1Extranodal involvement in young patients with diffuse large B-cell lymphoma: distribution, prognostic value and treatment options显示文摘Objective: Extranodal involvement represents a peculiar presentation of diffuse large B-cell lymphoma(DLBCL). Previous studies have suggested that older patients are more prone to extranodal involvement. This study retrospectively addressed the distribution, prognostic value and treatment options of extranodal involvement in young patients with DLBCL.Methods: A total of 329 patients were enrolled according to the inclusion requirements. The effects of gender,extranodal involvement, age-adjusted international prognostic index(aa IPI), rituximab infusion and radiotherapy on patient outcomes were evaluated.Results: Among these patients, 59% presented extranodal involvement in 16 anatomic sites. More than one instance was linked to many poorer clinical characteristics and poorer survival compared with either nodal disease or one instance. In patients with one extranodal lesion, multivariate analysis revealed that the site of extranodal involvement, but not the aa IPI or rituximab infusion, was independently related to the outcome, and radiotherapy had a negative influence on survival.Conclusions: Extranodal involvement is common in younger patients and exhibits a ubiquitous distribution.The site of extranodal involvement is of strong prognostic significance. 2Association between 15 known or potential breast cancer susceptibility genes and breast cancer risks in Chinese women显示文摘Objective:There are many hereditary breast cancer patients in China,and multigene panel testing has been a new paradigm of genetic testing for these patients and their relatives.However,the magnitude of breast cancer risks related to multiple breast cancer susceptibility genes are largely unknown in Chinese women.Methods:We screened pathogenic variants in 15 established or potential breast cancer susceptibility genes from 8,067 consecutive Chinese female breast cancer patients and 13,129 Chinese cancer-free female controls.These breast cancer patients were unselected for age at diagnosis or family history.Results:We found that pathogenic variants in TP53[odds ratio(OR):16.9,95%confidence interval(CI):5.2–55.2];BRCA2(OR:10.4,95%CI:7.6–14.2);BRCA1(OR:9.7,95%CI:6.3–14.8);and PALB2(OR:5.2,95%CI:3.0–8.8)were associated with a high risk of breast cancer.ATM,BARD1,CHEK2,and RAD51D were associated with a moderate risk of breast cancer with ORs ranging from 2-fold to 4-fold.In contrast,pathogenic variants of NBN,RAD50,BRIP1,and RAD51C were not associated with increased risk of breast cancer in Chinese women.The pathogenic variants of PTEN,CDH1,and STK11 were very rare,so they had a limited contribution to Chinese breast cancer.Patients with pathogenic variants of TP53,BRCA1,BRCA2,and PALB2 more often had earlyonset breast cancer,bilateral breast cancer,and a family history of breast cancer and/or any cancer.Conclusions:This study provided breast cancer risk assessment data for multiple genes in Chinese women,which is useful for genetic testing and clinical management of Chinese hereditary breast cancer.Fenfen Fu Dongjie Zhang Li Hu Senthil Sundaram Dingge Ying Ying Zhang Shuna Fu Juan Zhang Lu Yao Ye Xu Yuntao Xie 2022Cancer Biology & Medicine2022,19,2:3
3Programmable deaminase-free base editors for G-to-Y conversion by engineered glycosylase显示文摘Current DNA base editors contain nuclease and DNA deaminase that enables deamination of cytosine(C)or adenine(A),but no method for guanine(G)or thymine(T)editing is available at present.Here we developed a deaminase-free glycosylase-based guanine base editor(gGBE)with G editing ability,by fusing Cas9 nickase with engineered N-methylpurine DNA glycosylase protein(MPG).By several rounds of MPG mutagenesis via unbiased and rational screening using an intron-split EGFP reporter,we demonstrated that gGBE with engineered MPG could increase G editing efficiency by more than 1500 fold.Furthermore,this gGBE exhibited high base editing efficiency(up to 81.2%)and high G-to-T or G-to-C(i.e.G-to-Y)conversion ratio(up to 0.95)in both cultured human cells and mouse embryos.Thus,we have provided a proof-of-concept of a new base editing approach by endowing the engineered DNA glycosylase the capability to selectively excise a new type of substrate.Huawei Tong Nana Liu Yinghui Wei Yingsi Zhou Yun Li Danni Wu Ming Jin Shuna Cui Hengbin Li Guoling Li Jingxing Zhou Yuan Yuan Hainan Zhang Linyu Shi Xuan Yao Hui Yang 2023National Science Review2023,10,8:1

6PtNiCu nanowires with advantageous lattice-plane boundary for enhanced ethanol electrooxidation显示文摘It is generally accepted that the interface effect and surface electronic structure of catalysts have vital impact on catalytic properties.Understanding and tailoring the atomic arrangement of interface structure are of great importance for electrocatalysis.Herein,we proposed a simple method to synthesize etching-PtNiCu nanowires(e-PtNiCu NWs)enclosed by both(110)and(100)facets evolving from PtNiCu nanowires(PtNiCu NWs)mainly with(111)facets by selectively etching process.After acetic acid etching treatment,the e-PtNiCu NWs possess high total proportions(88.3%)of(110)and(100)facets,whereas the(111)facet is dominant in PtNiCu NWs(64%)by qualitatively and quantitatively evaluation.Combining the structure characterizations and performance tests of ethanol electrooxidation reaction(EOR),we find that the e-PtNiCu NWs display remarkably performance for EOR,which is nearly 4.5 times and 1.5 times enhancement compared with the state-of-the-art Pt/C catalyst,as well as 2.2 and 1.4 times of PtNiCu NWs,in specific activity and mass activity,respectively.The improved performance of e-PtNiCu NWs is attributed to synergistic catalytic effect between(110)and(100)facets that not only significantly decreases the onset potentials of adsorbed CO(CO_(ads))but also favors the oxidation of CO_(ads)on the surface of catalyst.Furthermore,thermodynamics and kinetic studies indicate that the synergistic effect of both(110)and(100)facets in e-PtNiCu NWs can decrease the activation energy barrier and facilitate the charge transfer during the reaction.This work provides a promising approach to construct catalysts with tunable surface electronic structure towards efficient electrocatalysis.Shuna Li Yao Wang Yunrui Li Xu Fang Yujie Liu Mingxuan Li Zhen Wang Yufeng Gao Haixiao Sun Fei Gao Xin Zhang Xiaoping Dai 2022Nano Research2022,15,4:0
