Clinical significance of extended high-risk human papillomavirus genotyping and viral load in cervical cancer and precancerous lesions显示文摘Persistent infections with specific high-risk human papillomavirus(HR-HPV)strains are the leading cause of cervical cancer and precancerous lesions.HPV-16 and HPV-18 are associated with more than 70%of cervical cancer. However, with recent widespread vaccination efforts against cervical cancer, the infection rates of HPV-16 and HPV-18 have decreased across all age groups, while the infection rates of other HR-HPV strains have increased. The non-16/18 HR-HPV strains play an important role in cervical lesions. These strains can be identified with extended genotyping, and the 2019 American Society for Colposcopy and Cervical Pathology(ASCCP)guidelines recommended an HPV-based testing to assess the risk of cervical disease in patients. We reviewed and analyzed the clinical benefits of applying extended HR-HPV genotyping, which was published by the International Agency for Research on Cancer(HPV-16,18,31,33,35,39,45,51,52,56,58,59,66,and 68),to cervical cancer screening. This review concluded that cervical cancer screening needs to include extended HR-HPV genotyping. The examination of extended HR-HPV genotyping in cervical intraepithelial lesions and cervical cancers can help guide clinical practices. | Pingping Su Jincheng Ma Lirui Yu Shuting Tang Pengming Sun | 2023 | Gynecology and Obstetrics Clinical Medicine2023,3,1: | 2
Genomic evidence of lung carcinogenesis associated with coal smoke in Xuanwei area, China显示文摘Xuanwei area, in southwestern China, harbors the highest female lung cancer rate in the country(Supplementary Table 1) [1]. Epidemiological studies have shown that lung cancer incidence in Xuanwei area is associated with the use of different types of local coal for household cooking and heating [2]. The genomic landscape of Xuanwei female adenocarcinoma(XWFA), the most distinctive feature of lung cancer in Xuanwei, has yet to be elucidated systematically. Here,we provide the fundamental resource of genomic datasets for further exploration of XWFA molecular mechanisms and development of targetable therapy. | Honglei Zhang Chao Liu Li Li Xu Feng Qing Wang Jihua Li Shaobin Xu Shuting Wang Qianlu Yang Zhenghai Shen Jinhua Su Xiaosan Su Ruifen Sun Xuhong Zhou Junliang Wang Yongchun Zhou Baowei Jiao Wanbao Ding Xianbao Cao Yue Wang Yunchao Huang Lianhua Ye | 2021 | National Science Review2021,8,12: | 2
Vagal-mAChR4 signaling promotes Friend virus complex(FV)-induced acute erythroleukemia显示文摘Erythroleukemia belongs to acute myeloid leukemia(AML)type 6(M6),and treatment remains difficult due to the poor prognosis of the disease.Friend virus(FV)is a complex of two viruses:Friend murine leukemia virus(F-MuLV)strain along with a defective spleen focus-forming virus(SFFV),which can induce acute eryth-roleukemia in mice.We have previously reported that activation of vagalα7 nicotinic acetylcholine receptor(nAChR)signaling promotes HIV-1 transcription.Whether vagal muscarinic signaling mediates FV-induced erythroleukemia and the underlying mechanisms remain unclear. In this study, sham and vagotomized mice were intraperitoneally injected with FV.FV infection caused anemia in sham mice, and vagotomy reversed this change.FV infection increased erythroblasts ProE,EryA,and EryB cells in the spleen, and these changes were blocked by vagotomy. In bone marrow,FV infection reduced EryC cells in sham mice,an effect that was coun-teracted by vagotomy.FV infection increased choline acetyltransferase(ChAT)expression in splenic CD4^(+)and CD8þT cells, and this change was reversed by vagotomy.Furthermore, the increase of EryA and EryB cells in spleen of FV-infected wild-type mice was reversed after deletion of ChAT in CD4^(+)T cells. In bone marrow,FV infection reduced EryB and EryC cells in sham mice, whereas lack of ChAT in CD4^(+)T cells did not affect this change.Activation of muscarinic acetylcholine receptor 4(mAChR4)by clozapine N-oxide(CNO)significantly increased EryB in the spleen but decreased the EryC cell population in the bone marrow of FV-infected mice.Thus,vagal-mAChR4 signaling in the spleen and bone marrow synergistically promotes the pathogenesis of acute erythroleukemia. We uncover an unrecognized mechanism of neuromodulation in erythroleukemia. | Shuting Song Zhekai Lin Caiqi Zhao Jing Wen Jie Chen Shitao Xie Huaxin Qi Jianhua Wang Xiao Su | 2023 | Virologica Sinica2023,38,3: | 0
De novo transcriptome assembly and metabolomic analysis of three tissue types in Cinnamomum cassia显示文摘Objective:The barks, leaves, and branches of Cinnamomum cassia have been historically used as a traditional Chinese medicine, spice, and food preservative,in which phenylpropanoids are responsible compounds. However phenylpropanoid biosynthesis pathways are not clear in C.cassia.We elucidated the pathways by descriptive analyses of differentially expressed genes related to phenylpropanoid biosynthesis as well as to identify various phenylpropanoid metabolites. Methods:Chemical analysis, metabolome sequencing, and transcriptome sequencing were performed to investigate the molecular mechanisms underlying the difference of active components content in the barks, branches and leaves of C.cassia.Results:Metabolomic analysis revealed that small amounts of flavonoids, coumarine, and cinnamaldehyde accumulated in both leaves and branches.Transcriptome analysis showed that genes associated with phenylpropanoid and flavonoid biosynthesis were downregulated in the leaves and branches relative to the barks. The observed differences in essential oil content among the three tissues may be attributable to the differential expression of genes involved in the phenylpropanoid and flavonoid metabolic pathways. Conclusion:This study identified the key genes in the phenylpropanoid pathway controling the flavonoid, coumarine, and cinnamaldehyde contents in the barks, branches and leaves by comparing the transcriptome and metabolome. These findings may be valuable in assessing phenylpropanoid and flavonoid metabolites and identifying specific candidate genes that are related to the synthesis of phenylpropanoids and flavonoids in C.cassia. | Hongyang Gao Huiju Zhang Yuqing Hu Danyun Xu Sikai Zheng Shuting Su Quan Yang | 2023 | Chinese Herbal Medicines2023,15,2: | 0
Effect of copper tolerant Elsholtzia splendens on bacterial community associated with Commelina communis on a copper mine spoil显示文摘Facilitation, or positive plant–plant interaction, has received increasing concern from ecologists over the last two decades. Facilitation may occur through direct mitigation of severe environments or indirect mediation by a third participant from the same or different trophic levels. The copper(Cu) tolerant species Elsholtzia splendens facilitates the establishment and growth of co-occurring Commelina communis through indirect enrichment of microbial activity. However, whether and how E. splendens impacts the microbial community that is associated with C. communis is less known. We characterized the soil bacterial community in the rhizosphere of C. communis in the absence and presence of E.splendens using PCR-DGGE(polymerase chain reaction-denaturing gradient gel electrophoresis) and sequencing. The result showed that the richness of the bacterial community increased, but diversity and evenness remained similar, in the presence of E. splendens.Chloroflexi, Acidobacteria and Proteobacteria were the most dominant bacteria. The relative abundance of dominant and minor bacterial groups showed distinctly different responses to E. splendens. Principal component analysis and redundancy analysis indicated that variation of the bacterial community was determined by multiple factors and might be driven by the tested soil parameters collectively, or alternatively changed through plant root exudates or other microorganisms. Our results enhance the understanding of how the bacterial community associated with a beneficiary plant responds to a benefactor plant and suggests that the changes of bacterial community composition may have far-reaching influence on plant–soil feedback and the aboveground plant community in the long run. | Ruyi Yang Fuyu Guo Jing Li Nannan Su Zongyuan Shao Shuting Zan | 2016 | Journal of Environmental Sciences2016,28,8: | 0
