1Comprehensive analysis of NAC transcription factors and their expression during fruit spine development in cucumber (Cucumis sativus L.)显示文摘The cucumber(Cucumis sativus L.)is an important vegetable crop worldwide,and fruit trichomes or spines are an important trait for external fruit quality.The mechanisms underlying spine formation are not well understood,but the plant-specific NAC family of transcription factors may play important roles in fruit spine initiation and development.In this study,we conducted a genome-wide survey and identified 91 NAC gene homologs in the cucumber genome.Clustering analysis classified these genes into six subfamilies;each contained a varying number of NAC family members with a similar intron–exon structure and conserved motifs.Quantitative real-time PCR analysis revealed tissue-specific expression patterns of these genes,including 10 and 12 that exhibited preferential expression in the stem and fruit,respectively.Thirteen of the 91 NAC genes showed higher expression in the wild-type plant than in its near-isogenic trichome mutant,suggesting their important roles in fruit spine development.Exogenous application of four plant hormones promoted spine formation and increased spine density on the cucumber fruits;several NAC genes showed differential expression over time in response to phytohormone treatments on cucumber fruit,implying their essential roles in fruit-trichome development.Among the NAC genes identified,12 were found to be targets of 13 known cucumber micro-RNAs.Collectively,these findings provide a useful resource for further analysis of the interactions between NAC genes and genes underlying trichome organogenesis and development during fruit spine development in cucumber.Xingwang Liu Ting wang Ezra Bartholomew Kezia Black Mingming Dong Yaqi Zhang Sen Yang Yanling Cai Shudan Xue Yiqun Weng Huazhong Ren 2018Horticulture Research2018,5,1:10
2Selection footprints reflect genomic changes associated with breeding efforts in 56 cucumber inbred lines显示文摘Cucumber selective breeding over recent decades has dramatically increased productivity and quality,but the genomic characterizations and changes associated with this breeding history remain unclear.Here,we analyzed the genome resequencing data of 56 artificially selected cucumber inbred lines that exhibit various phenotypes to detect trait-associated sequence variations that reflect breeding improvement.We found that the 56 cucumber lines could be assigned to group 1 and group 2,and the two groups formed a distinctive genetic structure due to the breeding history involving hybridization and selection.Differentially selected regions were identified between group 1 and group 2,with implications for genomic-selection breeding signatures.These regions included known quantitative trait loci or genes that were reported to be associated with agronomic traits.Our results advance knowledge of cucumber genomics,and the 56 selected inbred lines could be good germplasm resources for breeding.Bin Liu Dailu Guan Xuling Zhai Sen Yang Shudan Xue Shuying Chen Jing Huang Huazhong Ren Xingwang Liu 2019Horticulture Research2019,6,1:0
3High-density genetic mapping identified a major locus for environmental sex expression in pumpkin(Cucurbita moschata Duch.)显示文摘Long-day length and high temperature inhibit sex expression in pumpkin(Cucurbita moschata Duch.),and therefore directly impact the production potential.In this study,female flowering patterns in photoperiod-insensitive(PPIS)and photoperiod-sensitive(PPS)germplasms differed significantly in a moderately long day and high temperature environment.However,both germplasms exhibited a similar response in short day with either low temperature or high temperature environment.Photoperiod sensitivity led to this difference in sex expression between the germplasms.For the traits of 1st female flowering node(FFFN)and number of female flowers(NFF),high-density linkage map construction and quantitative trait locus(QTL)mapping were performed using SLAF-seq technology and 162 F_(2) individuals generated from PPIS and PPS.In total,4655 SLAFs were selected and mapped on 20 linkage groups(LGs).The total map length was 2502.01 cM with an average interval distance of 0.75 cM.Major QTLs for both FFFN and NFF were detected on LG6 with intervals of 7.89 and 17.67 cM and PVE values of 30.5%and 22.9%,respectively.Further analyses of the major locus for FFFN revealed 73 protein-coding genes.Among them,4 were related to sex expression,photoperiod flowering,and hormone response.An InDel(insertion and deletion)marker partially correlated with FFFN of the F_(2) population was also developed.Our study identified the QTL for the sex expression response to environmental factors using the high-density linkage map.The identified candidate genes and markers will provide useful information about the molecular interaction between the environment and sex expression and for marker-assisted selection of pumpkin environment-insensitive resources.Hafiz Muhammad Khalid Abbas Hexun Huang Tingquan Wu Rui Wang Hu Du Sen Lu Shudan Xue Chunpeng Yao Qingmin Jin Yujuan Zhong 2022Horticultural Plant Journal2022,8,5:0
