| 4 | Efficient production of pronuclear embryos in breeding and nonbreeding season for generating transgenic sheep overexpressing TLR4显示文摘Background: Brucella is a zoonotic Gram-negative pathogen that causes abortion and infertility in ruminants and humans. TLR4 is the receptor for LPS which can recognize Brucella and initiate antigen-presenting cell activities that affect both innate and adaptive immunity. Consequently, transgenic sheep over-expressing TLR4 are an suitable model to investigate the effects of TLR4 on preventing Brucellosis. In this study, we generated transgenic sheep overexpressing TLR4 and aimed to evaluate the effects of different seasons(breeding and non-breeding season) on superovulation and the imported exogenous gene on growth.Results: In total of 43 donor ewes and 166 recipient ewes in breeding season, 37 donor ewes and 144 recipient ewes in non-breeding season were selected for super-ovulation and injected embryo transfer to generate transgenic sheep.Our results indicated the no. of embryos recovered of donors and the rate of pronuclear embryos did not show any significant difference between breeding and non-breeding seasons(P > 0.05). The positive rate of exogenous TLR4 tested were 21.21 % and 22.58 % in breeding and non-breeding season by Southern blot. The expression level of TLR4 in the transgenic sheep was 1.5 times higher than in the non-transgenic group(P < 0.05). The lambs overexpressing TLR4 had similar growth performance with non-transgenic lambs, and the blood physiological parameters of transgenic and non-transgenic were both in the normal range and did not show any difference.Conclusions: Here we establish an efficient platform for the production of transgenic sheep by the microinjection of pronuclear embryos during the whole year. | 6 | Integration of filter membrane and Ca_(2)Nb_(3)O_(10) nanosheets for high performance flexible UV photodetectors显示文摘We report that the integration of filter membrane and Ca_(2)Nb_(3)O_(10)nanosheets(FM@CNO) UV photodetector(UV PD) shows high performance and excellent flexibility. The Ca_(2)Nb_(3)O_(10)nanosheets were prepared by a facile solid-state reaction and liquid exfoliation process. The Ca_(2)Nb_(3)O_(10)nanosheets can be integrated into the pores of a filter membrane via a simple vacuum filtration method. The FM@CNO UV PD shows high performance under 300 nm light illumination at 5 V bias, including high responsivity(0.08 AW^(-1)), high detectivity(1.1 × 10^(12)Jones), high UV/visible rejection ratio(3.86 × 10^(3)) and fast speed(0.12/1.24 ms). Furthermore, the FM@CNO device exhibits excellent flexibility after many bending cycles.In addition, the FM@CNO array device was used as a pixel array detector for UV imaging. | 8 | PIK3CA mutation in Chinese patients with lung squamous cell carcinoma显示文摘Objective: To investigate PIK3CA mutation in Chinese patients with lung squamous cell carcinoma (LSCC) and explore their relationship with clinicopathological profiles. Methods: Tumor samples from 123 cases of LSCC were included in this study. PIK3CA mutations in exon 9 and 20 were screened by pyrosequencing and confirmed by clone sequencing or amplification refractory mutation system (ARMS). Denaturing performance liquid chromatography (DHPLC) was employed for evaluation of EGFR mutation in exon 19, 21 and KRAS mutation. Results: PIK3CA mutations were found in 3 (2.4%) patients. The mutation type included E545K, E452Q and H1047R. Of these three patients, one coupled with EGFR mutation, and the other two coupled with PIK3CA amplification. All the three patients shared the same clinicopathologic characteristics: male, less than 60 years old, had smoke history, stage III and carried wild-type KRAS. Conclusions: The frequency of PIK3CA mutation is low in Chinese patients with LSCC.
