1肝癌高发区不同饮用水类型中微囊藻毒素含量调查显示文摘为了解肝脏促癌剂微囊藻毒素(MC)在肝癌高发区饮用水中的污染情况,于1992至1993年对江苏海门居民饮用水中藻类及其产生的MC进行了检测,经鉴定在海门沟河水中颤藻是目前已被证明能产生MC的最常见的藻属;在65个水样中有2份经高效液相和液相色谱/质谱分析确定含有MC。进一步于1994年7月在江苏海门采集不同饮用水源共989份,采用高敏感度酶联免疫方法测定MC含量。结果在沟塘水、河水、浅井水和深井水中MC的阳性率(>50pg/ml)分别为17.3%,31.9%,4.3%,0.0%;阳性样本中MC平均含量分别为101,160,68,0。在沟塘水和河水中MC的阳性率和阳性样本中的平均含量均显著大于浅井水和深井水,提示应对饮用水中MC与肝癌的关系作进一步研究。陈刚 俞顺章 卫国荣 陈公超 徐小英 黄裕华 Ken-ichi Harada Yoshio Ueno 1996中华预防医学杂志1996,30,1:59
2微卫星标记中的无效等位基因显示文摘微卫星标记以其独有的优点广泛应用于遗传学研究,但无效等位基因(nullalleles)的存在与潜在影响是其最大缺陷之一,在研究工作中并未得到足够重视。本文在综述国内外相关文献的基础上,明确了微卫星无效等位基因的概念与特点,对其可能的产生原因、频率估算方法、相关分析软件及其对群体遗传学、亲本分析等研究结果的影响进行述评,以期对无效等位基因有较为全面、深入的了解。微卫星无效等位基因的产生与SSR侧翼序列的变异(点突变、插入或缺失)及引物结合位点有关,其与同工酶标记中的无效等位基因有本质区别,并非基因本身的自然属性。虽然微卫星无效等位基因具有普遍性、复杂性和隐匿性等特点,但完全可以通过Hardy-Weinberg平衡检验、亲子代基因型分析和重新设计引物等方法认识、检测并估算其频率。无效等位基因会对遗传学相关研究结果造成显著影响,如降低群体遗传多样性,加大群体间遗传分化;降低亲本分析排除率,甚至可能造成亲本分析结果的错误与混乱。今后研究工作中,我们应对无效等位基因予以足够重视并谨慎对待,从标记位点选择、无效等位基因数据调整及重新设计引物分析等多个方面尽可能减少和避免其影响,以获得最真实的分析结果。文亚峰 Kentaro Uchiyama 韩文军 Saneyoshi Ueno 谢伟东 徐刚标 Yoshihiko Tsumura 2013生物多样性2013,21,1:42
3Th17 plasticity and its changes associated with inflammatory bowel disease显示文摘CD4 T helper(Th) cell differentiation into distinct T cell subsets is critical to the normal function of the immune system. Until recently,the paradigm held that na?ve T cells differentiated into distinct subsets under the guidance of environmental cues(e.g.,cytokines) and that once polarized,these cells were committed to a particular functional state. However,the existence of transdifferentiated T cell populations,which express signature transcription factors and cytokines associated with more than one Th subset,challenges the immutability of T helper subsets and suggests that plasticity is a feature of multifaceted immune responses. How this process impacts immune dysregulation in diseases such as inflammatory bowel diseases(IBD) and the machinery that underlies this process is far from fully understood. Interleukin(IL)-17 secreting helper T(Th17) cells have been heavily implicated in tissue-specific immune pathology including murine models of IBD,human Crohn's disease and ulcerative colitis. Plasticity within this subset is suggested by the existence of IL-17 secreting cells,which,can also secrete interferon-γ,the signature cytokine for Th1 cells or,can co-express the anti-inflammatory transcription factor forkhead box p3,a signature transcription factor of regulatory T cells. 5Direct imaging of the near field and dynamics of surface plasmon resonance on gold nanostructures using photoemission electron microscopy显示文摘Localized surface plasmon resonance(LSPR)can be supported by metallic nanoparticles and engineered nanostructures.An understanding of the spatially resolved near-field properties and dynamics of LSPR is important,but remains experimentally challenging.We report experimental studies toward this aim using photoemission electron microscopy(PEEM)with high spatial resolution of sub-10 nm.Various engineered gold nanostructure arrays(such as rods,nanodisk-like particles and dimers)are investigated via PEEM using near-infrared(NIR)femtosecond laser pulses as the excitation source.When the LSPR wavelengths overlap the spectrum of the femtosecond pulses,the LSPR is efficiently excited and promotes multiphoton photoemission,which is correlated with the local intensity of the metallic nanoparticles in the near field.Thus,the local field distribution of the LSPR on different Au nanostructures can be directly explored and discussed using the PEEM images.In addition,the dynamics of the LSPR is studied by combining interferometric time-resolved pump-probe technique and PEEM.Detailed information on the oscillation and dephasing of the LSPR field can be obtained.The results identify PEEM as a powerful tool for accessing the near-field mapping and dynamic properties of plasmonic nanostructures.Quan Sun Kosei Ueno Han Yu Atsushi Kubo Yasutaka Matsuo Hiroaki Misawa 2013Light(Science & Applications)2013,2,1:14

7Pegylated interferon plus ribavirin for genotype Ib chronic hepatitis C in Japan显示文摘AIM:To evaluate the efficacy of pegylated interferon α-2b(peg-IFNα-2b) plus ribavirin(RBV) therapy in Japanese patients with chronic hepatitis C(CHC) genotype Ib and a high viral load.METHODS:One hundred and twenty CHC patients(58.3% male) who received peg-IFNα-2b plus RBV therapy for 48 wk were enrolled.Sustained virological response(SVR) and clinical parameters were evaluated.RESULTS:One hundred(83.3%) of 120 patients completed 48 wk of treatment.53 patients(44.3%) achieved SVR.Early virological response(EVR) and end of treatment response(ETR) rates were 50% and 73.3%,respectively.The clinical parameters(SVR vs non-SVR) associated with SVR,ALT(108.4 IU/L vs 74.5 IU/L,P = 0.063),EVR(76.4% vs 16.4%,P < 0.0001),adherence to peg-IFN(≥ 80% of planned dose) at week 12(48.1% vs 13.6%,P = 0.00036),adherence to peg-IFN at week 48(54.7% vs 16.2%,P < 0.0001) and adherence to RBV at week 48(56.1% vs 32.1%,P = 0.0102) were determined using univariate analysis,and EVR and adherence to peg-IFN at week 48 were determined using multivariate analysis.In the older patient group(> 56 years),SVR in females was significantly lower than that in males(17% vs 50%,P = 0.0262).EVR and adherence to Peg-IFN were demonstrated to be the main factors associated with SVR.CONCLUSION:Peg-IFNα-2b plus RBV combination therapy demonstrated good tolerability in Japanese patients with CHC and resulted in a SVR rate of 44.3%.Treatment of elderly female patients is still challenging and maintenance of adherence to peg-IFNα-2b is important in improving the SVR rate.Takayuki Kogure Yoshiyuki Ueno Koji Fukushima Futoshi Nagasaki Yasuteru Kondo Jun Inoue Yasunori Matsuda Eiji Kakazu Takeshi Yamamoto Hiroyoshi Onodera Yutaka Miyazaki Hiromasa Okamoto Takehiro Akahane Tomoo Kobayashi Yutaka Mano Takao Iwasaki Motoyasu Ishii Tooru Shimosegawa 2008World Journal of Gastroenterology2008,14,47:7
8Shared and distinct roles of T peripheral helper and T follicular helper cells in human diseases显示文摘The interactions of CD4^(+)T cells and B cells are fundamental for the generation of protective antibody responses,as well as for the development of harmful autoimmune diseases.Recent studies of human tissues and blood samples have established a new subset of CD4^(+)B helper T cells named peripheral helper T(Tph)cells.Unlike T follicular helper(Tfh)cells,which interact with B cells within lymphoid organs,Tph cells provide help to B cells within inflamed tissues.Tph cells share many B helper-associated functions with Tfh cells and induce B cell differentiation toward antibody-producing cells.The differentiation mechanism is also partly shared between Tph and Tfh cells in humans,and both Tfh and Tph cells can be found within the same tissues,including cancer tissues.However,Tph cells display features distinct from those of Tfh cells,such as the expression of chemokine receptors associated with Tph cell localization within inflamed tissues and a low Bd-6/Blimp1 ratio.Unlike that of Tfh cells,current evidence shows that the target of Tph cells is limited to memory B cells.In this review,we first summarize recent findings on human Tph cells and discuss how Tph and Tfh cells play shared and distinct roles in human diseases.Hiroyuki Yoshitomi Hideki Ueno 2021Cellular & Molecular Immunology2021,18,3:7

11Selective decrease in colonic CD56^+ T and CD161^+ T cells in the inflamed mucosa of patients with ulcerative colitis显示文摘AIM: To investigate the role of local colonic mucosal NK receptor-positive T (NKR+ T) cells in the regulation of intestinal inflammation, we analyzed the population and function of these cells in ulcerative colitis (UC). METHODS: Colonic mucosal tissues were obtained from colonoscopic biopsies of the descending colon from 96 patients with UC (51 endoscopically uninfl amed, 45 inflamed) and 18 normal controls. Endoscopic appearance and histologic score at the biopsied site were determined by Matts’ classification. A single cell suspension was prepared from each biopsy by collagenase digestion. Two NKR+ T cell subsets, CD56+ (CD56+CD3+) T cells and CD161+ (CD161+CD3+) T cells, were detected by flow cytometric analysis. Intracellular cytokine analysis for anti-in? ammatory cytokine interleukin-10 (IL-10) was performed by in vitro stimulation with phorbol-myristate- acetate (PMA) and ionomycin. RESULTS: CD56+ T cells and CD161+ T cells are present in the normal human colon and account for 6.7% and 21.3% of all mononuclear cells, respectively. The populations of both CD56+ T cells and CD161+ T cells were decreased significantly in the inflamed mucosa of UC. In contrast, the frequency of conventional T cells (CD56-CD3+ cells and CD161-CD3+ cells) was similar among the patient and control groups. The populations of NKR+ T cells were correlated inversely with the severity of infl ammation, which was classif ied according to the endoscopic and histologic Matts’ criteria. 12Association between Diabetes Mellitus and Oral Health Status in Japanese Adults显示文摘Aim The objective of this study was to analyze the oral health among Japanese adults, with and without diabetes mellitus. Methodology The subjects were 518 community residents aged 20 to 91 years in Japan, who participated in the 'Akita health and nutrition survey' and the 'Akita dental disease survey', conducted in 2006. The surveys comprised a self-administered questionnaire, along with medical and dental examinations. Results Using the community periodontal index (CPI), the mean numbers of sextants presenting codes 0, 1 and 2 were significantly lower in diabetics than non-diabetics among the 59 years or younger age group. Although the mean numbers of sextants with codes 0, 1 and 2 among the 60 to 69 years age group were lower, and sextants with a code X among the 59 years or younger age group were higher in diabetics than non-diabetics, no statistically significant differences were detected. The tendency of lower mean numbers of natural teeth and functional tooth units in diabetics than non-diabetics was observed, however no differences were statistically significant. 13Fulminant hepatic failure in a case of autoimmune hepatitis in hepatitis C during peg-interferon-alpha 2b plus ribavirin treatment显示文摘A 27-year-old Caucasian female with hepatitis C virus (HCV) infection treated with interferon (IFN) who developed severe autoimmune hepatitis (AIH) is described. The infecting viral strain was of genotype Ib and the pre-treatment HCV viral load was at a high level. The patient was treated with pegylated IFN-alpha 2b and ribavirin,and her HCV-RNA became negative at wk 12,but after that she developed fulminant hepatic failure. The patient recovered after steroid pulse therapy consisting of methylprednisolone 1000 mg/d for three days which was administered twice. 15Familial pancreatic cancer: Concept, management and issues显示文摘Familial pancreatic cancer (FPC) is broadly defined as two first-degree-relatives with pancreatic cancer (PC) and accounts for 4%-10% of PC. Several genetic syndromes, including Peutz-Jeghers syndrome, hereditary pancreatitis, hereditary breast-ovarian cancer syndrome(HBOC), Lynch syndrome, and familial adenomatous polyposis (FAP), also have increased risks of PC, but the narrowest definition of FPC excludes these known syndromes. When compared with other familial tumors, proven genetic alterations are limited to a small proportion (<20%) and the familial aggregation is usually modest. However, an ethnic deviation (Ashkenazi Jewish>Caucasian) and a younger onset are common also in FPC. In European countries, 'anticipation' is reported in FPC families, as with other hereditary syndromes; a trend toward younger age and worse prognosis is recognized in the late years. The resected pancreases of FPC kindred often show multiple pancreatic intraepithelial neoplasia (Pan IN) foci, with various K-ras mutations, similar to colorectal polyposis seen in the FAP patients. As with HBOC patients, a patient who is a BRCA mutation carrier with unresectable pancreatic cancer (accounting for 0%-19% of FPC patients) demonstrated better outcome following platinum and Poly (ADP-ribose) polymerase inhibitor treatment. Western countries have established FPC registries since the 1990 s and several surveillance projects for highrisk individuals are now ongoing to detect early PCs. Improvement in lifestyle habits, including non-smoking, is recommended for individuals at risk. 16青藏高原唐古拉山南北坡夏季风降水特征的对比分析显示文摘根据 GAME- Tibet加强观测期间 ( 1 998年 5~ 9月 )所得到的位于青藏高原唐古拉山北坡的 D1 0 5站和南坡的 WADD站夏季风降水资料 ,对唐古拉山南北坡夏季风降水特征进行了初步的对比分析 .结果表明两地夏季风降水频率较高 .在 1 998年 6月 2 5日~ 9月 1 9日 ,WADD站的降水量比 D1 0 5站的降水量多 48.8% ,而发生的降水次数在 WADD站比 D1 0 5站多 1 0次 ,两者的累积降水次数基本上差不多 .同时 ,WADD站平均降水强度高于 D1 0 5站 .这两地的夏季风降水存在明显的活跃期和中断期 ,7月中旬夏季风降水都不活跃 .大多数情况下 ,这两地的日降水量的变化有较为一致的趋势 ,表明两地降水可能受同一降水过程的控制 .9月初以后 。杨梅学 姚檀栋 田立德 Ken'ichi UENO 2000应用气象学报2000,11,2:5
171998年夏季(7,8月)藏北降水的某些特征分析显示文摘利用GAME Tibet加强观测期间所得到的青藏高原不同地点的降水量资料 ,初步分析了 1 998年 7月和 8月夏降风降水随纬度和海拔高度的变化特征。研究表明藏北高原夏季风降水的分布是纬度效应和海拔高度效应综合作用的结果。 7月份降水量的纬度效应要比高度效应明显 ,而 8月份的降水量不但具有一定的纬度效应 ,也具有一定的海拔高度效应。 7月~ 8月份的降水量主要表现为随纬度的分布特征 ,但它也受到海拔高度效应的修正 ,即由于地形对降水的再分配过程 ,使得某些测点降水随海拔高度的升高而增加。由于青藏高原地表状况复杂 ,其上的降水显然应当同时受到各种因素的影响和制约 ,因此要简单地区分各因子的作用是较困难的。杨梅学 姚檀栋 Ken'ichi UENO 2000资源科学2000,22,6:5
18Treatment of colorectal carcinoids:A new paradigm显示文摘It is often difficult to evaluate the grade of malignancy and choose an appropriate treatment for colorectal carcinoids in clinical settings. Although tumor size and depth of invasion are evidently not enough to stratify the risk of this rare tumor, the present guidelines or staging systems do not mention other clinicopathological variables. Recent studies, however, have shed light on the impact of lymphovascular invasion on the outcome of colorectal carcinoids. It has been revealed that the presence of lymphovascular invasion was among the strongest risk factors for metastasis along with tumor size and depth of invasion. Furthermore, tumors smaller than 1 cm, within submucosal invasion and without lymphovascular invasion, carry minimal risk for metastasis with 100% 5-year survival in the studies from Japan as well as from the USA. This would suggest that these tumors could be curatively treated by endoscopic resection or transanal local excision. On the other hand, colorectal carcinoids with either lymphovascular invasion or tumor size larger than 1 cm carry the risk for metastasis equivalent to adenocarcinomas. Therefore, it should be emphasized that histological examination of lymphovascular invasion is mandatory in the specimens obtained by endoscopic resection or transanal local excision, as this would provide useful information for determining the need for additional radical surgery with regional lymph node dissection.
