1Anti-inflammatory and Antimicrobial Effects of Anthocyanin Extracted from Black Soybean on Chronic Bacterial Prostatitis Rat Model显示文摘Objective: To investigated the anti-inflammatory and antimicrobial effects of anthocyanins extracted from black soybean on the chronic bacterial prostatitis(CBP) rat model.Methods: The SpragueDawley rats were divided into 4 groups,including control,ciprofloxacin,anthocyanins and anthocyanins with ciprofloxacin groups(n=8 in each group).Then,drip infusion of bacterial suspension(Escherichia coli Z17O_2:K_1:H^-) into Sprague-Dawley rats was conducted to induce CBP.In 4 weeks,results of prostate tissue,urine culture,and histological analysis on the prostate were analyzed for each group.Results: The use of ciprofloxacin,anthocyanins,and anthocyanins with ciprofloxacin showed statistically significant decreases in bacterial growth and improvements in the reduction of prostatic inflammation compared with the control group(P<0.05).The anthocyanins with ciprofloxacin group showed a statistically significant decrease in bacterial growth and improvement in prostatic inflammation compared with the ciprofloxacin group(P<0.05).Conclusions: These results suggest that anthocyanins may have anti-inflammatory and antimicrobial effects,as well as a synergistic effect with ciprofloxacin.Therefore,we suggest that the combination of anthocyanins and ciprofloxacin may be effective in treating CBP to obtain a higher rate of treatment success.Byung Il Yoon Woong Jin Bae Yong Sun Choi Su Jin Kim U Syn Ha Sung-Hoo Hong Dong Wan Sohn Sae Woong Kim 2018Chinese Journal of Integrative Medicine2018,24,8:11
2The association between the genetic polymorphisms of LMP2/LMP7 and the outcomes of HCV infection among drug users显示文摘Objective:To investigate a possible association of LMP2/LMP7 genes with chronic hepatitis C virus(HCV) infection,and to assess whether LMP2/LMP7 genes could influence the outcomes of HCV infection among drug users.Methods:Genomic DNAs of 362 anti-HCV sero-positive drug users and 225 control drug users were extracted from the peripheral blood leukocytes.The sero-positive patients were divided into those who had persistent infection and those who had spontaneously cleared the infection.Polymorphisms of LMP genes were determined by PCR combined with restriction fragment length polymorphism(RFLP).Results:The distribution of LMP2 genotypes among the control,persistent infection and spontaneous clearance groups were not different.However,the LMP7 codon 145 Gln/Lys,Lys/Lys,and Gln/Lys+Lys/Lys genotypes were found significantly more frequent in the persistent infection group than in control group(OR=1.75,95%CI=1.06~2.90;OR=3.16,95%CI=1.23-8.12;OR=1.94,95%CI=1.21-3.12,respectively).Similarly,the frequencies of the codon 145 Gln/Lys,Lys/Lys,and Gln/Lys+Lys/Lys genotypes were found significantly more frequent in the persistent infection group than in the spontaneous clearance group(OR=1.64,95%CI=1.04-2.57;OR=2.40,95%CI=1.09-5.28;OR=1.76,95%CI=1.152.69,respectively).Stratified analysis indicated that combined genotype Gln/Lys+Lys/Lys of the LMP7 gene was related to an increasing susceptibility to HCV infection(OR=1.91,95%CI=1.02-3.55;OR=2.19,95%CI=1.243.89;OR=1.91,95%CI=1.05-3.48,OR=2.86,95%CI=1.41-5.78,respectively)and the risk of persistent HCV infection(OR=1.94,95%CI=1.12-3.34;OR=2.02,95%CI=1.21-3.38;OR=1.78,95%CI=1.12-2.85,OR=2.23,95%CI=1.09-4.58,respectively)among>30-year-old,males,the injection drug user(IDU)subjects and/or the shorter duration drug users(≤5 y).Conclusion:These results suggest that polymorphism of the LMP7 gene may have an influence on the outcomes of HCV infection,and is one of the factors accounting for the genetic susceptibility to HCV infection among drug users.Qian Cui Yongxiang Zhang Jing Su Chao Shi Na Lei Keqin Ding Jun Li Rongbin Yu L u Wang Ning Wang 2010The Journal of Biomedical Research2010,24,5:6

9Suppression of Oxidative Stress of Modified Gongjin-Dan (WSY^1075) in Detrusor Underactivity Rat Model Bladder Outlet Induced by Obstruction显示文摘Objective: To investigate the anti-oxidative stress and preventive effect of modified Gongjin-dan(WSY-1075) in a detrusor underactivity rat model. Methods: Rats were randomly allocated to three groups: shamoperated(control), bladder outlet obstruction-induced detrusor underactivity(BOO-DU), and BOO-DU with WSY-1075(WSY) groups. WSY-1075 was oral y administrated to rats 200 mg daily for 2 weeks prior to the operation and 4 weeks after the operation. Bladder outlet obstruction was surgical y induced in rats by ligation around the urethra avoiding total obstruction. Cystometrography was conducted on rats in each group for examination of bladders. Results: Compared with the control group, bladder outlet obstruction led to a significant increase in oxidative stress with consequent changes to molecular composition, and decrease in maximal detrusor pressure(P<0.05). WSY-1075 treatment significantly suppressed oxidative stress and prevented degenerative and dysfunctional changes in bladder, as compared with BOO-DU group(P<0.05). 19N^(6)-methyladenosine methyltransferase plays a role in hypoxic preconditioning partially through the interaction with lncRNA H19显示文摘N^(6)-methyladenosine(m6A),a m ethylation in the N6 position of adenosine especially in the mRNA,exerts diverse physiological and pathological functions.However,the precise role of m 6A methylation in hypoxic preconditioning(HPC)is still unknown.Here,we observed that HPC treatm ent protected H9c2 cells against H2O2-induced injury,upregulated the m 6A level in the total RNA and the expression of methyltransferase like 3(METTL3),methyltransferase like 14(METTL14),and long noncoding RNA(lncRNA)H19.Either knockdown of METTL3 or METTL14 notably reversed the HPC-induced enhancement of cell viability,anti-apoptosis ability,and H19 expression.M ethylated RNA im m unoprecipitation(IP)indicated that knockdown of METTL3 or METTL14 decreased m 6A level in the lncRNA H19.Gain-of-function assay dem onstrated that H19 overexpression could partially rescue the decreased protection mediated by METTL3 or METTL14 knockdown in HPCtreated H9c2 cells.RNA binding protein im m unoprecipitation(RIP)assay showed that METTL3 and METTL14 could directly bind w ith H19.Our study identified a novel pattern of posttranscriptional regulation in HPC treatm ent.Since METTL3,METTL14,and lncRNA H19 were involved in HPC protection,they could be considered as potential biomarkers and therapeutic targets in HPC-derived cardiac rehabilitation and therapeutic approaches.Yamin Su Rongfeng X u Rui Zhang Yangyang Qu Wenjie Zuo Zhenjun Ji Haihua Geng Min Pan Genshan Ma 2020Acta Biochimica et Biophysica Sinica2020,52,12:1
