| 1 | Probiotic metabolites from Bacillus coagulans GanedenBC30^(TM) support maturation of antigen-presenting cells in vitro显示文摘AIM:To study the effects of probiotic metabolites on maturation stage of antigen-presenting immune cells.METHODS:Ganeden Bacillus coagulans 30(GBC30) bacterial cultures in log phase were used to isolate the secreted metabolite(MET) fraction.A second fraction was made to generate a crude cell-wall-enriched fraction,by centrifugation and lysis,followed by washing.A preparation of MET was subjected to size exclusion centrifugation,generating three fractions:< 3 kDa,3-30 kDa,and 30-200 kDa and activities were tested in comparison to crude MET and cell wall in primary cultures of human peripheral blood mononuclear cell(PBMC) as a source of antigen-presenting mononuclear phagocytes.The maturation status of mononuclear phagocytes was evaluated by staining with monoclonal antibodies towards CD14,CD16,CD80 and CD86 and analyzed by flow cytometry.RESULTS:Treatment of PBMC with MET supported maturation of mononuclear phagocytes toward both macrophage and dendritic cell phenotypes.The biological activity unique to the metabolites included a reduction of CD14+ CD16+ pro-inflammatory cells,and this property was associated with the high molecular weight metabolite fraction.Changes were also seen for the dendritic cell maturation markers CD80 and CD86.On CD14dim cells,an increase in both CD80 and CD86 expression was seen,in contrast to a selective increase in CD86 expression on CD14bright cells.The co-expression of CD80 and CD86 indicates effective antigen presentation to T cells and support of T helper cell differentiation.The selective expression of CD86 in the absence of CD80 points to a role in generating T regulatory cells.CONCLUSION:The data show that a primary mechanism of action of GBC30 metabolites involves support of more mature phenotypes of antigen-presenting cells,important for immunological decision-making. | Kathleen F Benson Kimberlee A Redman Steve G Carter David Keller Sean Farmer John R Endres Gitte S Jensen | 2012 | World Journal of Gastroenterology2012，18，16： | 8 |

| 3 | Air quality monitoring using mobile microscopy and machine learning显示文摘Rapid,accurate and high-throughput sizing and quantification of particulate matter(PM)in air is crucial for monitoring and improving air quality.In fact,particles in air with a diameter of≤2.5μm have been classified as carcinogenic by the World Health Organization.Here we present a field-portable cost-effective platform for high-throughput quantification of particulate matter using computational lens-free microscopy and machine-learning.This platform,termed c-Air,is also integrated with a smartphone application for device control and display of results.This mobile device rapidly screens 6.5 L of air in 30 s and generates microscopic images of the aerosols in air.It provides statistics of the particle size and density distribution with a sizing accuracy of~93%.We tested this mobile platform by measuring the air quality at different indoor and outdoor environments and measurement times,and compared our results to those of an Environmental Protection Agency–approved device based on beta-attenuation monitoring,which showed strong correlation to c-Air measurements.Furthermore,we used c-Air to map the air quality around Los Angeles International Airport(LAX)over 24 h to confirm that the impact of LAX on increased PM concentration was present even at 47 km away from the airport,especially along the direction of landing flights.With its machinelearning-based computational microscopy interface,c-Air can be adaptively tailored to detect specific particles in air,for example,various types of pollen and mold and provide a cost-effective mobile solution for highly accurate and distributed sensing of air quality. | Yi-Chen Wu Ashutosh Shiledar Yi-Cheng Li Jeffrey Wong Steve Feng Xuan Chen Christine Chen Kevin Jin Saba Janamian Zhe Yang Zachary Scott Ballard Zoltán Göröcs Alborz Feizi Aydogan Ozcan | 2017 | Light(Science & Applications)2017，6，1： | 5 |

| 5 | Sex-specific differences in white matter microvascular integrity after ischaemic stroke显示文摘background and purpose Functional outcomes after ischaemic stroke are worse in women,despite adjusting for differences in comorbidities and treatment approaches.White matter microvascular integrity represents one risk factor for poor long-term functional outcomes after ischaemic stroke.The aim of the study is to characterise sex-specific differences in microvascular integrity in individuals with acute ischaemic stroke.Methods A retrospective analysis of subjects with acute ischaemic stroke and brain MRI with diffusion-weighted(DWI)and dynamic-susceptibility contrast-enhanced(DSC)perfusion-weighted imaging obtained within 9 hours of last known well was performed.In the hemisphere contralateral to the acute infarct,normal-appearing white matter(NAWM)microvascular integrity was measured using the K_(2) coefficient and apparent diffusion coefficient(ADC)values.Regression analyses for predictors of K_(2) coefficient,DWI volume and good outcome(90-day modified Rankin scale(mRS)score<2)were performed.results 105 men and 79 women met inclusion criteria for analysis.Despite no difference in age,women had increased NAWM K_(2) coefficient(1027.4 vs 692.7×10^(-6)/s;p=0.006).In women,atrial fibrillation(β=583.6;p=0.04)and increasing NAWM ADC(β=4.4;p=0.02)were associated with increased NAWM K_(2) coefficient.In multivariable regression analysis,the K_(2) coefficient was an independent predictor of DWI volume in women(β=0.007;p=0.01)but not men.Conclusions In women with acute ischaemic stroke,increased NAWM K_(2) coefficient is associated with increased infarct volume and chronic white matter structural integrity.Prospective studies investigating sex-specific differences in white matter microvascular integrity are needed. | Mark R Etherton Ona Wu Pedro Cougo Svetlana Lorenzano Hua Li Lisa Cloonan Mark J R J Bouts Arne Lauer Ken Arai Eng H Lo Steve K Feske Karen L Furie Natalia S Rost | 2019 | Stroke & Vascular Neurology2019，4，4： | 2 |
