| 1 | A leaf shape mutant provides insight into PINOID Serine/Threonine Kinase function in cucumber(Cucumis sativus L.)显示文摘Optimizing leaf shape is a major challenge in efforts to develop an ideal plant type. Cucumber leaf shapes are diverse;however, the molecular regulatory mechanisms underlying leaf shape formation are unknown. In this study, we obtained a round leaf mutant(rl) from an ethyl methanesulfonate-induced mutagenesis population. Genetic analysis revealed that a single recessive gene, rl, is responsible for this mutation. A modified Mut Map analysis combined linkage mapping identified a single nucleotide polymorphism within a candidate gene,Csa1 M537400, as the mutation underlying the trait.Csa1 M537400 encodes a PINOID kinase protein involved in auxin transport. Expression of Csa1 M537400 was significantly lower in the rl mutant than in wild type, and it displayed higher levels of IAA(indole-3-acetic acid) in several tissues. Treatment of wild-type plants with an auxin transport inhibitor induced the formation of round leaves,similar to those in the rl mutant. Altered expression patterns of several auxin-related genes in the rl mutant suggest that rl plays a key role in auxin biosynthesis,transport, and response in cucumber. | 12 | Deformation behavior and fluid action of quartz veins in the Xuelongshan metamorphic complex,Western Yunnan显示文摘The Xuelongshan metamorphic complex underwent Cenozoic sinistral strike-slip shearing and exhumation from deep to shallow in the southeast margin of the Tibet Plateau.We studied the deformation behavior and fluid action of quartz veins in the metamorphic complex in depth based on macroscopic observation,electron backscatter diffraction fabric,and fluid inclusion analysis.The mylonitic samples are characterized by strongly elongated and orientated porphyroclasts and a finegrained matrix with a shear strain of 2.4.The ultramylonitic samples are characterized by a strongly fine-grained matrix with a shear strain of 5.8.The{m}glide controls the quartz deformation in the mylonites and ultramylonites.As the dominant recovery mechanism,subgrain rotation dynamic recrystallization reduces the grain size and promotes rheological weakening.The porphyroclasts restrict the crystal orientation of the recrystallized quartz matrix.The mylonitic quartz veins have abundant single-phase and two-phase fluid inclusions.However,the ultramylonitic quartz veins have a few fluid inclusions,most of which are small and irregular toward the grain boundaries.The temperature and pressure of fluid activity in quartz veins are 550–500℃and 391–218 MPa for ultramylonitic quartz veins,with a depth of 14.2–7.9 km and 450–400℃and 236–91 MPa for mylonitic quartz veins,with a depth of 8.6–3.3 km.The deformation process changes the shape of fluid inclusions from mylonitization to ultramylonitization and causes fluid leakage,an increase in local fluid pressure at the boundary,and hydrolysis weakening,promoting lattice dislocation glide and recrystallization,which is directly related to regional continuous shear deformation and exhumation. | Shuting WANG Shuyun CAO Lefan ZHAN Xuemei CHENG Yanlong DONG Xiaowen LI Wenyuan LI | 2023 | Science China Earth Sciences2023,66,9: | 0 |
| 13 | A mutation in CsKTN1 for the Katanin p60 protein results in miniature plant in cucumber,Cucumis sativus L.显示文摘Katanin,a microtubule shearing protein,plays an important role in plant architecture formation.However,little is known about its mechanisms in regulating plant architecture in cucumber.In the present study,through EMS mutagenesis,we identified a novel micro-plant(mp)mutant in the North China type inbred line CCMC that may be of value for cucumber breeding.The size and number of stem cells were altered in the mp mutant.Through bulked segregant analysis(BSA)sequencing approach combined with genetic mapping,the mp locus was delimited to an interval of 130.9-kb.Multiple lines of evidence suggested that the mp mutation is due to a single nucleotide polymorphism in Csa7G435510 that is predicted to encode the Katanin p60 subunit protein(CsKTN1).The expression levels of CsKTN1 decreased significantly in all tissues except the tendril of mp mutant.Subcellular localization showed that both wild-type and mutant CsKTN1 proteins were located in cell membrane,cytoplasm and nucleus of tobacco leaf cells.The mutant protein lost part of its ability to bind and shear microtubule in vitro.These findings provide new insight into the regulatory function of microtubule-shearing protein,Katanin p60,in plant architecture of cucumber. | MengFei Song WenYuan Fu YuHui Wang Feng Cheng MengRu Zhang JinFeng Chen QunFeng Lou | 2022 | Vegetable Research2022,2,1: | 0 |
