| 1 | Physiological responses of osteoblasts to cyclic stretching and the change of intracellular calcium concentration显示文摘The development of bone tissues is regulated by mechanical stimulation. Cyclic stretching was applied to the osteoblasts that were delivered from rat calvarie. The results showed that stretching at 500 me increased the cell prolifera-tion while loading at 1000 me and 1500 me inhabited cell growth. Loading also increased the adhesive force between cells and substrate as well as spreading areas of osteobalsts. Furthermore, the fluorescence probe Fluo-3/AM was used to investigate the effect of stretching stimulation on the intra-cellular calcium concentration of osteoblasts. The intracellu-lar calcium concentration of osteoblasts that were stretched at 500 me for 5 min was 92.9% higher than the control. After being treated with the panax ontoginseng saponins, the stretched osteoblasts still expressed 28.6% higher intracellu-lar calcium concentration than that of the control, which proved that both the influx of extracellular calcium and the release of intracellular calcium store were involved in the increase of intracellular calcium concentration when os-teoblasts responded to the cyclic stretching. | 2 | PDMS microchannel fabrication technique based on microwire-molding显示文摘Micro-flow channel is basic functional component of microfluidic chip,and every step-forward of its construction technique has been receiving concern all over the world.This article presents a not-complicated but flexible method for fabrication of micro-flow channels.This method mainly utilizes the conventional molding capability of polydimethylsiloxane(PDMS) and widespread commercial microwires as templates.We have fabricated out some conventional types of microchannels with different topological shapes,as examples for the demonstration of this flexible fabrication route which was not dependent on the stringent demands of photolithographical or microelectromechanical system(MEMS) techniques.The smooth surface,high-intensity,and high flexibility of the wires made it possible to create many types of topological structures of the two-dimensional or three-dimensional microchannel or channel array.The geometric shape of the cross-section of thus forming microchannel in PDMS was the negative of that of embedded-in microwire,in high-fidelity if suitable measures were taken.More-over,such a microchannel fabrication process can easily integrate the conductivity and low resistivity of the metal wire to create micro-flow devices that are suitable for the electromagnetic control of liquid or the temperature regulation in the microchannel.Furthermore some preliminary optical analysis was provided for the observation of thus forming rounded microchannel.Based on this molding strategy,we even made some prototypes for functional microflow application,such as microsolenoids chip and temperature control gadgets.And an experiment of forming a droplet in the cross channel further confirmed the feasibility and applicability of this flexible microchannel forming technique. | JIA YueFei JIANG JiaHuan MA XiaoDong LI Yuan HUANG HeMing CAI KunBao CAI ShaoXi WU YunPeng | 2008 | Chinese Science Bulletin2008,53,24: | 5 |
| 3 | Auxin-induced AUXIN RESPONSE FACTOR4 activates APETALA1 and FRUITFULL to promote flowering in woodland strawberry显示文摘Flowering time is known to be regulated by numerous pathways,such as the autonomous,gibberellin,aging,photoperiod-mediated,and vernalization pathways.These regulatory mechanisms involve both environmental triggers and endogenous hormonal cues.Additional flowering control mechanisms mediated by other phytohormones,such as auxin,are less well understood.We found that in cultivated strawberry(Fragaria×ananassa),the expression of auxin response factor4(FaARF4)was higher in the flowering stage than in the vegetative stage.Overexpression of FaARF4 in Arabidopsis thaliana and woodland strawberry(Fragaria vesca)resulted in transgenic plants flowering earlier than control plants.In addition,FveARF4-silenced strawberry plants showed delayed flowering compared to control plants,indicating that FaARF4 and FveARF4 function similarly in regulating flowering.Further studies showed that ARF4 can bind to the promoters of the floral meristem identity genes APETALA1(AP1)and FRUITFULL(FUL),inducing their expression and,consequently,flowering in woodland strawberry.Our studies reveal an auxin-mediated flowering pathway in strawberry involving the induction of ARF4 expression. | Xiangxiang Dong Yanjun Li Yuhan Guan Shaoxi Wang He Luo Xiaoming Li He Li Zhihong Zhang | 2021 | Horticulture Research2021,8,1: | 4 |

| 5 | A novel in vitro angiogenesis model based on a microfluidic device显示文摘Angiogenesis is very important for many physiological and pathological processes. However, the molecular mechanisms of angiogenesis are unclear. To elucidate the molecular mechanisms of angiogenesis and to develop treatments for 'angiogenesis-dependent' diseases, it is essential to establish a suitable in vitro angiogenesis model. In this study, we created a novel in vitro angiogenesis model based on a microfluidic device. Our model provides an in vivo-like microenvironment for endothelial cells (ECs) cultures and monitors the response of ECs to changes in their microenvironment in real time. To evaluate the potential of this microfluidic device for researching angiogenesis, the effects of pro-angiogenic factors on ECs proliferation, migration and tube-like structure formation were investigated. Our results showed the proliferation rate of ECs in 3D matrix was significantly promoted by the pro-angiogenic factors (with an increase of 59.12%). With the stimulation of pro-angiogenic factors gradients, ECs directionally migrated into the Matrigel from low concentrations to high concentrations and consequently formed multi-cell chords and tube-like structures. | 20 | Colchicine Inhibited the Expression of Tissue Inhibitor of Metalloprotenase-1 and Interleukin-6 in Cultured Activated Hepatic Stellate Cells显示文摘Cultured HSCs were treated colchicine with different concentrations for 12 h, respectively. The effects of colchicine on HSCs growth were measured by MTT assay. Effects of colchicine on gene expression of HSCs were analysed by using a self-made oligonucleotide microarray. Colchicine inhibited HSCs growth in a dose-dependent manner. After 12 h of treatment with 6.25 mg/L of colchicine, the expression of tissue inhibitor of metalloprotenase-1 (TIMP-1) and the expression of interleukin-6 (IL-6) in HSCs were down-regulated by 2.3 folds and 2.1 folds, respectively.
