1Real world effectiveness of PCSK-9 inhibitors combined with statins versus statins-based therapy among patients with very high risk of atherosclerotic cardiovascular disease in China(RWE-PCSK study)显示文摘BACKGROUND The efficacy and safety of proprotein convertase subtilisin/kexin type 9(PCSK-9) inhibitors were confirmed by several clinical trials, but its effectiveness in routine clinical practice in China has not been evaluated. This study aims to describe the real world effectiveness of PCSK-9 inhibitors combined with statins compared with statins-based therapy among patients with very high risk of atherosclerotic cardiovascular disease(ASCVD).METHODS This is a multi-center observational study, enrolled patients from 32 hospitals who underwent percutaneous coronary intervention(PCI) from January to June in 2019. There are 453 patients treated with PCSK-9 inhibitors combined with statins in PCSK-9 inhibitor group and 2,610 patients treated with statins-based lipid lowering therapies in statins-based group. The lipid control rate and incidence of major adverse cardiovascular events(MACE) over six months were compared between two groups.A propensity score-matched(PSM) analysis was used to balance two groups on confounding factors. Survival analysis using Kaplan-Meier methods was applied for MACE.RESULTS In a total of 3,063 patients, 89.91% of patients had received moderate or high-intensity statins-based therapy before PCI, but only 9.47% of patients had low-density lipoprotein cholesterol(LDL-C) levels below 1.4 mmol/L at baseline. In the PSM selected patients, LDL-C level was reduced by 42.57% in PCSK-9 inhibitor group and 30.81%(P < 0.001) in statins-based group after six months. The proportion of LDL-C ≤ 1.0 mmol/L increased from 5.29% to 29.26% in PCSK-9 inhibitor group and 0.23% to 6.11% in statins-based group, and the proportion of LDL-C ≤ 1.4 mmol/L increased from 10.36% to 47.69% in PCSK-9 inhibitor group and 2.99% to 18.43% in statins-based group(P < 0.001 for both). There was no significant difference between PCSK-9 inhibitor and statins-based treatment in reducing the risk of MACE(hazard ratio = 2.52, 95% CI: 0.49-12.97, P = 0.250).CONCLUSIONS In the real world, PCSK-9 inhibitors combined with statins could significantly reduce LDL-C levels among patients with very high risk of ASCVD in China. 2Synthesis and in vitro Antimycobacterial Activity of Moxifloxacin Methylene and Ethylene Isatin Derivatives显示文摘A series of novel moxifloxacin methylene and ethylene isatin derivatives with remarkable improvement in lipophilicity, compared to the parent moxifloxacin, was designed, synthesized and characterized by 1H NMR, MS and HRMS. These derivatives were initially evaluated for their in vitro antimycobacterial activity against M. smegmatis CMCC 93202. Compounds 3a―3f, 5a, 5f and 5j were chosen for the further evaluation of their in vitro activity against Mycobacterium tuberculosis(MTB) H37Rv ATCC 27294 and MDR-MTB 09710. All the target com pounds[minimum inhibitory concentration(MIC): 0.39―>16 μg/mL] were far more active than rifampin(MIC: 2.0―>256 μg/mL), but less active than moxifloxacin(MIC: 0.1―1.0 μg/mL) against the three tested strains. 3Control of fibrosis and hypertrophic scar formation via glycolysis regulation with IR780显示文摘Background:Hypertrophic scars(HS)represent one of the most common clinical challenges due to unsatisfactory therapeutic results.HS formation is associated with the abnormal activation of fibroblasts and their excessive fibrotic behavior.Glycolysis dysregulation has been shown to participate in the incidence and progression of various fibrotic diseases and shows potential as a means of controlling HS formation.This work aimed to discuss the impact of augmented glycolysis on HS and to propose a method for controlling HS formation through glycolysis regulation.Methods:Here,augmented glycolysis was confirmed together with enhanced fibrotic activity in both HS fibroblasts(HFs)and HS tissues,and the suppression of glycolysis also attenuated fibroblast activation.We also introduced IR780,a heptamethine cyanine dye,to regulate glycolysis for the control of HS formation.Results:In vitro,cell studies indicated that IR780 significantly down-regulated glycolysis and suppressed the fibrotic activity of HFs.In vivo,the intralesional injection of IR780 into rabbit HS models led to the downregulation of glycolysis and the control of HS formation.Furthermore,IR780 accumulated preferentially in activated fibroblasts in both in vitro and in vivo studies,and thus specifically downregulated glycolysis and efficiently controlled fibrosis by targeting activated fibroblasts.Conclusions:This work identified a strategy for controlling fibrosis and HS formation from the perspective of glycolysis regulation with IR780 targeting of activated fibroblasts.Xinxian Meng Zhixi Yu Wanyu Xu Jun Chai Shuo Fang Peiru Min Yunsheng Chen Yixin Zhang Zheng Zhang 2022Burns & Trauma2022,10,1:2
4Structural variation and parallel evolution of apomixis in citrus during domestication and diversification显示文摘Apomixis,or asexual seed formation,is prevalent in Citrinae via a mechanism termed nucellar or adventitious embryony.Here,multiple embryos of a maternal genotype form directly from nucellar cells in the ovule and can outcompete the developing zygotic embryo as they utilize the sexually derived endosperm for growth.Whilst nucellar embryony enables the propagation of clonal plants of maternal genetic constitution,it is also a barrier to effective breeding through hybridization.To address the genetics and evolution of apomixis in Citrinae,a chromosome-level genome of the Hongkong kumquat(Fortunella hindsii)was assembled following a genome-wide variation map including structural variants(SVs)based on 234 Citrinae accessions.This map revealed that hybrid citrus cultivars shelter genome-wide deleterious mutations and SVs into heterozygous states free from recessive selection,which may explain the capability of nucellar embryony in most cultivars during Citrinae diversification.Analyses revealed that parallel evolution may explain the repeated origin of apomixis in different genera of Citrinae.Within Fortunella,we found that apomixis of some varieties originated via introgression.In apomictic Fortunella,the locus associated with apomixis contains the FhRWP gene,encoding an RWP-RK domain-containing protein previously shown to be required for nucellar embryogenesis in Citrus.We found the heterozygous SV in the FhRWP and CitRWP promoters from apomictic Citrus and Fortunella,due to either two or three miniature inverted transposon element(MITE)insertions.A transcription factor,FhARID,encoding an AT-rich interaction domain-containing protein binds to the MITEs in the promoter of apomictic varieties,which facilitates induction of nucellar embryogenesis.This study provides evolutionary genomic and molecular insights into apomixis in Citrinae and has potential ramifications for citrus breeding.Nan Wang Xietian Song Junli Ye Siqi Zhang Zhen Cao Chenqiao Zhu Jianbing Hu Yin Zhou Yue Huang Shuo Cao Zhongjie Liu Xiaomeng Wu Lijun Chai Wenwu Guo Qiang Xu Brandon S.Gaut Anna M.G.Koltunow Yongfeng Zhou Xiuxin Deng 2022National Science Review2022,9,10:1

6Effect of Low Temperature Treatment on Agrobacterium-mediated Transformation of Maize Seed Buds显示文摘[Objectives]This study was conducted to improve the genetic transformation rate of maize. [Methods]The seeds of maize ' Zheng 58' as the experimental material were germinated and treated under freezing temperature of-18 ℃ for 30 and 35 min. The EGFP gene was transformed into the growth points of these seed buds by Agrobacterium tumefaciens EHA105. The transformation effect was determined by fluorescence protein detection on transformed buds. [Results]After a certain period of treatment at-18 ℃ following germination and transformation of maize bud growth points with A. tumefaciens,although the survival rate of maize buds was reduced,the percentage of transformed cells significantly increased. 8Effects of Acetylacetone Solution Soaking on Agrobacterium - transformed Maize Seed Buds显示文摘[Objectives]The effect of acetosyringone seed soaking on the transformation of maize seed buds was analyzed,so as to improve the genetic transformation efficiency of maize and to provide technical support for transgenic breeding of maize.[Methods]The seeds of the'Zheng 58'maize inbred line were used as experimental materials.When the seeds were germinated,AS was added to the water at concentrations of 70,140,210,and 280μmol/L,respectively,and the seeds germinated without the addition of AS served as the CK.The Agrobacterium-mediated method was used to transform bud growth points of maize seeds,and green fluorescent protein detection was performed on the young shoots transformed with EGFP(enhanced green fluorescent protein)gene.The effect of soaking seeds with acetosyringone solution on the transformation of maize bud growth points by Agrobacterium was studied according to the detection results.[Results]Soaking seeds in acetosyringone solutions for germination had the effect of inhibiting the germination of maize seeds and inhibiting sprout elongation,and the higher the concentration of acetosyringone,the stronger the inhibition.When the concentration of acetosyringone solution was 280μmol/L,the germination rate of seeds was only 36.2%of the CK,while soaking seeds with 70-140μmol/L acetosyringone solution for germination could not only ensure a higher germination rate of maize seeds,but also significantly increased the transformation efficiency of maize bud growth points.When the seeds were soaked with 70μmol/L acetosyringone solution for germination,the positive rate of transformed maize buds was the highest,reaching 32.1%.[Conclusions]When maize bud growth points were used as the receptor of Agrobacterium transformation,soaking seeds with 70-140μmol/L acetosyringone for germination basically did not affect the germination of seeds,and was beneficial to the activation of Agrobacterium,thereby promoting the transformation.Mengyu LYU Fushuang DONG Shuo ZHOU Jianfang CHAI He ZHAO Yongwei LIU Fan YANG Bo JIAO Xiaoyu LI Junmin ZHANG Haibo WANG 2021Agricultural Biotechnology2021,10,3:0

10Dynamic evaluation of a scaled-down heat pipe-cooled system during start-up/shut-down processes using a hardware-in-the-loop test approach显示文摘Micro-mobile heat pipe-cooled nuclear power plants are promising candidates for distributed energy resource power genera-tors and can be flexibly deployed in remote places to meet increasing electric power demands.However,previous steady-state simulations and experiments have deviated significantly from actual micronuclear system operations.Hence,a transient analysis is required for performance optimization and safety assessment.In this study,a hardware-in-the-loop(HIL)approach was used to investigate the dynamic behavior of scaled-down heat pipe-cooled systems.The real-time features of the HIL architecture were interpreted and validated,and an optimal time step of 500 ms was selected for the thermal transient.The power transient was modeled using point kinetic equations,and a scaled-down thermal prototype was set up to avoid mod-eling unpredictable heat transfer behaviors and feeding temperature samples into the main program running on a desktop PC.A series of dynamic test results showed significant power and temperature oscillations during the transient process,owing to the inconsistency of the rapid nuclear reaction rate and large thermal inertia.The proposed HIL approach is stable and effective for further studying of the dynamic characteristics and control optimization of solid-state small nuclear-powered systems at an early prototyping stage.Jiao‑Long Deng Tian‑Shi Wang En‑Ping Zhu Shuo Yuan Xiao‑Jing Liu Xiang Chai 2023Nuclear Science and Techniques2023,34,11:0
11RNA interference targeting ω-secalin genes differentially affects the processing quality in a wheat T1BL·1RS translocation line显示文摘Wheat-rye T1BL·1RS translocation lines are widely used,especially in China,but their processing quality is generally poor.An interfering expression vector targeting theω-secalin genes was constructed with the 1Bx7 seed-specific promoter.Biolistic-mediated genetic transformation of the wheat cultivar KN199 carrying the T1BL·1RS translocation generated 10 transgenic lines.Two representative transgenic lines,8-2 and 13-7,were selected for analysis.Compared with the control,the two transformants showed an up to 4.5-fold decrease in totalω-secalins and various levels of decrease inω-gliadins,γ-gliadins,and low-molecular-weight glutenins.A decrease in high molecular weight(HMW)glutenin 1Bx7 was detected only in 8-2,owing possibly to promoter methylation.Increased levels ofα-gliadins were observed in both transformants,but increased levels of HMW glutenins were observed only in 13-7.Line 13-7 showed increases in gluten index,Zeleny sedimentation value,stabilization time,and maximum resistance.Its bread volume was 849.6 mL,an 11.9%increase over that of the control.Line 8-2 showed decreases in these parameters,but its total cake-making quality score was 88,an 17.3%increase over that of the control.The study demonstrates that the same RNAi construct may produce different effects on wheat processing quality and highlights the influence of the vector promoter in RNA interference.Shuo Zhou Cuimian Zhang He Zhao Mengyu Lyu Fushuang Dong Yongwei Liu Fan Yang Haibo Wang Jianfang Chai 2021The Crop Journal2021,9,2:0
12Capture efficiency and bias from the defect dynamics near grain boundaries in BCC Fe using mesoscale simulations显示文摘The capture efficiency describes the capability of a sink,such as a grain boundary(GB),dislocation,and void,to absorb point defects(PDs).The bias defines the difference in capture efficiency between the absorption of a vacancy and dumbbell at a sink.Complete kinetic information on PDs,including diffusion barriers and diffusion orientations,as well as accurate saddle points,are needed to determine the capture efficiency and bias at a sink accurately,which is computationally demanding.In the present study,the Self-Evolving Atomistic Kinetic Monte Carlo(SEAKMC)method was used to investigate the defect dynamics of PDs near different types of grain boundaries(GBs)(with both 100 and 110 families)accurately in body-centered cubic(BCC)iron(Fe).The capture efficiency,sink strength,and bias factor of different types of GBs were determined in Fe,which,different from traditional rate theory estimation,showed a distinct capture efficiency,sink strength,and bias in different GBs.The results demonstrate a strong positive correlation between the capture efficiency and the GB strain width,instead of the GB misorientation,GB energy,or GB-PD binding energy,which have been investigated previously.This work provides valuable insight into the radiation-induced microstructural evolution of GBs.Jun Chai Shuo Jin Ziang Yu Haixuan Xu Guang-Hong Lu 2021Journal of Materials Science & Technology2021,,34:0

14Study on Differentiation of Growing Points in Corn Sprouts into Multiple Stems显示文摘[Objectives] This study was conducted to improve the efficiency of genetic transformation using growing points of corn sprouts. [Methods] A mixed liquid of plant hormones 6-BA and KT at a suitable concentration and ratio was added dropwise to the growing points of corn sprouts, followed by culture under appropriate conditions. [Results] Corn sprouts could be induced to form multiple plants, thereby improving the transformation efficiency of growing points in corn sprouts. 15Copy number variation sequencing for diagnosis of cytomegalovirus infection based low-depth whole-genome sequencing technology in fetus:Three cases and literature review显示文摘Objective:To summarize the application value of copy number variant sequencing(CNV-seq)in the detection of fetal chromosome and cytomegalovirus load.Methods:The study analyzed the clinical basic data,relevant laboratory tests,treatment process,and outcomes of three patients with positive cytomegalovirus load detected by CNV-seq for fetal chromosomes and cytomegalovirus load,and literature review was done simutaneoubly.Results:In all three cases,the amniotic fluid cytomegalovirus load was less than 105 Copies/ml,and there were no significant neurological abnormalities observed during pregnancy or postpartum follow-up.There is no literature review on the application of CNV-seq technology in the detection of cytomegalovirus infection,only literature reports on genome analysis of CMV-DNA in confirmed patients were available.Conclusion:CNV-seq can be used to detect cytomegalovirus load,which may have a certain degree of predictive value for fetal outcome.CNV-seq can simultaneously detect fetal chromosomes and pathogenic microorganisms,which is of great significance for the prevention and control of birth defects.CHAI Shi-wei CHEN Ze-jun LIU Chun-tao CHEN Su HE Gui-lin CHEN Yue-fen WANG Rui-xia ZHU Xin LING Yi GU Shuo 2023Journal of Hainan Medical University2023,29,14:0
