| 2 | MicroRNA-548 down-regulates host antiviral response via direct targeting of IFN-λ1显示文摘Interferon(IFN)-mediated pathways are a crucial part of the cellular response against viral infection.Type III IFNs,which include IFN-λ1,2 and 3,mediate antiviral responses similar to Type I IFNs via a distinct receptor complex.IFN-λ1 is more effective than the other two members.Transcription of IFN-λ1 requires activation of IRF3/7 and nuclear factor-kappa B(NF-κB),similar to the transcriptional mechanism of Type I IFNs.Using reporter assays,we discovered that viral infection in-duced both IFN-λ1 promoter activity and that of the 3′-untranslated region(UTR),indicating that IFN-λ1 expression is also regulated at the post-transcriptional level.After analysis with microRNA(miRNA)prediction programs and 3′UTR targeting site assays,the miR-NA-548 family,including miR-548b-5p,miR-548c-5p,miR-548i,miR-548j,and miR-548n,was identified to target the 3′UTR of IFN-λ1.Further study demonstrated that miRNA-548 mimics down-regulated the expression of IFN-λ1.In contrast,their inhibitors,the complemen-tary RNAs,enhanced the expression of IFN-λ1 and IFN-stimulated genes.Furthermore,miRNA-548 mimics promoted infection by enterovirus-71(EV71)and ve-sicular stomatitis virus(VSV),whereas their inhibitors significantly suppressed the replication of EV71 and VSV.Endogenous miRNA-548 levels were suppressed during viral infection.In conclusion,our results sug-gest that miRNA-548 regulates host antiviral response via direct targeting of IFN-λ1,which may offer a poten-tial candidate for antiviral therapy. | Yongkui Li Jiajia Xie Xiupeng Xu Jun Wang Fang Ao Yushun Wan Ying Zhu | 2013 | Protein & Cell2013,4,2: | 22 |

The method is to discretizee independently the PDEs in different directions with symplectic schemes, so that the multisymplectic schemes can be constructed by concatenating those symplectic schemes. By this method, we can construct multisymplectic schemes, including some widely used schemes with an accuracy of any order. The desorption percentages are lower than 28% of the sorbed 1, 4-DCB after desorbing for 6 days. The sorption and desorption isotherms are well fitted to Freundlich equation, whose parameter 1/n ranges from 0.055 to 0.527. Moreover, the parameter 1/n of the desorption isotherm is significantly lower than that of the sorption isotherm, but the parameter logK increases on contrary to 1/n. The desorption isotherms are very well fitted to Langmuir equation, whose Qm decreases in the order: TP>FP> P. The apparent partition coefficients (Kp) increase with increasing sorption time or decreasing aqueous equilibrium concentration of DCB. | 13 | Effect of selective uterine artery embolization on symptomatic uterine fibroids显示文摘Objective: To evaluate the outcomes of selective uterine artery embolization (SUAE) in treatment of symptomatic uterine fibroids using Color Doppler ultrasonography (CDUS). Methods: As the procedure SUAE for fibroids, prospective data of the initial 65 consecutive women treated from March 2007 to September 2009 were collected. The follow-up period from 1 week to 30 months, using questionnaires, we investigated the fibroid-related physical and psychological symptoms, and the cumulative rates of symptom control, gynecologic interventions, and complications. The volumes of uterine and uterine fibroids were calculated by CDUS pre- and post-SUAE for 6 months. And we tested the levels of internal hormone including FSH, LH and E2 before and after SUAE for 3, 6 months. Results: SUAE was performed successfully in all patients. There was no peri-operative morbidity in all procedures. Compared with pre-SUAE volumes of uterine [(322±112) cm3] and uterine fibroids [(125±46) cm3], their volumes in post-SUAE were (144±72) cm3 and (51±11) cm3 (P<0.01), and reduction rate was 58% and 61%, respectively. At 3, 6 months after SUAE, levels of FSH, LH, and E2 were not decreased than that of in pre-SUAE (P>0.05). After SUAE for 24 months, the fibroids related physical and psychological symptoms such as bleeding, pain and quality of life were significantly improved (P<0.01). 90.8% (59/65) women had resolution of symptoms and no significant post-procedural symptoms. Parts of patients discharge necrotic fibroids. 86% (34/40) complications were minor, requiring no therapy. 9.2% (6/65) had significant post-procedural symptoms, requiring therapy in hospital or out-patient department. Conclusion: SUAE contributes to a long-term significant improvement of all investigated uterine fibroid-related symptoms and markedly improves women's health-related quality of life. SUAE that no impair on changes of internal hormone, under suitable conditions, represents an attractive alternative to hysterectomy for the treatment of symptomatic fibroids and is a minimally invasive, new method of good clinical prognosis.
