| 1 | Expressed sequence tags analysis revealing the taxonomic position and fatty acid biosynthesis in an oleaginous green microalga,Myrmecia incisa Reisigl(Trebouxiophyceae,Chlorophyta)显示文摘cDNA library of Myrmecia incisa H4301 was constructed using λ phage vectors.The library consisted of 1.5×10 6 clones with a recombination rate of 90%,and 1942 clones were randomly sequenced.All 1854 readable expressed sequence tags(ESTs) were clustered into 596 non-redundant sequences(NRSs),among which 126 NRSs were from 1384 ESTs,showing a high redundancy.Among the 596 NRSs,30 were ribosomal RNA,and 152 significantly matched with those available in NCBI database and JGI Genome Portal,the latter were divided into nine subcategories.Overall,59 NRSs were involved in photosynthesis,the respiratory electron transport chain,ATP synthesis,oxidation reduction,fatty acid biosynthesis,glucose metabolism,protein metabolism,and small molecular metabolism,suggesting that these genes were abundantly transcribed during energy and substance metabolism.Acyl-carrier protein,ferrodoxin and fatty acid elongase genes obtained from this cDNA library enabled presumption of a possible biosynthesis pathway of ArA in M.incisa.Codon usage analysis of 142 NRSs with 17798 codons in the predicted coding regions showed that the average G+C content level of the total codons was 55.39%,and that of the third position in base trimers was 66.42%,indicating a strong bias toward cytosine and/or guanosine in this algal genome.Among all synonymous codons,NAG was most favored,while NUA was most avoided.Phylogenic trees inferred from ribulose-1,5-bisphosphate carboxylase/oxygenase small subunit genes and the extra partial sequences of 18S rRNA obtained from this library demonstrated that M.incisa belonged to Trebouxiophyceae and was significantly clustered with M.incisa SAG 2007,Lobosphaera tirolensis,M.bisecta,and Parietochloris incisa,but was clearly distant from P.pseudoalveolaris and P.alveolar.Transmission electron microscopy revealed pyrenoids traversed by many parallel thylakoids membranes,while starch grains were only clearly observed when cells were grown under nitrogen stress.Based on combined investigation of the phylogeny and morphological characteristics,it is proposed that M.incisa be kept in the genus Myrmecia in which there might be two parallel groups,one living freely and another symbiotic species. | OUYANG LongLing DU DaoHai YU ShuiYan LI ChunYang ZHANG ChengWu GAO HongJian ZHOU ZhiGang | 2012 | Chinese Science Bulletin2012,57,25: | 4 |
| 2 | Lamprey buccal gland secretory protein-2 (BGSP-2) inhibits human T lymphocyte proliferation显示文摘Lamprey is a representative of the agnathans, the most ancient class of vertebrates. Parasitic lampreys secrete anticoagulantfrom their buccal glands and prevent blood coagulation of host fishes. We identified a buccal gland secretory protein-2(BGSP-2) from a buccal gland cDNA library of Lampetra japonica. The full-length BGSP-2 gene was cloned and the recombinantBGSP-2 protein was generated. The role of BGSP-2 on lymphocyte proliferation was studied by examining its effects onhuman T lymphocytes. We found that lamprey BGSP-2 was able to effectively block the proliferation of T cells in vitro by inducingG_1/S cell cycle arrest. Furthermore, it inhibited the proliferation of human T lymphocytes stimulated by phytohemagglutinin(PHA) at a minimum concentration of 0.1μg/ml. | 3 | Edge detection algorithm based on ICA-domain shrinkage in noisy images显示文摘We propose a robust edge detection method based on ICA-domain shrinkage(independent component analysis) . It is known that most basis functions extracted from natural images by ICA are sparse and similar to localized and oriented receptive fields,and in the proposed edge detection method,a target image is first transformed by ICA basis functions and then the edges are detected or reconstructed with sparse components. Furthermore,by applying a shrinkage algorithm to filter out the components of noise in ICA-domain,we can readily obtain the sparse components of the original image,resulting in a kind of robust edge detection even for a noisy image with a very low SN ratio. Morphological and molecular studies, however, have resulted in conflicting views with regard to their interrelationships. To clarify the phylogenetic relationships between them, 48 orthologous protein-coding gene families were analyzed. Even as the analysis of 34 nuclear gene families supported the monophyly of cyclostomes, the analysis of 14 mitochondrial gene families suggested a closer relationship between lampreys and gnathostomes compared to hagfishes. Lampreys were sister group of gnathostomes. The results of this study sup- ported the cyclostomes. Choice of outgroup, tree-making methods, and software may affect the phylogenetic prediction, which may have caused much debate over the subject. | 16 | Clinical significance of serum IgM and IgG levels in COVID-19 patients in Hubei Province,China显示文摘Background:There have been many studies about coronavirus disease 2019(COVID-19),but the clinical signifi-cance of quantitative serum severe acute respiratory syndrome-coronavirus-2(SARS-CoV-2)-specific IgM and IgG levels of COVID-19 patients have not been exhaustively analyzed.We aimed to investigate the time profiles of these IgM/IgG levels in COVID-19 patients and their correlations with clinical features.Methods:A multicenter clinical study was conducted from February 20 to March 52020.It involved 179 COVID-19 patients(108 males and 71 females)from five hospitals in Huangshi in Hubei Province,China.To detect SARS-CoV-2-specific IgM/IgG,quantitative antibody assays(two-step indirect immunoassays with direct chemilu-minescence technology)based on the nucleocapsid protein(NP)and spike protein 1(S1)were used.For normally distributed data,means were compared using the t-test,��2-test,or exact probability method.For categorical data,medians were compared using Mann-Whitney U test.Results:The median age was 57(44-69)years(58[38-69]for males and 57[49-68]for females).The median duration of positive nucleic acid test was 22.32(17.34-27.43)days.The mortality rate was relatively low(3/179,1.68%).Serum SARS-CoV-2-specific IgG was detected around week 1 after illness onset,gradually increased until peaking in weeks 4 and 5,and then declined.Serum IgM peaked in weeks 2 and 3,then gradually declined and returned to its normal range by week 7 in all patients.Notably,children had milder respiratory symptoms with lower SARS-CoV-2-specific IgM/IgG levels.The duration of positive nucleic acid test in the chronic obstructive pulmonary disease(COPD)group was 30.36(18.99-34.76)days,which was significantly longer than that in the non-COPD group(21.52[16.75-26.51]days;P=0.025).The peak serum SARS-CoV-2-specific IgG was sig-nificantly positively correlated with the duration of positive nucleic acid test.The incidence rate of severe and critical cases in the IgM hi group(using the median IgM level of 29.95 AU/mL as the cutofffor grouping)was about 38.0%(19/50),which was twice as much as that in the IgM lo group(18.4%;9/49).The patients with positive chest imaging and lymphocytopenia(<1×10^(9)/L)had a higher SARS-CoV-2-specific IgM level.Conclusions: Quantitative SARS-CoV-2-specific IgM and IgG levels are helpful for the diagnosis, severity classifi- cation, and management of COVID-19 patients, and they should be monitored in each stage of this disease. | Zhenjiang Bai Qing Li Qinghui Chen Changming Niu Yu Wei Hanpeng Huang Wei Zhao Nian Chen Xin Yao Qiang Zhang Chuanyong Mu Jian Feng Chuanlong Zhu Zhuo Li Ming Ding Binhui Feng Chaochao Jin Xiang Lu Yi Yang Shuiyan Wu Xiaochen Shu Haibo Qiu YingZi Huang | 2022 | Journal of Intensive Medicine2022,2,1: | 0 |
| 17 | Electrically driven single microwire-based single-mode microlaser显示文摘Engineering the lasing-mode oscillations effectively within a laser cavity is a relatively updated attentive study and perplexing issue in the field of laser physics and applications. Herein, we report a realization of electrically driven single-mode microlaser, which is composed of gallium incorporated zinc oxide microwire (ZnO:Ga MW) with platinum nanoparticles (PtNPs, d ~ 130 nm) covering, a magnesium oxide (MgO) nanofilm, a Pt nanofilm, and a p-type GaN substrate. The laser cavity modes could resonate following the whispering-gallery mode (WGM) among the six side surfaces by total internal reflection, and the single-mode lasing wavelength is centered at 390.5 nm with a linewidth of about 0.18 nm. The cavity quality factor Q is evaluated to about 2169. In the laser structure, the usage of Pt and MgO buffer layers can be utilized to engineer the band alignment of ZnO:Ga/GaN heterojunction, optimize the p-n junction quality and increase the current injection. Thus, the well-designed device structure can seamlessly unite the electron-hole recombination region, the gain medium, and optical microresonator into the PtNPs@ZnO:Ga wire perfectly. Such a single MW microlaser is essentially single-mode regardless of the gain spectral bandwidth. To study the single-mode operation, PtNPs working as superabsorber can engineering the multimode lasing actions of ZnO:Ga MWs even if their dimensions are typically much larger than that of lasing wavelength. | 18 | Lifting surface reconstruction of Au(100)by tellurium adsorption显示文摘The Au(100)surface has been a subject of intense studies due to excellent catalytic activities and its model character for surface science.However,the spontaneous surface reconstruction buries active Au(100)plane and limits practical applications,how to controllably eliminate the surface reconstruction over large scale remains challenging.Here,we experimentally and theoretically demonstrate that simple decoration of the Au(100)surface by tellurium(Te)atoms can uniquely lift its reconstruction over large scale.Scanning tunneling microscopy imaging reveals that the lifting of surface reconstruction preferentially starts from the boundaries of distinct domains and then extends progressively into the domains with the reconstruction rows perpendicular to the boundaries,leaving a Au(100)-(1×1)surface behind.The Au(100)-(1×1)is saturated at~84%±2%with respect to the whole surface at a Te coverage of 0.16 monolayer.With further increasing the Te coverage to 0.25 monolayer,the Au(100)-(1×1)surface becomes reduced and overlapped by a well-ordered(2×2)-Te superstructure.No similar behavior is found for Te-decorated Au(111),Cu(111),Cu(100)surfaces,nor for the decorated Au(100)with other elements.This result may pave the way to design Au-based catalysts and,as an intermediate step,even potentially open a new route to constructing complex transition metal dichalcogenides. | Erwen Zhang Huan Lu Wei Zhang Shuiyan Cao Yang Wang Rong Rong Ying Liu Zhuhua Zhang Ming Yang Yanpeng Liu Wanlin Guo | 2023 | Nano Research2023,16,5: | 0 |
