1New status of the infrared beamlines at SSRF显示文摘There are two infrared beamlines at the Shanghai synchrotron radiation facility(SSRF)-BL01B and BL06B.BL01B was the first infrared beamline of the National Facility for Protein Science in Shanghai at SSRF,which is dedicated to synchrotron infrared microspectroscopy.It utilizes bending magnet radiation and edge radiation as light sources.Diffraction-limited spatial resolution is reached in the infrared microspectroscopy experiment.BL01B has been in operation for approximately five years since it opened in January 2015.In the past few years,many meaningful results have been published by user groups from various disciplines,such as biomacromolecule materials and pharmaceutical,environmental,and biomedical sciences.In addition,a new infrared beamline station BL06B is under construction,which is optimized for the mid-infrared and far-infrared band.BL06B is equipped with a vacuum-type Fourier transform infrared spectrometer,infrared microscope,custom longworking-distance infrared microscope,infrared scanning near-field optical microscope,and mid-infrared Mueller ellipsometer.The purpose is to serve experiments with high vacuum requirements and spatial resolution experiments,as well as those that are in situ and time-sensitive,such as high-pressure and atomic force microscopy infrared experiments.The station can be used for research in biomaterials,pharmacy,geophysics,nanotechnology,and semiconductor materials.Xiao-Jie Zhou Hua-Chun Zhu Jia-Jia Zhong Wei-Wei Peng Te Ji Yue-Cheng Lin Yu-Zhao Tang Min Chen 2019Nuclear Science and Techniques2019,30,12:3

3Paris saponin VII,a Hippo pathway activator,induces autophagy and exhibits therapeutic potential against human breast cancer cells显示文摘Dysregulation of the Hippo signaling pathway seen in many types of cancer is usually associated with a poor prognosis.Paris saponin VII(PSVII)is a steroid saponin isolated from traditional Chinese herbs with therapeutic action against various human cancers.In this study we investigated the effects of PSVII on human breast cancer(BC)cells and its anticancer mechanisms.We showed that PSVII concentration-dependently inhibited the proliferation of MDA-MB-231,MDA-MB-436 and MCF-7 BC cell lines with IC_(50) values of 3.16,3.45,and 2.86μM,respectively,and suppressed their colony formation.PSVII(1.2-1.8μM)induced caspase-dependent apoptosis in the BC cell lines.PSVII treatment also induced autophagy and promoted autophagic flux in the BC cell lines.PSVII treatment decreased the expression and nuclear translocation of Yes-associated protein(YAP),a downstream transcriptional effector in the Hippo signaling pathway;overexpression of YAP markedly attenuated PSVII-induced autophagy.PSVII-induced,YAP-mediated autophagy was associated with increased active form of LATS1,an upstream effector of YAP.The activation of LATS1 was involved the participation of multiple proteins(including MST2,MOB1,and LATS1 itself)in an MST2-dependent sequential activation cascade.We further revealed that PSVII promoted the binding of LATS1 with MST2 and MOB1,and activated LATS1 in the BC cell lines.Molecular docking showed that PSVII directly bound to the MST2-MOB1-LATS1 ternary complex.Microscale thermophoresis analysis and drug affinity responsive targeting stability assay confirmed the high affinity between PSVII and the MST2-MOB1-LATS1 ternary complex.In mice bearing MDA-MB-231 cell xenograft,administration of PSVII(1.5 mg/kg,ip,4 times/week,for 4 weeks)significantly suppressed the tumor growth with increased pLATS1,LC3-II and Beclin 1 levels and decreased YAP,p62 and Ki67 levels in the tumor tissue.Overall,this study demonstrates that PSVII is a novel and direct Hippo activator that has great potential in the treatment of BC.Yu-chen Xiang Peng Peng Xue-wen Liu Xin Jin Jie Shen Te Zhang Liang Zhang Fang Wan Yu-liang Ren Qing-qing Yu Hu-zi Zhao Yuan Si Ying Liu 2022Acta Pharmacologica Sinica2022,43,6:1

7Infrared microspectroscopy beamline BL06B at SSRF显示文摘The infrared microspectroscopy beamline(BL06B) is a phase Ⅱ beamline project at the Shanghai Synchrotron Radiation Facility(SSRF). The construction and optical alignment of BL06B were completed by the end of 2020. By 2021, it became accessible to users. The synchrotron radiation infrared(SRIR) source included edge radiation(ER) and bending magnet radiation(BMR). The extracted angles in the horizontal and vertical directions were 40 and 20 mrad, respectively. The photon flux, spectral resolution, and focused spot size were measured at the BL06B endstation, and the experimental results were consistent with theoretical calculations. SRIR light has a small divergence angle, high brightness, and a wide wavelength range. As a source of IR microscopy, it can easily focus on a diffraction-limited spatial resolution with a high signal-to-noise ratio(SNR). The paper describes the progress of visible (red) VCSELs in particular. 9Effect of Down-regulated Expression of cPouV and cNanog on Pluripotency of Chicken(Gallus gallus) Embryonic Stem Cells(cESCs)显示文摘To explore the pluripotency maintenance and update the functional influence of pluripotency genes c Nanog and c Pou V in chicken( Gallus gallus) embryonic stem cells( c ESCs),the stable RNAi vectors p Super-c Nanog and p Super-c Pou V constructed previously were used to transfect c ESCs. The mRNA levels of two target genes were detected with real-time PCR. These two genes were down-regulated since the 48 thh and the down-regulation continued with the extension of time,the interference efficiency reached 65% at 96thhour( P < 0. 05). With the down-regulation of c Nanog or c Pou V gene,c ESCs showed differentiation and proliferation rate of these cells slowed down,the domed colony of these cells disappeared gradually when the edge of colony became irregular. At 96 thhour after transfection,the alkline phosphatase( AKP) and stage-specific embryonic antigen-1( SSEA-1) were not be detected in c Nanog gene-knocked out c ESCs,but it was done in that with c Pou V gene-knocked out. The c Pou V-suppressing c ESCs were again transfected with p Super-c Nanog,the pluripotency markers AKP and SSEA-1 were both not found expressing at the 48 thhour. The results showed that c Pou V and c Nanog genes played an important role in maintaining pluripotency and self- renewal in c ESCs,and c Nanog gene was dominant. 10Effects of hydrogen charging and deformation on tensile properties of a multi-component alloy for nuclear applications显示文摘In this study,the influence of hydrogen charging and deformation on the tensile behavior of a 60Fe-12Cr-10Mn-15Cu-3Mo multi-component alloy was investigated using electron microscopy and positron annihilation lifetime spectroscopy.The results show that hydrogen-induced vacancy clusters found in the electrochemically charged hydrogen specimens are responsible for crack initiation.Upon ingress to the microstructure,hydrogen promotes the formation of cell-structured dislocations that are beneficial for the improvement of tensile strength.In addition,hydrogen embrittlement can be mitigated by dislocations that can hinder hydrogen mobility in the deformed specimens.Te Zhu Zhi-Hong Zhong Koichi Sato Ya-Min Song Feng-Jiao Ye Qian-Qian Wang Ye Dong Peng Zhang Run-Sheng Yu Bao-Yi Wang Alfonso H.W.Ngan Xing-Zhong Cao Qiu Xu 2022Tungsten2022,4,3:0
