3Dynamic characteristics of an intestine capsule robot with variable diameter显示文摘This paper proposes an intestine capsule robot with variable diameter. It is driven by external rotating magnetic field and has the function of automatic radial clearance compensation. An external magnetic field generating device and a capsule robot with variable diameter are developed. Radial dynamic balance equation and kinematics equation of the capsule robot traveling inside flexible elasto-wall environment are established, and dynamic characteristics of the capsule robot inside flexible elasto-wall pipe and rigid wall pipe environments are analyzed and compared. Simulations and experiment results show that its dynamic characteristics in both pipes are almost the same. Under the action of radial clearance compensation, fluid dynamic pressure around outer surface of the capsule robot and its propulsion, as well as its adaptability to diameter change range, are significantly improved. Its propulsion and moving speed are controlled by adjusting rotating angular speed of the rotating magnetic field. 4Targeted mutagenesis of amino acid transporter genes for rice quality improvement using the CRISPR/Cas9 system显示文摘High grain protein content(GPC) reduces rice eating and cooking quality(ECQ). We generated OsAAP6 and OsAAP10 knockout mutants in three high-yielding japonica varieties and one japonica line using the CRISPR/Cas9 system. Mutation efficiency varied with genetic background in the T_0 generation, and GPC in the T_1 generation decreased significantly,owing mainly to a reduction in glutelin content. Amylose content was down-regulated significantly in some Osaap6 and all Osaap10 mutants. The increased taste value of these mutants was supported by Rapid Visco Analysis(RVA) profiles, which showed higher peak viscosity and breakdown viscosity and lower setback viscosity than the wild type. There were no significant deficiencies in agronomic traits of the mutants. 5Development of a Drilling and Coring Test-bed for Lunar Subsurface Exploration and Preliminary Experiments显示文摘Drill sampling has been widely employed as an effective way to acquire deep samples in extraterrestrial exploration. A novel sampling method, namely, flexible-tube coring, was adopted for the Chang'e mission to acquire drilling cores without damaging stratification information. Since the extraterrestrial environment is uncertain and different from the terrestrial environment, automated drill sampling missions are at risk of failure. The principles of drilling and coring for the lunar subsurface should be fully tested and verified on earth before launch. This paper proposes a test-bed for conducting the aforementioned experiments on earth. The test-bed comprises a rotary-percussive drilling mechanism, penetrating mechanism, drilling medium container, and signal acquisition and control system. For granular soil, coring experiments indicate that the sampling method has a high coring rate greater than 80%. For hard rock, drilling experiments indicate that the percussive frequency greatly affects the drilling efficiency. A multi-layered simulant composed of granular soil and hard rock is built to test the adaptability of drilling and coring. To tackle complex drilling media, an intelligent drilling strategy based on online recognition is proposed to improve the adaptability of the sampling drill. 6Drilling Power Consumption and Soil Conveying Volume Performances of Lunar Sampling Auger显示文摘The sampling auger used in lunar sampling and return mission is to transmit power and convey soil, and its performance is the key factor of the whole mission. However, there is currently a lack of the optimization research on soil conveying volume and power consumption models in auger structure design. To provide the drilled object, the simulation lunar soil, whose physical and mechanical property is the same as the real soil, is made by reducing soil void ratio. The models are formulated to analyze the influence of auger structure parameters on power consumption and soil conveying volume. To obtain the optimized structure parameters of auger, the multi-objective optimization functions of the maximum soil conveying volume and minimum power consumption are developed. To verify the correctness of the models, the performances of different augers drilling simulation soil are tested. The test results demonstrate that the power consumption of optimized auger is the lowest both in theory and test, and the experimental results of soil conveying volume are in agreement with theoretical analysis. Consequently, a new method for designing a lunar sampling auger is proposed which includes the models of soil conveying volume and transportation power consumption, the optimization of structure parameters and the comparison tests. 8An Improved Genetic Algorithm for Test Cases Generation Oriented Paths显示文摘This paper discusses a method that can automatically generate test cases for selected paths using a special genetic algorithm. The special algorithm is called Queen-bee evolutionary genetic algorithm(QBEA). In this algorithm, sequences of operators iteratively executes for test cases to evolve to target paths. The best chromosome called queen among the current population is crossover with drones selected according to a certain crossover probability, which enhances the exploitation of searching global optimum. 9Drilling load modeling and validation based on the filling rate of auger flute in planetary sampling显示文摘Some type of penetration into a subsurface is required in planetary sampling. Drilling and coring, due to its efficient penetrating and cuttings removal characteristics, has been widely applied in previous sampling missions. Given the complicated mechanical properties of a planetary regolith, suitable drilling parameters should be matched with different drilling formations properly.Otherwise, drilling faults caused by overloads could easily happen. Hence, it is necessary to establish a drilling load model, which is able to reveal the relationships among drilling loads, an auger's structural parameters, soil's mechanical properties, and relevant drilling parameters. A concept for the filling rate of auger flute(FRAF) is proposed to describe drilling conditions. If the FRAF index under one group of drilling parameters is less than 1, this means that the auger flute currently removes cuttings smoothly. Otherwise, the auger will be choked with compressed cuttings. In drilling operations, the drilling loads on the auger mainly come from the conveyance action, while the drilling loads on the drill bit primarily come from the cutting action. Experiments in one typical lunar regolith simulant indicate that the estimated drilling loads based on the FRAF coincide with the test results quite well. 11Aptamer-wrapped gold nanoparticles for the colorimetric detection of omethoate显示文摘Organophosphorous pesticide(OP) contamination has serious adverse effects on human health and the environment. Due to the toxicity of OPs and the threat presented by their accidental or intentional release in populated areas, the determination and monitoring of these OPs in food products and environment is of great importance. OPs are present in very small quantities and therefore, methods for their detection need to be highly sensitive and selective. Here, we aimed to develop a simple and selective aptamer-based colorimetric assay for the detection of omethoate, which is one of the commonly used OPs. The principle of the assay is that single-stranded DNA(ss DNA)-wrapped gold nanoparticles(Au NPs) are resistant to salt-induced aggregation. By employing an 'artificial antibody' organophosphorous pesticide-binding aptamer(OBA) as the recognition element, aptamer-wrapped Au NPs(Au-apta) show high selectivity towards omethoate, resulting in the disconnection of aptamers from Au NPs and the aggregation of Au NPs. As there is a significant color change from the interparticle plasmon coupling during the aggregation of Au NPs, the established assay showed good linearity between 0.1 and 10 μmol/L, with a low detection limit of 0.1 μmol/L. Other OPs such as profenofos, phorate, and isocarbophos would not interfere with the detection of omethoate despite having similar structures. 12The RING E3 ligase CLG1 targets GS3 for degradation via the endosome pathway to determine grain size in rice显示文摘G-protein signaling and ubiquitin-dependent degradation are both involved in grain development in rice,but how these pathways are coordinated in regulating this process is unknown.Here,we show that Chang Li Geng 1(CLG1),which encodes an E3 ligase,regulates grain size by targeting the Gγprotein GS3,a negative regulator of grain length,for degradation.Overexpression of CLG1 led to increased grain length,while overexpression of mutated CLG1 with changes in three conserved amino acids decreased grain length.We found that CLG1 physically interacts with and ubiquitinats GS3which is subsequently degraded through the endosome degradation pathway,leading to increased grain size.Furthermore,we identified a critical SNP in the exon 3 of CLG1 that is significantly associated with grain size variation in a core collection of cultivated rice.This SNP results in an amino acid substitution from Arg to Ser at position 163 of CLG1 that enhances the E3 ligase activity of CLG1 and thus increases rice grain size.Both the expression level of CLG1 and the SNP CLG1163S may be useful variations for manipulating grain size in rice.Wensi Yang Kun Wu Bo Wang Huanhuan Liu Siyi Guo Xiaoyu Guo Wei Luo Shengyuan Sun Yidan Ouyang Xiangdong Fu Kang Chong Qifa Zhang Yunyuan Xu 2021Molecular Plant2021,14,10:6
13SARS-CoV-2 promotes RIPK1 activation to facilitate viral propagation显示文摘Coronavirus disease 2019(COVID-19),caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),is the ongoing global pandemic that poses substantial challenges to public health worldwide.A subset of COVID-19 patients experience systemic inflammatory response,known as cytokine storm,which may lead to death.Receptor-interacting serine/threonine-protein kinase 1(RIPK1)is an important mediator of inflammation and cell death.Here,we examined the interaction of RIPK1-mediated innate immunity with SARS-CoV-2 infection.We found evidence of RIPK1 activation in human COVID-19 lung pathological samples,and cultured human lung organoids and ACE2 transgenic mice infected by SARS-CoV-2.Inhibition of RIPK1 using multiple smallmolecule inhibitors reduced the viral load of SARS-CoV-2 in human lung organoids.Furthermore,therapeutic dosing of the RIPK1 inhibitor Nec-1s reduced mortality and lung viral load,and blocked the CNS manifestation of SARS-CoV-2 in ACE2 transgenic mice.Mechanistically,we found that the RNA-dependent RNA polymerase of SARS-CoV-2,NSP12,a highly conserved central component of coronaviral replication and transcription machinery,promoted the activation of RIPK1.Furthermore,NSP12323L variant,encoded by the SARS-CoV-2 C14408T variant first detected in Lombardy,Italy,that carries a Pro323Leu amino acid substitution in NSP12,showed increased ability to activate RIPK1.Inhibition of RIPK1 downregulated the transcriptional induction of proinflammatory cytokines and host factors including ACE2 and EGFR that promote viral entry into cells.Our results suggest that SARS-CoV-2 may have an unexpected and unusual ability to hijack the RIPK1-mediated host defense response to promote its own propagation and that inhibition of RIPK1 may provide a therapeutic option for the treatment of COVID-19.Gang Xu Ying Li Shengyuan Zhang Haoran Peng Yunyun Wang Dekang Li Taijie Jin Zhuohao He Yilun Tong Chunting Qi Guowei Wu Kangyun Dong Jizhou Gou Yang Liu Tongyang Xiao Jing Qu Liang Li Liang Liu Ping Zhao Zheng Zhang Junying Yuan 2021Cell Research2021,31,12:6
14A novel miRNA negatively regulates resistance to Glomerella leaf spot by suppressing expression of an NBS gene in apple显示文摘Glomerella leaf spot(GLS)of apple(Malus×domestica Borkh.),caused by Glomerella cingulata,is an emerging fungal epidemic threatening the apple industry.Little is known about the molecular mechanism underlying resistance to this devastating fungus.In this study,high-throughput sequencing technology was used to identify microRNAs(miRNAs)involved in GLS resistance in apple.We focused on miRNAs that target genes related to disease and found that expression of a novel miRNA,Md-miRln20,was higher in susceptible apple varieties than in resistant ones.Furthermore,its target gene Md-TN1-GLS exhibited the opposite expression pattern,which suggested that the expression levels of Md-miRln20 and its target gene are closely related to apple resistance to GLS.Furthermore,downregulation of MdmiRln20 in susceptible apple leaves resulted in upregulation of Md-TN1-GLS and reduced the disease incidence.Conversely,overexpression of Md-miRln20 in resistant apple leaves suppressed Md-TN1-GLS expression,with increased disease incidence.We demonstrated that Md-miRln20 negatively regulates resistance to GLS by suppressing Md-TN1-GLS expression and showed,for the first time,a crucial role for miRNA in response to GLS in apple.Yi Zhang Qiulei Zhang Li Hao Shengnan Wang Shengyuan Wang Wenna Zhang Chaoran Xu Yunfei Yu Tianzhong Li 2019Horticulture Research2019,6,1:4

16Machine learning-based fast frequency response control for a VSC-HVDC system显示文摘HVDC system can realize a very fast frequency response to the disturbed system under a contingency because its active power control is decoupled from the frequency deviation.However,most of existing HVDC frequency control strategies are coupled with system primary frequency control and secondary frequency control.Since the traditional system frequency control is dominated by the thermal generators,the advantage of the fast response of the HVDC system is not made fully used.The development of a frequency response estimation based on a machine learning algorithm provides another approach to improve the frequency response capability of the HVDC system.Different from other frequency deviation tracking strategies,a machine learning based HVDC frequency response control can directly increase the power flow of a HVDC system by estimation of the system generator or load lost.In this paper,a fast frequency response control using a HVDC system for a large power system disturbance based on the multivariate random forest regression(MRFR)algorithm is proposed.The simulation is carried out with an integrated power system model based on the North American interconnections.The simulation results indicate that the proposed MRFR based frequency response control can significantly improve the frequency low point during an event,while stabilizing the frequency in advance.Kaiqi Sun Huangqing Xiao Shengyuan Liu Yilu Liu 2021CSEE Journal of Power and Energy Systems2021,7,4:3
17Changing provenance of late Cenozoic sediments in the Jianghan Basin显示文摘The Yangtze River is one of the most important components of the East Asia river system.In this study,sediments in the Jianghan Basin,middle Yangtze River,were selected for trace element and rare earth element(REE)measurements,in order to decipher information on the change of sediment provenance and evolution of the Yangtze River.According to the elemental variations,the late Cenozoic sediments of the Jianghan Basin could be divided into four parts.During 2.68e2.28 Ma and 1.25e0 Ma,provenance of the sediments was consistent,whereas sediments were derived from variable sources during2.28e1.25 Ma.Comparison of the elemental compositions between the Pliocene and Quaternary sediments revealed a change in sediment source from a more felsic source area to a more basic source area around the PlioceneeQuaternary boundary.Input from the Emeishan LIP should account for this provenance change.Based on the provenance analysis of sediments in the Jianghan Basin,we infer that the Yangtze River developed into a large river with its drainage basin extended to the Emeishan LIP no later than the PlioceneeQuaternary boundary.Lei Shao Shengyuan Yuan Chang'an Li Chunguo Kang Wenjing Zhu Yindi Liu Jietao Wang 2015Geoscience Frontiers2015,6,4:3

19Multi-state autonomous drilling for lunar exploration显示文摘Due to the lack of information of subsurface lunar regolith stratification which varies along depth, the drilling device may encounter lunar soil and lunar rock randomly in the drilling process. To meet the load safety requirements of unmanned sampling mission under limited orbital resources, the control strategy of autonomous drilling should adapt to the indeterminable lunar environments. Based on the analysis of two types of typical drilling media(i.e., lunar soil and lunar rock), this paper proposes a multi-state control strategy for autonomous lunar drilling. To represent the working circumstances in the lunar subsurface and reduce the complexity of the control algorithm, lunar drilling process was categorized into three drilling states: the interface detection, initiation of drilling parameters for recognition and drilling medium recognition. Support vector machine(SVM) and continuous wavelet transform were employed for the online recognition of drilling media and interface, respectively. Finite state machine was utilized to control the transition among different drilling states. To verify the effectiveness of the multi-state control strategy, drilling experiments were implemented with multi-layered drilling media constructed by lunar soil simulant and lunar rock simulant. 20Gene expression microarray analysis of the spinal trigeminal nucleus in a rat model of migraine with aura显示文摘Cortical spreading depression can trigger migraine with aura and activate the trigeminal vascular system. To examine gene expression profiles in the spinal trigeminal nucleus in rats following cor-tical spreading depression-induced migraine with aura, a rat model was established by injection of 1 M potassium chloride, which induced cortical spreading depression. DNA microarray analysis re-vealed that, compared with the control group, the cortical spreading depression group showed seven upregulated genes-myosin heavy chain 1/2, myosin light chain 1, myosin light chain (phos-phorylatable, fast skeletal muscle), actin alpha 1, homeobox B8, carbonic anhydrase 3 and an un-known gene. Two genes were downregulated-RGD1563441 and an unknown gene.
