1Osteoclast-derived apoptotic bodies couple bone resorption and formation in bone remodeling显示文摘Bone remodeling is precisely coordinated by bone resorption and formation.Apoptotic osteoclasts generate large amounts of apoptotic bodies(ABs)marking the end of the bone resorption phase,whereas the functions of osteoclast-derived ABs remain largely unknown.Here,we identified the molecular profile of ABs derived from osteoclasts at distinct differentiation stages and investigated their corresponding functions.ABs were isolated from apoptotic bone marrow macrophages,preosteoclasts,and mature osteoclasts induced by staurosporine.Proteomic signature analysis with liquid chromatography-tandem mass spectrometry suggested marked protein cargo differences among the different ABs.Further bioinformatic analysis showed that the proteomic signatures of the ABs were highly similar to those of their parental cells.Functionally,pOC-ABs induced endothelial progenitor cell differentiation and increased CD31hiEmcnhi endothelial cell formation in a murine bone defect model via their PDGF-BB cargo.mOC-ABs induced osteogenic differentiation of mesenchymal stem cells and facilitated osteogenesis via RANKL reverse signaling.In summary,we mapped the detailed proteomic landscapes of ABs derived from osteoclasts and showed that their potential biological roles are important in coupling bone formation with resorption during bone remodeling.Qinyu Ma Mengmeng Liang Yutong Wu Fei Luo Zaisong Ma Shiwu Dong Jianzhong Xu Ce Dou 2021Bone Research2021,9,1:8
2Natural Feature-based Visual Servoing for Grasping Target with an Aerial Manipulator显示文摘Aerial transportation and manipulation have attracted increasing attention in the unmanned aerial vehicle field,and visual servoing methodology is widely used to achieve the autonomous aerial grasping of a target object.However,the existing marker-based solutions pose a challenge to the practical application of target grasping owing to the difficulty in attaching markers on targets.To address this problem,this study proposes a novel image-based visual servoing controller based on natural features instead of artificial markers.The natural features are extracted from the target images and further processed to provide servoing feature points.A six degree-of-freedom(6-DoF)aerial manipulator system is proposed with differential kinematics deduced to achieve aerial grasping.Furthermore,a controller is designed when the target object is outside a manipulator's workspace by utilizing both the degrees-of-freedom of nnmanned aerial vehicle and manipulator joints.Thereafter,a weight matrix is used as basis to develop a multi-tasking visual servoing framework to integrate the controllers inside and outside the manipulator's workspace.Lastly,experimental results are provided to verify the effectiveness of the proposed approach.Bin Luo Haoyao Chen Fengyu Quan Shiwu Zhang Yunhui Liu 2020Journal of Bionic Engineering2020,17,2:4
3The experimental rules of mica as a reference sample of AFM/FFM measurement显示文摘For the friction measurements with AFM/FFM, usually the relative values of friction signal can be obtained. In order to compare the micro-tribological properties of different samples, mica is often used as an reference sample for friction measurement. However, due to the friction force of new cleaved mica surface is unstable, it is urged to systematically investigate the tribological properties of mica to design the experimental rules of the reference sample mica for friction measurements. Experimental results show that the friction of mica varies with the cleaving time, humidity and surface state of tip. The friction measured with different tips on mica varies in the range of ±15%. For a new tip, the friction increases with the tip’s wear and then becomes stable. For new cleaved mica, the friction increases within thefirst two hours and then keeps unchanged. 4Bone-targeted pH-responsive cerium nanoparticles for anabolic therapy in osteoporosis显示文摘Antiresorptive drugs are widely used for treatment of osteoporosis and cancer bone metastasis,which function mainly through an overall inhibition of osteoclast.However,not all osteoclasts are“bone eaters”;preosteoclasts(pOCs)play anabolic roles in bone formation and angiogenesis through coupling with osteoblasts and secreting platelet derived growth factor-BB(PDGF-BB).In this study,a bone-targeted pH-responsive nanomaterial was designed for selectively eliminating mature osteoclasts(mOCs)without affecting pOCs.Biocompatible cerium nano-system(CNS)was guided to the acidic extracellular microenvironment created by mOCs and gained oxidative enzymatic activity.Oxidative CNS decreased the viability of mOCs through accumulating intracellular reactive oxygen species and enhancing calcium oscillation.Non-acid secreting anabolic pOCs were thus preserved and kept producing PDGF-BB,which lead to mesenchymal stem cell osteogenesis and endothelial progenitor cell angiogenesis via PI3K-Akt activated focal adhesion kinase.In treating osteoporotic ovariectomized mice,CNS showed better protective effects compare with the current first line antiresorptive drug due to the better anabolic effects marked by higher level of bone formation and vascularization.We provided a novel anabolic therapeutic strategy in treating bone disorders with excessive bone resorption.Ce Dou Jianmei Li Jian He Fei Luo Tao Yu Qijie Dai Yueqi Chen Jianzhong Xu Xiaochao Yang Shiwu Dong 2021Bioactive Materials2021,6,12:3
5Phosphorylation inhibition of protein-tyrosine phosphatase 1B tyrosine-152 induces bone regeneration coupled with angiogenesis for bone tissue engineering显示文摘A close relationship has been reported to exist between cadherin-mediated cell-cell adhesion and integrin-mediated cell mobility,and protein tyrosine phosphatase 1B(PTP1B)may be involved in maintaining this homeostasis.The stable residence of mesenchymal stem cells(MSCs)and endothelial cells(ECs)in their niches is closely related to the regulation of PTP1B.However,the exact role of the departure of MSCs and ECs from their niches during bone regeneration is largely unknown.Here,we show that the phosphorylation state of PTP1B tyrosine-152(Y152)plays a central role in initiating the departure of these cells from their niches and their subsequent recruitment to bone defects.Based on our previous design of a PTP1B Y152 region-mimicking peptide(152RM)that significantly inhibits the phosphorylation of PTP1B Y152,further investigations revealed that 152RM enhanced cell migration partly via integrinαvβ3 and promoted MSCs osteogenic differentiation partly by inhibiting ATF3.Moreover,152RM induced type H vessels formation by activating Notch signaling.Demineralized bone matrix(DBM)scaffolds were fabricated with mesoporous silica nanoparticles(MSNs),and 152RM was then loaded onto them by electrostatic adsorption.The DBM-MSN/152RM scaffolds were demonstrated to induce bone formation and type H vessels expansion in vivo.In conclusion,our data reveal that 152RM contributes to bone formation by coupling osteogenesis with angiogenesis,which may offer a potential therapeutic strategy for bone defects.Yong Tang Keyu Luo Yin Chen Yueqi Chen Rui Zhou Can Chen Jiulin Tan Moyuan Deng Qijie Dai Xueke Yu Jian Liu Chengmin Zhang Wenjie Wu Jianzhong Xu Shiwu Dong Fei Luo 2021Bioactive Materials2021,6,7:3
6A functional missense variant in ITIH3 affects protein expression and neurodevelopment and confers schizophrenia risk in the Han Chinese population显示文摘The Psychiatric Genomics Consortium(PGC)has recently identified 10 potential functional coding variants for schizophrenia.However,how these coding variants confer schizophrenia risk remains largely unknown.Here,we investigate the associations between eight potential functional coding variants identified by PGC and schizophrenia in a large Han Chinese sample(n=4022 cases and 9270 controls).Among the eight tested single nucelotide polymorphisms(SNPs),rs3617(a missense variant,p.K315 Q in the ITIH3 gene)showed genome-wide significant association with schizophrenia in the Han Chinese population(P=8.36×10-16),with the same risk allele as in PGC.Interestingly,rs3617 is located in a genomic region that is highly evolutionarily conserved,and its schizophrenia risk allele(C allele)was associated with lower ITIH3 mRNA and protein expression.Intriguingly,mouse neural stem cells stably overexpressing ITIH3 with different alleles of rs3617 exhibited significant differences in proliferation,migration,and differentiation,suggesting the impact of rs3617 on neurodevelopment.Subsequent transcriptome analysis found that the differentially expressed genes in neural stem cells stably overexpressing different alleles of rs3617 were significantly enriched in schizophrenia-related pathways,including cell adhesion,synapse assembly,MAPK and PI3 K-AKT pathways.Our study provides convergent lines of evidence suggesting that rs3617 in ITIH3 likely affects protein function and neurodevelopment and thereby confers risk of schizophrenia.Kaiqin Li Yifan Li Junyang Wang Yongxia Huo Di Huang Shiwu Li Jiewei Liu Xiaoyan Li Rong Liu Xiaogang Chen Yong-Gang Yao Ceshi Chen Xiao Xiao Ming Li Xiong-Jian Luo 2020Journal of Genetics and Genomics2020,47,5:1

8Effects of Long-term Site-specific Fertilization on Soil Physical and Chemical Properties and Nutrients in Dry Farmland显示文摘In order to investigate the variation in soil physical and chemical properties and nutrients in the mountainous areas in southern Ningxia, and to provide a theoretical basis for fertilization management in local farmland, the soil p H, total salt content,crop root length, root weight, soil organic matter, available nitrogen, total nitrogen, total phosphorous and total potassium in different fertilization treatments were measured from 2010 to 2016. Multiple comparisons of the data were performed using Duncan's new multiple range test. The results indicated that in the 0-20 cm soil layer, soil p H value and total salt content changed in different patterns, and varied greatly from 2010 to 2016(P<0.05). The changes of both root length and root weight of millet over time fitted S-shaped curves. The root length and root weight in the four fertilization treatments(Treatment 2 to Treatment5) increased faster than those in the control(Treatment 1). The soil organic matter content in all the five treatments gradually increased from 2010 to 2016. The content of alkaline hydrolyzable nitrogen in soil rapidly increased in the first two to three years of the experiment, followed by a slow increase or decrease in 2013, and then raised rapidly again from 2014 to 2016.The soil total nitrogen content varied significantly from 2010 to 2016. The total phosphorus content in soil changed in a different pattern from that of total nitrogen content. 9Diversity Evaluation of Proso Millet Germplasm Resources in Dryland显示文摘[Objective] This study was to provide a theoretical basis for the selection of the parents of proso millet breeding in dry land of south Ningxia,with the aim to improve the production of hilly proso millet in Ningxia hilly area. [Method]The genetic diversity of 46 accessions of dryland proso millet germplasm at 11 agronomic traits were analyzed by using principal component analysis,Shannon-weaver diversity index and non-weighted matching arithmetic average clustering method( UPGMA). Cluster analysis was also performed. [Result] The proso millet resources mainly had green inflorescence color,panicle spikes,and yellow grain color. The maximum diversity index of proso millet was 2. 03. The variation coefficient of grain weight per plant was 19. 23. In all the principal components,the main information concentrated on the first 4 principal components with the cumulative contribution rate reaching 83. 34%. The tested materials were clustered into 3 categories at euclidean distance of 0. 984 4.
