4A one-step reverse-transcription recombinase aided PCR assay for the rapid and sensitive detection of human enteroviruses显示文摘Human enteroviruses(HEVs)include many different types that cause a wide range of diseases,and an effective method of genus-level identification has therefore significant clinical implications.However,quantitative real-time reverse transcription polymerase chain reaction(qRT-PCR),the gold-standard method,still has shortfalls in diagnostic sensitivity and timeliness.Here we established a one-step real-time reverse-transcription recombinase-aided PCR assay(RT-RAP)to detect HEV fragment within an hour.The RT-RAP assay showed a detection limit of 5 copies/μL using recombinant plasmids and was extensively verified using 15 HEV strains.Among 15 types of HEV(species A-C),the sensitivity of RT-RAP was approximately 2-8 folds lower than that of the qRT-PCR in 9 types,and no-cross reaction with other viruses was observed.RT-RAP was further applied to analyze CSF and fecal specimens;the clinical performance demonstrated that the RT-RAP and the commercial qRT-PCR kit provided consistent results.These results indicated that RT-RAP assay may be a promising approach for rapid and sensitive detection of HEV.Xiuli Sun Huanhuan Lu Yanqing Tie Mengchuan Zhao Ruiqing Zhang Zhenlu Sun Guohao Fan Fengyu Li Fengyu Tian Yaxin Hu Mengyi Zhang Xinxin Shen Xuejun Ma Zhishan Feng 2023Biosafety and Health2023,5,2:0
5Electrochemical C-H/N-H cross-coupling of 2-phenylindolizines with phenothiazines to synthesize novel N-aryl phenothiazine derivatives显示文摘A facile and elegant method for synthesis of novel N-aryl phenothiazine derivatives from 2-phenylindolizines and phenothiazines through direct electrochemical oxidation has been developed.This approach was performed smoothly at room temperature without external oxidant and catalyst.Cyclic voltammetry and in situ FTIR techniques were applied to analyze the cross-coupling process of phenothiazines and 2-phenylindolizines,which helped to select the appropriate reaction potential.Under the optimized conditions,a broad range of substrates were well tolerated,affording the desired products in moderate to excellent isolated yields(up to 91%)with high regioselectivity.Meanwhile,a plausible mechanism involving a radical pathway has been proposed.Chenglong Feng Xin Liu Yuanbin She Zhenlu Shen Meichao Li 2023Chinese Chemical Letters2023,34,6:0
6Diastereoselective Construction of α-Fluoroalkyl Cycloalkanols via Radical Alkylation of α-Fluoroalkyl Ketones显示文摘α-Fluoroalkyl (Rf) alcohols are privileged motifs in drugs and pharmaceutically active compounds.As such,it is highly desirable to develop efficient methods for assembling these scaffolds.Herein,a visible-light-induced cascade radical cyclization of alkenyl fluoroalkyl ketones is developed,producing a variety of decorate α-Rf cycloalkanols in promising yields with up to >20 : 1 dr selectivity.A radical chain mechanism involving an intramolecular radical addition to the α-Rf carbonyl group and a subsequent intermolecular hydrogen atom transfer (HAT) has been proposed.Density functional theory (DFT) calculations indicate that a fluorine effect contributes to the radical addition to carbonyls by lowering the π* (C=O) orbital energy of CORf and energy barrier of the HAT between alkoxy radicals and THF,which may be valuable for controllable transformations of fluorinated molecules.Yi Liu Ji Li Yulu Zhou Hanliang Zheng Zhenlu Shen Gangguo Zhu 2023Chinese Journal of Chemistry2023,41,23:0
