An Integrated Analysis of Risk Factors of Cognitive Impairment in Patients with Severe Carotid Artery Stenosis显示文摘Objective To investigate cognitive dysfunction in patients with carotid artery stenosis(CAS) and potential risk factors related to cognitive-especially memory-dysfunction. Methods Forty-seven patients with carotid artery stenosis were recruited into our study cohort. The Mini-Mental State Examination(MMSE) and the Montreal Cognitive Assessment(MoCA) were adopted to assess cognitive function, the Wechsler Memory Scale(WMS) to assess memory function, high-resolution MRI and enhanced ultrasound to evaluate carotid plaques, and computed tomography perfusion(CTP) imaging to evaluate intracranial blood perfusion. Single-factor analysis and multiple-factor regression analysis were used to analyze potential risk factors of cognitive impairment. Results Mini-Mental State Examination test results showed that 22 patients had cognitive impairment, and MoCA test results showed that 10 patients had cognitive impairment. Analysis of various risk factors indicated that the average memory quotient of female patients was higher than that of males(P = 0.024). The cognitive and memory performance of those with an educational background above high school were significantly better than those of participants with high school or lower(P = 0.045). Patients with abnormal intracranial perfusion performed worse on the MMSE test(P = 0.024), and their WMS scores were significantly lower(P = 0.007). The MMSE scores and the memory quotients were significantly lower in patients with a history of cerebral infarction(MMSE, P = 0.047, memory quotient score, P = 0.018). Conclusion A history of cerebral infarction and abnormal cerebral perfusion are associated with decline in overall cognitive function and memory in patients with carotid stenosis. Being female and having an educational background above high school may be protective factors in the development of cognitive dysfunction.LUO Ru Tao WANG Pei Jiong DENG Xiao Feng ZHOU Shu Jie ZHAO Meng QIAN Jing ZHANG Dong WANG Rong ZHANG Yan 2018Biomedical and Environmental Sciences2018,31,11:17
Optical tuning of exciton and trion emissions in monolayer phosphorene显示文摘Monolayer phosphorene provides a unique two-dimensional(2D)platform to investigate the fundamental dynamics of excitons and trions(charged excitons)in reduced dimensions. However, owing to its high instability, unambiguous identification of monolayer phosphorene has been elusive.Consequently, many important fundamental properties, such as exciton dynamics, remain underexplored. We report a rapid, noninvasive, and highly accurate approach based on optical interferometry to determine the layer number of phosphorene, and confirm the results with reliable photoluminescence measurements.Furthermore,we successfully probed the dynamics of excitons and trions in monolayer phosphorene by controlling the photo-carrier injection in a relatively low excitation power range.Based on our measured optical gap and the previously measured electronic energy gap,we determined the exciton binding energy to be~0.3 eV for the monolayer phosphorene on SiO_(2)/Si substrate, which agrees well with theoretical predictions.A huge trion binding energy of~100 meV was first observed in monolayer phosphorene, which is around five times higher than that in transition metal dichalcogenide(TMD)monolayer semiconductor, such as MoS_(2).The carrier lifetime of exciton emission in monolayer phosphorene was measured to be,220 ps,which is comparable to those in other 2D TMD semiconductors. Our results open new avenues for exploring fundamental phenomena and novel optoelectronic applications using monolayer phosphorene.Jiong Yang Renjing Xu Jiajie Pei Ye Win Myint Fan Wang Zhu Wang Shuang Zhang Zongfu Yu Yuerui Lu 2015Light(Science & Applications)2015,4,1:8
Intestinal dysbiosis exacerbates the pathogenesis of psoriasis-like phenotype through changes in fatty acid metabolism显示文摘The intestinal microbiota has been associated with host immunity as well as psoriasis;however, the mechanism of intestinal microbiota regulating psoriasis needs to be demonstrated systematically. Here,we sought to examine its role and mechanism of action in the pathogenesis of psoriasis. We found that the severity of psoriasis-like skin phenotype was accompanied by changes in the composition of the intestinal microbiota. We performed co-housing and fecal microbial transplantation(FMT)experiments using the K14-VEGF transgenic mouse model of psoriasis and demonstrated that the transfer of intestinal microbiota from mice with severe psoriasis-like skin phenotype exacerbated psoriasiform skin inflammation in mice with mild symptoms, including increasing the infiltration and differentiation of Th17,and increased the abundance of Prevotella, while decreasing that of Parabacteroides distasonis,in the colon. These alterations affected fatty acid metabolism, increasing the abundance of oleic and stearic acids.Meanwhile, gentamicin treatment significantly reduced the abundance of Prevotella and alleviated the psoriasis-like symptoms in both K14-VEGF mice and imiquimod(IMQ)-induced psoriasis-like mice.Indeed, administration of oleic and stearic acids exacerbated psoriasis-like symptoms and increased Th17 and monocyte-derived dendritic cell infiltration in the skin lesion areas in vivo,as well as increased the secretion of IL-23 by stimulating DCs in vitro. At last,we found that, treatment of PDE-4 inhibitor alleviated psoriasis-like phenotype of K14-VEGF mice accompanied by the recovery of intestinal microbiota, including the decrease of Prevotella and increase of Parabacteroides distasonis.Overall, our findings reveal that the intestinal microbiota modulates host metabolism and psoriasis-like skin inflammation in mice, suggesting a new target for the clinical diagnosis and treatment of psoriasis.Qixiang Zhao Jiadong Yu Hong Zhou Xiaoyan Wang Chen Zhang Jing Hu Yawen Hu Huaping Zheng Fanlian Zeng Chengcheng Yue Linna Gu Zhen Wang Fulei Zhao Pei Zhou Haozhou Zhang Nongyu Huang Wenling Wu Yifan Zhou Jiong Li 2023Signal Transduction and Targeted Therapy2023,8,2:2
Associations of Daytime Napping with Incident Cardiovascular Diseases and Hypertension in Chinese Adults:A Nationwide Cohort Study显示文摘Objective This study aimed to examine the associations of daytime napping with incident risks of cardiovascular diseases(CVDs)and hypertension(HTN).Methods Data for napping and CVD outcomes in 25 provinces were collected from baseline(2010)and three waves of follow-up(2012-2017)investigations of the China Family Panel Studies.Cox frailty models with random intercepts for the surveyed provinces were used to assess the longitudinal effects of daytime napping on CVD and HTN.Results Compared with non-nappers,30+min nappers had higher risks of CVD and HTN,while no significant associations were observed among<30 min nappers.Incident risks among 30-to<60-min nappers increased by 22%[hazard ratio(HR)1.22,95% confidence interval(CI)1.08-1.39]for CVD and 21%(1.21,1.04-1.41)for HTN,respectively, with corresponding HRs of CVD and HTN of 1.27(1.09-1.47)and 1.38(1.16-1.65)among≥60 min nappers.Nap-associated CVD risks varied by subgroups, with stronger associations in participants with lower body mass index(<24 kg/m^(2)),physically inactive persons, smokers, and participants with longer nighttime sleep(≥7 h/night).Significant effects of daytime napping were observed on rural and northern residents only, highlighting great regional variations in CVD risks associated with napping habits. Conclusions This cohort study revealed strong evidence that long daytime napping(≥30 min)is associated with an increased incidence of cardiovascular events.WANG Lu WANG Ke LIU Lin Jiong ZHANG Yuan Yuan SHU Hai Nan WANG Kai ZHOU Pei Xuan ZHANG Yun Quan 2022Biomedical and Environmental Sciences2022,35,1:2
Specific pupylation as IDEntity reporter(SPIDER)for the identification of protein-biomolecule interactions显示文摘Protein-biomolecule interactions play pivotal roles in almost all biological processes. For a biomolecule of interest, the identification of the interacting protein(s)is essential. For this need, although many assays are available, highly robust and reliable methods are always desired.By combining a substrate-based proximity labeling activity from the pupylation pathway of Mycobacterium tuberculosis and the streptavidin(SA)-biotin system,we developed the Specific Pupylation as IDEntity Reporter(SPIDER)method for identifying protein-biomolecule interactions.Using SPIDER,we validated the interactions between the known binding proteins of protein,DNA,RNA,and small molecule. We successfully applied SPIDER to construct the global protein interactome for m^(6)A and m RNA,identified a variety of uncharacterized m^(6)A binding proteins, and validated SRSF7 as a potential m^(6)A reader. We globally identified the binding proteins for lenalidomide and Cob B.Moreover,we identified SARS-CoV-2-specific receptors on the cell membrane.Overall,SPIDER is powerful and highly accessible for the study of proteinbiomolecule interactions.He-Wei Jiang Hong Chen Yun-Xiao Zheng Xue-Ning Wang Qingfeng Meng Jin Xie Jiong Zhang ChangSheng Zhang Zhao-Wei Xu Zi-Qing Chen Lei Wang Wei-Sha Kong Kuan Zhou Ming-Liang Ma Hai-Nan Zhang Shu-Juan Guo Jun-Biao Xue Jing-Li Hou Zhe-Yi Liu Wen-Xue Niu Fang-Jun Wang Tao Wang Wei Li Rui-Na Wang Yong-Jun Dang Daniel MCzajkowsky JianFeng Pei Jia-Jia Dong Sheng-Ce Tao 2023Science China(Life Sciences)2023,66,8:0
Interleukin-37 promotes colitis-associated carcinogenesis via SIGIRR-mediated cytotoxic T cells dysfunction显示文摘Interleukin-37b(hereafter called IL-37)was identified as fundamental inhibitor of natural and acquired immunity. The molecular mechanism and function of IL-37 in colorectal cancer(CRC)has been elusive. Here,we found that IL-37 transgenic(IL-37tg)mice were highly susceptible to colitis-associated colorectal cancer(CAC)and suffered from dramatically increased tumor burdens in colon.Nevertheless,IL-37 is dispensable for intestinal mutagenesis, and CRC cell proliferation, apoptosis, and migration.Notably,IL-37 dampened protective cytotoxic T cell-mediated immunity in CAC and B16-OVA models.CD8^(+)T cell dysfunction is defined by reduced retention and activation as well as failure to proliferate and produce cytotoxic cytokines in IL-37tg mice, enabling tumor evasion of immune surveillance. The dysfunction led by IL-37 antagonizes IL-18–induced proliferation and effector function of CD8+T cells, which was dependent on SIGIRR(single immunoglobulin interleukin-1 receptor-related protein). Finally,we observed that IL-37 levels were significantly increased in CRC patients, and positively correlated with serum CRC biomarker CEA levels, but negatively correlated with the CD8+T cell infiltration in CRC patients. Our findings highlight the role of IL-37 in harnessing antitumor immunity by inactivation of cytotoxic T cells and establish a new defined inhibitory factor IL-37/SIGIRR in cancerimmunity cycle as therapeutic targets in CRC.Zhen Wang Fan-lian Zeng Ya-wen Hu Xiao-yan Wang Fu-lei Zhao Pei Zhou Jing Hu Yuan-yuan Xiao Zhong-lan Hu Ming-feng Guo Xiao-qiong Wei Xiao Liu Nong-yu Huang Jun Zhang Shu-wen Chen Juan Cheng Hua-ping Zheng Hong Zhou Qi-xiang Zhao Chen Zhang Yan Hao Song Zou Yi-yue Gui Jia-dong Yu Lin-na Gu Cheng-cheng Yue Hao-zhou Zhang Wen-ling Wu Yi-fan Zhou Xi-kun Zhou Guo-bo Shen Xiu Teng Jiong Li 2022Signal Transduction and Targeted Therapy2022,7,2:0
