Shuai Yu Qiao-Yan Shen Ju-Qing Zhang Wei Yue Rui Zhang Xin He Sha Peng Shi-Qiang Zhang Na Li Ming-Zhi Liao Jin-Lian Hua 2021Zoological Research2021,42,3:5

13Conophylline Promotes the Proliferation of Immortalized Mesenchymal Stem Cells Derived from Fetal Porcine Pancreas (iPMSCs)显示文摘Conophylline, is a bis (indole) alkaloid consisting of two pentacyclic aspidosperma skeletons, isolated from Tabernaemontana divaricata, which has been found to induce β-cell differentiation in rat pancreatic acinar carcinoma cells and in cultured rat pancreatic tissue. However, the precise role of conophylline in the growth and survival of immortalized pancreatic mesenchymal stem cells (iPMSCs) derived from fetal porcine pancreas were not understood at present. To determine whether this molecule is involved in controlling the proliferation of iPMSCs, we examined the effects of conophylline on iPMSCs. We found that conophylline can robustly stimulate iPMSCs proliferation, even promote their potential differentiation into islet-like clusters analyzed by cell counting, morphology, RT-PCR and real-time PCR, Western blotting, glucose-stimulated insulin release and insulin content analysis. The effects of conophylline were inhibited by LY294002, which is the inhibitor of the PI3K pathway. 14Clinicopathologic characteristics and therapeutic responses of Chinese patients with non-small cell lung cancer who harbor an anaplastic lymphoma kinase rearrangement显示文摘Introduction: The rearrangement of the anaplastic lymphoma kinase(ALK) gene accounts for approximately 1%–6% of lung adenocarcinoma cases and deines a molecular subgroup of tumors characterized by clinical sensitivity to ALK inhibitors such as crizotinib. This study aimed to identify the relationship between ALK rearrangement and the clinico?pathologic characteristics of non?small cell lung cancer(NSCLC) and to analyze the therapeutic responses of crizotinib and conventional chemotherapy to ALK rearrangement in NSCLC patients.Methods: A total of 487 lung cancer patients who underwent testing for ALK rearrangement in our department were included in this study. ALK rearrangement was examined by using fluorescence in situ hybridization(FISH) assay.Results: Among the 487 patients, 44(9.0%) were diagnosed with ALK rearrangement by using FISH assay. In 123 patients with adenocarcinoma who were non?smokers and of a young age(≤58 years old), the frequency of ALK rearrangement was 20.3%(25/123). Short overall survival(OS) was associated with non?adenocarcinoma tumor type(P = 0.006), poorly diferentiated tumors(P al growth factor rece= 0.001), advanced?stage tumors(P < 0.001), smoking history(P ptor(EGFR)(P = 0.008), and wild?type epidermrter time to cancer p= 0.008). Moreover, patients with poorly diferentiated and advanced?stage tumors had a shorogression compared with those with well diferentiated(P = 0.023) and early?stage tumors(P = 0.001), respectively.Conclusions: ALK?rearranged NSCLC tends to occur in younger individuals who are either non?smokers or light smokers with adenocarcinoma. 16Investigation of Coal Fueled Chemical Looping Combustion Using Fe_3O_4 as Oxygen Carrier:Influence of Variables显示文摘Chemical-looping combustion (CLC) is a novel combustion technique with inherent CO2 separation.Magnetite (Fe3O4) was selected as the oxygen carrier.Shenhua coal (Inner Mongolia,China),straw coke and natural coke were used as fuels for this study.Influences of operation temperatures,coal to Fe3O4 mass ratios,and different kinds of fuels on the reduction characteristics of the oxygen carrier were investigated using an atmosphere thermogravimetric analyzer (TGA).Scanning electron microscopy (SEM) was used to analyse the characteristic of the solid residues.Experimental results shown that the reaction between the coal and the oxygen carrier become strong at a temperature of higher than 800℃.As the operation temperature rises,the reduction conversion rate increases.At the temperatures of 850oС,900℃,and 950℃,the reduction conversion rates were 37.1%,46.5%,and 54.1% respectively.However,SEM images show that at the temperature of higher than 950℃,the iron oxides become melted and sintered.The possible operation temperature should be kept around 900℃.When the mass ratios of coal to Fe3O4 were 5/95,10/90,15/85,and 20/80,the reduction conversion rates were 29.5%,40.8%,46.5%,and 46.6% respectively.With the increase of coal,the conversion rate goes up.But there exist an optimal ratio around 15/85.Comparisons based on different kinds of fuels show that the solid fuel with a higher volatile and a more developed pore structure is conducive to the reduction reactivity of the oxygen carrier.Xiaoyan Sun Wenguo Xiang Sha Wang Wendong Tian Xiang Xu Yanji Xu Yunhan Xiao 2010Journal of Thermal Science2010,19,3:3
17Benzimidazoles induce concurrent apoptosis and pyroptosis of human glioblastoma cells via arresting cell cycle显示文摘Glioblastoma multiforme(GBM)is the most malignant and lethal primary brain tumor in adults accounting for about 50%of all gliomas.The only treatment available for GBM is the drug temozolomide,which unfortunately has frequent drug resistance issue.By analyzing the hub genes of GBM via weighted gene co-expression network analysis(WGCNA)of the cancer genome atlas(TCGA)dataset,and using the connectivity map(CMAP)platform for drug repurposing,we found that multiple azole compounds had potential anti-GBM activity.When their anti-GBM activity was examined,however,only three benzimidazole compounds,i.e.flubendazole,mebendazole and fenbendazole,potently and dose-dependently inhibited proliferation of U87 and U251 cells with IC50 values below 0.26μM.Benzimidazoles(0.125−0.5μM)dose-dependently suppressed DNA synthesis,cell migration and invasion,and regulated the expression of key epithelial-mesenchymal transition(EMT)markers in U87 and U251 cells.Benzimidazoles treatment also dose-dependently induced the GBM cell cycle arrest at the G2/M phase via the P53/P21/cyclin B1 pathway.Furthermore,the drugs triggered pyroptosis of GBM cells through the NF-κB/NLRP3/GSDMD pathway,and might also concurrently induced mitochondria-dependent apoptosis.In a nude mouse U87 cell xenograft model,administration of flubendazole(12.5,25,and 50 mg·kg^(−1)·d^(−1),i.p,for 3 weeks)dose-dependently suppressed the tumor growth without obvious adverse effects.Taken together,our results demonstrated that benzimidazoles might be promising candidates for the treatment of GBM.Li-wen Ren Wan Li Xiang-jin Zheng Jin-yi Liu Yi-hui Yang Sha Li Sen Zhang Wei-qi Fu Bin Xiao Jin-hua Wang Guan-hua Du 2022Acta Pharmacologica Sinica2022,43,1:3
18Complete mitochondrial genome of the Japanese snapping shrimp Alpheus japonicus(Crustacea:Decapoda:Caridea):Gene rearrangement and phylogeny within Caridea显示文摘The complete sequence of the mitochondrial genome of the Japanese snapping shrimp Alpheus japonicus Miers(Crustacea:Decapoda:Caridea) is presented here.A comparative analysis based on the currently available mitochondrial genomic data revealed many previously unknown characteristics of the mitochondrial genomes of caridean shrimps.The A.japonicus mitochondrial genome is 16487 bp long and contains the typical set of 37 metazoan genes.The gene arrangements in the mitochondrial genomes of four previously studied carideans(Macrobrachium rosenbergii,M.nipponense,M.lanchesteri and Halocaridina rubra) were found to be identical to the pancrustacean ground pattern;thus,it was considered that gene rearrangements probably did not occur in the suborder Caridea.In the present study,a translocation of the trnE gene involving inversion was found in Alpheus mitochondrial genomes.This phenomenon has not been reported in any other crustacean mitochondrial genome that has been studied so far;however,the translocation of one transfer RNA gene(trnP or trnT) was reported in the mitochondrial genome of Exopalaemon carinicauda.When the ratios of the nonsynonymous and synonymous substitutions rates(Ka/Ks) for the 13 protein coding genes from two Alpheus species(A.japonicus and A.distinguendus) and three Macrobrachium species(M.rosenbergii,M.nipponense,M.lanchesteri) were calculated,the Ka/Ks values for all the protein coding genes in Alpheus and Macrobrachium mitochondrial genomes were found to be less than 1(between 0.0048 and 0.2057),indicating that a strong purification selection had occurred.The phylogenetic tree that was constructed based on the mitochondrial protein coding genes in the genomes of nine related species indicated that Palaemonidae and Alpheidae formed a monophyly and shared a statistically significant relationship,(Palaemonidae+Alpheidae)+Atyidae,at the family level.SHEN Xin LI Xiao SHA ZhongLi YAN BinLun XU QiHua 2012Science China(Life Sciences)2012,55,7:3

20Preconcentration of Ultra-trace Cadmium with Nanometer-size TiO_2 Colloid and Determination by GFAAS with Slurry Sampling显示文摘A novel method of ultra-trace Cd(II) preconcentration with nanometer-size TiO2 colloid and determination by graphite furnace atomic adsorption spectrometry(GFAAS) with slurry sampling was first advanced in this paper. The adsorption efficiency of nanometer-size TiO2 colloid for ultra-trace Cd(II) could reach above 96% in a short time when the pH value was between 5 and 6. Other problems were also studied,such as adsorption capacity,nanometer-size TiO2 colloid dosage,effect of coexistent ions. The detection limit(3σ) and the relative standard deviation (R.S.D) of this method were 4.46?10-3 μg/L and 1.30%(n=7),respectively.
