1A targeted sequencing approach to find novel pathogenic genes associated with sporadic aortic dissection显示文摘Aortic dissection(AD) is a heterogeneous genetic disease of the aorta with high mortality and poor prognosis. However, only few genetic causes of AD have been explored till date. After conducting a broad literature review focused on identifying potential pathogenic pathways, we designed a panel containing 152 AD-associated genes to conduct massively parallel targeted nextgeneration sequencing of 702 sporadic aortic dissection patients and 163 matched healthy controls. After validation by Sanger sequencing, we identified 21 definitely pathogenic and 635 likely pathogenic variants in 61.25%(430/702) of patients. In these patients, 34.88%(150/430) harbored more than one variant that was either definitely or likely to be pathogenic. Among the candidate genes, we identified 546 likely pathogenic variants in 47.72%(335/702) of patients. Importantly, we identified 94 lossof-function(LOF) variants in 45 genes in AD patients, but only five LOF variants in the controls(P=1.34×10^(-4)). With a burden test, we highlighted RNF213 as an important new gene for AD pathogenesis. We also performed transcriptome sequencing of human aorta tissues to evaluate the expression levels of these newly identified genes. Our study has compiled a comprehensive genetic map of sporadic AD in the Han Chinese population. 2Variants of genes encoding collagens and matrix metalloproteinase system increased the risk of aortic dissection显示文摘Aortic dissection(AD) is a devastating,heterogeneous condition of aorta.The homeostasis between collagens and matrix metalloproteases(MMPs)/tissue inhibitors of MMPs(TIMPs) system in the extracellular matrix plays an important role for structure and functions of aorta.However,our knowledge on association between variants of genes in this system and pathogenesis of AD is very limited.We analyzed all yet known coding human genes of collagens(45 genes),MMPs/TIMPs(27genes) in 702 sporadic AD patients and in 163 matched healthy controls,by using massively targeted next-generation and Sanger sequencing.To define the pathogenesis of potential disease-causing candidate genes,we performed transcriptome sequencing and pedigree co-segregation analysis in some genes and generated Col5a2 knockout rats.We identified 257 pathogenic or likely pathogenic variants which involved 88.89%(64/72) genes in collagens-MMPs/TIMPs system and accounted for 31.05%(218/702) sporadic AD patients.In them,84.86%patients(185/218) carried one variant,12.84%two variants and 2.30%more than two variants.Importantly,we identified 52 novel probably pathogenic loss-of-function(LOF) variants(20 nonsense,16 frameshift,14 splice sites,one stop-loss,one initiation codon) in 11.06%(50/452) AD patients,which were absent in 163controls(P=2.5×10^(-5)).Transcriptome sequencing revealed that identified variants induced dyshomeostasis in expression of collagens-TIMPs/MMPs systems.The Col5α2^(-/-) rats manifested growth retardation and aortic dysplasia.Our study provides a first comprehensive map of genetic alterations in collagens-MMPs/TIMPs system in sporadic AD patients and suggests that variants of these genes contribute largely to AD pathogenesis.Zongzhe Li Chengming Zhou Lun Tan Peng Chen Yanyan Cao Chenze Li Xianqing Li Jiangtao Yan Hesong Zeng Dao-Wu Wang Dao-Wen Wang 2017Science China(Life Sciences)2017,60,1:4
3Calibration of the solar radio spectrometer显示文摘This paper shows some improvements and new results of calibration of Chinese solar radio spectrometer by analyzing the daily calibration data recorded in the period of 1997-2007. First, the calibration coefficient is fitted for three bands (1.0-2.0 GHz, 2.6-3.8 GHz, 5.2-7.6 GHz) of the spectrometer by using the moving-average method confined by the property of the daily calibration data. By this calibration coefficient, the standard deviation of the calibration result was less than 10 sfu for 95% frequencies of 2.6-3.8 GHz band in 2003. This result is better than that calibrated with the constant coefficient. Second, the calibration coefficient is found in good correlation with local air temperature for most frequencies of 2.6-3.8 GHz band. 9Immediate and legacy effects of snow exclusion on soil fungal diversity and community composition显示文摘Background:Soil fungi play crucial roles in ecosystem functions.However,how snow cover change associated with winter warming affects soil fungal communities remains unclear in the Tibetan forest.Methods:We conducted a snow manipulation experiment to explore immediate and legacy effects of snow exclusion on soil fungal community diversity and composition in a spruce forest on the eastern Tibetan Plateau.Soil fungal communities were performed by the high throughput sequencing of gene-fragments.Results:Ascomycota and Basidiomycota were the two dominant fungal phyla and Archaeorhizomyces,Aspergillus and Amanita were the three most common genera across seasons and snow manipulations.Snow exclusion did not affect the diversity and structure of soil fungal community in both snow-covered and snow-free seasons.However,the relative abundance of some fungal communities was different among seasons.Soil fungal groups were correlated with environmental factors(i.e.,temperature and moisture)and soil biochemical variables(i.e.,ammonium and enzyme).Conclusions:These results suggest that the season-driven variations had stronger impacts on soil fungal community than short-term snow cover change.Such findings may have important implications for soil microbial processes in Tibetan forests experiencing significant decreases in snowfall.Li Zhang Yuzhi Ren Kaijun Yang Zhijie Li Bo Tan Yang Liu Han Li Chengming You Sining Liu Lixia Wang Rui Yin Jian Zhang Zhenfeng Xu 2021Forest Ecosystems2021,8,2:1

11Blasting Technology Research in Construction of Fenghuoshan Tunnel in Permafrost显示文摘The Fenghuoshan Tunnel is an important project in Qinghai-Tibet Railway. How to protect permafrost during tunnelling is one of key technology. There are two kinds of influences on permafrost because of the blasting construction, firstly, blasting results in the damage of the surrounding rock, producing cranny in surrounding rock or making existing cranny spreading even connecting, secondly, the explosion produces gas with high temperature and high pressure, tremendous heat energy influences the heat balance of permafrost. To minimize the effect on permafrost during explosion construction, this paper has done research for relevant explosion scheme based on the theoretic analysis and spot tests. Pre-crack explosion is adopted on the hollow segment, and smooth explosion is adopted on the tunnel body segment, whole section excavation is adopted on the tunnel body segment with grade Ⅳ surrounding rock, and the face steps excavation is adopted on the tunnel body segment with grade Ⅴ surrounding rock. Explosion parameters were optimised finally. The maximal explosion vibration velocity in the surrounding rock is controlled in 5~10 cm/s. To solve the heat disturbance problem, brine mud is adopted to block up explosion holes in order to coll and dustproof. 12Nitrogen addition mediates the response of foliar stoichiometry to phosphorus addition: a meta-analysis显示文摘Background:Changes in foliar nitrogen(N)and phosphorus(P)stoichiometry play important roles in predicting the efects of global change on ecosystem structure and function.However,there is substantial debate on the efects of P addition on foliar N and P stoichiometry,particularly under diferent levels of N addition.Thus,we conducted a global meta-analysis to investigate how N addition alters the efects of P addition on foliar N and P stoichiometry across different rates and durations of P addition and plant growth types based on more than 1150 observations.Results:We found that P addition without N addition increased foliar N concentrations,whereas P addition with N addition had no efect.The positive efects of P addition on foliar P concentrations were greater without N addition than with N addition.Additionally,the efects of P addition on foliar N,P and N:P ratios varied with the rate and duration of P addition.In particular,short-term or low-dose P addition with and without N addition increased foliar N concentration,and the positive efects of short-term or low-dose P addition on foliar P concentrations were greater without N addition than with N addition.The responses of foliar N and P stoichiometry of evergreen plants to P addition were greater without N addition than with N addition.Moreover,regardless of N addition,soil P availability was more efective than P resorption efciency in predicting the changes in foliar N and P stoichiometry in response to P addition.Conclusions:Our results highlight that increasing N deposition might alter the response of foliar N and P stoichiometry to P addition and demonstrate the important efect of the experimental environment on the results.These results advance our understanding of the response of plant nutrient use efciency to P addition with increasing N deposition.Chengming You Changhui Peng Zhenfeng Xu Yang Liu Li Zhang Rui Yin Lin Liu Han Li Lixia Wang Sining Liu Bo Tan Paul Kardol 2021Ecological Processes2021,10,1:0
13Do root modules still exist after they die?显示文摘Background:The terminal branch orders of plant root systems are increasingly known as an ephemeral module.This concept is crucial to recognize belowground processes.However,it is unknown if root modules still exist after they die?Methods:The decomposition patterns of the first five root orders were observed for 3 years using a branch-order classification,a litter-bag method and sequential sampling in a common subalpine tree species(Picea asperata)of southwestern China.Results:Two root modules were observed during the 3-year incubation.Among the first five branch orders,the first three order roots exhibited temporal patterns of mass loss,nutrients and stoichiometry distinct from their woody mother roots throughout the experimental period.This study,for the first time,reported the decomposition pattern of each individual root order and found a similar decomposition dynamic among ephemeral root branches in a forest tree species.Conclusions:Results from this study suggest that root modules may also exist after death,while more data are needed for confirmation.The findings may further advance our understanding of architecture-associated functional heterogeneity in the fine-root system and also improve our ability to predict belowground processes.Jihong Li Chengming You Li Zhang Han Li Bo Tan Yang Liu Lixia Wang Sining Liu Zhenfeng Xu 2021Forest Ecosystems2021,8,2:0
14Degree of Humification in Fresh Litter from a Subtropical Mixed Plantation Forest in Southwest China显示文摘The humification degree of fresh litter directly controls the accumulation of soil humus derived from plant litter,but very little information on this process is available.Planted forests are well known to restrict soil fertility,which is often indicated by the soil humus level.In this study,fresh litter was collected during different plant phenological stages during 2016 and 2017 in a mixed plantation in Southwest China.The values of hue coefficientΔlogK(absorbance ratio of 400 nm and 600 nm on a logarithmic scale),optical density E4/E6(absorbance ratio of 400 nm and 600 nm)and A600/C(absorbance at 600 mm per mg of carbon per ml of extraction)and the concentration of extractable humus carbon(HC)were determined in four litter components(foliar,twigs,reproductive organs and miscellaneous)of the dominant species(Pterocarya stenoptera,Quercus acutissima,Cunninghamia lanceolata and Toxicodendron vernicifluum).All of the litter components exhibited obvious humification characteristics,and showed the highest concentration of extractable HC during the leaf maturation period.The miscellaneous and foliar litters showed greater humification than the other litter types.The components of Pterocarya stenoptera litter exhibited greater degrees of humification than those of the other species,with lowerΔlogK and E4/E6 values and higher A600/C values.The litter from coniferous and evergreen species showed lower humification than that from broad-leaf and deciduous species regardless of the litter component examined.The present results provide new insights into the management of plantations and theoretical data to accurately improve the quality of plantations and maintain soil fertility under a global change.Fujia Wu Chengming You Jie Du Xiangjun Pei Ningfei Lei Bo Tan Huixing Song 2021Phyton-International Journal of Experimental Botany2021,90,2:0

16Nitrogen addition promotes foliar litterfall and element return in a subtropical forest,southwestern China显示文摘Nitrogen deposition has a considerable impact on biogeochemical cycling in terrestrial ecosystems.However,how litter production and element return respond to N addition remains poorly understood in nitrogen-rich subtropical regions.In this study,a 4-year nitrogen addition experiment explored its eff ects on foliar litter production and carbon,nitrogen and phosphorus in a subtropical Michelia wilsonii forest.A clear seasonal pattern in foliar litterfall was observed,regardless of nitrogen treatments,with a peak in spring and a smaller one in autumn.Foliar litter increased with increasing nitrogen but did not aff ect litter carbon concentrations and often decreased nitrogen and phosphorous concentrations.The eff ect of nitrogen addition was dependent on time(month/year).Carbon,nitrogen and phosphorous return showed similar bimodal seasonal patterns.Nitrogen addition increased carbon and nitrogen return but did not aff ect phosphorous.Our results suggest that the addition of nitrogen stimulates carbon and nutrient return via litterfall.Yue Zhang Shichen Xiong Chengming You Sining Liu Lixia Wang Li Zhang Han Li Bo Tan Yang Liu Zhenfeng Xu 2023Journal of Forestry Research2023,34,4:0
