2Bone biomaterials and interactions with stem cells显示文摘Bone biomaterials play a vital role in bone repair by providing the necessary substrate for cell adhesion,proliferation, and differentiation and by modulating cell activity and function. In past decades, extensive efforts have been devoted to developing bone biomaterials with a focus on the following issues:(1)developing ideal biomaterials with a combination of suitable biological and mechanical properties;(2)constructing a cell microenvironment with pores ranging in size from nanoscale to submicro-and microscale;and(3) inducing the oriented differentiation of stem cells for artificial-to-biological transformation. Here we present a comprehensive review of the state of the art of bone biomaterials and their interactions with stem cells. Typical bone biomaterials that have been developed, including bioactive ceramics, biodegradable polymers, and biodegradable metals, are reviewed, with an emphasis on their characteristics and applications. The necessary porous structure of bone biomaterials for the cell microenvironment is discussed,along with the corresponding fabrication methods. Additionally, the promising seed stem cells for bone repair are summarized, and their interaction mechanisms with bone biomaterials are discussed in detail.Special attention has been paid to the signaling pathways involved in the focal adhesion and osteogenic differentiation of stem cells on bone biomaterials. 3Accelerated degradation of HAP/PLLA bone scaffold by PGA blending facilitates bioactivity and osteoconductivity显示文摘The incorporation of hydroxyapatite(HAP)into poly-L-lactic acid(PLLA)matrix serving as bone scaffold is expected to exhibit bioactivity and osteoconductivity to those of the living bone.While too low degradation rate of HAP/PLLA scaffold hinders the activity because the embedded HAP in the PLLA matrix is difficult to contact and exchange ions with body fluid.In this study,biodegradable polymer poly(glycolic acid)(PGA)was blended into the HAP/PLLA scaffold fabricated by laser 3D printing to accelerate the degradation.The results indicated that the incorporation of PGA enhanced the degradation rate of scaffold as indicated by the weight loss increasing from 3.3%to 25.0%after immersion for 28 days,owing to the degradation of high hydrophilic PGA and the subsequent accelerated hydrolysis of PLLA chains.Moreover,a lot of pores produced by the degradation of the scaffold promoted the exposure of HAP from the matrix,which not only activated the deposition of bone like apatite on scaffold but also accelerated apatite growth.Cytocompatibility tests exhibited a good osteoblast adhesion,spreading and proliferation,suggesting the scaffold provided a suitable environment for cell cultivation.Furthermore,the scaffold displayed excellent bone defect repair capacity with the formation of abundant new bone tissue and blood vessel tissue,and both ends of defect region were bridged after 8 weeks of implantation.Cijun Shuai Wenjing Yang Pei Feng Shuping Peng Hao Pan 2021Bioactive Materials2021,6,2:8
4Physical stimulations and their osteogenesis-inducing mechanisms显示文摘Physical stimulations such as magnetic,electric and mechanical stimulation could enhance cell activity and promote bone formation in bone repair process via activating signal pathways,modulating ion channels,regulating bonerelated gene expressions,etc.In this paper,bioeffects of physical stimulations on cell activity,tissue growth and bone healing were systematically summarized,which especially focused on their osteogenesis-inducing mechanisms.Detailedly,magnetic stimulation could produce Hall effect which improved the permeability of cell membrane and promoted the migration of ions,especially accelerating the extracellular calcium ions to pass through cell membrane.Electric stimulation could induce inverse piezoelectric effect which generated electric signals,accordingly up-regulating intracellular calcium levels and growth factor synthesis.And mechanical stimulation could produce mechanical signals which were converted into corresponding biochemical signals,thus activating various signaling pathways on cell membrane and inducing a series of gene expressions.Besides,bioeffects of physical stimulations combined with bone scaffolds which fabricated using 3D printing technology on bone cells were discussed.The equipments of physical stimulation system were described.The opportunities and challenges of physical stimulations were also presented from the perspective of bone repair.Cijun Shuai Wenjing Yang Shuping Peng Chengde Gao Wang Guo Yuxiao Lai Pei Feng 2018International Journal of Bioprinting2018,4,2:3

8Construction and Characterization of a Novel Recombinant Attenuated and Replication-Deficient Candidate Human Adenovirus Type 3 Vaccine:'Adenovirus Vaccine Within an Adenovirus Vector'显示文摘Human adenoviruses(HAd Vs)are highly contagious and result in large number of acute respiratory disease(ARD)cases with severe morbidity and mortality.Human adenovirus type 3(HAd V-3)is the most common type that causes ARD outbreaks in Asia,Europe,and the Americas.However,there is currently no vaccine approved for its general use.The hexon protein contains the main neutralizing epitopes,provoking strong and lasting immunogenicity.In this study,a novel recombinant and attenuated adenovirus vaccine candidate against HAd V-3 was constructed based on a commercially-available replication-defective HAd V-5 gene therapy and vaccine vector.The entire HAd V-3 hexon gene was integrated into the E1 region of the vector by homologous recombination using a bacterial system.The resultant recombinants expressing the HAd V-3 hexon protein were rescued in AD293 cells,identified and characterized by RT-PCR,Western blots,indirect immunofluorescence,and electron microscopy.This potential vaccine candidate had a similar replicative efficacy as the wild-type HAd V-3 strain.However,and importantly,the vaccine strain had been rendered replication-defective and was incapable of replication in A549 cells after more than twentygeneration passages in AD293 cells.This represents a significant safety feature.The mice immunized both intranasally and intramuscularly by this vaccine candidate raised significant neutralizing antibodies against HAd V-3.Therefore,this recombinant,attenuated,and safe adenovirus vaccine is a promising HAd V-3 vaccine candidate.The strategy of using a clinically approved and replication-defective HAd V-5 vector provides a novel approach to develop universal adenovirus vaccine candidates against all the other types of adenoviruses causing ARDs and perhaps other adenovirus-associated diseases.Yuqian Yan Shuping Jing Liqiang Feng Jing Zhang Zhiwei Zeng Min Li Shan Zhao Junxian Ou Wendong Lan Wenyi Guan Xiaowei Wu Jianguo Wu Donald Seto Qiwei Zhang 2021Virologica Sinica2021,36,3:1

11Robust Centralized CFAR Detection for Multistatic Sonar Systems显示文摘This paper proposes a novel centralized Constant false alarm ratio(CFAR) detector for multistatic sonar systems. The detector employs the idea of Variability index(VI) CFAR detection, to adaptively select the matched detection algorithm in diversified undersea environments. All the echo data from mutistatic sonar receivers are transmitted into the centralized fusion center. Firstly, the background statistics of reference cells from different nodes are analyzed. Then choose one appropriate centralized detection algorithm according to the background statistics, which refers to the centralized Cell averaging CFAR(CA-CFAR), greatest of CFAR, order statistic CFAR detection algorithms.The performance of the proposed detector is analyzed by computer simulation and measured sonar data. 17A multi-scale porous scaffold fabricated by a combined additive manufacturing and chemical etching process for bone tissue engineering显示文摘It is critical to develop a fabrication technology for precisely controlling an interconnected porous structure of scaffolds to mimic the native bone microenvironment.In this work,a novel combined process of additive manufacturing(AM)and chemical etching was developed to fabricate graphene oxide/poly(L-lactic acid)(GO/PLLA)scaffolds with multiscale porous structure.Specially,AM was used to fabricate an interconnected porous network with pore sizes of hundreds of microns.And the chemical etching in sodium hydroxide solution constructed pores with several microns or even smaller on scaffolds surface.The degradation period of the scaffolds was adjustable via controlling the size and quantity of pores.Moreover,the scaffolds exhibited surprising bioactivity after chemical etching,which was ascribed to the formed polar groups on scaffolds surfaces.Furthermore,GO improved the mechanical strength of the scaffolds.Cijun Shuai Youwen Yang Pei Feng Shuping Peng Wang Guo Anjie Min Chengde Gao 2018International Journal of Bioprinting2018,4,2:1
