1Detection of bursts in neuronal spike trains by the mean inter-spike interval method显示文摘Bursts are electrical spikes firing with a high frequency,which are the most important property in synaptic plasticity and information processing in the central nervous system. However,bursts are difficult to identify because bursting activities or patterns vary with phys-iological conditions or external stimuli. In this paper,a simple method automatically to detect bursts in spike trains is described. This method auto-adaptively sets a parameter (mean inter-spike interval) according to intrinsic properties of the detected burst spike trains,without any arbitrary choices or any operator judgment. When the mean value of several successive inter-spike intervals is not larger than the parameter,a burst is identified. By this method,bursts can be automatically extracted from different bursting patterns of cultured neurons on multi-electrode arrays,as accurately as by visual inspection. Furthermore,significant changes of burst variables caused by electrical stimulus have been found in spontaneous activity of neuronal network. 2Cell type-specific proteomics uncovers a RAF15-SnRK2.6/OST1 kinase cascade in guard cells显示文摘Multicellular organisms such as plants contain various cell types with specialized functions.Analyzing the characteristics of each cell type reveals specific cell functions and enhances our understanding of organization and function at the organismal level.Guard cells(GCs)are specialized epidermal cells that regulate the movement of the stomata and gaseous exchange,and provide a model genetic system for analyzing cell fate,signaling,and function.Several proteomics analyses of GC are available,but these are limited in depth.Here we used enzymatic isolation and flow cytometry to enrich GC and mesophyll cell protoplasts and perform in-depth proteomics in these two major cell types in Arabidopsis leaves.We identified approximately 3,000 proteins not previously found in the GC proteome and more than600 proteins that may be specific to GC.The depth of our proteomics enabled us to uncover a guard cell-specific kinase cascade whereby Raf15and Snf1-related kinase2.6(SnRK2.6)/OST1(open stomata 1)mediate abscisic acid(ABA)-induced stomatal closure.RAF15 directly phosphorylated SnRK2.6/OST1 at the conserved Ser175 residue in its activation loop and was sufficient to reactivate the inactive form of SnRK2.6/OST1.ABAtriggered SnRK2.6/OST1 activation and stomatal closure was impaired in raf15 mutants.We also showed enrichment of enzymes and flavone metabolism in GC,and consistent,dramatic accumulation of flavone metabolites.Our study answers the long-standing question of how ABA activates SnRK2.6/OST1 in GCs and represents a resource potentially providing further insights into the molecular basis of GC and mesophyll cell development,metabolism,structure,and function.Hongliang Wang Yubei Wang Tian Sang Zhen Lin Rongxia Li Weiwei Ren Xin Shen Bing Zhao Xiao Wang Xuebin Zhang Shaoqun Zhou Shaojun Dai Honghong Hu Chun-Peng Song Pengcheng Wang 2023Journal of Integrative Plant Biology2023,65,9:0
3Development of a monitoring and warning system based on optical fiber sensing technology for masonry retaining walls and trees显示文摘Hong Kong has a long history of applying masonry retaining walls to provide horizontal platforms and stabilize man-made slopes.Due to the sub-tropical climate,some masonry retaining walls are colonized by trees.Extreme weather,such as typhoons and heavy rains,may cause rupture or root failure of those trees,thus resulting in instability of the retaining walls.A monitoring and warning system for the movement of masonry retaining walls and sway of trees has been designed with the application of fiber Bragg grating(FBG)sensing technology.The monitoring system is also equipped with a solar power system and 4G data transmission devices.The key functions of the proposed monitoring system include remote sensing and data access,early warning,and real-time data visualization.The setups and working principles of the monitoring systems and related transducers are introduced.The feasibility,accuracy,serviceability and reliability of this monitoring system have been checked by in-site calibration tests and four-month monitoring.Besides,a two-level interface has been developed for data visualization.The monitoring results show that the monitored masonry retaining wall had a reversible movement up to 2.5 mm during the monitoring period.Besides,it is found that the locations of the maximum strain on trees depend on the crown spread of trees.Peichen Wu Daoyuan Tan Shaoqun Lin Wenbo Chen Jianhua Yin Numan Malik An Li 2022Journal of Rock Mechanics and Geotechnical Engineering2022,14,4:0
