2Preventive effect of glutamine on intestinal barrier dysfunction induced by severe trauma显示文摘AIM: To investigate the mechanism underlying intestinalbarrier function damage after severe trauma and thetherapeutic effect of glutamine.METHODS: Burned patients, and animal models of severstrauma replicated by hemorrhagic shock combined withendotoxin infusion and burn injury, were included in a serialexperiment. Effects of oral glutamine on intestinal barrierfunction were observed in scalded rets. Parametersmeasured in these experiments were as follows: plasmalevels of diamine oxidase (DAO), tumor necrosis factor(TNFα), endotoxin (LPS), and lactate as well as D-lactateby biochemical methods, lactose/mannitol (L/M) ratio inurine by SP-3400, and pathological examination of intestinalmucosa under light microscopy.RESULTS: Plasma DAO activity was significantly increasedafter injury. There was a negative correlation betweenplasma DAO and intestinal mucosal DAO or pHi ( r= -0.93,plasma 0.80 ± 0.93,2.83 ± 1.71, 1.14 ± 0.64,2.36 ± 2.06 and2.49± 1.67 vs intestinal 0.52± 0.12,0.34 ± 0.03,0.45 ± 0.18,0.37± 0.26 and 0.41 ± 0.07; r = - 0.533, plasma 0.87 ± 0.75,1.89± 1.13, 1.21 ± 0.23,3.03 ± 2.61 and 4.70 ± 1.22 Vs pHi7.03± 0.05,7.05 ± 0.06,7.14 ± 0.096,7.20 ± 0.08 and 7.05 ±0.07; P < 0.01-0.05). Damage of intestinal mucosa was found bypathological examination. Intestinal barrier function wasimproved to a certain extent by oral glutamine in scaldedrats.CONCLUSION: Intestinal barrier function was damaged in theearly stage after trauma. 3New vacuum solar telescope and observations with high resolution显示文摘The New Vacuum Solar Telescope(NVST) is a one meter vacuum solar telescope that aims to observe fine structures on the Sun. The main goals of NVST are high resolution imaging and spectral observations, including measurements of the solar magnetic field. NVST is the primary ground-based facility used by the Chinese solar research community in this solar cycle. It is located by Fuxian Lake in southwest China, where the seeing is good enough to perform high resolution observations. We first introduce the general conditions at the Fuxian Solar Observatory and the primary science cases of NVST. Then, the basic structures of this telescope and instruments are described in detail. 4A double-blind,randomized,placebo-and positive-controlled phase III trial of 1% benvitimod cream in mild-to-moderate plaque psoriasis显示文摘Background:Benvitimod cream,a novel synthetic small molecule,was effective in treating mild-to-moderate plaque psoriasis.We conducted a phase III clinical trial to assess the efficacy and safety of benvitimod cream in patients with mild-to-moderate plaque psoriasis.Methods:We randomly assigned 686 patients(2:1:1)to receive 1%benvitimod cream,0.005%calcipotriol ointment or placebo twice a day for 12 weeks.The primary efficacy end points were the percentage of patients with a 75%or greater reduction from baseline in the psoriasis area and severity index(PASI 75)score and with a score of 0 or 1 in static physician’s global assessment(sPGA)at week 12.Results:The results showed that 50.4%of patients in the benvitimod group achieved PASI 75,which was significantly higher than that in the calcipotriol(38.5%,P<0.05)and placebo(13.9%,P<0.05)groups.The proportion of patients achieving an sPGA score 0 or 1 was 66.3%in the benvitimod group and 63.9%in the calcipotriol group,which were both significantly higher than that in the placebo group(34%,P<0.05).In the long-term follow-up study,50.8%of patients experienced recurrence.After retreatment with 1%benvitimod,73.3%of patients achieved an sPGA score of 0 or 1 again at week 52.Adverse events included application site irritation,follicular papules,and contact dermatitis.No systemic adverse reactions were reported.Conclusion:During this 12-week study,benvitimod cream was demonstrated with high effectiveness and safety in patients with mild-to-moderate plaque psoriasis.Lin Cai Gen-Hui Chen Qian-Jin Lu Min Zheng Yu-Zhen Li Jin Chen Jie Zheng Fu-Ren Zhang Jian-Bin Yu Sen Yang Fu-Qiu Li Sheng-Xiang Xiao Qiu-Ning Sun Jin-Hua Xu Xing-Hua Gao Hong Fang Tian-Wen Gao Fei Hao Quan-Zhong Liu Ya-Ting Tu Ruo-Yu Li Bao-Xi Wang Dan-Qi Deng Qing-Shan Zheng Hong-Xia Liu Jian-Zhong Zhang 2020Chinese Medical Journal2020,,24:11
5Immunological inhibition of transplanted liver allografts by adeno-associated virus vector encoding CTLA4Ig in rats显示文摘BACKGROUND: Blockade interaction between CD28 and B7 with CTLA4Ig has been shown to induce experimental transplantation tolerance. In order to prolong the inhibitory effect of CTLA4Ig, a recombinant adeno- associated virus vector pSNAV expressing CTLA4Ig was constructed, and its effects on transplanted liver allografts were investigated. METHODS: The pSNAV-CTLA4Ig construct was infused into partial liver allografts of rats via the portal vein during transplantation. CTLA4Ig expression in the transplanted livers was detected with reverse transcriptase- polymerase chain reaction (RT-PCR) analysis and immunohistochemistry. Furthermore, real-time quantita- tive PCR was used to measure the expression of IL-2, IFN-γ, IL-4 and IL-10 in the allografts. RESULTS: The expression of CTLA4Ig in the partial allograft was detected successfully and pSNAV-CTLA4Ig improved the survival rate of rats after liver transplantation. Agarose gel analysis of RT-PCR products indicated the presence of CTLA4Ig in the pSNAV-CTLA4Ig treatment group. Cytokines expressed in allografts on day 7 after orthotopic liver transplantation showed that IL-2, IFN-γ, IL-4 and IL-10 mRNA levels decreased in transplant recipients treated with pSNAV-CTLA4Ig compared with those treated with pSNAV-LacZ (1.62±0.09, 1.52±0.11, 1.50± 0.07 and 1.43±0.07 versus 1.29±0.09, 1.32±0.07, 1.34±0.06 and 1.35±0.04, respectively).CONCLUSIONS: pSNAV-CTLA4Ig effectively expressed CTLA4Ig in liver allografts. CTLA4Ig improved the pathological findings after liver transplantation. CTLA4Ig induced immune tolerance of liver transplantation, and the mechanism involved induced alteration of Th1 and Th2 cytokine transcripts. 6MAPPING OF THE RICE (ORYZA SATIVA L.) THERMO-SENSITIVE GENIC MALE STERILE GENE TMS5 WITH EST AND SSR MARKERS显示文摘With the cDNA suppression subtraction hybridization method, a spikelet-specific cDNA library was constructed that expressed at meiosis stage in rice. A total of 121 cDNA fragments were selected from the library and used as EST (expressed se-quence tags) markers to detect the polymorphism between Annong N, a normal fertile Indica rice line and Annong S-1, its spontaneous mutant with thermo-sensitive genic male sterility, using the RFLP (restriction fragment length polymorphism) technique. HN57, one of the EST probes, could detect poly-morphism between them. The results of segregation analysis with the F2 population developed from An-nong S-1 and Annong N indicate that HN57 co-seg- regates with the thermo-sensitive genic male-sterility controlled by tms5, the recessive gene in Annong S-1. This marker is located on the 31.2-cM region of the chromosome 2 of RGP (rice genome research pro-gram) genetic map. To further determine the location of tms5, 80 SSR (simple sequence repeat) markers around this region were developed, and 12 of them were polymorphic. 7Effectiveness of glucocorticoid therapy in patients with severe coronavirus disease 2019:protocol of a randomized controlled trial显示文摘Background:At the end of 2019,a novel coronavirus outbreak causative organism has been subsequently designated the 2019 novel coronavirus(2019-nCoV).The effectiveness of adjunctive glucocorticoid therapy in the management of 2019-nCoV-infected patients with severe lower respiratory tract infections is not clear,and warrants further investigation.Methods:The present study will be conducted as an open-labeled,randomized,controlled trial.We will enrol 48 subjects from Chongqing Public Health Medical Center.Each eligible subject will be assigned to an intervention group(methylprednisolone via intravenous injection at a dose of 1-2 mg/kg/day for 3 days)or a control group(no glucocorticoid use)randomly,at a 1:1 ratio.Subjects in both groups will be invited for 28 days of follow-up which will be scheduled at four consecutive visit points.We will use the clinical improvement rate as our primary endpoint.Secondary endpoints include the timing of clinical improvement after intervention,duration of mechanical ventilation,duration of hospitalization,overall incidence of adverse events,as well as rate of adverse events at each visit,and mortality at 2 and 4 weeks.Discussion:The present coronavirus outbreak is the third serious global coronavirus outbreak in the past two decades.Oral and parenteral glucocorticoids have been used in the management of severe respiratory symptoms in coronavirus-infected patients in the past.However,there remains no definitive evidence in the literature for or against the utilization of systemic glucocorticoids in seriously ill patients with coronavirus-related severe respiratory disease,or indeed in other types of severe respiratory disease.In this study,we hope to discover evidence either supporting or opposing the systemic therapeutic administration of glucocorticoids in patients with severe coronavirus disease 2019.Yuan-Yuan Qin Yi-Hong Zhou Yan-Qiu Lu Feng Sun Sen Yang Vijay Harypursat Yao-Kai Chen 2020Chinese Medical Journal2020,,9:8

9Heat shock protein 90 promotes RNA helicase DDX5 accumulation and exacerbates hepatocellular carcinoma by inhibiting autophagy显示文摘Objective:Hepatocellular carcinoma(HCC),the main type of liver cancer,has a high morbidity and mortality,and a poor prognosis.RNA helicase DDX5,which acts as a transcriptional co-regulator,is overexpressed in most malignant tumors and promotes cancer cell growth.Heat shock protein 90(HSP90)is an important molecular chaperone in the conformational maturation and stabilization of numerous proteins involved in cell growth or survival.Methods:DDX5 m RNA and protein expression in surgically resected HCC tissues from 24 Asian patients were detected by quantitative real-time PCR and Western blot,respectively.The interaction of DDX5-HSP90 was determined by molecular docking,immunoprecipitation,and laser scanning confocal microscopy.The autophagy signal was detected by Western blot.The cell functions and signaling pathways of DDX5 were determined in 2 HCC cell lines.Two different murine HCC xenograft models were used to determine the function of DDX5 and the therapeutic effect of an HSP90 inhibitor.Results:HSP90 interacted directly with DDX5 and inhibited DDX5 protein degradation in the AMPK/ULK1-regulated autophagy pathway.The subsequent accumulation of DDX5 protein induced the malignant phenotype of HCC by activating theβ-catenin signaling pathway.The silencing of DDX5 or treatment with HSP90 inhibitor both blocked in vivo tumor growth in a murine HCC xenograft model.High levels of HSP90 and DDX5 protein were associated with poor prognoses.Conclusions:HSP90 interacted with DDX5 protein and subsequently protected DDX5 protein from AMPK/ULK1-regulated autophagic degradation.DDX5 and HSP90 are therefore potential therapeutic targets for HCC.Ting Zhang Xinrui Yang Wanping Xu Jing Wang Dawei Wu Zhixian Hong Shengxian Yuan Zhen Zeng Xiaodong Jia Shanshan Lu Rifaat Safadi Sen Han Zhihong Yang Leonard M.Neckers Suthat Liangpunsakul Weiping Zhou Yinying Lu 2021Cancer Biology & Medicine2021,18,3:7
10Improving the electrical performance of resistive switching memory using doping technology显示文摘In this paper, improvements of resistive random access memory (RRAM) using doping technology are summarized and analyzed. Based on a Cu/ZrO2/Pt device, three doping technologies with Ti ions, Cu, and Cu nanocrystal, respectively, are adopted in the experiments. Compared to an undoped device, improvements focus on four points: eliminating the electroforming process, reducing operation voltage, improving electrical uniformity, and increasing device yield. In addition, thermal stability of the high resistance state and better retention are also achieved by the doping technology. 12A novel approach for designing efficient broadband photodetectors expanding from deep ultraviolet to near infrared显示文摘Broadband photodetection(PD)covering the deep ultraviolet to near-infrared(200–1000 nm)range is significant and desirable for various optoelectronic designs.Herein,we employ ultraviolet(UV)luminescent concentrators(LC),iodinebased perovskite quantum dots(PQDs),and organic bulk heterojunction(BHJ)as the UV,visible,and near-infrared(NIR)photosensitive layers,respectively,to construct a broadband heterojunction PD.Firstly,experimental and theoretical results reveal that optoelectronic properties and stability of CsPbI3 PQDs are significantly improved through Er3+doping,owing to the reduced defect density,improved charge mobility,increased formation energy,tolerance factor,etc.The narrow bandgap of CsPbI3:Er3+PQDs serves as a visible photosensitive layer of PD.Secondly,considering the matchable energy bandgap,the BHJ(BTP-4Cl:PBDB-TF)is selected as to NIR absorption layer to fabricate the hybrid structure with CsPbI3:Er3+PQDs.Thirdly,UV LC converts the UV light(200–400 nm)to visible light(400–700 nm),which is further absorbed by CsPbI3:Er3+PQDs.In contrast with other perovskites PDs and commercial Si PDs,our PD presents a relatively wide response range and high detectivity especially in UV and NIR regions(two orders of magnitude increase that of commercial Si PDs).Furthermore,the PD also demonstrates significantly enhanced air-and UV-stability,and the photocurrent of the device maintains 81.5%of the original one after 5000 cycles.This work highlights a new attempt for designing broadband PDs,which has application potential in optoelectronic devices.Nan Ding Yanjie Wu Wen Xu Jiekai Lyu Yue Wang Lu Zi Long Shao Rui Sun Nan Wang Sen Liu Donglei Zhou Xue Bai Ji Zhou Hongwei Song 2022Light(Science & Applications)2022,11,5:5
13Epigenetic changes of pituitary tumor-derived transforming gene 1 in pancreatic cancer显示文摘BACKGROUND:Pancreatic cancer is a devastating disease with abnormal genetic changes.The pituitary tumor- derived transforming gene(PTTG)is considered to be implicated in the tumorigenesis of cancers when the gene is epigenetically transformed.In this study,we investigated the relationships between aberrant expression and epigenetic changes of the PTTG1 gene in pancreatic cancer. METHODS:We chose 4 cell lines(PANC-1,Colo357,T3M-4 and PancTuⅠ)and pancreatic ductal adenocarcinoma (PDAC)tissues.After using restriction isoschizomer endonucleases(MspⅠ/HpaⅡ)to digest the DNA sequence (5'-CCGG-3'),we performed PCR reaction to amplify the product.And RT-PCR was applied to determine the gene expression. RESULTS:The mRNA expression of the PTTG1 gene was higher in pancreatic tumor than in normal tissue.The gene was also expressed in the 4 PDAC cell lines.The methylation states of the upstream regions of the PTTG1 gene were almost identical in normal,tumor pancreatic tissues and the 4 PDAC cell lines.Some(5'-CCGG-3')areas in the upstream region of PTTG1 were methylated,while some others were unmethylated. 14Clinical significance of nerve growth factor and tropomyosin-receptor-kinase signaling pathway in intrahepatic cholangiocarcinoma显示文摘AIM:To investigate the correlation between nerve growth factor-tropomyosin-receptor-kinase(NGF-TrkA)signaling pathway and prognosis in intrahepatic cholangiocarcinoma(IHCC).METHODS:NGF and TrkA expression in 83 samples of IHCC was assessed by immunohistochemistry.Correlations between NGF-TrkA expression and clinicopathological features were analyzed byχ2 test.Moreover,we evaluated the association between NGF-TrkA and overall survival by univariate and multivariate analysis.With experiments in vitro,we investigated the crucial role of NGF-TrkA on proliferation and invasion of IHCC cells with recombinant NGF-βstimulation.RESULTS:We found that NGF and TrkA expression was significantly related with differentiation(P=0.024)and intraneural invasion(P=0.003),respectively.Additionally,double higher expression of NGF and TrkA was identified as an independent prognostic factor in IHCC(P=0.003).Moreover,we demonstrated that NGF-TrkA signaling pathway can promote IHCC proliferation and invasion.CONCLUSION:NGF-TrkA double higher expression is an independent prognostic factor in IHCC.NGF-TrkA pathway can promote IHCC progression,indicating that NGF-TrkA may become a potential drug target.Xiao-Qing Yang Yun-Fei Xu Sen Guo Yi Liu Shang-Lei Ning Xiao-Fei Lu Hui Yang Yu-Xin Chen 2014World Journal of Gastroenterology2014,20,14:4

METHODS:Four cell lines(PANC-1,Colo357,T3M-4 and PancTuⅠ)and pancreatic ductal adenocarcinoma(PDAC) tissues were used for mRNA detection.After restriction isoschizomer endonucleases(MspⅠ/HpaⅡ)were used to digest the DNA sequence(5'-CCGG-3'),PCR was made to amplify the product.And RT-PCR was applied to determine the expression of the gene. RESULTS:The mRNA expression of the ECT2 gene was higher in pancreatic tumor tissue than in normal tissue. The gene was also expressed in the 4 PDAC cell lines.The methylation states of the upstream regions of the ECT2 gene were almost identical in normal,tumor pancreatic tissues,and the 4 PDAC cell lines.Some of the 5'-CCGG-3' areas in the upstream region of ECT2 were methylated, while others were unmethylated.CONCLUSIONS:The oncogene ECT2 is overexpressed in pancreatic tumor tissues as verified by RT-PCR detection. 17The causes and processes responsible for rocky desertification in karst areas of southern China显示文摘The karst region of southern China is a fragile ecological zone with widespread rocky desertification. This paper describes the rocky desertification occurring in this region in terms of both natural and anthropogenic factors. During geological time periods, the region's changing environment governed the natural rocky desertification processes, whereas during historical and modern times, anthropogenic processes have been superimposed on these natural processes. Human activities have accelerated and exacerbated rocky desertification. The period from the beginning to the middle of the Qing dynasty was an important transitional period in which human activities began to exert a particularly strong influence on rocky desertification. Since then, the effect of anthropogenic factors has increasingly exceeded the effect of natural factors. The rocky desertification process in southern China's karst region combines surface ecological processes (including vegetation degradation and loss, soil erosion, surface water loss, and bedrock solution) with a reduction of the land's biological productivity, leading to degradation that produces rocky desert. 18Signalling pathways in autism spectrum disorder:mechanisms and therapeutic implications显示文摘Autism spectrum disorder(ASD)is a prevalent and complex neurodevelopmental disorder which has strong genetic basis.Despite the rapidly rising incidence of autism,little is known about its aetiology,risk factors,and disease progression.There are currently neither validated biomarkers for diagnostic screening nor specific medication for autism.Over the last two decades,there have been remarkable advances in genetics,with hundreds of genes identified and validated as being associated with a high risk for autism.The convergence of neuroscience methods is becoming more widely recognized for its significance in elucidating the pathological mechanisms of autism.Efforts have been devoted to exploring the behavioural functions,key pathological mechanisms and potential treatments of autism.Here,as we highlight in this review,emerging evidence shows that signal transduction molecular events are involved in pathological processes such as transcription,translation,synaptic transmission,epigenetics and immunoinflammatory responses.This involvement has important implications for the discovery of precise molecular targets for autism.Moreover,we review recent insights into the mechanisms and clinical implications of signal transduction in autism from molecular,cellular,neural circuit,and neurobehavioural aspects.Finally,the challenges and future perspectives are discussed with regard to novel strategies predicated on the biological features of autism.Chen-Chen Jiang Li-Shan Lin Sen Long Xiao-Yan Ke Kohji Fukunaga Ying-Mei Lu Feng Han 2022Signal Transduction and Targeted Therapy2022,7,8:3
19Application of air insufflation to prevent clinical pancreatic fistula after pancreaticoduodenectomy显示文摘AIM:To introduce an air insufflation procedure and to investigate the effectiveness of air insufflation in preventing pancreatic fistula(PF).of 185 patients underwent pancreaticoduodenectomy(PD)at our institution,and 74 patients were not involved in this study for various reasons.The clinical outcomes of 111 patients were retrospectively analyzed.The air insufflation test was performed in 46 patients to investigate the efficacy of the pancreaticojejunal anastomosis during surgery,and 65 patients who did not receive the air insufflation test served as controls.Preoperative assessments and intraoperative outcomes were compared between the 2 groups.Univariate and multivariate analyses were performed to identify the risk factors for PF.RESULTS:The two patient groups had similar baseline demographics,preoperative assessments,operative factors,pancreatic factors and pathological results.The overall mortality,morbidity,and PF rates were1.8%,48.6%,and 26.1%,respectively.No significant differences were observed in either morbidity or mortality between the two groups.The rate of clinical PF(grade B and grade C PF)was significantly lower in the air insufflation test group,compared with the nonair insufflation test group(6.5%vs 23.1%,P=0.02).Univariate analysis identified the following parameters as risk factors related to clinical PF:estimated blood loss;pancreatic duct diameter≤3 mm;invagination anastomosis technique;and not undergoing air insufflation test.By further analyzing these variables with multivariate logistic regression,estimated blood loss,pancreatic duct diameter≤3 mm and not undergoing air insufflation test were demonstrated to be independent risk factors.CONCLUSION:Performing an air insufflation test could significantly reduce the occurrence of clinical PF after PD.Not performing an air insufflation test was an independent risk factor for clinical PF.Hui Yang Xiao-Fei Lu Yun-Fei Xu Hong-Da Liu Sen Guo Yi Liu Yu-Xin Chen 2015World Journal of Gastroenterology2015,21,6:2
